1.CHANGES OF c fos GENE IN PERIAQUEDUCTAL GRAY OF AUDIOGENIC SEIZURES PRONE RATS FOLLOWING AUDIOGENIC SEIZURES

Objective\ To find out the relationship between the periaqueductal gray (PAG) and the audiogenic seizures. Methods The neuronal(Nissl)staining method and the immunocytochemical method were used to identify the Fos postive neurons in the PAG of the audiogenic seizures prone rats(P77PMC) after audiogenic seizures. Results There were lots of Fos positive neurons in the dorsolateral, dorsal and caudoventrolateral subdivisions of the PAG in P77PMC rats two hours post seizure, and the percentage of Fos positive neurons was significant higher than that of rats in control groups. Conclusion A large number of neurons in the PAG of the audiogenic seizures prone rats participated in the process of the audiogenic seizures.
2.QUANTITATIVE ANALYSIS OF SUBSTANCE PCONTAINING NERVE FIBERS AND CELLS IN THE NUCLEUS OF DORSOLATERAL FUNICULUS OF THE RAT

Using immunocytochemical method and image analysis computer system, quantitative analysis of substance P (SP) containing nerve fibers and neurons in the nucleus of dorsolateral funiculus (NDLF) was studied in the rat spinal cord. Under light microscope, in the white substance of the whole length of the spinal cord, a concentrated triangular or cuneiform area of SP-immunoreactive positive fibers and cells could be observed in the NDLF region. Using the image anlysis computer system (IBAS 2000) the outline of the whole SP stained area in the NDLF was drawn with a light point of the Mouse and this whole area was measured and named as the reference area (REFAREA). We selected the optimal gray level of the image in NDLF region by the Interactive Segment as the threshold. The sum of the SP labelled areas was called the field area (FAREA). The ratio of the FAREA to REFAREA (AREA%) represented the percentage of density to area of the SP labelled substance in NDLF. The mean value of REFAREA, FAREA, and AREA% are: 47931.38?m~2, 21995.25?m~2, and 45.89% in the cervical segments; 31575.15 ?m~2, 7646.85?m~2, and 24.22% in the thoracic region; 26889.89?m~2, 8470.66?m~2, and 31.50% in the lumbar and sacral segments, respectively. In short, according to the density of SP positive fibers and neurons in NDLF and the size of their distributive area, the sequence of density and size from highest to lowest is: cervical, lumbar, sacral and thoracic segments, with the lowest one at thoracic 8-10 segments.
3.THE ORIGINS OF THE DESCENDING PRO-PRIOSPINAL TRACT AND THE CONNEC-TIONS OF THE NUCLEUS DORSOLA-TERAL FUNICULUS IN THE SPINAL CORD OF THE RAT

HRP solution was injected into the right side of the thoracic spinal cord of the rat with a micropipette to observe the neurons which give rise to the descending propiospinal tract. Labeled neurons were distributed throughout the bilateral Rexed's laminae Ⅰ-Ⅹ of the spinal cord in the segments rostral to the site of HRP application. The greatest number of the labeled cells were consistently found in lamina Ⅶ (including the intermediolateral nucleus), and then in laminae Ⅷ and Ⅳ. Our results verified the existence of the nucleus of the dorsolateral funiculus and its intraspinal connections in the spinal cord of the rat. This nucleus gives rise to the descending projections to the lower spinal segments.
4.SYMPATHETIC PREGANGLIONIC NEURONS IN THE RABBIT SPINAL CORD PROJECTING TO THE SUPERIOR CERVICAL GANGLION——HRP STUDY

