3.Oxaliplatin and teniposide inhibited proliferation and induced apoptosis of gastric cancer cell line BGC-823
Tumor 2010;(1):31-35
Objective:To investigate the effects of oxaliplatin (OXA) and teniposide (VM-26) on proliferation and apoptosis of gastric cancer cell line BGC-823.Methods:MTT assay was used to examine the inhibition rate of cell growth when cells were treated at various concentrations of OXA and VM-26 alone or in combination. The apoptosis was examined by flow cytometry (FCM). The protein expression of cell apoptosis associated proteins caspase-9 and livin were examined by immunocytochemistry. Results:OXA or VM-26 effectively inhibited the growth of BGC-823 gastric cancer cells in a concentration-dependent manner at certain range of concentrations. The inhibitory rate of combined treatment with OXA and VM-26 was significantly higher than that of OXA or VM-26 alone (P<0.05). The value of combination index (CI) was 0.46. The rate of apoptosis cells induced by 1.25 μg/mL OXA was 6.13%, 13.86% and 21.48% at 12, 24 and 48 h. The apoptosis rate induced by 0.625 μg/mL VM-26 was 4.60%, 10.72% and 17.07% for 12, 24 and 48 h. In combined treatment group the apoptosis rate was 11.73%, 24.14% and 44.75% at 12, 24 and 48 h, respectively. The difference was significant between combined treatment group and single drug treatment group (P<0.05). Immunnocytochemical analysis showed that the expression of caspase-9 protein was up-regulated after being exposed to OXA (1.250 μg/mL) or OXA plus VM-26 (0.750 μg/mL), while the expression rate of livin protein was down-regulated. There were significant differences among different treatment groups as well as between treatment groups and control group (P<0.05).Conclusion:OXA combined with VM-26 has synergistic effects in inhibiting the growth and inducing apoptosis of gastric cancer BGC-823 cells.
4.Research on evaluation system and impact factors for public interest nature in public hospitals
Journal of Shanghai Jiaotong University(Medical Science) 2009;29(6):737-740
Objective To establish the evaluation system and explore the possible impact factors for public interest nature in public hospitals. Methods One hundred and twenty-five public hospitals in Zhejiang Province were served as sample hospitals, and related data of year 2004 to year 2006 were collected. The Delphi expert investigation method, variation coefficient method and TOPSIS method were employed to screen the impact factors for public interest nature and build up the evaluation system for public hospitals, and stepwise regression analysis was conducted for possible impact factors of public interest nature. Results The evaluation system mainly included five aspects of quantity, quality, efficiency, mean charge of medical service and social satisfaction to public hospitals. The two most important factors that influenced the public interest nature were the number of beds and the number of equipments valued above 10 000 RMB. Conclusion The hospital scale is the most important impact factor for the nature of public interest in public hospitals. Therefore, the scale of public hospitals should be properly designed so as to ensure the public interest nature in public hospitals.
5.Empirical analysis of economies of scale in public hospitals of Zhejiang Province
Journal of Shanghai Jiaotong University(medical Science) 2010;30(1):91-93
Objective To analyse the economies of scale in public hospitals in Zhejiang Province. Methods One hundred and twenty-five public hospitals in Zhejiang Province with qualification no less than class two level two were selected as sample hospitals. Douglas production function was employed to analyse the relationship between investment and output in sample hospitals between 2004 to 2006, with two types of production models established by medical service and incomes in hospitals as dependent variables. Results From 2004 to 2006, the medical service and incomes of sample hospitals were in line with economies of scale. The production increase rate was higher than investment increase rate. The medical service, incomes, total assets and number of staff in class three hospitals were higher than those in class two hospitals (P =0.000). Conclusion Economies of scale is the ultimate incentive to continuous outstretch for hospitals. Hospitals should rationally control the scale so as to avoid problems in medical service caused by overstretch.

Tianjin Medical Journal 2017;45(8):830-832,前插1
Objective To study the effect of neural stem cell transplantation on neurological behavior and expressions of Caspase-3 and Caspase-9 in rats with traumatic brain injury (TBI). Methods A total of 108 SD rats were randomly divided into the sham group (n=36), traumatic brain injury group (TBI, n=36) and neural stem cell transplantation group (NSC, n=36). TBI model of rats was established by using improved Feeney method. Rats of NSC group were injected neural stem cells in cortex after traumatic brain injury was established for 1 week, and then nurological severity scores (NSS) of rats were evaluated at 7 days, 14 days and 21 days after cell transplantation in TBI and NSC groups, respectively. Rats of sham group were only cut the scalp and exposed the skull without striking. The expressions of Caspase-3 and Caspase-9 in brain tissues were detected by immunohistochemistry and Western blot method. Results Compared with NSC group at the same time points, NSS was decreased significantly in TBI group (P<0.01). Compared with the sham group, the optical density (OD) values of Caspase-3 and Caspase-9 positive expressions were increased significantly in TBI group (P<0.05). Compared with TBI group at the same time points, the OD values of Caspase-3 and Caspase-9 positive expressions were decreased significantly in NSC group (P<0.05). Conclusion The neural stem cell transplantation can promote the recovery of brain injury by down-regulating the expressions of Caspase-3 and Caspase-9.
