Objective To investigate the mechanism of anti-tumor in lung cancer patients and their effects to immune system undergoing with microwave ablation treatment, radioimmunotherapy of 131Ⅰ tumor cells human mouse chimeric monoclonal antibody injection (131Ⅰ-chTNT) and combined treatment of two anterior method. Methods The 50 cases of lung cancer were divided into three groups randomly,17 cases were in the group of simple microwave ablation treatment,15 cases were in the group of simple radiotherapy immunotherapy,18 cases were in the group of combined microwave ablation treatment with 131Ⅰ-chTNT radioimmunoassay.During the study, the T lymphocyte subsets, the activity of NK cells, the expressing of interleukin (IL)-2,IL-10,IL-12,the changing of the interferon γ(IFN-γ) and tumor necrosis factor α (TNF-α) were detected and compared of before and after treatment on all the patients. Results The CD4+ value,CD4+/CD8+ value and the activity of NK cells in these three groups after treatment were higher than that of before treatment (P < 0.05),there was statistic difference between the combined treatment group and two simple treatment groups (P < 0.01).The expression of IL-2,IL-12,IFN-γ TNF-α in three group patients after treatment were higher than that of before treatment, which had statistical significance (P < 0.05),while there were no significant differences between groups (P > 0.05).Conclusion The method combined microwave ablation treatment with 131Ⅰ-chTNT radioimmunoassay has efficiency to improve the immune function, which could improve the comprehensive therapeutic effect of lung cancer excellently.

Chinese Acupuncture & Moxibustion 2016;36(5):552-556
Nowadays extensive attention has been paid on the research of acupuncture academic schools, however, a widely accepted method of partition of acupuncture academic schools is still in need. In this paper, the methods of partition of acupuncture academic schools in the history have been arranged, and three typical methods of"partition of five schools" "partition of eighteen schools" and "two-stage based partition" are summarized. After adeep analysis on the disadvantages and advantages of these three methods, a new method of partition of acupuncture academic schools that is called "three-stage based partition" is proposed. In this method, after the overall acupuncture academic schools are divided into an ancient stage, a modern stage and a contemporary stage, each schoolis divided into its sub-school category. It is believed that this method of partition can remedy the weaknesses ofcurrent methods, but also explore a new model of inheritance and development under a different aspect through thedifferentiation and interaction of acupuncture academic schools at three stages.

5.Role of caspase activation in butyrate-induced apoptosis of HT-29 colon carcinoma cells

Chinese Journal of Pathophysiology 2000;0(10):-
AIM:Sodium butyrate has antitumor effects on colon cancer cells such as inhibiting cell growth and promoting differentiation and apoptosis. The aim of this study is to investigate whether sodium butyrate induces apoptosis in human colon cancer cell line HT-29 and to examine the intracellular mechanisms involved, especially the role of caspase activation in the process.METHODS:HT-29 cells were cultured to logarithmic phase before treatment with sodium butyrate at concentration of 5.0 mmol/L and caspase inhibitors at the concentration of 20 ?mol/L.The latter were added in the medium ahead of sodium butyrate for 1 h.Then, the staining of Annexin V-FITC and PI were used to analyze HT-29 apoptosis and the dye JC-1 was applied to detect mitochondrial membrane potential by flow cytometry.Caspase activity within the cells was measured respectively using a specific caspase activity assay kit and a microplate reader.RESULTS:Preincubation of HT-29 cells with sodium butyrate significantly increased apoptosis [(35.40?0.70)%] and decreased mitochondrial membrane potential(5.53?0.91).This effect was blocked when pretreatments were enforced with z-VAD-fmk,z-DEVD-fmk and z-LEHD-fmk.The apoptosis percentages were(1.33?0.59)%,(1.40?0.53)% and(1.27?0.91)%,respectively and mitochondrial membrane potentials were 9.80?1.15,10.23?0.50 and 10.33?1.02,respectively.However,the role of reduction by z-IETD-fmk,which presented the apoptosis percentage of(32.10?2.33)% and mitochondrial membrane potential of 5.93?1.31,was not observed. An enhancement of caspase-3 and-9 activities(2-3-fold)but no change of caspase-8 activity was confirmed.CONCLUSION:Apoptosis of HT-29 colon carcinoma cells induced by sodium butyrate is tightly linked to caspase activation via mitochondrial pathway other than tumor necrosis factor-alpha and has the potential to inhibit proliferation and thereby may contribute to the progression of colon cancer.
6.Establishment and evaluation of indirect competitive enzyme-linked immunosorbent assay for detecting the concentration of trypsiongen activation peptides
International Journal of Laboratory Medicine 2006;0(03):-
Objective To establish an ELISA method for detection of the concentration of TAP in human urine samples. Methods TAP-BSA was used as coating antigen. The TAP was a competitor to TAP-BSA. They reacted to the limited amount of monoclonal antibody against TAP.Results The optimal concentration of the coating antigen was 250ng/ml.The dilutions of monolclonal antibody against TAP and sheep anti-mouse IgG were 25?g/ml and 1∶4 000 respectively. The optimal range was from 0.69 to 1 000ng/ml.The assay provided a sensitivity of 0.69ng/ml.The coefficients of variation of intra-assay and inter-assay were 9.10% and 10.33% respectively. The average recovery rate of TAP was 97.70%.Conclusion The indirect competitive enzyme-linked immunosorbent assay is established.
8.Key factors in design of case report form.
Acta Pharmaceutica Sinica 2015;50(11):1452-1455
Case report form (CRF) is a key document for data collection in clinical trials. A well-designed CRF is required for database construction, data accuracy, data query/cleaning, CRF completion and statistical analysis. A well-defined process or SOP should be in place for CRF design. Data collection should fully meet the demand of study protocol. The layout of CRF should be clear with well-structured fields and standard coding for fields.

Chinese Journal of Blood Transfusion 1988;0(04):-
Objective To investigate the allelic polymorphism of Human Leukocyte Antigen(HLA)-DRB1*04 in Southern and Northern Chinese Han populations.Method A total of 104 and 168 samples of HLA-DRB1*04 alleles from Southern and Northern Chinese Han populations respectively were genotyped at high-resolution by PCR-SBT,and then the DRB1*0406/0449 ambiguous allele pairs were identified via high-resolution PCR-SSP.The allelic distribution of DRB1*04 in Southern and Northern Chinese Han populations were compared with other different populations by chi-square test. Results All 56 ambiguous allelic pairs of DRB1*0406/0449 were identified as DRB1*0406.A total of 5 alleles were observed in Southern Chinese Han population and the most frequent alleles were DRB1*0405(47.32%),DRB1*0406(24.24%)and DRB1*0403(16.35%).There were 10 alleles tested in Northern Chinese Han population, and the major alleles were DRB1*0405(40.48%),DRB1*0406(18.45%) and DRB1*0403(17.26%).The overall distribution of DRB1*04 alleles in Southern and Northern Chinese Han differed significantly from that of the Caucasian,African American,Hispanics, and Asian and Pacific Islanders, but not from the Korean. Conclusions The overall distribution of HLA-DRB1*04 alleles differed significantly between Southern and Northern Chinese Han populations, but the major alleles DRB1*0405,DRB1*0406 and DRB1*0403 showed the same distribute characteristic. The distribution of DRB1*04 alleles in different populations has their own characteristics.
