1.In Vitro Effect of Fungicides, Plant Extracts and Smoke on Conidial Germination of Fusarium oxysporum Root Rot Pathogen of Piper betle.

Five fungicides such as rovral, bavistin, cupravit, dithane M-45 and thiovit were tested against conidial germination of Fusarium oxysporum. Dithane M-45 was the most effective against the fungus. Rests of the fungicides were more or less effective in the inhibition of conidial germination after 5~30 minutes immersion in 500~2500 ppm concentration. Five plant extracts(leaf extracts of Ocimum sanctum, Lantana camera, Calotropis procera, Azadirachta indica and Vinca rosea) found to be more or less effective against the fungus. 'Dhup' somke have good inhibitory effect on conidial germination of F. oxysporum.

Effect of ethanolic extracts of Lawsonia inermis, Azadirachta indica, Vinca rosea, Tagetes patula, Ocimum sanctum, Colocasia antiquorum, Adhatoda vasica, Moringa oleifera, Datura metel and Curcuma longa leaf on conidial germination, mycelial growth and sporulation of Aspergillus flavus, A. niger and A. fumigatus were examined. The conidial germination of A. flavus and A. fumigatus were most inhibited by the extract of L. inermis, while that of A. niger was inhibited by A. indica. Other tested plant extracts have a good effect on conidial germination on the selected fungi. The highest mycelial growth of A. flavus (37 mm) was found in V. rosea, but in case of A. niger and A. fumigatus it (38 and 39 mm) was found in D. metel. The lowest (4, 9 and 6 mm) respectively mycelial growth of these fungi found in L. inermis. The highest sporulation (75 x 10(4)/ml) of A. flavus was counted in V. rosea, but in case of A. niger and A. fumigatus those (45 x 10(4) and 55 x 10(4)/ml) were in D. metel and the lowest (5 x 10(4), 12 x 10(4) and 9 x 10(4)/ml) respectively sporulation of these fungi counted in L. inermis plant extract medium.

Twenty plant extracts were tested against mycelial growth, sclerotium formatiom and dry weight of mycelium with sclerotia of Sclerotium rolfsii Sacc. The highest(90 mm) mycelial growth was measured in Adhatoda vasica, Tegetes erecta, Allium cepa, and Curcuma longa. The lowest(25 mm) was in Azadirachta indica. No mycelial growth was found in any concentration of cow, buffalo, and goat urine. The highest(90 mm) and the lowest(15 mm) mycelial growth were measured in Biomil and Urea, respectively. No mycelial growth was observed in Zinc. The highest(60 mm) and the lowest(2 mm) mycelial growth were recorded in Macuprex(Dodine; 65% WP) and Boron(100% Boric acid and 17% Boron) respectively. Mycelial growth was totally inhibited in Rovral(Iprodione; 50% WP).

A newly synthesized Nickel (II) tyrosine complex was screened as potential antimicrobial agent against a number of medically important bacteria (Bacillus subtilis, Streptococcus beta-haemolytica, Escherichia coli, Shigella dysenterae) and fungi (Aspergillus fumigatus, Candida albicans, Aspergillus niger, Aspergillus flavus, Penicillium sp.) strains. were used for antifungal activity. The antimicrobial activity was evaluated using the Agar Disc method. Moreover, the minimum inhibitory concentration of the complexes was determined against the same pathogenic bacteria and the values were found between 4~64 microg ml(-1). Brine shrimp bioassay was carried out for cytotoxicity measurements of the complexes. The LC50 values were calculated after probit transformation of the resulting mortality data and found to be 6 microg ml(-1).

5.Juvenile Polyps in Bangladeshi Children and Their Association with Fecal Calprotectin as a Biomarker

Purpose:
Colonoscopy is considered the most reliable method for the diagnosis of juvenile polyps. However, colonoscopic screening is an invasive and expensive procedure. Fecal calprotectin (FCP), a marker of intestinal inflammation, has been shown to be elevated in patients with polyps. Therefore, this study aimed to evaluate FCP as a screening biomarker for the diagnosis of juvenile polyps.
Methods:
This cross-sectional, observational study was conducted at the Pediatric Gastroenterology and Nutrition Department, Bangabandhu Sheikh Mujib Medical University, Dhaka, Bangladesh. For children with polyps, colonoscopic polypectomy and histopathology were performed. FCP levels were analyzed before and 4 weeks after polypectomy in all patients. Information was recorded in a datasheet and analyzed using the computer-based program SPSS.
Results:
The age of the children was between 2.5 and 12 years. Approximately 93% of the polyps were found in the rectosigmoid region. Children with juvenile polyps had elevated levels of FCP before polypectomy that subsequently normalized after polypectomy. The mean FCP levels before and after polypectomy were 277±247 µg/g (range, 80–1,000 µg/g) and 48.57±38.23 µg/g (range, 29–140 µg/g) (p<0.001), respectively. The FCP levels were significantly higher in patients with multiple polyps than in those with single polyps. Moreover, mean FCP levels in patients with single and multiple polyps were 207.6±172.4 µg/ g and 515.4±320.5 µg/g (p<0.001), respectively.
Conclusion
Colonic juvenile polyps were found to be associated with elevated levels of FCP that normalized after polypectomy. Therefore, FCP may be recommended as a noninvasive screening biomarker for diagnosis of colonic juvenile polyps.