The purpose of this research work is to find out the longitudinal distribution of the preganglionic neurons which project to the superior cervical sympathetic ganglion (SCSG). Experiments were performed on 12 adult rabbits and a monkey. Under sodium pentobarbital anesthesia, 20 microliters of 10% HRP were injected slowly into the rabbit's SCSG. In the monkey, 20 microliters of 15% HRP was injected.After a postoperative survival time of 3~6 days, the animals were perfused through,the ascending aorta with a cold fixative mixture composed of 2% paraformaldehyde and 1.25% glutaraldyhyde in 0.1mol phosphatebuffer at pH 7.4. The spinal cord segments C_1~L_3 were cut serially in transverse plane on a cryostat at 48 micra.The HRP reaction product was demonstrated according to Mesulam's (1976) benzidine blue reaction method, and counterstained with neutral red.HRP labeled neurons in the spinal cord were located exclusively on the side ipsilateral to the injected SCSG. The total number of labeled cells were 7908 in 12 rabbits, but the number of labeled cells varied from animal to animal. The highest amount was 1690 (423~#) and the lowest amount was 71 (425~#). The longitudinal distribution of the labeled cells in 12 rabbits was 12 segaments of the spinal cord (C_6~T_9), but the largest proportion (86.18%) of them was concentrated from T_1 to T_4, especially at the level of T_2 and T_3 (56.22%), and with a peak at T_2 (29.10%).In cross section of the spinal cord. HRP-labeled cells were concentrated in four cell groups, they are: nucleus intermediolateralis pars principalis (ILp), nucleus intermediolateralis pars funicularis (ILf), nucleus intercalatus (IC) and nucleus intercalatus pars paraependymalis (ICpe). The latter is subdivided into dorsal portion and ventral portion. HRP positive cells were mainly located in the ILp, In 12 rabbits about 92.99% cells were located in it. A small portion of labeled cells(6.25%) were seen in the ILf. A few labeled neurons could be detected within,the IC (0.68%) and ICpe (0.08%). Furthermore, occasionally, very few labeled cells were found at the dorsol portion of the anterior horn.In the monkey, generally speaking, the pattern of the distribution of labeled cells was the same as the rabbit.
5.THE DISTRIBUTION OF NEURONS CONSTITUTING THE SUPERIOR MESENTERIC PLEXUS OF THE CAT—A HRP STUDY

The origin of nerves innervating the small intestine and oral part of the colon of the cat was studied by means of the retrograde axonal transport of HRP. Twelve adult cats were used in this experiment. The proximal cut end of the superior mesenteric plexus was immersed in HRP solution, in order to localize the neurons which contribute fibers to this plexus. Neurohistochemical procedures were processed according to Mesulam's tetramethylbenzidine (TMB) method. HRP labeled cells or fibers can be identified bilaterally in the following places: 1. The dorsal motor nucleus of the vagus nerve; about 80% of the labeled cells were found in a region from 0.4mm below to 2.0 mm above the obex, approximately at the rostral two-thirds of the middle segment of this nucleus. 2. The reticular formation of medulla; only a few cells labeled and 92% of them were located in an area from 0.2, mm to 2.7 mm above the obex. 3. The coeliac-superior mesenteric ganglion complex; almost all of the labeled cells were concentrated in the caudal part of it, near the origin of the superior mesenterie plexus. 4. The nodose ganglion; most of the oval or round labeled cells were of medium or small size, their diameters vary from 30 to 40?m, and the average total numbers of the labeled cells were 661 in the left and 695 in the right ganglion respectively. 5. The spinal ganglia; small round or oval labeled cells (25~45 ?m in diameter) were found in the left T_2 to L_5 and right T_2 to L_4 ganglia, with the most heavily labeled segments at T_(13) and L_1 (left, mean 365; right, mean 34% of all labeled cells). The average total number of the labeled Cells were 1872 in the left and 1698 in the right side. 6. Transganglionic labeled fibers were seen in the gracile nuclei of one cat, suggesting that HRP molecules are transported retrogradely in the peripheral process and anterogradely in the centrally projecting process of the same neuron of spinal ganglia.
6. Prevalence of breast and gynecological diseases in female workers in the administrative office and workshop of an automobile part factory

