1.Suppression of cartilage injury in osteoarthritis by IL-1Ra ex vivo gene therapy in rabbit

Objective To assess the effect of IL 1Ra gene transfer on amelioration of symptoms and cartilage injury of osteoarthritis (OA) in rabbit. Methods Thirty New Zealand rabbits were divided into 3 groups. Synovial membrane was harvested from the left knee, and synovial fibroblasts were isolated and cultured. Fibroblasts from each rabbit were traanfected using the retrovirus with either IL 1Ra gene (group 3) or marker gene (group 2) in vitro . OA was induced in the right knee of all rabbits by anterior cruciate ligament transection (ACLT). The autologous cells tranfected with IL 1Ra gene or marker gene were respectively injected into the right knee of group 3 and 2. At the end of 2nd and 4th week, IL 1Ra level in synonival fluid (SF) of the right knee were determined by ELISA. The results were evaluated by grading local pain stimulating response, change of gait, joint swelling, and range of joint motion. The degree of injury to the cartilage was graded by the staining of India ink and pathological examination. Results The IL 1Ra level in the right knee SF of group 3 were 20 16?1 8 ng/ml and 4 82?0 52 ng/ml at the end of 2 and 4 weeks after gene delivery, respectively, while that in the group 2 and group 1 was very low. In IL 1Ra gene transfection group, the symptoms of the joint injury and the degree of cartilage lesion were less severe compared with control group. Conclusion This study showed that IL Ra transfer could raise IL 1Ra level in knee joints, thereby alleviate the symptoms and cartilage injury in experimental osteoarthritis
2.A clinical analysis of reninoma-induced hypertensive crisis associated with reversible posterior encephalopathy syndrome

Objective Reninoma is a rare benign tumor of the renal juxtaglomerular cell apparatus that causes hypertension and hypokalemia via hypersecretion of renin,while it is extremely rare that reninoma induced hypertensive crisis with reversible posterior encephalopathy syndrome (RPES).To improve the clinical understanding for this disease,we conducted a case-analysis.Methods To analyze the clinical and pathological data of a case of reninoma-induced hypertensive crisis with reversible posterior encephalopathy syndrome,who was admitted to Peking University First Hospital in November,2007 and follow-up.Results This was a 16-year old female patient,onset with suddenly spasm with loss of consciousness,while blood pressure stepped up to 210/140 mm Hg( 1 mm Hg =0.133 kPa),and the head magnetic resonance imaging (MRI) revealed “multiple long-T2 signal”,and hypopotassaemia(2.8-3.2 mmol/L),urine protein positive,ultrasoundcardiogram revealed left ventricular hypertrophy,laboratory study revealed hyperreninism (38.23ng · ml-1 · h-1,normal range 0.07-1.15 ng· ml-1 · h-1 ) and hyperaldosteronism(660.9 ng/L,normal range 60-174 ng/L),abdominal CT-Scan revealed a mass at right kidney,blood pressure achieved safety range and the head MRI was rechecked and revealed “the abnormal long-T2 signal disappeared”.The clinical diagnosis was reninoma induced hypertensive crisis with RPES.The tumor was resected and the pathologic diagnosis was reninoma.The patient remained normotensive in the postoperative period without any medication.Conclusions Reninoma represents a rare but surgically curable cause of hypertension,thus the clinical suspicion of it is very important in young patients.If the diagnosis is confirmed,positive treatment must be done immediately to improve the prognosis.The most common cause of RPES is hypertension,and the diagnosis depends on the distinctive head MRI.There is always a good prognosis with the decline of blood pressure rapidly.
3.Suppression of chondrocyte apoptosis in the osteoarthritis by interleukin-1Ra ex vivo gene therapy

Objective To explore suppression of chondrocyte apoptosis by IL-1Ra gene transfer to the knees of experimental osteoarthritis (OA).Method ①Thirty New Zealand white rabbits were divided into 3 groups,partial synovectomy of the left knee had been done.Synovial fibroblasts (SF) were isolated and cultured.②Fibroblasts from each rabbit were transduced by using the retrovirus with either IL-1Ra gene (group 3) or marker gene to (group2) in vitro.③ OA was induced in right knee of rabbits by anterior cruciate ligament transection (ACLT).④ The autologous cells transduced with IL-1Ra gene or marker gene were injected into the right knee of group 3 or 2.⑤At the second week and at the forth week,IL-1Ra level in SF was determined by ELISA.⑥Cartilage section were stained for checking histologic changes and chondrocyte apoptosis were determined using TUNEL method.Results The IL-1Ra level in the SF of group 3 were (20.6?1.8)ng/ml and (4.82?0.52)ng/ml after 2 and 4 weeks,after gene delivered respectively,and the IL-1Ra level in the group 2 and group 1 were very lowat both 2 and 4 weeks.Approximately 40% of chondrocytes underwent apoptosis in group 2,whereas apoptosis were inhibited in group 3,only 18% of chondrocytes underwent apoptosis,and 2%~5% of chondrocytes underwent apoptosis in normal joint.Conclusion This study showes that a local increase IL-1Ra level in OA keen joints by intraartilcular injection of IL-1Ra gene transduced synovial cells can suppress articular cartilage chondrocyte apoptosis.

