Objective To investigate the correlated factors of prognosis for acute myeloid leukemia (AML).Methods A retrospective analysis of initial 182 patients with AML was performed to assess the relationship between prognostic factors including sex,age,white cell count at diagnosis,immunophenotype,cytogenetics,allo-HSCT and complete remission (CR) after one-course of induction and overall survival (OS) or event free survival (EFS).Results The median age of 182 patients was 49 (14-80) years old,and the median follow up was 9.7 (0.5-75.5) months.There were 107 patients achieved CR after one-course of induction (total response rate was 65.9 ％,120/182),age≥60 years.CD64 negative,unfavorable cyto-genetics and failing to achieve CR after one-course of induction were associated with lower total CR rate.Univariate analysis results demonstrated that age ≥60 years,unfavorable cytogenetics,CD11b expression and failing to achieve CR after one-course of induction had poor influence on 3-years OS and EFS,while the 3-years OS and EFS for patients with CD19 expression,CD64 expression and allo-HSCT were better than that of the patients with negative CD19 and CD64.Multivariate analysis suggested that age,cytogenetics,CD11b,CD64,allo-HSCT and CR after one-course of induction were independent prognostic factors for OS,while cytogenetics,CD64,allo-HSCT and CR after one-course of induction were important on EFS.Conclusions Prognostic analysis depending on AML corelation factors is benifit to individual therapy and can be applied to improve the survival.

Objective To investigate the effect of ovariectomy and estrogen on secretion function and number of pancreatic islet beta cell in low-dose streptozotocin-induced diabetic rats. Methods Thirty female SD rats were randomly divided into five groups: normal control(NC) group, streptozotocin(STZ) group, ovariectomized(OVX) group, OVX + STZ(OS) group and OVX+STZ+estradiol(OSE) group. OVX, OS and OSE groups underwent ovariectomy, while NC and STZ groups underwent just sham operation. After surgery, OSE group was treated subcutaneously with estradiol 0.2 mg/kg twice weekly. At the end of 3 weeks, STZ, OS and OSE groups were induced by a single intraperitoneal injection of 40 mg/kg STZ. Then eight days later, plasma glucose and insulin levels were tested. The insulin protein, the average beta cell area and the relative beta cell mass were tested by streptavidin peroxidase conjugation method (SP). The quantification of beta cell apoptosis was performed by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL). The expressions of proliferating cell nuclear antigen(PCNA) protein, Bax and Bcl-2 were tested. Results With the administration of low-dose STZ, the plasma glucose was significantly higher and the insulin secretion curve after glucose loading, △I30/△G30 and modified beta cell function index(MBCI) were lower in OVX group than in other groups(all P<0.05). At the same time, the insulin protein, the relative beta cell mass and the beta cell area were dramatically decreased(all P<0.05). The beta cell apoptotic index was increased (t = 2.957, P< 0.05), the expression ratio of Bcl-2/Bax was decreased (0.41±0.03 vs. 0.76±0.05, P<0.05). Estrogen replacement therapy could obviously inhibit these changes. Compared with OS group, glucose disturbances and insulin secretion were improved dramatically in OSE group(all P<0.05); the insulin content, the relative beta cell mass and the average beta cell area were all enhanced (all P<0.05); the beta cell apoptotic rate was decreased(t =2.482, P<0.05), and the expression tatio of Bcl-2/Bax was increased (0.71±0.05 vs. 0.41±0.03, P<0.05). Conclusions The ovx rats are significantly more susceptible to low-dose STZ toxicity than in control rats. Under the effect of STZ, the ability of insulin secretion of beta cell is obviously decreased, while apoptosis is increased, which induces a higher glucose level and a lower insulin secretion. Administration of estrogen may protect OVX rats from the metabolic disturbances.

