1.Micro-system engineering and SIMPP analysis method for teaching reform

In order to improve the teaching reform of higher education systematically and op-erably, and in order to improve the effectiveness of teaching reform, teaching reform in Higher Educa-tion in the micro system engineering and SIMPP analysis method were proposed. With questionnaire survey on classroom learning state of all students from traditional Chinese medicine Grade 2010-2013 of Beijing University as an example, the principle and operation steps of SIMPP analysis method was systematically discussed. According to the students' related behavior pattern and strategy analysis affected by teachers, the analysis of the results of interpretation method and application direction was illustrated. At the same time, through the monograph, nine aspects (academic diligence level, learning effectiveness of self cognition, learning interest, teacher's influence, self-study ability, class atten-dance, learning objectives, curriculum satisfaction, major satisfaction) were analyzed by SIMPP. By modeling the related factors and their relationship, the internal relations between different factors were cleared, and the students' behavior patterns were found, and the corresponding reform measures were designed. Research shows that the concept and methods of teaching reform in Higher Education in the micro system engineering will produce a positive impact on teaching reform, curriculum reform, and the integration of family education and school education.

3.Feature of Ventilation Induced Lung Injury with Different Tidal Volume in Neonatal Rats

Objective To investigate the feature of ventilation induced lung injury(VILI) with different tidal volume in neonatal rats.Methods Thirty-two neonatal rats were assigned to control group(without ventilation),conventional ventilation group(tidal volume 0.010 L?kg-1 for 5 h),hyperventilation 5 h group(tidal volume 0.025 L?kg-1 for 5 h),hyperventilation 3 h group(tidal volume 0.025 L?kg-1 for 3 h) randomly.After ventilation,the lungs were obtained to weigh,score about the degree of lung pathologic injury was count.The levels of IL-6 and IL-10 of lung tissue were detected by enzyme-linked immunosorbent assay.Results The VILI histopathology score in hyperventilation 5 h group,hyperventilation 3 h group,conventional ventilation group and control group were 9.63?1.40,4.40?1.06,6.50?1.85 and 0.00,respectively,the differences were significant among the 4 groups(P=0.000).IL-6 in hyperventilation 5 h group,hyperventilation 3 h group,conventional ventilation group and control group were(785.33?39.06) pg?g-1,(656.78?48.82) pg?g-1,(701.6?33.65) pg?g-1 and(635.02?65.78) pg?g-1,there were significant differences among the 4 groups(P=0.000).The IL-6 level was positively correlated with VILI histopathology score(r=0.78,P

Aim To observe the effects of Breviscapine (Bre) on the contractions induced by noradrenaline (NA) and high potassium in rabbit aorta strips and to investigate the relationships of these effects to the changes of intracellular free calcium( i). Methods The effects of Bre on the concentration-response curves for NA and high potassium, and on the transient contractions induced by NA and caffeine in Ca 2+-free medium and the sustained contraction by NA after replenishing Ca 2+ were surveyed using rabbit aorta strips; the changes of i increased by NA and high potassium in the presence of Bre were determined using fura-2/AM loaded cultured smooth muscle cells of rabbit aorta. Results Bre shifted the concentration-response curve for NA to right in a dose-dependent manner , but shifted that for high potassium to left; it inhibited the transient contraction induced by NA and caffeine in the Ca 2+-free medium and the sustained contraction induced by NA after replenishing Ca 2+; Bre inhibited the i increased by NA, but enhanced that increased by high potassium in the smooth muscle cells of rabbit aorta. Conclusion Bre inhibits the contraction induced by NA through its inhibition effects on Ca 2+-influx and Ca 2+-release ; it enhances the Ca 2+-influx induced by high potassium, but the mechanism by which Bre enhances the high potassium inducing Ca 2+ -influx is not known.
6.A novel 99Tcm labelled polypeptide analogue for interleukin-11 receptor imaging in a bone metastases model for prostate cancer

