1.A study of lymphoblast transformation in recurrent spontaneous abortion

This study is aimed to investigate the activity of lymphocytes of recurrent spontaneous aborters, their response to phytohemagglutinin (PHA) mitogen was studied by comparing with that of normal pregnancy and normal non-pregnancy women. The results are as follows: there is no or a significant decrease of suppression of lymphoblast transformation in recurrent spontaneous abortion comparing to that of normal pregnancy women. It should be considered all of three parameters: absolute counts of OD with PHA, non-PHA and stimulation index as a general analysis for lymphoblast transformation.

2.Effect of Vitexin on Restoration of Lymphocyte Transformation in Breast Cancer patients after Radiotherapy

60 breast cancer patients in the II and III phrase were treated on the regime: Surgery with after radiotherapy at the II radio Department, K Hospital in Ha Noi. These breast cancer patients after radiotherapy, Vitexin was used within continuous 2 months and 30 patients in the control group with placebo of 50mg Vitamine C. The number of lymphocytes were significantly increased after radiotherapy. In the presence of phytohemaglutinin, Vitexin also restores the number of lumphocytes to the level before radiotherapy significantly.

3.Research advances in the expression of co-stimulatory molecules and signaling pathways in γδT cells.

Human T lymphocytes are divided into αβT cells and γδT cells on the basis of the different expressions of T cell receptors. In recent years, the studies of the regulation of T cell activation and tolerance by co-stimulatory molecules and their signaling pathways in αβT cell have made remarkable progress; however, relatively fewer investigations have been performed on γδT cells. A clearer understanding of the roles of co-stimulatory molecules and their signaling pathways in the positive/negative regulation of γδT cells at different stages will provides new insights for the treatment of viral infections, cancer, autoimmune diseases, transplant rejection, and other conditions.

4.Effects of Different Stimulators on Proliferation of Peripheral Blood Lymphocyte Subsets.

OBJECTIVE:
To investigate the effects of different stimultors (PHA, PMA and IL-2) and culture systems (PBMC and whole blood) on the proliferation of human peripheral blood lymphocyte subsets, so as to provide the experimental basis for selecting the appropriate system according to the experimental purposes.
METHODS:
A total of 10 ml serum samples were collected from healthy volunteers (n=6). The 300 μl whole blood was directly used to detect lymphocyte subsets by flow cytometry. The 400 μl whole blood were inoculated respectively with 3 different stimulators at 37℃ and 5% CO2 for 60 h; Three different stimulators were also added to the PBMC which were isolated from 2 ml whole blood. Then the proliferation ability of lymphocyte subsets was analyzed by flow cytometry.
RESULTS:
After the PBMC were stimulated with PHA, CD4CD8CD3 lymphocytes were the most subset; The proportion of CD3CD4 T lymphocytes and CD3CD19 B lymphocytes decreased after being stimulated by PMA (P<0.01, P<0.05); the lymphocyte subset ratio had no significant change after being stimulated by IL-2. After the whole blood system was stimulated with PHA, the CD4/CD8 T lymphoblasts were main subsets, the counts of B lymphocytes and NK cells were reduced; after being stimulated with PMA, the number of CD8CD3 T lymphoblast and CD4CD8T lymphocytes increased, the B/NK cells were not distinguished with the surface markers; after the whole blood system was stimulated with IL-2, the proportion of NK cells significantly increased (P<0.05).
CONCLUSION
Lymphocyte proliferation stimulated by PMA is the fastest, while the effect of IL-2 on the lymphocyte subset proportion stimulated by IL-2 is the minimal. After being stimulated by PHA the division cycles of lymphocyte are the most.

5.Transmembrane Adaptor Proteins Positively Regulating the Activation of Lymphocytes.