Objective To describe the research status, analyze the existing problems and summarize development trend of basic experiment research in China by analyzing the papers publication year, publication journals, geographical distribution, fund status, cooperation situation and the main experimental methods or technologies.It was aimed at providing guidance for further research and development of nursing scientific research.Methods All literatures related to nursing professional basic experiment research were extracted by searching through the China National Knowledge Infrastructure, VIP Database for Chinese Technical Periodicals and Wan-fang database from January 2005 to November 2014.Bibliometric analysis methods were used to analyze the search results.Results 472 references were retrieved to meet the requirements.about 76.3％ (360/472) papers were published in the statistical source journals, 10.0％ (47/472) of the literatures were published in the nursing professional journals;68.9％ (325/472) of the literatures had funding source, 21.0％ (99/472) for national fund support, 35.4％ (167/472) were supported by provincial fund;literature cooperation degree was 4.38(2 065/472), the co-authored rate was 97.2％ (459/472).The level of research focused on tissue and cells, a few involved in molecular level.Conclusions All the literatures on the nursing professional basic experiment research show a increa~d trend year by year.Nursing professional journals' publication volume is not high for such research.Scientific research fund contributions remain to be improved.The interdisciplinary cooperation needs to be expanded.Nursing experimental research will provide a possible for discipline development.

Objective An ELISA-based assay for detecting alloantibodies against FⅧ was established to estimate the incidence of alloantibodies against FⅧ in treated patients with hemophilia A. Methods One hundred and forty patients with hemophilia A and 80 healthy controls were enrolled. Among hemophilia A patients, 84, 34 and 22 patients were in severe, moderate and mild conditions respectively. All patients were treated with plasma-derived FⅧ concentrates before. The titer wells were coated with MoAb against FⅧ which was developed in our laboratory. Then human recombinant FⅧ concentrates were applied. After incubation in room temperature for 2 hours, diluted plasma samples and HRP-conjugated goat anti-human IgG were added successively. Finally Absorbance (A490) were measured and recorded. Inhibitor activity against FⅧ for all plasma samples was measured by a modified Nijmegen assay simultaneously. Results The results showed that alloantibodies against FⅧ were found in 40.0% (56/140) patients by ELISA. And the alloantibody incidences in the severe and non-severe patients were 47.6% (40/84) and 28.5% (16/56)respectively. There was statistical significance between these two categories (x2 = 5.079, P ＜ 0.05 ). The FⅧ inhibitor activity was detected in 24.3% (34/140) patients by modified Nijmegen assay. The inhibitor incidences in the severe and non-severe patients were 33.3% (28/84) and 10.7% (6/56) respectively.There was statistical significance (x2 = 9.349, P ＜ 0.05). Twenty-five patients were positive for FⅧ alloantibodies by ELISA but had no FⅧ inhibitor activity by the modified Nijmegen assay. The positive rates of FⅧ alloantibodies and inhibitor activity were 40.0% (56/140) and 24.3% (34/140) respectively,which had significant difference (x2 = 15.75, P ＜ 0.01 ) and strong positive correlation ( rn = 0.59, P ＜0.01 ). Meanwhile the results deduced from these two tests shared a high consistency rate ( Kappa = 0.55,P ＜0.01 ). Conclusions The detection rate for alloantibodies against F Ⅷ is enhanced by our newlydeveloped ELISA. Our results suggest that the occurrence of the alloantibodies against F Ⅷ in Chinese hemophilia A patients is not rare and the alloantibody incidence is preponderant in the patients with severe hemophilia A compared with non-severe hemophilia A patients.

OBJECTIVE:To explore the feasibility of question-and-answer practice teaching in the pharmacy department of our hospital. METHODS:The practice teaching of pharmacy department was taken as a pilot,and the question-and-answer practice teaching was used in emergency and outpatient pharmacies,inpatient pharmacy,PIVAS,drug warehouse,clinical pharmaceutics room,preparation room and drug testing laboratory. The effects of the practice teaching on internships,teachers,pharmacy depart-ments and patients were excavated. RESULTS & CONCLUSIONS:Satisfaction of teachers,pharmacy,patients and clinics for the question-and-answer practice teaching was 100%,and satisfaction of internships was 94.5%. The question-and-answer practice teaching has improved their professional knowledge and competence,resolved difficulty of patients about drug counseling and helped the improvement of overall business level in department.

