1.Flavonoids in Shuanghuanglian Injection

AIM: To investigate the chemical constituents of Shuanghuanglian Injection(Flos lonicerae japonicae, Radix scutellariae, Fructus forsythiae). METHODS: Chromatography and spectral analysis were used respectively to isolate and identify the constituents. RESULTS: Six compounds were isolated from Shuanghuanglian Injection which included baicalin(Ⅰ)、quercitrin(Ⅱ)、hyperoside(Ⅲ)、rutin(Ⅳ)、quercetin(Ⅴ) and luteolin(Ⅵ). CONCLUSION: The other five compounds were first isolated and identified in Shuanghuanglian Injection except baicalin(Ⅰ).
2.Quantitative analysis of interleukine-2 receptor ? mRNA expression in ankylosing sondylitis by fluorescence quantitative reverse transcription polymerase chain reaction and clinical significance

Objective To construct and evaluate the real-time fluorescence quantitative polymerase chain reaction for detecting IL-2R? mRNA in ankylosing spondylitis(AS) based on TaqMan technique.Methods A pair of primers and a TaqMan probe were designed by sequence in GenBank.Total RNA isolated from the fresh peripheral blood monocytes (PBMC) of homo sapiens was amplified by the real-time FQ-RT-PCR.The product was collected by agarose gel electrophoresis and sequencing analysis then ligated with pUCm-T.The recombined plasmid was transcribed to cRNA by T7 RNA polymerase in order to prepare serial standard materials.A new method was created to quantify IL-2R? mRNA in ideal condition.Sensitivity, reproducibility and efficiency were evaluated and used, combined with sIL-2R,for clinical application of AS.Results The linear range was (7~107) cells/ml.The intra-and-inter-assay coefficient variation was 8.4% and 9.6% respectively. Recombined plasmid contained the target fragment was specific and accurate by BLAST.Standard reference was constructed successfully.The RT-PCR product in AS with HLA-B27 positive groups was higher than that with HLA-B27 negative groups and health controls (P0.05).The sensitivity of IL-2R? mRNA was 96.7%.Conclusions Real Time FQ-RT-PCR of IL-2R? mRNA is constructed successfully.This is an easy,rapid,sensitive, accurate and reliable method for quantifing IL-2R? mRNA. There is highly statistical significance, compared with sIL-2R, on the expression of IL-2R? mRNA and inflammatory states between AS and control group and effective information for administration of patients.
3.Retrospective Analysis of Cesarean Hysterectomy Treated with Intervention of Postpartum Hemorrhage in 9 Cases

Objective To investigate the indication, occasion and risk predication of cesarean hysterectomy and interventional treatment. Methods The clinical data of 9 patients with low cesarean section postpartum hemorrhage that underwent hysterectomy after interventional treatment from January 2012 to January 2013 were analyzed which included surgical indication, disease characteristics, procedure time, bleeding volume and other parameters. Results The most common surgical indication for cesarean that caused postpartum hemorrhage was dangerous placenta previa, accounting for 56%, followed byⅢdegree of placental abruption and preeclampsia. The surgical bleeding of cesarean accounted for 62%-70%of the total amount of bleeding. The total time of rescue was 7.4 ± 1.4 hours. The total amount of bleeding was 7 100 ± 2 250 mL. The occurrence of coagulation disorder was significantly correlated with surgical time and amount of blood loss. Conclusion For postpartum hemorrhage caused by cesarean section with risk factors such as scar uterus, placenta previa, placental abruption, many pregnant prolific and others, we must relax indications of hysterectomy. With future treatment potential, the early intervention of postpartum hemorrhage also plays an important role in reducing hysterectomy rate, reducing intraoperative and postoperative blood loss.
4.Studies on Safeguard Measures of the Shortage of Essential Medicines Based on Database Model

Objective: By building database model, to put forward measures to solve the shortage of basic drugs. Methods: There were five factors influencing medicine shortage have been researched, collecting data, using SPSS 18.0 software to build the database model of different parts, and then analyzed the existing problems and inner link, scientifically came up with the basic framework and main solutions of guarantee measures on shortages of essential medicines. Results: The database model of country and six areas in it have been built, and the influence factors of drug shortage in different region have been analyzed, and raised five aspects measures to secure reliable supplies of drugs, including selection, bid and procurement; production and flow; reserve and adjust;pricing and pay;use training. Conclusion: This study extends the application of database, enriches research ideas of drug policy, and the policy put forward to avoid drug shortages to a certain extent.
5.rhG-CSF increases the expression of fibronectin in the marginal zone of the cerebral infarction in rats

AIM: To explore whether the administration of rhG-CSF improve the neurological function after focal cerebral ischemia and observe the expression of fibronectin. METHODS: Injection of 10 ?g?kg -1?d -1 rhG-CSF for 5 days was given subcutaneously to rats undergoing middle cerebral artery occlusion (MCAO). Neurological severity scores (NSS) test was performed. The expression of fibronectin and Brdu were observed by immunohistochemistry. To visualize the cellular colocalization of Brdu and fibronectin, glial fibrillary acidic protein (GFAP) and fibronectin, double fluorescent staining were used. RESULTS: ① NSS at 7th day, 14th day and 21th day of groups undergoing MCAO treated with rhG-CSF were 4.00?0.89, 3.83?1.17, 3.50?1.38, respectively. 6.Clinical efficacy of Yaotongning capsule in the treatment of lumbar osteoarthritis patients