7.Transverse Cerebellar Diameter as a Sonographic Parameter for Estimating Gestational Age: an Experience From Yemeni Population

Introduction: Estimation of gestational age (GA) is clinically crucial for managing pregnancy and assessing the foetal anatomy, growth and development. Transverse cerebellar diameter (TCD) has been reported as an accurate tool for dating the pregnancy. This study aimed to determine the accuracy of foetal TCD for dating the pregnancy and to construct a reference chart for GA of Yemeni foetuses. Methods: We conducted this prospective cross-sectional study among 400 Yemeni pregnant women between 18 and 40 weeks of gestation provided that they were with known last menstrual period and singleton normal pregnancies. Sonographic TCDs were measured for each foetus. The mean TCD was measured for gestational weeks separately, and a polynomial regression model was then used to predict the GA by TCD. Results: There was a robust correlation between GA and TCD (r = 0.995, p <0.001). The coefficient of determination was 0.989, which indicates that the TCD explains 98.9% of GA change. Conclusion: TCD is an
accurate tool to estimate the GA in second and third trimesters, including final weeks, among Yemeni pregnant women as in developed countries. The narrow range between the 5% and 95% percentiles is a significant indication of the accuracy of the TCD in estimating the GA. Therefore, clinicians are recommended to use TCD for estimating the GA and should be trained on measuring it accurately.
8.Frequency of Gallbladder Stones Among Patients Underwent Abdominal Ultrasound in a Tertiary Hospital in Sana’a City, Yemen.

Introduction: The Gallbladder stone (GBS) disease is most commonly asymptomatic that may lead to several
complications such as ascending cholangitis and obstructive jaundice. In this study the frequency of gallbladder stones among patients referred for abdominal ultrasound at the University of Science and Technology
hospital (USTH), Sana’a – Yemen, have been estimated during the period between January and June 2013.
Methods: This study is a record-based and conducted at the radiology department in USTH, on cases
underwent abdominal ultrasound during the period from January – June 2013. Information were collected
from abdominal ultrasonography reports. Results: In this study 4935 patients’ records are included. Of them,
2541 were males and 2394 were females. The frequency of patients with GBS was 5.53%. Multiple stones
were observed in 3.57% of patients and 4.34% patients had large stones with size ≥ 5 mm. Females had
significantly higher frequency of GBS (8.0%: 191/2394) than males (3.2%: 82/2541) (P < 0.001). It was found
that, no significant difference between males and females in harboring small stones (< 5mm) (P = 0.251).
However, significantly higher frequency of large GBS (≥ 5 mm) was found among females compared to
males (P < 0.001). The frequencies of GBS, small size of GBS and large size of GBS have significantly increased
with increasing age (P < 0.001). Conclusion: In this study it was found that females had significantly higher
frequency of GBS than males. No significant difference between males and females in harboring small stones.
There was a significantly higher frequency of large GBS was found among females compared to males. The
frequencies of GBS, small size of GBS and large size of GBS have significantly increased with increasing age.
9.Synthesis, antitubercular activity, and molecular docking studies of Benzyl-modified 8-hydroxyquinolines

Background: Infection with Mycobacterium tuberculosis, the causative agent of TB, is responsible for one of the global epidemics. Thus, new drugs are needed that do not confer cross-resistance with currently administered front-line therapeutics. Quinoline-based natural products and synthetic derivatives have been extensively explored for antitubercular activity.
Objective: The main goal of this study was to prepare a collection of benzylated 8-hydroxyquinoline derivatives through synthesis and assess their antitubercular activity along with a molecular docking study to clarify their biological mechanism of action.
Methodology: The benzylated 8-hydroxyquinoline derivatives were synthesized using Williamson synthesis methods. Antitubercular activity was assessed against fast replicating M. tuberculosis H??Rv using Microplate Alamar Blue Assay (MABA) and non-replicating cultures using Low-Oxygen Recovery Assay (LORA). Molecular docking studies were carried out against enoyl-acyl carrier protein reductase (InhA).
Results: Five benzylated 8-hydroxyquinoline derivatives were synthesized in moderate yields and characterized using NMR spectroscopy. MABA and LORA assays indicate compounds 3-5 as the most inhibitory derivatives with MIC90's ranging from 6.38 to 54.28 ?M. Molecular docking against InhA showed modest 90 binding energies for compounds 4 (-8.5 kcal/mol) and 5 (-8.6 kcal/mol).
Conclusion: Findings suggest a rationale for the further evolution of this promising series of antitubercular quinoline small molecules. Structure-activity analysis shows that an 8-benzyl moiety with chlorine atom/s is important for improved activity against replicating and non-replicating M. tb. H??Rv. This is also supported by our in silico studies.
