1.VEGFR-3 expression in esophageal carcinoma and its significance

Objective To investigate of the expression of vascular endothelial growth factor receptor 3 (VEGFR-3) and its relationship with the prognosis of esophageal carcinoma. Methods VEGFR-3 in 156 cases of esophageal carcinoma tissues was detected by immunohistochemistry. The follow-up data for 3 years in 47 cases were analyzed. Results The VEGFR-3 expression in esophageal carcinoma tissues with lymph node metastasis was higher than that in those without lymph node metastasis. VEGFR-3 expression was associated with the clinicopathological stages. Analysis of the follow-up data also showed that there was a statistically positive correlation between VEGFR-3 expression and lymph node metastasis in primary tumors (P

Objective:Peptides were designed on the basis of high conservative regions of amino acid sequences and structures of the SEs,three-dimension structure of P72 was constructed.Methods: Bioinformatics analysis softwares such as Vector NTI 10.3, InsightII 2000,Discovery Studio 1.7 were used to analyse and predict the space structure of P72.Results:three-dimensional domains of the peptide P72 from SEA, SEB and SEC were quite similar, Peptide P72 was far away from TCRVβchain and MHC class II molecule.Conclusion:The inhibitory activity of peptide P72 may not due to binding to MHCⅡ and TCRVβchain.The exact mechanism of inhibitory activity of P72 should be explored.
4.Investigation, analysis and countermeasures of death attitude and death education needs of undergraduate nursing students in different grades

Objective:To comparatively analyze the changes regulation of death attitude and death education needs for undergraduate nursing students in different grades.Methods:From March to June in 2019, 766 undergraduate nursing students from Batch 2015 to Batch 2018 of a medical college were collected in cluster sampling. The investigation was conducted by using the scale of Death Attitude Profile-revised in Chinese version and the scale of death education needs. SPSS 22.0 was used for statistical analysis, LSD test for measurement data, and chi-square test for counting data.Results:There were increasing trends of scores from freshman to junior in dimensions of fear of death, approach-oriented death acceptance and escape-oriented death acceptance ( P < 0.05), and the scores decreased in senior, but without statistical difference compared with junior ( P > 0.05). In neutral death acceptance dimension, the score decreased significantly in the second year ( P = 0.001), and then returned back to the original level in the fourth year. No significantly statistical difference was found in scores of different grades in death escape dimension ( P = 0.038). The score of death education need in the fourth year was the highest ( P < 0.05) and was positively correlated with the score of neutral death acceptance dimension ( P = 0.007). Conclusion:With the increase of school year, nursing students' positive attitude for death demonstrates positive trend, while the level of negative attitude of death are becoming balanced. The fourth year is suggested to be the right time to conduct death education immediately.
5.Effects of Jianpi Yangxue Qufeng Formula on Aquaporin-3 in Model Mice Skin Tissue with Chronic Eczema

Objective To observe the effects of Jianpi Yangxue Qufeng Formula (JPYXQF) on the AQP3 in mice with chronic eczema, and explore mechanism of action. Methods Fifty healthy male mice were randomly divided into 5 groups, namely normal group, model group, positive medicine group and JPYXQF high and low dose groups. Low-dose DNCB and Sennae Fominm were used to establish mice models of chronic eczema with spleen deficiency. JPYXQF groups were treated by JPYXQF for gavage, while the positive medicine group was treated by levocetirizine hydrochloride for gavage. The expression of AQP3 in mice skin tissue was detected by immunohistochemical method. At the same time, the pathological changes of skin were observed. Results The pathology of mice skin lesion showed that JPYXQF has certain recovery effects on the inflammation injury of skin lesion. Compared with the normal group, expression of AQP3 over expressed in model group. Compared with the model group, the expression of AQP3 in all treatment groups significantly decreased, and the staining intensity decreased. In the model group, the average optical density of AQP3 was significantly higher than that in the normal group (P<0.01). Compared with the model group, the treatment groups can reduce the expression of AQP3 in mice skin tissues (P<0.05). Conclusion JPYXQF can reduce the over expression of AQP3 in skin lesion, which is probably its mechanism for the treatment of chronic eczema.
6.Correlation of the levels of serum betatrophin, adiponectin, and interleukin-1β in newly diagnosed type 2 diabetes with different stages of urinary albumin