Objective To assess the feasibility of a novel 99Tcm labelled polypeptide analogue for interleukin-11 receptor ( IL11R) imaging in a bone metastases model for prostate cancer. Methods A novel circular polypeptide analogue of IL11 ( c( CGRRAGGSC ) ) was indirectly labeled with 99Tcm and the product was named as 99Tcm-DTPA-IL11 RR. The labeling efficiency, radiochemical purity and stability of the product were measured with paper chromatography and high performance liquid chromatography (HPLC). The biodistribution of 99Tcm-DTPA-IL11RR was investigated in 28 ICR normal mice. The organ radioactivity was measured as percentage activity of injection dose per gram of tissue ( %ID/g). The models of bone metastases from prostate cancer were established at the tibias of BALB/c nude mice bearing human prostate cancer PC-3 cells. The tumor bearing ( n= 5 ) and standard closed fracture nude mice models underwent both 99Tcm-DTPA-IL11RR and 99Tcm-methylene diphosphonate (MDP) scintigraphy study. The images were acquired at 0.5, 1,2, 4, 6, 8, 24 h after intravenous injection of the tracers. The competitive inhibition imaging was perfomed in three tumor bearing mice. One-way variance analysis was used. Results The labeling efficiency was 90.7%. The radiochemical purity of 99Tcm-DTPA-IL11RR in normal saline solution was (99.57 ±0.09)%, (99.29 ±0.18)%, (98.95 ±0.78)%, (98.67 ±0.11)%, (96.53 ±0.91)%, (95.20±0.70)%, (88.38 ±0.22)% and (36.17±1.29)% at room temperature after0, 1,2, 3, 4, 6, 8 and 24 h, respectively. The tracer radiochemical purity in the blood of ICR mice remained over 90% at 37 C for 6 h. The labeling compounds were excreted mainly through kidney. The peak uptake of bone ( ( 1.910 ±0.109) %ID/g) and liver ( (0.366 ±0.030) %ID/g) was at 4 h after injection. In the tumor bearing mice, the uptake of spine marrow and large joints of extremities was mild. The highest uptake at tumor region was at 4 h and persistent at 6-8 h after injection. The tumor to non-tumor ratios (T/NT) were 1.17 ±0.17, 2.20 ±0.29, 3.20 ±0.15, 3.67 ±0.23, 13.61±0.85, 9.45 ±0.37 and 3.33 ±0.30 at 0.5,1, 2, 4, 6, 8 and 24 h, respectively (F=621.54, P<0.05). In the standard closed fracture models,high uptake of 99Tcm-MDP was shown at the fracture site, with no increased 99Tcm-DTPA-IL11RR uptake noted. The tumor uptake was significantly depressed after a pre-injection of the unlabeled polypeptide analogue. Conclusions The synthesis of 99Tcm-DTPA-IL11RR is stable and the labeling efficiency is high. It may be a potential molecular probe in metastatic bone imaging for prostate cancer.
7.Application of parametric image processing of contrast-enhanced ultrasound imaging in diagnosis of ovarian masses

ObjectiveTo investigate application value of parametric image processing in contrastenhanced ultrasound imaging in diagnosis of ovarian masses. MethodsFifty cases with ovarian masses underwent routine ultrasound and contrast-enhanced ultrasound imaging using a new dedicated parametric image processing software SonoLiver to analyze patterns of vascular formation and blood stream perfusion in the ovarian mass tissues on a digital video recorder in real time, compare their morphological characteristics of time-intensity curve (TIC) and dynamic vascular patterns (DVP) curve, and analyze quantitatively all indicators generated by SonoLiver.ResultsIn ultrasound imaging of the 50 cases, there were 24 cases (86%) displaying mainly blue lowly-enhanced imaging in those with benign masses and 15 cases (68%)displaying mainly red highly-enhanced imaging in those with malignant masses, with statistical significance (P <0. 01 ). There was significantly different characteristics of TIC and DVP between patients with benign and malignant masses. In 23 cases with benign masses, their DVP were significantly higher above the baseline than in those with malignant ones ( P = 0. 000), and in 15 cases with malignant lesions, their DVP were much shorter below the baseline than in those with benign ones, with statistical significance (P <0.05). The intensity of contrast medium, the time to reach its peak intensity and average transit time were all significantly higher in those with malignant masses than in those with benign ones (all P < 0. 05 ). But, no statistical difference in the time of initial increasing between the two groups was found (P > 0. 05). ConclusionsThere is significant difference in TIC and DVP of ultrasound imaging between benign and malignant masses, which if combined with contrast enhanced ultrasound parametric image processing can provide a more visualized quantitative information of benign and malignant ovarian masses with SonoLiver software.