Objective To analyze the effect of Yaotongning capsule in the treatment of lumbar osteoarthritis patients.Methods From March 2013 to March 2015 in our hospital, 80 cases of lumbar osteoarthritis patients according to the digital table method were divided into two groups: the control group and the experimental group.The patients of the control group were given Bitongning, and the patients of the experimental group were given Yaotongning capsule, and the therapeutic effect, serum indexes and Japanese Orthopaedic Association ( JOA ) of two groups were compared. Results The curative effect of the experimental group was higher than control group(95.00%vs.67.50%) , the JOA score of the experimental group was higher than control group [(27.46 ±1.07)points vs.(21.06 ±1.89)points] (P<0.05).After treatment, the MMP-3(65.28 ±4.37) ng/L and IL-1β(12.43 ±1.01) ng/L of the experimental group were significantly lower than MMP-3(79.56 ±5.36) ng/L, IL-1β(16.44 ±1.03) ng/L of the control group, and the difference was significant (P<0.05).Conclusion The lumbar osteoarthritis patients with Yaotongning capsule has obvious effect, and the function of lumbar intervertebral joint improves obviously, and the serum index is normal, safe and reliable.

8.Construction of murine model of TAO by immunification with recombinant extracellular domain of human thyrotropin receptor in cationic liposomes

Background Thyroid-associated ophthalmopathy (TAO) is a kind of clinically common and incurable ocular disease,and its incidence is at top place.The etiology and pathologic mechanism of TAO are still unknown because of shortness of replicative animal models and difficulty to acquire the ocular tissues in the early stage of the disease.To better understand the pathogenesis of TAO and investigate effective treatable measures, an appropriate animal model should be developed.Objective This study was to immunize female BALB/c mice with the recombinant plasmid of human thyroid-stimulating hormone receptor (TSHR) extracellular domain in cationic liposomes for the establishement of TAO models.Methods Thirty-two 6-to 8-week-old female BALB/c mice were randomly assigned to four groups according to computer random allocation.pcDNA3.1 +/hTSHR289 of 100 μg in an adjuvant cationic liposomes was injected via anterior tibialis muscle and peritoneal cavity separately in the recombinant plasmid injection group in 0, 3,6 weeks, and pcDNA3.1 or cationic liposomes was injected in the liposomes injection group or the blank plasmid group in the same way, respectively, and normal saline solution was injected in the blank control group.Body weight of the mice was measued before and 1 month,2,3 and 4 months after initial injection.The manifestations were observed after modeling.The mice were sacrificed 17 weeks after initial injection,and the histopathology examination was carried out on the thyroid gland and orbital tissue.The heart blood was collected from the mice,and serum contents of total thyroxin 4 (TT4) and thyroid-stimulating hormone (TSH)were assayed by ELISA.Results Protrusion, eyelid swell and keratitis occurred in 12 eyes of 6 mice in the recombinant plasmid injection group after immunization.A significant difference in the body weight of the mice was found among the blank control group, blank plasmid group, liposomes injection group and recombinant plasmid injection group (Fgroup =3.425, P =0.028), and the body weight was considerably reduced in the recombinant plasmid injection group in comparison with the blank control group, blank plasmid group,liposomes injection group (Ftime =0.838 ,P=0.023).The serum levels of TT4 were (7.75±1.00), (7.96±0.76), (6.76±1.10) and (4.43±2.88) μg/dlin the blank control group, liposomes injection group, blank plasmid group, and recombinant plasmid injection group, and those of TSH were (6.36±2.58),(4.83±3.96),(6.63±1.71) and (1.60 ±1.76) ng/ml, showing significant differences among the groups (F =7.150, P<0.001;F =5.521, P<0.01) , and the serum levels of TT4 and TSH were remarkably lower in the recombinant plasmid injection group than those of the blank control group,liposomes injection group and blank plasmid group (all at P < 0.05).Histopathology revealed the lymphocyte infiltration of thyroid gland in 6 mice and proliferation of orbital adipose tissue, infiltration of lymphocytes and mastocytes,deposition of hyaluronic acid as well as swell, breakage and inflammatory cell infiltration of extraocular muscle in 15 eyes of the recombinant plasmid injection group.Conclusions A murine model of TAO can be successfully induced by immunization with recombination plasmid pcDNA3.1 +/hTSHR289 and cationic liposomes.The histopathology characteristics and ocular findings of the animal models are similar to human TAO.

To observe the efficacy and safety of mini-percutaneous nephrolithotomy with Neodymium: Yttrium Aluminum Garnet (Nd-YAG) laser in the treatment for upper urinary tract stones, from December 2005 to September 2006, 31 patients with renal stones, 15 patients with ureteral stones and 7 patients with renal and ureteral stones underwent mini-percutaneous nephrolithotomy with Nd-YAG laser by combination of rigid ureteroscope and flexible ureteroscope under B-ultrasound guidance. Clinical data including operation time, lithotripsy time, complications and stone-free rate were analyzed retrospectively. Our study showed that the percutaneous renal access (F14-F18) was successfully established under B-ultrasound guidance in all cases. Immediate phase: lithotripsy was performed in 47 cases through single tract, and in one case through two tracts. Delayed phase II lithotripsy was done in 5 cases of renal stones. Operation time ranged from 55 to 180 min with an average time of 100+/-15 min. Lithotripsy time was from 25 to 135 min and the average lithotripsy time was 65+/-11 min. No severe complications occurred in our series. Complex renal stones were cleared in 34 of 38 cases (89.5%). All ureteral stones were completely removed in 15 cases (100%). It was concluded that mini-percutaneous nephrolithotomy with Nd:YAG laser for the treatment of upper urinary tract stones by combination of rigid ureteroscope and flexible ureteroscope has the advantages of effectiveness, mini-invasion, shorter operative time and safety.
