1.A comparison of knowledge awareness rates and influencing factors of clo-norchiasis among residents in 4 counties(cities)of Hunan Province

Objective To understand the status of knowledge awareness rates and influencing factors of clonorchiasis among the residents in 4 counties(cities)of Hunan Province,so as to provide the references for formulating the prevention and control strategy of clonorchiasis in the province. Methods A total of 1224 subjects were selected by the multi-stage cluster sampling method and investigated with questionnaires and face-to-face interviews. Results The clonorchiasis knowledge awareness rates of the residents were 15.67% in Yongzhou City,9.94% in Yueyang City,13.25% in Wangcheng County,and 21.96% in Tongd-ao County,respectively. In the whole province,the clonorchiasis knowledge awareness rates of the residents were 44.67%, 56.43%,60.95%,and 42.46% in the etiology,clinical manifestations,epidemiological characteristics and daily prevention,re-spectively. The logistic regression showed that the influencing factors of clonorchiasis knowledge awareness rates were the daily dining place,whether or not having the medical personnel in the family,the degree of attention to the relevant news reports, and the history of parasitic diseases. Conclusions The level of clonorchiasis than that in other areas,but the overall level of clonorchiasis knowledge awareness of the residents in Hunan Province is not high. Therefore,the relevant authority should strengthen the health education in clonorchiasis prevention knowledge including personal daily food hygienic knowledge. knowl-edge awareness rate of the residents in the west-southern areas of Hunan Province is higher
2.Effect of metacentric particles with bisoprolol fumarate pesos on the elderly patients with heart failure with ventricular arrhythmia treatment

Objective To explore clinical effect of metacentric particles with bisoprolol fumarate pesos on the elderly patients with heart failure with ventricular arrhythmia treatment.Methods In August 2013 to April 2016, 102 cases of elderly patients in our hospital heart failure with ventricular arrhythmia were selected as research object, according to touch the ball method, they were divided into control group and experimental group, each group of 51 cases, control group was given bisoprolol fumarate pesos, on this basis, the experimental group was combined use of metacentric particles.Two groups of clinical curative effect were compared .Results The experimental group and control group rate of sinus heart rate to maintain statistical comparison of the experimental group was higher than control group ,the difference was statistically significant(P<0.05),group total effective rate was significantly higher than the control group ( P <0.05 ) , the incidence of adverse reactions were significantly lower than the control group (P<0.05),the control group and experimental group heart rate compared before treatment, there was no statistically significant difference, but after treatment, the experimental group was lower than the control group, the differe nce was statistically significant (P <0.05). Conclusion Metacentric particles with bisoprolol fumarate pesos for elderly patients with heart failure with ventricular arrhythmia in clinical curative effect is distinct, high safety.
3.Detection of cell apoptosis induced by 125I brachytherapy in nude mice bearing lung adenocarcinoma using 99Tcm-Annexin V imaging combined with diffusion weighted MRI

Objective To detect apoptosis of transplanted lung adenocarcinoma cells in nude mice after 125I brachytherapy by 99Tcm-Annexin V combined with diffusion weighted MRI (MR-DWI).Methods Twenty-five BALB/c-nu nude mice models subcutaneously transplanted with A549 cells were divided into experimental group (EG,n=13) and control group (CG,n=12) by random number table method.One 125I seed with apparent activity of (24.8±6.3) MBq was implanted into each mouse in EG,while CG underwent cold seed implantation.Both of 99Tcm-Annexin V imaging and MR-DWI were performed within 7-10 d after brachytherapy,then all mice were sacrificed and tumor cell apoptosis was detected by terminal oxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL) immunofluorescence,Survivin expression was assayed by SP.Two-sample t test,x2 test and Pearson correlation analysis were used for data analysis.Results Positive rate of cell apoptosis by 99Tcm-Annexin V imaging was 69.2%(9/13) and 8.3%(1/12) respectively in EG and CG (x2 =12.73,P<0.01).The uptake ratio of 99Tcm-Annexin V,apparent diffusion coefficient(ADC) value and apoptosis index(M) of the tumor in EG were 2.91±0.85,(2.03±0.44)×10-3 mm2/s and (49± 18) %,respectively,which were significantly higher than those of CG (1.26 ± 0.37,(1.29 ± 0.21) ×10-3 mm2/s and (11±4)% respectively,t=5.930,5.452,7.606,all P<0.05).Survivin expression in EG and CG was (46± 13) % and (15±7) % respectively (t =5.158,P<0.05).The value of ADC was correlated with AI and uptake ratio(r=0.756,0.788,both P<0.05).Uptake ratio was correlated with AI (r=0.754,P<0.05),while Survivin expression was negatively correlated with AI (r =-0.772,P<0.05).Conclusions Down-regulation of Survivin expression may play an important role in apoptosis induced by 125I brachytherapy.99Tcm-Annexin V combined with MR-DWI could effectively evaluate apoptosis of lung adenocarcinoma cells in a non-invasive way,thus it might be helpful in evaluation of early efficacy of 125I brachytherapy.
4.Effects of acetamidophenol on auditory in mice.

