1.Expression of hypoxia-inducible factor 1 alpha, vascular endothelial growth factor and angiopoie-tin-2 in tissues of condyloma acuminatum in pregnant women

Objective To investigate the expression and significance of hypoxia-inducible factor 1 alpha (HIF- 1α), vascular endothelial growth factor (VEGF) and angiopoietin-2 (Ang-2) in tissues of condy- loma acuminatum (CA) in pregnant women. Methods Tissue specimens were obtained from the lesions of 30 pregnant women with CA and 30 non-pregnant women with CA, and from the vulva of 15 normal preg- nant women. By immunohistochemical staining, the expressions of HIF -1α, VEGF and Ang-2 were detected in these specimens. Results The expression rates of HIF -1α,VEGF and Ang-2 were 86.67%, 93.33% and 83.33% in pregnant women with CA, respectively, 63.33%, 66.67% and 53.33% in non-pregnant women with CA, respectively, and 0, 6.67% and 0 in normal women, respectively. Enhanced expressions of HIF -1α,VEGF and Ang-2 were observed in pregnant women with CA compared with the latter two groups (P < 0.05). In pregnant women with CA, a significant correlation was noted between the expression of HIF -1α and VEGF (r = 0.412, P < 0.01) and between the expression of Ang-2 and VEGF (r = 0.460, P < 0.01). Further more, the expression of HIF -1α, VEGF and Ang-2 positively correlated with each other in non-preg- nant women with CA. Conclusion In tissues of CA in pregnant women, HIF-1α,Ang-2 and VEGF are over expressed, which may be related to angiogenesis.
2.Effects of propfol and midazolam for sedation effect of central venous puncture

Objective To compare the effect of propofol and Midazolam for the vein puncture and indicators of respiratory cycle in the e-mergency center. Methods According to different anaesthetic,100 patients who needed for central venepuncture in patients,were randomly divided into Propofol group (50 cases) and Midazolam group (50 cases),given different dose input,and evaluated by RSS(Ramsay Sedation Scale,RSS) . In terms of the onset time and recovery time of consciousness,the central venous puncture were performed with all the patients. Results There was no significant difference in the process of sedation effect on respiration and circulation of the patients between 2 groups. And the differences between 2 groups with the time of reaching RSS-5 were no statistical significance,while the differences of consciousness recovery time between 2 groups were statistical significance. Conclusion The appropriate dosage and velocity of Propofol and Midazolam can quickly achieve central vein puncture,with slight influence on respiratory and circulatory system and no serious adverse reaction. But the onset time and consciousness recovery time of the Propofol which has a good controllability was shorter than those of Midazolam.
3.Determination of Deltamethrin Residue in Water by Rephase High Performance Liquid Chromatogrophy

Objective An analytic method with rephase high pressure liquid chromatogrophy(RP-HPLC) was developed to determine the residue of deltamethrin in water.Methods Water sample was filtrated and determined by RP-HPLC.Chromatographic column was YMC C_(18)(250 mm?4.6 mm,5?m),mobile phase was methanol:water=90:10(V/V),the flowing rate was 0.8 ml/min, DAD detecting wavelength was 205 nm.Results The regression equation was y=655.6 x-1.8,r=0.999 99.The lowest detectable concentration was 2.9?g/L.The recovery rate was 98.0%-102.0%and the relative standard deviation was 0.29%-0.48%. Conclusion The method is proved to be satisfactory in precision,accuracy and sensitivity and can be used for the rapid determination of deltamethrin residue in water.
4.Protective Effects and Mechanisms of Anisodamine on Experimental Liver Injury

Objective To investigate the protective effects and the mechanisms of anisodamine on experimental liver damage. Methods The experimental liver injury model was established by ANIT and CCl 4.Studies were made after ANIT or CCl 4 administration,and the control group compared with the experimental groups which were treated by anisodamine on the pathologic morphology, biochemical indices and the contents of Ca 2+ ,MDA and the ability of total antioxidation in the liver.Effects of anisodamine on sleeping time of the mice toxicated by given sodium phenobarbital were also determined.Results The elevation of ALT, ALP, BiL, CHE and the decrease of serum protein in ANIT or CCl 4 liver damage were significantly improved by treatment with anisodamine .It also remarkably diminished the hepato-cellular and chole-epthelial-cellular degeneration and necrosis induced by ANIT or CCl 4.The contents of Ca 2+ ,MDA and the ability of total antioxidation in the liver were all decreased by treatment with anisodamine. The sleeping time induced by sodium phenobarbital in the toxicated mice was reduced by anisodamine. Conclusion Anisodamine have significant protective effects on liver injury of intrahepatic cholestasis and chemical hepatitis and the mechanism may be associated with blocking M-receptor and enhancing antioxidation and antitoxic activity in liver.
5.Simultaneous Determination of Pinoresinol Diglucoside and Chlorogenic Acid in Traditional Decoction and Its Solid Decoction of Sam Soup by HPLC

Objection To estabish a HPLC methold for determining the contents of pinoresinol diglucoside and chlorogenic acid in Sam Soup and its solid decoction. Methods Chromatographic column was Ultimate XB-C18 reversed-phase chromatography column (250 mm×4.6 mm, 5 μm). Mobile phase was acetonitrile-0.4% glacial acetic acid with gradient elutim. The flow rate was 1.0 mL/min, detection wavelength was 277 nm, and column temperature was 40 ℃. Results The pinoresinol diglucoside and showed good linear relationship in the range of 5.0-50.0 μg, r =0.999 9 (n =6), and chlorogenic acid showed linear relationship in the range of 5.4-54.0 μg, r=0.999 6 (n=6). The average recoveries of pinoresinol diglucoside and chlorogenic acid were 98.75% (RSD=1.39%) and 101.11% (RSD=2.69%), respectively. Conclusion The established method was accurate and reliable. The contents of pinoresinol diglucoside and chlorogenic acid in solid decoction are higher than traditional decoction of Sam Soup.
6.Inhibition of peroxiredoxin Ⅱ on human intervertebral disc cells cultured in vitro