Objective:
To investigate the prevalence of breast and gynecological diseases in female workers in the administrative office and workshop of an automobile part factory, and to provide a basis for promoting the health of female workers.
Methods:
In March 2015, color ultrasound examination of the breast, uterus, and bilateral adnexa was performed for all female workers, and routine gynecological examination, routine leucorrhea examination, and thinprep liquid-based cytology test were performed for married female workers. The detection rates of breast and gynecological diseases in female workers in the administrative office and workshop were analyzed.
Results:
In the 314 female workers, the overall detection rate of breast and gynecological diseases was 86.31%; the detection rate of abnormal breast ultrasound results was 72.93%, mainly breast hyperplasia; the detection rate of abnormal gynecological ultrasound results was 12.14%, mainly hysteromyoma, pelvic effusion, and uterus-rectum fossa effusion. The overall detection rate of cervicitis or vaginitis in married female workers was 66.86%. The univariate analysis showed that compared with those in the administrative office, the female workers in the workshop had significantly higher detection rates of breast hyperplasia and grade 2 breast lesions on the Breast Imaging-Reporting and Data System (BI-RADS) and significantly lower detection rates of hysteromyoma and grade 3 breast lesions on the BI-RADS (all
7. Predict response to decitabine in patients with myelodysplastic syndrome and related neoplasms

Objective:
To identify clinical and molecular signatures for predicting response to decitabine (DAC) in patients with myelodysplastic syndrome (MDS) and related neoplasms.
Methods:
The clinical characteristics of 109 patients with MDS and related neoplasms who were treated with DAC were analyzed retrospectively and the next target sequencing was performed to define recurrently mutated genes in these disease samples, to examine the association of the clinical and molecular signatures with response to DAC treatment.
Results:
Of 109 MDS and related neoplasms patients, there were 70 males and 39 females, the median age was 61 years old (ranges: 17-85 years old) . According to the international prognostic scoring system (IPSS) , 46 cases were included in the relatively low risk group (low risk and intermediate-1 risk) , 63 in the relative high risk group (intermediate-2 and high risk) . There were 21 cases with complex karyotype, 17 chromosome 7 abnormality and 17 monosomal karyotype. The median courses of DAC treatment was 4 (2-11) . A total of 74 patients achieved response (67.9%) and 30 (27.5%) achieved complete response (CR) . Univariate analysis found that CR was higher in patients with high risk of IPSS, complex karyotypes, monosomal karyotypes, chromosome 7 abnormality, and platelet doubling after one cycle of DAC treatment. Patients with TP53 gene mutation were more likely to receive CR, 10 of 15 patients with TP53 mutations achieved CR. 8.Application of integrated theory of health behavior change in pelvic floor muscle exercise of patients with pelvic floor dysfunction after early cervical cancer surgery

Objective:To explore the effect of nursing intervention based on the integration theory of health behavior change in pelvic floor muscle exercise of patients with pelvic floor dysfunction (PFD) after early cervical cancer surgery.Methods:A total of 134 patients with pelvic floor dysfunction after early cervical cancer surgery were selected and divided into test group ( n=67) and control group ( n=67) by random digits table method. The test group received three months of pelvic floor muscle exercise based on the theory of healthy behavior change. The control group received conventional pelvic floor rehabilitation care. The Pelvic Floor Distress Inventory-Short Form 20(PFDI-20), Pelvic Floor Impact Questionnaire-7(PFIQ-7), General Self-Efficacy Scale (GSES) and Social Support Rating Scale (SSRS) were evaluated before the intervention (T0), After one month of intervention (T1) and at the end of the intervention (T2). Results:The results of repeated measurement analysis of variance showed significant group/time interaction between PFDI-20 and PFIQ-7 scores ( Finteraction=29.196,13.213, both P<0.05); significant group/time interaction between GSES and SSRS scores ( Finteraction value was 25.664,70.240, both P<0.05). At the end of the intervention, the scores of PFDI-20 and PFIQ-7 in the test group were 66.54±14.04 and 73.20±7.41, which were lower than 77.47±15.91 and 78.39±8.51 in the control group over the same period. The difference was statistically significant ( t value was -4.216, -3.765, both P<0.01); the GSES and SSRS scores of the test group were 2.86±0.30 and 50.37±2.45, which were higher than the control group (2.51±0.34 and 46.42±2.82), the differences between the groups were statistically significant ( t value was 6.447, 8.662, both P<0.01). Conclusion:The nursing intervention plan based on the integration theory of health behavior change can significantly improve the self-efficacy, social support level and quality of life of patients with PFD after early cervical cancer surgery, and enhance the short-term and long-term pelvic floor rehabilitation effect of patients.
9.Dosimetric effects of prone immobilization devices combined with a belly-board in intensity-modulated radiotherapy for gynecologic cancers