OBJECTIVE:
The purpose of this investigation was to observe the effects of the Acetaminophen on auditory in mice.
METHOD:
Regardless of male and female mice, healthy seven-year-old C57 mice were randomly divided into Acetaminophen low-dose group (150 mg/kg), Acetaminophen medium-dose group (300 mg/kg), Acetaminophen high-dose group(600 mg/kg) and control group. Then mice were tested for ABR to observe the changes of ABR's threshold in the zero, second, fourth and ninth day separately. The change of cochlea hair cells morphology was studied by immunofluorescent labeling. And HPLC detects the concentration of Acetaminophen in endolymph of mice cochlea.
RESULT:
After 30 minutes following administration, the Acetaminophen in endolymph of mice cochlear can be assayed by high performance liquid chromatography (HPLC). Acetaminophen increased the hearing thresholds compared to the control group. Hearing thresholds increased significantly in the Acetaminophen at 9 d,compared to the control group (P < 0.05). After administered medium-dose and high-dose of Acetaminophen, on the 9th day with the time of giving medicine to all the groups strengthens, the ABR thresholds heightens. And the average threshold is (44.75 +/- 16.00) dB, (50.00 +/- 11.00) dB respectively. Hair cells damages could be observed in experimental group after operations.
CONCLUSION
Acetaminophen can pass through blood-labyrinth barrier to the inner ear. These data demonstrate that taking certain acetaminophen can induce hearing impairment in mice.

5.Soluble P-selectin and L-selectin levels in serum of colorectal cancer patients and its clinical significance

Background and purpose:Colorectal cancer is a kind of common digestive malignancies, which seriously threaten the human health. Although modern diagnostic and treatment technology has developed rapidly, the incidence and mortality of colorectal cancer continue to show an increasing tendency in recent years, and early diagnosis and prevention of colorectal cancer liver metastases are important to increase the survival rate of patients and to improve the prognosis. P-selectin and L-selectin is attracting much attention in cancer research ifeld recently, and the change of their expression and mediated adhesion play an important role in tumor cell metastasis, but their relationship between the occurrence and clinical stage of colorectal cancer rarely reported. This study aimed to explore whether the serum P-selectin and L-selectin levels of colorectal cancer patients were correlated with clinical and pathological features and the situation before and after surgery.Methods:A total number of 132 cases of colorectal cancer patients and 100 healthy subjects with gender and age-matched were enrolled. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum P-selectin and L-selectin concentrations in 132 cases before and after surgery, 100 healthy subjects were enrolled as the control group, the P-selectin and L-selectin levels were analyzed combined with theclinical and pathological features.Results:Serum P-selectin and L-selectin levels in patients with colorectal cancer before surgery were signiifcantly higher than those in the healthy control group (75.2±13.3vs 26.6±9.6, 89.2±12.7vs 33.9±8.3,P<0.05) ; Compared with the patients before surgery, 1 week after surgery, the P-selectin and L-selectin levels were not significantly decreased (63.8±11.1vs 75.2±13.3, 71.5±10.9vs 89.2±12.7,P>0.05); however, significantly decreased after 3 months (34.8±10.4vs 75.2±13.3, 40.1±9.5vs 89.2±12.7,P<0.01); This paper studied patients with colorectal cancer, the higher clinical stage, higher P-selectin and L-selectin levels, and their expression levels with tumor invasion depth, Dukes stage, histological grade and lymph node metastasis were related, but were not correlated with age, gender and tumor site. Meanwhile, P-selectin and L-selectin levels were not signiifcantly correlated in the same serum of patients with colorectal cancer (r=4.114,P>0.05).Conclusion:Serum P-selectin and L-selectin levels to some extent relfect the invasion of colorectal cancer, the degree of inifltration and lymph node metastasis, and which can be an important indicator in the development and prognosis of colorectal cancer.
6.Optimizing the Ultrasonic Extraction of Gardenia Oils from Gardenia Jasminoides Ellis by Response Surface Methodology

