1.Establishment and identification of a ClC-3/MMTV-PyMT hybrid mouse strain

Objective To establish a CLCN3/MMTV-PyMT double transgenic mouse model of spontaneous breast tumor with simultaneously overexpressing ClC-3.Method CLCN3 transgenic mice were crossed with MMTV-PyMT spon-taneous mammary tumor model mice.The genotype was determined by PCR.The expression of ClC-3 in tissues was detec-ted by immunofluorescence and Western blot.Results CLCN3 and MMTV-PyMT transgenic mice were bred and CLCN3/MMTV-PyMT hybrid mouse model was successfully established.The ClC-3 expression in CLCN3/MMTV-PyMT hybrid mice was higher than that in the MMTV-PyMT mice, assessed by immunofluorescence and Western blot analysis.Conclu-sions Transgenic mouse models of spontaneous breast cancer with simultaneously overexpressing ClC-3 are successfully es-tablished.The double transgenic mice provide a good animal model for further research of ClC-3 in tumor growth and metas-tasis.
2."Music oxygen bar" on psychological resilience of breast cancer patients at early postoperative rehabilitation stage

Objective To investigate the effect of "music oxygen bar" on resilience in patients at early breast cancer postoperative rehabilitation stage.Methods From February 2016 to July 2016,180 patients with breast cancer modified radical mastectomy were randomly divided into observation group and control group, with 90 in each group. In the control group,rehabilitation guidance from case trained nurses separately at the patient's bedside after surgery. In the observation group,on the basis of the control group,patients at the first day of morning,afternoon add one "music oxygen bar",collective rehabilitation guidance and answering questions;after discharge,each week patients received review and invited to participate the "music oxygen bar". Before and after the interventions after operation,patients were investigated by Connor-Davidson Resilience Scale (CD-RISC) to evaluate self-resilience in two groups patients.Results After 4 weeks intervention,the scores of observation group were higher than that of control group in all items of CD-RISC,with self-fulfillment was (2.44±0.59),no discouragement at fail (2.39±0.61)(t=3.10,1.98;P<0.05).Conclusions The implementation of the "music oxygen bar" for breast cancer patients can provide effective psychological support,and improve psychological resilience for the early postoperative rehabilitation patients.
3.Research Progress on Animal Model for Cancer-related Fatigue

Cancer-related fatigue (CRF) is a common symptom which associated with cancer itself or cancer treatments. CRF will not only interrupt the patient's treatment, but also affect the patient's quality of life seriously. To date, the pathogenesis of CRF is still unclear, and recognized effective treatment is lacking. It is the precondition of establishing standardized animal models for CRF to find effective treatments. This paper summarizes the animal model of CRF in order to find suitable methods, and distinguish the effects of different interventions.
4.Surveillance for avian influenza virus in environments in Liaoning Province

Objective :
To learn the epidemic distribution of avian influenza virus in external environments in Liaoning Province,and to provide evidence for the prevention and control of avian influenza.
Methods :
The environmental samples were collected monthly during 2016 and 2017(including samples from emergency monitoring in June to August,2016 and March to May,2017)from live poultry markets,live poultry wholesale markets,large-scale poultry farms(households),free-range poultry famers and poultry processing factories in Liaoning Province. Real-time polymerase chain reaction assay was used to detect nucleic acid of Influenza A as well as H5,H7 and H9 subtypes in the environmental samples. The distribution of avian influenza virus in external environments in Liaoning Province was analyzed.
Results :
A total of 4 037 environmental samples were collected and detected from 2016 to 2017,there were 177 copies of type A avian influenza virus and the positive rate of avian influenza A virus was 4.38%. The positive rate in 2017 was 6.26%, which was higher than 2.40% in 2016(P<0.05). H9 subtype had the highest positive rate of 3.07%;H7 subtype was first detected in 2017. The positive rates of avian influenza virus from the first to fourth quarters of a year were 8.54%,4.88%,2.17% and 1.45%,respectively. The positive rates of avian influenza virus in live poultry markets were 8.08%,the highest among different sites,and the subtypes were mainly H9. The positive rates of avian influenza virus in samples of poultry cage and poultry washing sewage were 23.47% and 15.96%. H5 and H9 subtypes were detected in all types of samples,and H7 subtypes or mixed types were detected in samples of feces,poultry cage,poultry drinking water and chopping board.
Conclusion
The subtypes of avian influenza virus in the environments of Liaoning Province were mostly H9 and H5,and the H7 was first detected in 2017. Live poultry markets should be the key monitoring sites,especially in winter and spring.
5.Analysis of clinical phenotype and genetic variants among four Chinese pedigrees affected with Waardenburg syndrome.