Objective We aimed to investigate the community nurses' awareness of health policy,related behaviors and paths of learning.Methods A total of 150 community nurses were enrolled in this study by answering a self-completion questionnaire.The survey contents included general conditions of nurses,awareness,related behaviors and paths of learning of community health policy.Results 91.7％ of the respondents believed that heahh policy was important or very important,but only 10.4％ of the respondents understood health policy fully.71.5％ said that they would lecture patients health policy actively and 70.8％ would lecture their colleagues actively.51.4％ respondents would like to take part in policy-making if they were allowed.Education background,professional title and the awareness of health policy were factors of community nurses' policy related behaviors.They learned health policy by working meeting,training and documents.Conclusions It is necessary to improve their school record and adjust their professional title,to strengthen the publicity of health policy by broaden channels of publicity.By these,community nurses' awareness of health policy would be improved,their policy related behaviors would be promoted.Further,the health policy would be well delivered and implemented.

17β-estradiol(E2), progesterone (P) and testosterone (T) were investigated for their effects on the proliferation and differentiation of primary rat calvarial osteoblasts in vitro. Rat calvarial osteoblasts were cultured with l0-10 mol/L E2, 10-9-10-6 mol/L P and l0-10 mol/L T for 20days. Cell proliferation was determined by 3H-thymidine incorporation and cell counting. Cell differentiation was examined by measuring alkaline phosphatase (ALP) activity, osteocalcin gene expression and production, bone nodule formation in different periods of culture. Our results showed that no effect of three sex hormones was observed on cell proliferation, but, the responses of cell differentiation to the hormones were more or less different. Among these three hormones used in this study, P appeared to have multi-stimulating effect on cell differentiation. Effect of T seemed not so significant as that of P on cell differentiation. Although ALP activity and osteocalcin production were increased significantly by T, it had slight effect on osteocalcin mRNA and bone nodule formation. Besides, E2 did not demonstrate any effect on cell differentiation. It is concluded that the proliferation of rat calvarial osteoblasts was independent of the presence of sex hormones. However, the differentiation of these cells were stimulated in different levels and to different extent either by P or T. P appeared to be a multi-stimulator on differentiation of such cells.

Objective To explore the caregiver burden and psychological resilience of patients with dilated cardiomyopathy and the correlation between the two factors.Methods Totally 96 caregivers of dilated cardiomyopathy patients were selected by convenience sampling and investigated for their level of burden and psychological resilience with the Caregiver Burden Inventory (CBI) and Conor-Davidson Resilience Scale (CD-RISC). The correlation between the two factors was then analyzed.Results The CBI score of these caregivers was (47.19±10.38), significantly higher than the average level nationwide (24.2±5.8). Their CD-RISC score was (63.07±13.18), lower than (65.4±13.9) of China's community population. There was apparent negative correlation between caregiver burden and psychological resilience (r=-0.62,P<0.01). The total score of resilience was negatively related to the burden of development constraint, social burden and emotional burden (r=-0.72, -0.65, -0.51;P<0.01).Conclusions The caregivers of dilated cardiomyopathy patients have a heavy burden, which is negatively correlated with their psychological resilience. Therefore, measures should be taken in clinical work to optimize the caregivers' psychological resilience, reduce their burden, and improve their quality of care.

Objective: To explore the effects of checklist for nursing handover in emergency department. Methods: A total of 48 emergency department nurses were recruited by convenient sampling method. We implemented nursing bedside handover checklist four months, the quality of the nursing handover and nurses′ mastery of the patients′ condition and the patients′ satisfaction were compared before and after the implementation. Results: After four-month intervention, the score of the nursing handover quality increased from 60.39±3.22 to 67.73±3.09, the difference was statistically significant (t=-14.377, P<0.01). The score of the nurses′ mastery of the patients′ condition increased from 23.89±2.34 to 27.08±1.82, and there was significant difference (t=-7.287, P<0.01). Patients′ satisfaction improved from 91.163% (111/121) to 96.67% (116/120), the difference was statistically significant (χ²=10.66, P<0.05). Conclusions Implementation of nursing bedside-handover checklist in emergency department can reduce individual cognitive defects, decrease the information missing of the nursing handover,improve the quality of nursing work and ensure the safety of patients.
