Objective To investigate the mechanism by which electro-acupuncture (EA) promotes revascularization in the brain after focal cerebral ischemia and reperfusion.Methods The Sprague-Dawley rat model of focal cerebral ischemia was made by filament occlusion. The rats were randomly divided into a control group, a model group, and an EA group. The model and EA groups were each divided into 5 subgroups receiving reperfusion 1, 3,7, 14 or 21 days after ischemia. EA was given at the bilateral Hegn point (LI 4) in the EA group. The expression of stromal cell-derived factor-1α(SDF-1α) mRNA was detected using a RT-PCR in the 3, 7 and 14 day subgroups.The immunohistochemical method was employed to detect the expression of SDF-1α protein. Results Compared with the control group, expression of SDF-1α protein increased significantly in the model and EA groups. Compared with the model group, the expression of SDF-1α mRNA increased significantly in the 3, 7 and 14 day subgroups.SDF-1α protein expression and microvessel count increased slightly but not significantly in the 1d subgroup, but the increases were significant in the 3, 7, 14 and 21 day subgroups.Conclusions EA may promote angiogenesis in an ischemic area of the cortex by increasing the expression of SDF-1αmRNA and its protein after focal cerebral ischemia and reperfusion.

Study of kidney deficiency syndrome may utilize the modern molecular biology technique, such as gene chip, representational difference analysis (RDA) and genes sequence analysis, to clone the different genes of kidney deficiency syndrome from single TCM syndrome type of the disease, to investigate the inner connection between the various phenotypes of kidney deficiency syndrome and their genes expression pattern and their function products, then to definite the related genes of kidney deficiency syndrome. The research method may also be used for the study of other TCM syndrome types.

Objective To investigate characteristics of emotional intelligence(EI)and the ways of occupational stress coping in nurses as well as the predictive validity of EI on coping strategies.Methods 464 nurses from general hospitals were collected and evaluated with psychometric instruments including:Wong's Emotional Intelligence Scale(WEIS)and Nurse Ways of Coping Questionnaire.Data were conducted by Logistic regress analysis according to key themes.Results (1)Nurses' demographic characteristics had effects to the ways of occupational stress coping and emotional intelligence.However,there was no statistical difference of El varied with education levels(P＞0.05).(2)Both problems solving and positive recognition are main coping styles of nurses towards occupational stress.Either avoidance or self-reproach was seldom coping strategies of nurses toward occupational pressures(2.60±0.55;2.60±0.75;1.37±0.69;respectively).(3)El was correlated with coping styles(r=-0.123～0.253,P＜0.05,P＜0.01).Conclusion Four dimensions of EI positively or negatively predicted six kinds of occupational stress coping strategies separately.

AIM: To study the effects of M. vaccae on eosinophil apoptosis and Bcl-2 protein expression in lung tissues of asthmatic guinea pigs. METHODS: 30 guinea pigs were divided into normal saline (NS) group, asthma group and M. vaccae treatment group at random, every group included 10 guinea pigs. Guinea pigs in M. vaccae treatment group were injected intramuscularly with 22 5 ?g M. vaccae 10 days before OVA immunization. TdT-mediated dUTP nick end labeling (TUNEL) technique was used to investigate the apoptosis of eosinophils and immunohistochemistry method was used to study the expression of Bcl-2 protein in lung tissues. This study was designed to examine the effects of autophagy on the lipopolysaccharide(LPS)-induced acute lung injury (ALI)and to analyze the possible mechanism.Kunming mice of clean grade were randomly divided into 6 groups:control group(saline;ip);model group(LPS 10 mg/kg;ip);RAP group(rapamycin 6 mg/kg;ip);RAP+LPS group (RAP 6 mg/kg and LPS 10 mg/kg;ip);3-MA group (3-methyladenine 300 μg/kg;ip);and 3-MA+LPS group (3-MA 300 μg/kg and LPS 10 mg/kg;ip).6 h after LPS injection;mortality was checked.Neutrophil aggregation;and the expressions of TNF-α;LC3 and P62 in lung tissue were checked by fluo-rescence microscopy and Western blot.The results revealed a higher mortality;neutrophil infiltration and TNF-αexpression and significantly increased levels of autophagy marker LC3-II and P62 in LPS group;in RAP+LPS group;pretreatment with RAP notably reversed LPS-induced neutrophil infiltration and TNF-αexpression;LC3-II were further slightly increased;while P62 was significantly decreased;in 3-MA+LPS group;pretreatment of 3-MA slightly decreased LC3-II expression;P62;neutrophil infiltration and TNF-αremained almost the same to those in LPS group.These data suggesed that;in sepsis;autophagy flux in the lung tissue is partially blocked;and RAP help to alleviate LPS-induced lung injury;which might through promoting autophagosome maturation;smoothing autophagy flux;and eventually to mitigate pulmonary inflammation.

Objective To explore the feasibility of the hydrogel as a cell carrier for parathyroid cells.Methods Cryopreserved parathyroid cells were recovered and cultured until sufficient,then the cells were digested、centrifuged and pipetted to single-cell suspension.The chitosan solution,crosslinking agents and cell suspension were mixed to hydrogels by some ratio.After that,hydrogels covered by complete medium were incubated at 37 ℃ in constant temperature incubator.Meanwhile,the supernatant were collected for measuring the PTH level every other day and the viability and shape of cells in hydrogels was also evaluated by fluorescopy.Results Parathyroid cells were recovered and cultured successfully and the adherent cells were spindle-like.The hydrogel could be prepared at room temperature in less than 1 minutes.The fluorescent confocal microscopy showed that more than 90％ of the cells in the hydrogel kept viable after 24 hours and the proportion had no significant change after 3 days.Furthermore,PTH was detected in the supernatants.Conclusion The facilely prepared inexpensive hydrogel,which can maintain normal morphlology and function of cells,is of potential application to cell transplantation therapy for hypopatathyroidism.

Objective: To establish the quality control standards for Brains' King Oral solution. Methods: Rhizoma Gastrodia, Rhizoma et Radix Noto-pterygii in oral solution were identified by TLC. The content of gastrodin in Brain's King Oral Solution was determined by HPLC with external standard. The average recovery was 99.22% and RSD was 0.99%(n=6).Results: These methods are simple, accurate and specific.Conclusion: These methods can be used for the quality control of Brain's King Oral Solution.

Objective To investigate the Expression of interleukin-18(IL-18)and cyclooxygenase2(COX-2)in endometriosis.Methods Enzyme linked immunosorbent assay and innunohistochemistry were used to detrermine the levels of interleukin-18 in peritoneal fluid and serum,the expression difference of cyclooxygenase2 in ectopic endometrium with endometriosis and the normal control.Results The levels of interleukin-18 in peritoneal fluid and serum were higher than that of normal control,the expression of cyclooxygenase2 in ectopic endometrium with endometriosis were higher than that of normal control,and the degree in stage of Ⅲ、Ⅳ were higher than those in stage of Ⅰ、Ⅱ.Conclusions IL-18 and COX-2 is related to the endometriosis,the further studying of IL-18 and COX-2 may be helpful in the diagnosis and treatment of this disease.