This study aimed to optimize the ultrasonic extraction of gardenia oils from Gardenia jasminoides Ellis.by response surface analysis methodology (RSM).The duration of extraction,ratio of liquid to materials and extracting times were selected as impact factors based on single-factor experiment.RSM was adopted to learn the effects of the three factors on the extraction rate of Gardenia oil from Gardeniajasminoides Ellis.As a result,it was found that the duration of extraction was 28.7 min,while the ratio of liquid to materials was 11.62:1,and the number of extracting times were 3.36.Under this extracting condition,the average extraction rate of the oil was 13.5% according to three validation experiments,fitting the predicted value well.In conclusion,it was demonstrated that the ultrasonic extraction method effectively improved the extraction rate of gardenia oil,providing a certain basis for the development and utilization of gardenia oil.
7.Development of measurement method using TLD for workers occupation personally exposed to 125Ⅰ seed source in the implant

Objective To explore the method for measuring and calculating both absorbed dose and effective dose received in organ and tissues of occupational workers by using TLDs for the implantation of 125Ⅰ seed sources.Methods The experiments with 60Co γ-rays were carried out for the stability.A group of TLD chips was exposed to 125Ⅰ seed sources to establish standard dose curve for air kerma.During the 125Ⅰ seed implantation, the TLD chips were pasted to 13 locations like thyroid inside and outside the lead aprons worn by occupational workers to measure average absorbed dose and calculate the absorbed doses and effectives to organs and tissues.Results For 3 cases of prostate cancers with implantation of 125Ⅰ seeds, the worker's organs and tissues received the absorbed dose 0.02 -3.80 μ Gy and effective dose 0.06- 1.81 μSv outside lead aprons and the highest absorbed dose 2.35 μ Gy and effective 0.02 μSv inside lead aprons, respectively, with more than 65.9% of rays shielded.For 3 cases of brain cancers with implantation of 125Ⅰ seeds, the workers received the absorbed dose 0.23 - 11.31 μGy and effective dose 0.88 - 4.07 μSv outside lead aprons and the highest absorbed dose 2.22 μ Gy and effective dose 0.09 μSv inside lead aprons, respectively, with more than 54.5% of rays shielded.For 3 cases of lung cancers with implantation of 125Ⅰ seeds, the workers received the absorbed dose 0.03 - 14.78 μGy and effective dose 0.35 -7.59 μSv outside lead aprons and the highest absorbed dose 4.09 μGy and effective 0.22 μSv inside lead aprons, respectively, with more than 58.4% of rays shielded.For 2 cases of mediastinum cancers with implantation of 125Ⅰseeds, the workers received the absorbed dose 0.06 - 74.91 μGy and effective dose 0.83 - 17.96 μSv outside lead aprons and the highest absorbed dose 10.29 μGy and effective 0.5 μSv inside lead aprons, respectively, with more than 85% of rays shielded.For one case of ovary cancer with implantation of 125Ⅰ seeds, the worker received the absorbed dose 0.09 - 14.29 μGy and effective dose 2.40 - 4.50 μSv outside lead aprons and the highest absorbed dose 7.77 μGy and effective 0.12 μSv inside lead aprons, respectively, with more than 34% of rays shielded.For one case of eye cancer with implantation of 125Ⅰ seeds, the workers received the absorbed dose 2.2 -39.84 μGy and effective dose 4.48 - 10.06 μSv outside aprons and the highest absorbed dose 5.19 μGy and effective 0.16 μSv inside aprons, respectively, with more than 54.6 % of rays shielded.Conclusions The method of using TLDs to measure the doses to the occupational workers in the course of the implantation of 125Ⅰ seed sources is simple and easy to operate.It would be an effective approach to protecting medical workers in the case of brachytherapy.
8.Effects of RNA interference targeting CDC25a gene on proliferation of human liver cancer HepG2 cells

[ ABSTRACT] AIM:To investigate the effect of silencing cell division cycle 25a ( CDC25a) gene on the prolifera-tion of human hepatoma HepG2 cells.METHODS:CDC25a gene in human hepatoma HepG2 cells was silenced by RNA interference.Real-time PCR was applied to detect the expression of CDC25a, cyclin E and CDK2 at mRNA levels in the HepG2 cells.Western blotting was applied to detect the expression of CDC25a at protein level.In addition, MTT assay, Giemsa staining and flow cytometry were used to measure the proliferation of human hepatoma HepG2 cells.RESULTS:The expression of CDC25a at mRNA and protein levels in RNA silence group was lower than those in negative control group and normal control group (P<0.05).The mRNA expression of cyclin E and CDK2 in silence group was lower than that in negative control group and normal control group (P<0.05).The cell proliferation in silence group was lower than that in negative control group and normal control group ( P<0.05) .The results of flow cytometry revealed that the cells in silence group were blocked in G1 phase.CONCLUSION:Infection of LV-CDC25a-RNAi recombinant to the HepG2 cells effec-tively inhibits the CDC25a gene expression and the proliferation of human hepatoma cells, and arrests the cells in G1 phase, suggesting that CDC25a gene may be a key target for the treatment of liver cancer.
9.Effect of TanshinoneⅡA on Experimental Endometriosis in Rats