BACKGROUND:intervertebral disc degeneration can reduce nucleus pulposus cells,and peroxiredoxin II involved in the regulation of resist oxidation damage,cell division,differentiation,signal transduction and apoptosis.Peroxiredoxin Ⅱ has promotive effect on intervertebral disc degeneration,whereas the mechanism remains poorly understood.OBJECTIVE:To observe the effects of perexiredoxin Ⅱ on human intevertebral disc cells activity and type Ⅱ collagen synthesis in vitro.METHODS:Human degenerated human lumbar disc cells were cultured in vitro,and assigned into the control and peroxidase Ⅱ groups.Peroxidase Ⅱ with doses of 10,100 and 1 000 ng/L were added into the peroxidase Ⅱ groups.The cells were identified by immunohistochemical staining,and the cell proliferation was detected using cck-8 kit.Cell supematant was collected at days 3 and 7 after operation,and the expression of type Ⅱ collagen was measured by double-antibody sandwich enzyme-linked immunosorbent assay.RESULTS AND CONCLUSION:In vitro cultured human degenerative lumbar intervertebral disc nucleus pulposus cells by adding peroxidase increased with the dose-Ⅱ,the disc nucleus pulposus cells of the volume and type Ⅱ collagen synthesis gradually reduced(P < 0.01).Tips peroxidase Ⅱ on the intervertebral disc nucleus pulposus cells,the number and type Ⅱ collagen synthesis significantly inhibited in a dose-dependent manner.Thus speculated that peroxidase Ⅱ on the nucleus pulposus cells in vitro may lead to disc degeneration as a precipitating factor.
7.Comparison of value of MR and CT and different staging system in the diagnosis of nasopharyngeal carcinoma

Objective To evaluate the value of MR and CT examinations in the diagnosis of nasopharyngeal carcinoma (NPC) and compare 2008 staging system with 1992 staging system and 2002 UICC staging system for NPC. Methods MR and CT images of seventy-six cases with NPC were studied. According to 2008 staging system and taking MR as a standard, differences between these two examinations were evaluated under the new NPC staging system, and three staging system were compared by MR findings. Results MR was inconsistent with CT in eveluating invasion of medial pterygoid muscle(22,24 cases), lateral pterygoid muscle( 15, 11 cases), skull base(35, 32 cases) and intracranial fossa( 11,6 cases), but no statistical diffence existed ( P > 0. 05 ). There were statistical difference ( P < 0. 05 )between MR and CT in determining invasion of parapharyngeal space( 50, 61 cases), retropharyngeal lymph node metastasis(48, 23 cases), stage T1 (18, 11 cases), T2 (15, 22 cases), N0 (18, 24 cases) and N1(33, 27 cases) with differences of 11 cases, 25 cases, 7 cases, 7 cases, 6 cases and 6cases respectively.For invasion of parapharyngeal space, CT showed 11 cases more than MR while 5 cases were comfirmed as compression by local tumor and 6 cases were proved as retropharyngeal lymph node metastasis according to MR. For retropharyngeal lymph node metastasis, MR presented 25 cases more than CT. These two reasons above mainly caused the differences of T-staging and N-staging. For 2008 staging system, when compared with 1992 staging system, there were 9 cases upstaging and 1 case downstaging in T classification, 16 cases upstaging in N classification, and 15 cases upstaging and 1 case downstaging in clinical classification; and when compared with 2002 UICC staging system, there were 7 cases, 10 cases and 12 cases upstaging in T,N, and clinical staging respectively. Conclusions Compared with MR examination which was regarded as standard by 2008 staging system of NPC, there were some differences in demonstrating invasion of parapharyngeal space and retropharyngeal lymph node metastasis by CT. Compared to 1992 staging system and 2002 UICC staging system, 2008 staging system mainly made T and N classification of tumor upstage,resulting in upstaging in clinical classification.
8.Construction of prokaryotic expression vector of mouse CD25 extracellular domain

Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
9.Effects of anisodamine on the pressure of rats portal vein of liver fibrosis

Aim To investigate the effects of anisodamine on the pressure of portal vein of experimental liver fibrosis and its mechanisms of action. Methods The experimental liver fibrosis model was produced by CCl_4. The preventive group was treated ten weeks with anisodamine 7.0 mg?kg~(-1) ip once everyday. All therapeutic groups were treated six weeks with anisodamine 7.0 mg?kg~(-1) or 14.0mg?kg~(-1) ip once everyday. After CCl_4 injection for ten weeks, the pressure of portal vein,the content of NO in livers, and liver iNOS and eNOS mRNA expressions were detected with different methods.Results The pressure of portal vein was significantly reduced in anisodamine preventive group and anisodamine therapeutic groups. The liver content of NO and the expression of iNOS and eNOS were all inhibited by the treatment of anisodamine.Conclusion Anisodamine reduced the expressions of iNOS and eNOS to synthesis of NO in liver. As a result, the pressure of portal vein of fibrosis rats decreased. So Portal hypertension of liver fibrosis may be improved by anisodamine in patients.