Objective To explore protective effect of Heme oxygenase-1(HO-1) on irradiation-induced endothelial cell apoptosis.Methods Human endothelial cell line EA.hy926 were administered with or without HO-1 activator Copp and/or HO-1 inhibitor Znpp,respectively.Then,cells were treated with or without 8 Gy radiation.The HO-1 protein expression of cells were assessed with Western blotting and apoptosis of cells treated with irradiation were evaluated with flow cytometry.Moreover,cytochrome C releasing into cytosol were also determined by Western blotting. Results In PBS+R group,HO-1 protein expression of EA.hy926 was low posterior to irradiation.When cells were preconditioned with Copp and/or Znpp,then recieved with 8Gy irradiation,the HO-1 protein expression of EA.hy926 increased significantly in comparision with the PBS+R group(P
9.Repair of defects around the knee with reversed anterolateral thigh adipofascial flap

Objective To analyse clinical application of reversed anterolateral thigh adipefascial flap in repair of defects around the knee. Methods Reversed anterolateral thigh adipofascial flap was employed to repair defects of knee and upper shank in five patients from July 2006 to April 2007. The perforator arteries were detected and labelled with a hand held Doppler flowmeter before operation. The size of flaps containing perforator artery ranged from 6 cmx 8 cm to 12 cm×13 cm. After the descending branch was severed at the bifurcation of the lateral circumflex femoral artery, the flaps were elevated distal-ly to cover the defects of the knee and upper part of the shank. A split thickness graft was harvested to cover the flap. Results All the reversed anterolateral thigh adipofascial flaps survived well, with no complications occurred in donor site. Partial graft necrosis occurred in one patient and healed after conser-vative wound dressing change. Conclusion The reversed anterolateral thigh adipofascial flap is a good choice for repair of defects around the knee, for it can provide long thin vascular pedicle and maintain contour of the donor cite.
10.Effect of isoflurane on levels of PKA and PKC in hippocampus in rats

Objective To investigate the effect of isoflurane on the levels of protein kinase A (PKA) and protein kinase C (PKC) in hippocampus in rats. Methods Thirty-six 3-month-old male SD rats weighing 180-220 g were randomly divided into 3 groups ( n = 12 each): group Ⅰ underwent the cognitive function test without being pretreated with isoflurane inhalation (group C); group Ⅱ and Ⅲ inhaled 1.2% isoflurane for 4 h and underwent the cognitive function test 2 days and 2 weeks later respectively (group Ⅰso1,Iso2). Morris water maze was used to assess the cognitive function and the escape latency was recorded. The animals were killed immediately after the test.The hippocampus was isolated for determination of the expression and activities of PKA and PKC.Results The escape latency was significantly longer in group Ⅲ than in group Ⅰ.The expression of PKA and PKC was significantly down-regulated and the activities of PKA and PKC were significantly decreased in group Ⅱand Ⅲ as compared with group Ⅰ . There was no significant difference in the expression and activities of PKA and PKC between group Ⅱ and Ⅲ . Conclusion Four hour 1.2% isoflurane inhalation can decrease cognitive function by inhibiting the levels of PKA and PKC in hippocampus.