Objective To observe the effect of TanshinoneⅡA( TanⅡA) on endometriosis( EMS) rat model and study the possible mechanism. Methods EMS rat model was established by autologous endometrium transplantation. EMS rats were randomly divided into five groups: high-, medium-, low-dose TanⅡA group, mifepristone group and model control group( n=10), the other ten normal rats as normal control group.The rats in the model control group and normal control group were given 0.9% sodium chloride solution. The rats in the mifepristone group were given 25 mg . kg-1 . d-1 mifepristone. The rats in the high-, medium-, low-dose TanⅡA groups were given 30, 20 and 10 mg.kg-1.d-1 TanⅡA, respectively.Four weeks after the treatment, the volume of endometriotic implants in each rat was measured. Pathological changes of ectopic endometrium were observed under light microscope.Expression levels of Bcl-2, Bax and Caspase-9 in ectopic endomembrane of rats were determined by Western blotting. Results High and medium dose TanⅡA and mifepristone significantly reduced the volume of ectopic lesions(P<0.05).As compared with model control group, the inhibition rate of the ectopic lesions in low-, medium-, high-dose TanⅡA groups and mifepristone group was 17.81%, 57.78%, 58.88% and 64.12%, respectively.High and medium dose TanⅡA and mifepristone promoted atrophy of ectopic endometrium, inhibited the expression of Bcl-2 protein and promoted the expression of Bax and Caspase-9 in ectopic tissues( P<0.05) . Conclusion TanⅡA significantly decreases the size of ectopic lesions in rats, possibly through reducing the expression of Bcl-2 and increasing the expression of Bax and Caspase-9 to regulate cell apoptosis.
10.A Clinical Study on Kanglaite injection combined with comprehensive therapy in the treatment of patients with advanced non-small cell lung cancer

Objective To study the effect of Kanglaite combined with comprehensive therapy on advanced non-small cell lung cancer.Methods Sixty-one patients with advanced non-small cell lung cancer were randomly divided into treatment group ( n=31 ) and control group ( n=30 ).Both groups were given comprehensive therapy.Treatment group were additionally treated with intravenous injection of 200 ml Kanglaite.Clinical efficacy,quality of life,pain relief and adverse reactions of the two groups were observed.Results ( 1 ) Quality of life was improved in 20 cases (64.5% ),stabled in 8 cases (25.8%),declined in 3 cases ( 9.7% ) of treatment group,and in the control group there were 9 cases ( 30.0% ) improved,9 cases ( 30.0% ) stabilized,12 cases (40.0% ) declined respectively.Quality of life in treatment group was higher than in control group ( U=2.91,P<0.01 ).( 2 ) Pain relief:the number of patients with complete remission,partial remission,no change,and progression were 5 cases ( 16.1% ),16 cases ( 51.6 % ),16 cases (51.6% ),6 cases (19.4%) and 4 cases (12.9% ) in treatment group,and in control group they were 2 cases(6.7% ),9 cases (30.0% ),11 cases(36.7% ) and 8 cases(26.7% ) respectively.The effect of treatment on pain relief in treatment group was better than that in control group ( U=2.32,P<0.05 ).(3) Clinical efficacy:in the treatment group there were 12 cases (38.7%) with partial remission,14 cases (45.2%) stabilized,and 5 cases (16.1% ) progressed,and in control group the numbers were 8 cases (26.7% ),8 cases (26.7% ) and 14 cases (46.7% ) respectively.The clinical efficacy in treatment group was better than that in the control group( U=2.04,P<0.05).(4) There were significant difference on the change of white blood cell count and gastrointestinal reactions Ⅲ and Ⅳ degrees between treatment group and contrl group [22.6% (7/31) vs.53.3%(16/30),x2=6.139 P<0.05;19.4% (6/31) vs.46.7% (14/30),x2=5.161,P<0.05].Conclusion Kanglaite injection combined with comprehensive therapy can improve clinical efficacy of therapy for advanced non-small cell lung cancer,reduce the toxic adverse reaction,protect immunity system and improve the quality of life of patients.