Engagement of the immunoreceptors initiates signaling cascades resulting in lymphocyte activation and differentiation to effector cells, which are essential for the elimination of pathogens from the body. For the transduction of these immunoreceptor-mediated signals, several linker proteins termed transmembrane adaptor proteins (TRAPs) were shown to be required. TRAPs serve as platforms for the assembly and membrane targeting of the specific signaling proteins. Among seven TRAPs identified so far, LAT and LIME were shown to act as a positive regulator in TCR-mediated signaling pathways. In this review, we will discuss the functions of LAT and LIME in modulating T cell development, activation and differentiation.

6.The serum MOA concentration and response of lymphocyte transform in patients with breast cancer who treated by radiation and antioxidant agent - vitexin

A study on 115 patients with breast cancer treated by postoperative radiation and vitexin within 6 months has shown that there were changes of serum concentration of products of lipid peroxydation in patients with breast cancer in the different duration of treatment. An increase of serum MDA after radiation showed an increase of oxidation stress. Vitexin helped the establishment of oxidation balance only after a month of the treatment. The cell-mediated reduction of immunoresponse was a outcome of oxidation stress. The recovery of response of lymphocyte transform in the peripheral blood was corresponding to the improvement of oxidation balance by drug.

7.A Case of Allopurinol-Induced Fixed Drug Eruption Confirmed With a Lymphocyte Transformation Test.

Allopurinol is one of the causative drugs that induce fixed drug eruption (FDE). The lymphocyte transformation test (LTT) is a safe and reliable diagnostic procedure for drug allergy, but is reported to be rarely positive in patients with FDE. In the current case, we performed an LTT and successfully confirmed allopurinol as the offending drug. This case report suggests that an LTT should be an optional diagnostic tool for FDE or delayed reaction due to allopurinol.

9.Investigation of Cell Mediated Immunity in Patients with Behcet's Syndrome, Using the DNCB Sensitization.

Various immunologic investigations of 159 patients with Behqets syndrome undertaken included; the DNCB sensitization test, total T-cells and T-cell subsets and the lymphocyte transformation test using PHA. The percentage of positive esponsiveness to DNCB decreased in the order of possible (65%), suspected (60%), incomplete (37%) and complete type (37%), The number of patients with impaired LTT was larger in the group of patients with DNCB( + ) responsiveness (seventeen of 32) than in DNCB(+) group (six of 25), There was significant impairment of cell mediated immunity in Behcets syndrome compared to normal subjects, when analyzed by the impaired LTT and decreased proportions of helper T-cells.

10.Immunological aspects in patients with chronic active hepatitis--cellular immune responses.

We studied host immune parameters which might be related to the activity and the pathogenetic mechanism of chronic active hepatitis. The subjects consisted of 45 cases with hepatitis B virus surface antigen (HBsAg)-positive chronic active hepatitis (CAH), 44 HBsAg-negative CAH, 22 with inactive chronic hepatitis, and 45 cases of normal persons, hepatitis B virus (HBV) carriers, or the patients with acute myocardial infarction. The in vitro assay for the in vivo activated lymphocytes was performed by measuring spontaneous thymidine uptake (SLT) of lymphocytes isolated from peripheral blood. SLT was significantly (p less than 0.001) elevated in cases with HBsAg-positive (1227 +/- 806 cpm) and-negative CAH (1017 +/- 559 cpm) compared to the patients with inactive chronic hepatitis (347 +/- 79 cpm) and to the control group (320 +/- 106 cpm). SLT values observed in 7 cases with active disease (group I and II), in which remission and relapsing phase could be assessable, were elevated from 648 +/- 121 cpm in remission phase to 1548 +/- 606 cpm one to two weeks before the appearance of biochemical evidence (SGPT) of relapse. This pattern of SLT elevation, however, was not observed in patients with inactive hepatitis. Neither the abnormal distribution of T-celi subsets nor the presence of conventional HBV markers were related to the elevated SLT value. Our findings may therefore indicate that SLT might be useful in assessment of the disease activity in patients with CAH.
