1.Clinical analysis of 122 elderly cases of acute pancreatitis

Objective To investigate the clinical features and prognosis of elderly patients with acute panereatitis.Methods One hundred and twelve elderly patients with acute pancreatitis were included in our study.The clinical features and prognosis were compared with those of the 120 non -aged patients with acute pancreatitis during the same period.Results Cholecystolithiasis was the most common etiology of acute pancreatitis in both groups[81 cases(66.3%) in aged group,70 cases (58.3%) in non-aged group].Abdominal pain was the most common symptom in both groups, followed by fever.Two elderly cases were diagnosed only by autopsy because of mild abdominal pain and rapidly attack of shock.There were 31 cases with severe pancreatitis in aged group and 14 in non -aged group (P
2.The levels of serum bone alkaline phosphate in healthy children in Zhejiang Provincial Hospital of TCM

Objective To study the levels of bone alkaline phosphate (BALP) in healthy children in the specific hospital.Methods Totally 990 healthy children from 0-16 years old were selected,all of whom received physical examination in the Xiasha branch of Zhejiang Provincial Hospital of TCM during August 2013 to April 2014.Serum was collected and tested for BALP levels with a Beckman Coulter Immunoassay System UniCel DxI 800.Z-test was used to verify age and gender grouping,after which the 2.5th and 97.5th percentile values of serum BALP levels were calculated.Results The levels of serum BALP(bilateral 95% range) in healthy children were as follows:trottie stage (0-1 years old):37-300μg/L;infancy to school stage (> 1-9 years old):40-196 μg/L;puberty stage (>9-16 year-old):25-284 μg/L (male),30-199 μg/L (female).The serum BALP levels of male children showed a weak correlation with height,while the correlation for female children was not significant.Conclusions The levels of BALP in healthy children of the specific hospital and area were established.Serum BALP levels in children varied with age and sex.
3.Motilin expression in human thyroid and its clinical significance

Objective To observe the expression of motilin precursor mRNA and location of motilin in human thyroid and to explore the clinical significance.Methods RT-PCR,Sourthern blot and immuno- fluorescence histochemical techniques were applied to detect the expressions of motilin precursor mRNA and motilin in human thyroid and to compare the difference of motilin peptide and motilin mRNA expression among tissues of normal thyroid,nodular goiter and medullary thyroid carcinoama.Results(1)The results of RT-PCR and Southern blot showed that motilin precursor mRNA was expressed in human thyroid.(2)A large number of cells showing collocation of motilin with calcitonin were observed in human thyroid,suggesting that motilin was expressed in C cells.(3)Both the motilin immunoreactive(MTL-IR)and the motilin mRNA were significantly increased in medullary thyroid carcinoma(P0.05).(4)Western blot results showed that the expression of motilin in thyroid medullary carcinoma was significantly higher than that in normal thyroid. However no difference was observed between thyroid nodular goiter and normal thyroid tissue.Conclusion MTL- IR positive cells and motilin mRNA expression are found in human thyroid.Motilin is located in C cells of thyroid. The expressions of motilin and motilin precursor mRNA are significantly increased in medullary thyroid cancer,no difference between thyroid nodular goiter and normal thyroid tissue.
4.Clinical characteristics and prognosis of late-onset group B streptococcal sepsis in NICU

Objectives To investigate the clinical characteristics and prognosis of late-onset group B streptococcal (GBS) sepsis. Methods From Jan. 2007 to Dec. 2011, iffteen neonates diagnosed with late onset GBS sepsis at discharge from NICU were retrospectively analyzed, meanwhile, thirty-four neonates diagnosed with late onset non-GBS Gram-positive bacteria sepsis at discharge were selected as controls during the same period. Results There were signiifcant differences in occurrence rates of shortness of breath, convulsion and apnea between late onset non-GBS sepsis group and late onset GBS sepsis group (P<0.05). The percentages of neonates with white blood cell count (CSF)>100×106/L, high-sensitivity C-reaction protein (hsCRP)>100 mg/L and glucose in CSF<3.11 mmol/L in late onset GBS sepsis group were higher than those in late onset non-GBS sepsis group (P<0.05). GBS was sensitive to penicillin, ampicillin, ceftriaxone, piperacillin/tazobactam, levolfoxacin and vancomycin. The rates of GBS resistance to erythromycin and gentamycin were both 87.5%. There were signiifcant differences in occurrence rates of meningitis, hydrocephalus and ependymitis between late onset GBS sepsis group and late onset non-GBS sepsis group (P<0.05), while no difference in mortality was found between two groups (P>0.05). Conclusions The late onset GBS sepsis is in-sidious, atypical, with many complications and sequelae. It is important for the suspicious neonates to use effective antibiotics as early as possible.