5.Research Value of Multi-clone Cell Line: A Comment for the 30th Anniversary of J6-1 Human Leukemic Cell Line.

J6-1 cell line is the first leukemic cell line established in China. It is a multi-clone cell line infected with both EBV and HHV-6. Many cytokines, receptors and other genes were cloned from J6-1 cell line since its establishment 30 years ago. Valuable information on leukemic characteristics and functions were obtained from the studies on this cell line, which could be categorized into several research subjects. These achievements implied the unique research value of multi-clone cell lines. This comment focuses attention on research advance of the J6-1 leukemic cell line in 30 years, including heterogeneity and multi-cloning of J6-1 cells, survival mechanism of J6-1 cell populations, abnormal intercellalar communication of J6-1 cells with its significance and inspiration from J6-1 cell line.

6.Expressions of ADM andβ-catenin in human osteosarcoma and their clinical significance

ABSTRACT:Objective To study the expressions of adrenomedullin (ADM)andβ-catenin in human osteosarcoma of different differentiation grading and their clinical significance.Methods We chose 33 cases of human osteosarcoma with different pathological grading (14 cases of high differentiation grading and 1 9 cases of low grading)and 10 cases of human osteochondroma to detect the expressions of ADM protein,total (T)β-catenin and phosphorylated (P)β-catenin using immunohistochemical method. Results The expression of ADM was detected in the cytoplasm.In osteosarcomas of higher and lower differentiation grading,the positive rate of ADM was 57.1% (8/14)and 100% (1 9/1 9),respectively,with significant differences (P =0.003<0.05).The expression of T-β-catenin was observed in both the cytoplasm and the nucleus in higher differentiation grading and in only cytoplasm in lower grading.In osteosarcomas of higher and lower differentiation grading,the positive rate of T-β-catenin was 85.7%(12/14)and 94.7% (18/1 9),with no significant difference (P =0.561>0.05).The expression of P-β-catenin was detected in the cytoplasm.In osteosarcomas of higher and lower differentiation grading,the positive rate of P-β-catenin was 35.7% (5/14)and 78.9% (1 5/1 9),with a significant difference (P =0.029 <0.05).There was a significant correlation between ADM and P-β-catenin (r =0.424,P =0.025<0.05),but no significant correlation between ADM and T-β-catenin (P = 0.078 > 0.05 )or between T-β-catenin and P-β-catenin (P = 0.052 > 0.05 ).Conclusion The expressions of ADM and P-β-catenin proteins and the different distributions of expression of T-β-catenin protein in cytoplasm and/or nucleus are important indicators for judging the pathological grading of osteo-sarcoma.
7.Clinicopathological analysis of orbital neurilemoma:a report of 8 cases

Objective The purpose of this study is to study the clinical and pathological features of neurilemoma .Methods We observed the clinicopathologic features and immunohistochemical staining from eight patients with orbital neurilemoma between 2010.1~2012.12.Results Eight patients with classic neurilemoma were included in the study ,in which there were five males and three females ,aged between 21 and 63,mean age 35.The main symptom of the patients was exophthalmos ,including five cases of right eyes and three left eyes;2 cases of orbital floor and six above orbit ,lasting for one to ten years .The tumor diameter ranged between 1cm and 5 cm,an average of 3 cm,being pale and light yellow color .There were five cases of type Antoni A and one case of type was Antoni B among the six classic type neurilemoma .Two cases of ancient were neurilemoma ,in which one case was the histological structure of the classic type neurilemoma ,but there were more hypertrophy tumor cells , chromatin was coarse block atypia cells .The other one case with cells arranged disorderly ,which was mainly fine striated cells with scattered deeply stained atypia cells ,stromal transparent degeneration ,cystic degeneration .Im-munohistochemistry results showed that S -100(+),vimentin(+),ki67(-).Conclusion Antoni type B and ancient schwannoma are rare ,with complicated histologic characteristics .Combined with clinical features and im-munohistochemistry staining ,it can be diagnosed .
8.Effects ofLiuwei-Dihuang Pills combined with Ginkgo Biloba Leaves Extract Tablets on complications of type 2 diabetes mellitus