OBJECTIVE:
To explore the genetic basis for four Chinese pedigrees affected with Waardenburg syndrome (WS).
METHODS:
Four WS probands and their pedigree members who had presented at the First Affiliated Hospital of Zhengzhou University between July 2021 and March 2022 were selected as the study subjects. Proband 1, a 2-year-and-11-month female, had blurred speech for over 2 years. Proband 2, a 10-year-old female, had bilateral hearing loss for 8 years. Proband 3, a 28-year-old male, had right side hearing loss for over 10 years. Proband 4, a 2-year-old male, had left side hearing loss for one year. Clinical data of the four probands and their pedigree members were collected, and auxiliary examinations were carried out. Genomic DNA was extracted from peripheral blood samples and subjected to whole exome sequencing. Candidate variants were verified by Sanger sequencing.
RESULTS:
Proband 1, with profound bilateral sensorineural hearing loss, blue iris and dystopia canthorum, was found to have harbored a heterozygous c.667C>T (p.Arg223Ter) nonsense variant of the PAX3 gene, which was inherited from her father. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), the variant was classified as pathogenic (PVS1+PM2_Supporting+PP4), and the proband was diagnosed with WS type I. Proband 2, with moderate sensorineural hearing loss on the right side and severe sensorineural hearing loss on the left side, has harbored a heterozygous frameshifting c.1018_1022del (p.Val340SerfsTer60) variant of the SOX10 gene. Neither of her parents has harbored the same variant. Based on the ACMG guidelines, it was classified as pathogenic (PVS1+PM2_Supporting+PP4+PM6), and the proband was diagnosed with WS type II. Proband 3, with profound sensorineural hearing loss on the right side, has harbored a heterozygous c.23delC (p.Ser8TrpfsTer5) frameshifting variant of the SOX10 gene. Based on the ACMG guidelines, it was classified as pathogenic (PVS1+PM2_Supporting+PP4), and the proband was diagnosed with WS type II. Proband 4, with profound sensorineural hearing loss on the left side, has harbored a heterozygous c.7G>T (p.Glu3Ter) nonsense variant of the MITF gene which was inherited from his mother. Based on the ACMG guidelines, the variant was classified as pathogenic (PVS1+PM2_Supporting+PP4), and the proband was diagnosed with WS type II.
CONCLUSION
By genetic testing, the four probands were all diagnosed with WS. Above finding has facilitated molecular diagnosis and genetic counseling for their pedigrees.

6.Analysis of antigenicity-related gene evolution and vaccine-matching rate of influenza A virus prevalent strain in Liaoning province in 2018-2019

Objective:Analyze the hemagglutinin(HA) gene of influenza A strains in Liaoning Province from 2018 to 2019, calculate and evaluate the matching degree of influenza A strains and vaccine strains.Methods:49 influenza A strains from 2018 to 2019 were selected for sequencing, and the HA gene was analyzed for evolution; the mutation of antigen epitopes, glycosylation sites and receptor binding sites were studied; the matching degree between the epidemic strain and the vaccine was analyzed by pepidope model.Results:The 2018-2019 influenza pandemic season was concentrated in November-March, and the main pandemic strain was H1N1 pdm virus. Through evolutionary analysis, H1N1 pdm virus belonged to the 6B.1 branch, and the seasonal H3N2 strain was distributed in the 3C.2a branch, all of which belonged to the same branch as the vaccine strain this year. Some strains mutated at epitope and receptor binding sites. In some strains, glycosylation sites increased or were absent compared with vaccine strains, and no new glycosylation sites were present. The Pepitope model was used to evaluate the vaccine effect. H1N1 pdm virus vaccine of this year showed good protective effect, but the seasonal H3N2 vaccine strain showed negative protective effect for epidemic strain, which could not play an effective protective role. Conclusions:In 2018-2019, H1N1 epidemic strain in Liaoning Province had some variation, and the matching degree between H3N2 subtype epidemic strain and 2018-2019 vaccine strain was low. We should pay close attention to the gene variation of the epidemic strains and update the vaccine strains in time.
7.Mental health status of adolescents with adverse childhood experience and the influencing factors.

Adverse childhood experience (ACE) is potentially negative experience that occurs between 0 and 18 years old. The ACE adolescents have prominent mental health problems such as emotional regulation disorder, unstable interpersonal relationship, poor coping ability, and cognitive dysfunction. Until now, the factors affecting the mental health of ACE adolescents are not clear, but it is certain that the ecosystem in which ACE adolescents life affects their mental health. Specifically, the parent-child relationship, the school environment, the peer relationship in the micro-system, and the interaction between the parent-child relationship and other interpersonal relationship in the meso-system have been confirmed to be significantly related to the mental health of ACE adolescents. In the appearance system, the neighborhood cohesion, the level of family income, the educational level of parents and the different social and cultural background in the macro-system all have different degrees of impact on the development of ACE adolescents' psychological behaviors. In the diachronic system, the time and frequency of suffering from ACE have different effects on the mental health regarding the ACE adolescents.