267 newly-diagnosed patients with type 2 diabetes were divided into normoalbuminuria group [group N-UAlb, urinary albumin to creatinine ratio (UACR)<30 mg/g, n=103], microalbuminuria group(group M-UAlb, UACR 30~300 mg/g, n=88 ) , and macroalbuminuria group ( group L-UAlb, UACR>300 mg/g, n=76). The control group(group NC) consisted of 114 healthy individuals. Serum betatrophin, adiponectin(APN), and interleukin-1β( IL-1β) levels were determined with ELISA methods and the parameters of body mass index (BMI), estimated glomerular filtration rate(eGFR), HbA1C, fasting plasma glucose(FPG), OGTT 2h plasma glucose(2hPG), fasting insulin(FINS), OGTT 2h postprandial insulin(2hPINS), fasting C-peptide(FCP), homeostasis model assessment insulin resistant index(HOMA-IR), and blood lipid were collected. Compared with group NC, the serum betatrophin levels in patients with type 2 diabetes were obviously increased. In patients with type 2 diabetes, betatrophin levels increased along with the increase of UACR and there were significant differences in betatrophin among the three groups(P<0. 01). Betatrophin positively correlated with UACR, HbA1C, FPG, 2hPG, FINS, 2hPINS, HOMA-IR, TC, LDL-C, and TG( r were 0. 785, 0. 225, 0. 136, 0. 241, 0. 386, 0. 223, 0. 411, 0.216,0.193,and0.298,allP<0.05),and betatrophin were also positively correlated with APN and IL-1β(rwere 0. 643 and 0. 710, both P<0. 01). Stepwise multiple regression analysis showed that UACR, HbA1C, FINS, and TG were independent relevant factors affecting betatrophin levels.
7.Retrospective analysis on 57 cases of malignant tumor complicating herpes zoster

Objective To analyze the clinical characteristics of malignant tumor complicating herpes zoster.Methods Fifty-seven cases of malignant tumor complicating herpes zoster in our hospital from September 2003 to November 2014 were performed the retrospective analysis on the anti-tumor treatment history and the clinical characteristics of herpes zoster.Results Most of the cases were over 60 years old.Herpes zoster had the high incidence rate within 2 years of operation and radiochemotherapy.The on-set sites of herpes zoster were most commonly in the chest and back.Conclusion The factors of advanced age,surgery,radiochemo-therapy,poor nutritional status,etc.aggravate or promote the occurrence of herpes zoster in the patients with malignant tumor.
8.Effect of chemotherapy on function of isletβcells in patients with breast cancer

Objective To study the effect of chemotherapy on function of islet βcells in patients with breast cancer . Methods One hundred and fifty-six patients with breast cancer who had not received chemotherapy were enrolled from Sep 2012 to Aug 2013 at Chongqing Cancer Institute .One hundred and twenty-three cases completed 2 cycles of chemotherapy;one hundred and twelve cases completed 4 cycles;and, eighty-six cases completed 8 cycles.Height, weight, waist circum-ference(WC), hip circumference(HC) and blood pressure of each patient were measured .Waist-high ratio calculation (WHtR),waist-hip ratio(WHR) and body mass index(BMI) were calculated.All subjects underwent 75 g oral glucose tolerance test(OGTT)and insulin release test before chemotherapy,2 cycles,4 cycles and 8 cycles of chemotherapy, respec-tively.Besides, homeostasis model assessment of insulin resistance index ( HOMA-IR), Matsuda index and pancreatic βcells HOME(HOME-β) were calculated.Results Fasting blood glucose before chemotherapy and after 2 cycles, 4 cycles and 8 cycles of chemotherapy was (4.84 ±0.56), (4.78 ±0.61), (5.02 ±0.56) and (5.96 ±0.52) mmol/L, respec-tively.After therapy, the change in fasting blood glucose after 2 cycles and 4 cycles of chemotherapy was not statistically significant(P>0.05), but was significantly higher after 8 cycles than before therapy(P<0.05).2 h blood glucose before chemotherapy and after 2 cycles, 4 cycles and 8 cycles of chemotherapy was (5.43 ±0.50), (5.66 ±0.58), (8.30 ± 0.66) and (9.65 ±0.80) mmol/L, respectively.With chemotherapy treatment increased , the HOME-IR increased gradu-ally and the sharpest one was after 8 cycles, Matsuda index decreased gradually and the biggest drop was after 4 cycles,and HOME-βdecreased gradually after a transient increase .Conclusion Chemotherapy for breast cancer patients may lead to insulin resistance , pancreatic βcell function decline and the emergence of abnormal glucose tolerance .Four cycles of chemotherapy shoucd be followed by hypoglycemic drugs .
9.Application of extensively hydrolyzed protein formula for very low birth weight infants