Objective:To investigate the dosimetric effects of prone immobilization devices combined with a belly board (PIDBBs) in the intensity-modulated radiotherapy (IMRT) for gynecologic cancers.Methods:A total of 20 patients with cervical or endometrial cancer treated with radiotherapy in the Third Affiliated Hospital of Sun Yat-sen University from August 2020 to June 2021 were retrospectively analyzed. Two sets of body contours were outlined for each patient. One set of body contours did not contain the immobilization devices, and the other contour set included the immobilization devices. For each patient, doses were calculated for the two sets of contours using the same 7-field IMRT plan and were recorded as Plan without and Plan with. The dosimetric difference caused by the immobilization devices was assessed by comparing the parameter values in the dose-volume histograms (DVHs) and by plan subtraction. The Gafchromic EBT3 film and anthropomorphic phantom were used to verify the calculated doses. Results:The target coverage and average dose of Plan with were lower than those of Plan without. Specifically, the V50 Gy, V49 Gy, and Dmean of planning target volume (PTV) decreased by 19.75%, 7.99%, and 2.54% ( t = 8.96, 10.49, 22.09, P < 0.01), respectively. The V40 Gy, V30 Gy, V20 Gy, V15 Gy, and Dmean of skins increased by 51.79%, 51.05%, 45.72%, 33.63% and 10.80% ( t = -2.54, -5.63, -15.57, -24.06, -13.88, P < 0.01), respectively. Doses to other organs at risk (OARs) showed no significant differences. As indicated by the EBT3 measurements, the doses to skins of the abdomen and pelvis on the anthropomorphic phantom increased by approximately 37.24% ( t = 10.86, P<0.01). Conclusions:Although PIDBBs can effectively reduce the low dose to the small intestine, the radiation attenuation caused by them can reduce the PTV coverage of radiotherapy plans and increase the doses to abdominal and pelvic skins sharply, especially for patients requiring irradiation of the groin and perineum.
10.MRI for assessment of iron overload in patients with hematologic disease.

OBJECTIVECompare the characteristics of magnetic resonance imaging(MRI)liver T2*, cardiac T2* and serum ferritin on the assessment of iron overload.
METHODSA total of sixty-nine patients from November 2011 to June 2014 were enrolled in this study. Their cardiac and liver iron concentration levels were measured through MRI examination, with other clinical data were collected to perform statistical analysis.
RESULTSThe correlation between liver T2* and adjusted serum ferritin(ASF) was statistically significant(P=0.003). However, no significant correlation was found between cardiac T2* and liver T2*, ASF, respectively. According to the statistical analysis of the 69 cases, it is found that the number of iron overload cases diagnosed by liver T2* was 62 and 20 cases were severe iron overload (32.26%); the number of iron overload cases diagnosed by ASF was 47 and 14 cases were severe iron overload(29.79%), while the number of iron overload cases diagnosed by cardiac T2* was only 25 and no severe iron overload cases.
CONCLUSIONSince SF was affected by other factors, it cannot reflect the level of iron overload in human body objectively. Now, liver T2* has become the gold standard for assessment of iron overload because of its good reliability and repeatability. However, cardiac T2* cannot correctly be used as assessment for iron overload, and it is only a method of evaluating the level of cardiac iron deposition.