Objective To investigate the effects ofLiuwei-Dihuang Pills combined with Ginkgo Biloba Leaves Extract Tablets on diabetic complications in patients with type 2 diabetes mellitus (T2DM). Methods Seventy-eight patients with T2DM were randomized into a TCM treatment group and a conventional treatment group by random number table method, 39 in each group. The patients in the conventional treatment group were regularly using antihyperglycemic drugs or insulin. On the basis of conventional treatment group, the patients in the TCM treatment group were additionally treated withLiuwei-Dihuang Pills and Ginkgo Biloba Leaves Extract Tablets. Intima-media thickness (IMT) of internal carotid artery, vibration perception threshold (VPT) and diabetic retinopathy in both groups were detected before the treatment, 12 and 36 months after the treatment.Results The VPT in the left foot 12 months after the treatment in the TCM treatment group were significantly lower than that in the conventional treatment group (13.98 ±4.38 Vvs. 18.70 ±2.43 V;t=2.764, P=0.008). IMT of internal carotid artery (left: 0.81 ± 0.16 mmvs.0.70 ± 0.10 mm,t=3.120,P=0.003; right: 0.81 ± 0.17 mmvs. 0.73 ± 0.12 mm,t=2.286,P=0.026), VPT (left foot: 14.82 ± 6.45 Vvs. 20.63 ± 9.75 V,t=2.714, P=0.009; right foot: 16.73 ± 7.10 Vvs. 20.73 ± 10.35 V,t=2.001,P=0.048) and incidence of diabetic retinopathy (35.9%vs. 53.8%;χ2=5.804,P=0.016) 36 months after the treatment in the TCM treatment group were significantly lower than those in the conventional treatment group.ConclusionsLiuwei-Dihuang Pills combined with Ginkgo Biloba Leaves Extract Tablets can effectively reduce the progression of diabetic complications in patients with T2DM.
9.MRI features of joint tuberculosis of the appendicular skeleton in children

Objective To investigate the MRI features of children's joint tuberculosis of the appendicular skeleton, and to evaluate the diagnostic value of MR imaging. Methods Sixteen consecutive patients confirmed clinically and pathologically were examined with SE-T1WI, FSE-T2WI, FSE-PDWI and PD-SPIR in Philips Achieva 3.0T superconduct MR scanner. Axial, sagital and coronal images were obtained. Articular structure was all analyzed based on the affected parts and features of MRI. Results Hip joint and knee joint were most likely to suffer (each 25.00%). Metaphysis was affected in 13 patients with long signal intensity on T1WI and long signal intensity on T2WI. The changes of articular cartilage presented as abnormality of signal intensity in 6 patients with focal high signal intensity on T2WI. Thirteen patients presented as joint effusion. Synovial thickening in 4 patients showed moderate homogeneous signal on T1WI and slightly lower signal on T2WI. Articular soft tissue was involved in 16 patients, including the formation of cold abscess in 2 and cellulitis in 1 patient, showing long signal intensity on T1WI and long signal intensity on T2WI. Conclusion MRI has the advantage of being able to detect early abnormalities of bone marrow, cartilage damage and to evaluate joint effusion, synovial thickening and soft tissue lesions.
10.Chemical constituents of flavonoids in Lysimachia clethroide

AIM:To study the chemical constituents of Lysimachia clethroide Duby. METHODS:Chromatographic techniques,including MS and NMR,were used to isolate and purify the constituents based on column chromatography separation on silica gel,Sephadex LH-20 and Semipreparative RP-HPLC. RESULTS:Eleven flavonoids were obtained and elucidated as: kaempferol (1),kaempferol 3-O-?-D-glucopyranoside (2),kaempferol 3-O-?-D-galactoside (3),kaempferol 3-O-?-D-(6″-p-coumaroyl)-glucopyranoside (4),quercetin (5),Isorhamnetin (6) ,quercetin 3-O-?-D-galactoside (7),quercetin 3′-methoxy-3-O-?-D-galactoside (8),quercetin 3-O-?-D-(6″-p-coumaroyl)-galactoside (9),4′-methoxy-5,6-dihydroxyisoflavone-7-O-?-D-glucopyranoside (10),7-O-glucosyl liquiritigenin (11). CONCLUSION: Compounds 3,6 were isolated from this plant and compounds 8-11 were isolated from this genus for the first time.