Objective:To evaluate the clinical efficacy of extensively hydrolyzed protein formula(eHF) in very low birth weight(VLBW) infants.Methods:A prospective controlled signal-center trial was conducted in this study, the preterm infants with gestational age of 28-33 weeks and birth weight of 999-1 500 g who were hospitalized at Department of Neonatology, Zhuhai Maternal Hospital within the first 12 hours between January and December 2018, were selected.They were assigned into breast feeding group(HM) and formula feeding group according to the mothers′ disease and parents′ breastfeeding willingness.The formula feeding group was assigned into eHF group and preterm formula (PF)group according to the parents′ breastfeeding willingness.The infants discharging or dead before achieving full feeding, discharging within 28 days after birth, with congenital malformation (complex congenital heart disease, digestive system malformation, etc) and severe sepsis were rolled out. Chi- square test and One- Way ANOVA were used for statistical analysis.Prospective study was conducted among the 3 groups comparing the incidence of feeding intolerance, duration of meconium discharge, the time to regain birth weight and reach full enteral feeding, average hospital stay, incidence of necrotizing enterocolitis of newborn(NEC), cholestasis and extrauterine growth retardation(EUGR), the growth rate of head circumference, length and weight in the first 4 weeks of life, and blood biochemical indices at the first 2 weeks and 4 weeks of life. Results:A total of 102 infants were enrolled, 35 cases in the eHF group, 37 cases in the PF group and 30 cases in the HM group.Compared with the PF group [54.0%(20/37 cases)], the eHF group[22.0%(8/35 cases)] and the HM group [16.7%(5/30 cases)] had lower incidence of feeding intolerance, and the differences were statistically significant( χ2=7.366, 9.901, all P<0.05). The time to regain birth weight[(8.9±1.8) d, (9.1±1.4) d vs.(10.8±2.9) d], time for achieving full enteral feeding [(42.8±2.8) d, (42.3±3.3) vs.(45.5±3.4) d], the duration of meconium discharge [(7.2±1.8) d, (6.6±1.8) d vs.(8.7±2.1) d], and average hospital stay [(52.9±1.1) d, (52.3±1.2) d vs.(54.1±1.2) d]in the eHF group and HM group were shorter than those in PF group, and the differences were statistically significant(all P<0.05); and there was no statistically significant difference between the eHF group and the HM group(all P>0.05). There was no statistically significant difference in the incidence of incidence of NEC, cholestasis and EUGR, the growth rate of head circumfe-rence, length and weight in the first 4 weeks of life, the serum albumin, creatinine, urea nitrogen at first 2 weeks and 4 weeks of life among the 3 groups (all P>0.05). Compared with the PF group, the serum total bilirubin levels in the eHF group and the HM group were lower at 2 weeks [(109.4± 4.6) μmol/L, (110.2±1.0) μmol/L vs.(115.0±7.6) μmol/L]and 4 weeks after birth[(79.3±9.7) μmol/L, (80.0±1.7) μmol/L vs.(81.5±8.4)μmol/L], and the differences were statistically significant(all P<0.05), but no statistically significant difference was found between the eHF goup and the HM group(all P>0.05). Conclusions:For VLBW infants, eHF can reduce feeding intole-rance, promote defecation, achieve full feeding faster, promote bilirubin metabolism, shorten hospital stay, does not affect growth and development in short-term.

Objective:
To study the specific killing effect of CD4 membrane protein targeted chimeric antigen receptor modified T (CAR-T) cell.
Methods:
The second generation CD4 targeted chimeric antigen receptor containing 4-1BB costimulation domain was insert into lentiviral vector through recombinant DNA technology. Lentivirus was prepared and packaged by 293T cells with four plasmids. Beads activated T cells were transduced with lentivirus and the transduction efficiency was checked with Protein L and flow cytometry. T cell subsets and IFN-γ concentrations were detected with probe-tagged antibody and cytometric bead assay.
Results:
①The transduction efficiency of activated T cells with prepared lentivirus were 50.0%-70.0%. A subset of CD8+ T cell acquired dim expression of CD4 membrane protein after activation. CD4+T cell and CD8+CD4dim T cell were gradually killed by CD4 targeted CAR-T post lentivirus transduction. ②The kill efficacy of CD4 targeted CAR-T cell and control T cell toward KARPAS 299 T cell at an E∶T ratio of 8∶1 for 24 h was (96.9±2.1)% and (11.2±3.1)%, CAR-T cell has a higher killing efficacy than control T cell (