Succinic acid is widely used in the field of chemistry and food proces s. Compared to the traditional chemical synthesis, succinic acid production by fermentation has some benefits. The new process with competitive cost uses rene wable agricultural resources including green house gas CO 2 instead of fossil f uels and reduces pollution from chemical synthesis. In this article, research progr e ss in production of succinic acid by fermentation such as microorganisms, metabo lic pathways and its regulation, fermentation and isolation are briefly reviewed .
6.Construction of prokaryotic expression vector of mouse CD25 extracellular domain

Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
7.Inductive effect of muscular traumatic fluid on the proliferation and myogenesis of bone marrow mesenchymal stem cells

BACKGROUND: Bone marrow mesenchymal stem cells (BMMSCs) can be induced differentiating into osteoblasts and chondroblasts, DNA methylate depressant 5-azacytidine can induce BMMSCs expressing myogenic regulating factors: Myf5 and myopoietin, which involving in the differentiation of BMMSCs into myoblasts.OBJECTIVE: Muscular traumatic fluid containing the highest protein content was screened out and co-cultured with BMMSCs,in order to explore the inductive effect on the proliferation and myogeneis of BMMSCs.DESIGN : Standardized comparative study.SETTING .:At State Key Laboratory of Trauma, Bums and Combined Injury,Institute of Combined Injury, Medical College of Preventive Medicine, Third Military Medical University of Chinese PLA.MATERIALS: Muscular traumatic model was established on 18 Balb/C pure rats for the extraction of muscular traumatic fluid, the inductive effect of the fluid on BMMSC was then compared with 5-azacytidine.METHODS:This study was carried out at State Key Laboratory of Trauma,Burns and Combined Injury, Institute of Combined Injury, College of Preventive Medicine, Third Military Medical University of Chinese PLA from April 2001 and September 2003. Bradford colorimetric was used to detect the protein content in the muscular traumatic fluid, and the fluid with the highest protein was used to co-culture with BMMSC, whose effect on the proliferation of BMMSC was measured with MTT methods at day 0, 3, 6, 9, 12, 15.RT-PCR technique was used to detect the expression of myopoietin at day 6,with its myogenic effect compared with that of 5-azacytidine.fluid on BMMSC.eration of BMMSC: the proloferative activity of BMMSC in traumatic fluid genic effect of traumatic fluid on BMMSC: myopoietin could not be found expressed in traumatic fluid group, but strongly expressed in 5-azacytidine group.CONCLUSION: Muscular traumatic fluid can promote the proliferation of BMMSC, but has no myogenic effect.
8.Application of reticulocyte hemoglobin content in diagnosis of iron deficiency in premenopausal women

Objective To examine the application value of reticulocyte hemoglobin content (CHr)for diagnosing iron deficiency in premenopausal women.Methods The levels of CHr,hemoglobin (Hb), mean cellular volume(MCV),red cell distribution width (RDW) were measured on the ADVIA 120 (Bayer Diagnostics) automated hematology analyzer.Transferrin saturation (TS) and ferritin (SF) were measured on chemistry analyzer.Results CHr in iron deficiency without anemia group were significantly lower than that in the healthy control group (P<0.01)and significantly higher than that in iron deficiency anemia group(P<0.01).CHr in anemia of chronic disease group were significantly higher than that in iron deficiency anemia group(P<0.01).Receiver operator characteristic curve (ROC) analysis in diagnosis of iron dificiency without anemia demonstrated that the area under the curve for CHr,SF,RDW,MCV,Hb were 0.872,0.798,0.721,0.713,0.677,respectively (P<0.01).So CHr has a better overall sensitivity than SF,Hb,MCV and RDW in the diagnosis of iron deficiency without anemia.ROC also showed that the area under the curve for Hb,RDW,CHr,SF and MCV was 1.000,0.969,0.958,0.953 and 0.926,respectively (P<0.01) in iron deficiency anemia.Conclusion CHr is the early and sensitive predictor of iron deficiency in premenopausal women,especially for the diagnosis of iron deficiency without anemia.
9.Expression of α1-AT and VIEGF-C in human bronchoalveolar carcinoma

Objective To study the expressions of α1-AT and VEGF-C in human bronchoalveolarcarcinorrm, and the relation of the expression to the patholo~cM differentiation and clinical stage. Methods All 49 Darffin embedding samples of patients with bronchoalveolar carcinoma were studied. α1-AT and VEGF-C were detected by immunohistochemical SP method.Automated image analyzer was used to quantify α1-AT and VEGF-C expressions.Results The immunohistochemical positive stainings of α1-AT and VEGF-C in brown or dark brown were located in cytopla8m.The expression levels of α1-AT and VEGF-C were not related with the gender,age,tumor position and size,and histology subtypos(P>0.05).It Was found that the expression of α1-AT in patients with local lymph node metastasis was significantly lower than those without node metastasis(P<0.001).It was found that the expression of VEGF-C in patients with local node metastasis significantly higher than th08e without node metastasis(P<0.001).There Was a negative correlation between the expression level of α1-AT and the expression level of VEGF-C in bronchoalveolar carcinoma(r=-0.324,P<0.05).Conclusion α1-AT and VEGF-C could be secreted by bronehoalveolar carcinoma.Bronehoalveolar carcinoma with lower α1-AT expression and higher VEGF-C expression is more likely to have lymph node metastasis.Lower α1-AT expression and higher VEGF-C expression can participate in the mechanism of lymph node metastasis in carcinoma together.
10.Piezoelectric Gene Biosensors for Detection of Pseudomonas aeruginosa Using Signal Amplification Method with Cationic Conjugated Polymers

OBJECTIVE To discuss the feasibility of signal amplification method with cationic conjugated polymers(liposome) applied during the detection of Pseudomonas aeruginosa using piezoelectric gene biosensors.(METHODS) Oligonucleotide probe for P.aeruginosa was immobilized on the surface of gene sensor array and(hybridized) by PCR production of P.aeruginosa.After hybridization,liposome was added.The frequency shifts were recorded and compared with those ones of the control groups.RESULTS The frequency shifts were(significantly) increasing when adding liposome and the compatible concentration of liposome was 0.8?g/?l.(CONCLUSIONS) Liposome signal amplification is proved to be an effective method to amplify the piezoelectric(signal).
