1.Effects of 5-hydroxytryptamine on apoptosis of cultured human cytotrophoblast cells

Objective To investigate the effects of 5-hydroxytryptamine(5-HT) on apoptosis of cultured human cytotrophoblast cells and its possible mechanism involved in pathogenesis of pregnancy-induced hypertension syndrome.Methods Human cytotrophoblast cells from three term pregnancy(37-39 weeks) women were cultured and divided into four groups: normal control group,5-HT groups(0.1,1 and 10 ?mol?L~(-1)).The cells were cultivated for 3,6,12 and 24 h,respectively.Apoptotic index was measured by TdT-mediated biotinyated-dUTP nick end labeling(TUNEL) and morphological features of apoptotic cells were observed under electron microscope.Results When cells were cultivated for 3,6,12 and 24 h,compared with the normal control group(1.21?0.97,2.30?1.24,3.89?2.32 and 7.34?5.59,respectively),apoptotic indexes of cytotrophoblast cells in(10 ?mol?L~(-1)) 5-HT group were significantly increased(8.40?2.48,17.82?5.43,29.53?7.41,(45.56?)12.10,respectively)(P0.05).The significant morphological changes of apoptotic cytotrophoblast cells in various groups were found under electron microscope.Apoptotic cytotrophoblast cells were obviously compact and the chromatins were formed as mass and apoptotic bodies were also found under electron microscope.Conclusion 5-HT can promote the apoptosis of human cytotrophoblast cells and its mechanism may be correlated with pregnancy induced-hypertension syndrome.
2.Acupuncture for distal symmetric multiple peripheral neuropathy of diabetes mellitus: a randomized controlled trial.

OBJECTIVETo compare the clinical efficacy between acupuncture and intravenous administration of lipoic acid and alprostadil for distal symmetric multiple peripheral neuropathy of diabetes mellitus.
METHODSSixty patients were randomly divided into an acupuncture group (31 cases) and a medication group (29 cases). Patients in the two groups received basic treatment to control blood sugar within a safe range. On this basis, patients in the acupuncture group were treated with acupuncture at Geshu (BL 17), Weiwanxiashu (EX-B 3), Ganshu (BL 18), Pishu (BL 20), Shenshu (BL 23), Zusanli (ST 36), Taixi (KI 3), ashi points, etc. Patients in the medication group were treated with intravenous administration of lipoic acid (0.6 g) and alprostadil (10 mg). The treatment was given once a day, 10 days for a course of treatment; there was an interval of 2 days between courses, and totally 3 courses were given. The score of peripheral neuropathy of diabetes mellitus, the change of nerve conduction: velocity and clinical efficacy before and after treatment in the two groups were observed.
RESULTSAfter treatment, the score of peripheral neuropathy was significantly reduced in the two groups (both P < 0.05), which was more significant in the acupuncture group (P < 0.05). After treatment, the sensory nerve conduction velocity (SNCV) and motor nerve conduction velocity (MNCV) of median nerve and common peroneal nerve were significantly increased in the two groups (all P < 0.05). Compared between two groups, the SNCV and MNCV of common peroneal nerve in the acupuncture group were significantly superior to those in the medication group (both P < 0.05); the MNCV of median nerve in the acupuncture group was significantly superior to that in the medication group (P < 0.05); the SNCV of median nerve in the acupuncture group was not significantly different from that in the medication group (P > 0.05). The total effective rate was 83.9% (26/31) in the acupuncture group, which was significantly superior to 62.1% (18/29) in the medication group (P < 0.05).
CONCLUSIONAcupuncture and conventional medication both have satisfied effects for distal symmetric multiple peripheral neuropathy of diabetes mellitus, and acupuncture is superior to medication on improving clinical signs of sensory disorder, reflection disturbance and muscle weakness, nerve conduction and clinical curative effect.

3.Expression and significance of cyclinE in gastric cancer

Objective To investigate the expression and significance of cyclinE in gastric carcinoma. Methods We detected the expression of cyclinE in different pathologic types gastric cancer samples by immuno-histochemical staining technique (SP method). Results In 59 gastric cancer samples the positive rate of cyclinE expression was 55.93 %(33/59) and significantly higher than those of normal gastric mucosa 11.11 %(2/19) (P 0.05). Conclusions The high expression of cyclinE are associated with the progression of gastric cancer and probably related to the behavior of cellular biology.

Objective To investigate the action and the mechanisms of nerve growth factor(NGF) and its receptor(p75、TrkA) on the pathological alterations of the enteric nervous syst em in the rats cathartic colon. Methods The experimental animal model of cathartic colon was established by using rhubar b and phenolphthalein. Thirty Sprague Dawley (SD) rats were randomly divided int o the control group(10 rats) and cathartic colon model groups(10 rats with rhuba r b group; 10 rats with phenolphthalein group). The model group rats were fed with stimulant laxatives rhubarb and phenolphthalein respectively, and the control g roup rats were fed with distilled water. The rats were killed and the colon samp les were taken after 3 months. 5.Study of repair of bone defect using rabbit skeletal muscle stem cells transduced by AdrhBMP-2 gene

ObjectiveTo observe the effect of rabbit skeletal muscle stem cells (RSMSCs) modified by adenovirus mediated bone morphogenetic protein-2 (BMP-2) gene ex vivo in combination with demineralized bone matrix (DBM) on repair of longer bone defect in rabbit.MethodsModel of radial bone defects (20 mm) of rabbits was established. 50 rabbits were divided into 5 groups, group A (AdrhBMP-2 trusduced RSMSCs/DBM group), group B (adGFP trusduced RSMSCs/DBM group), group C (not trusduced RSMSCs/DBM group), group D (DBM group), and group E (untreated group). Roentgenographic, histologic, biomechanical, bone density of all animals were examined at the end of 4th and 6th week after surgery.ResultsAt 4th week, radial bone defects healed in group A. The healing rates from group A to group E were 100%, 50%, 33%, 0%, and 0% respectively at 6th week.ConclusionRSMSCs modified by AdrhBMP-2 ex vivo in combination with DBM can repair radial longer segemental defect in rabbit. It's possible to be used in the clinical treatment of longer segemental bone defect.
6.Introduction of traditional medicinal plants in Kyrgyzstan.

Kyrgyzstan is a mountainous country in the northeastern part of Central Asia which shares borders to the southeast with China. Due to their extreme environment and climate, there are a diverse range of species of plants. Many of the plants used in Kyrgyz folk medicine have not been studied using modern scientific techniques. This paper introduced the basic situation of medicinal herbs in Kyrgyzstan by comparing the differences traditional use between China and Kyrgyzstan, and looked for traditional medicinal plant research to provide basis for the development and cooperation of China and Kyrgyzstan.

7.Identification of bacteria and fungi by matrix-assisted laser desorption ionization-time of flight mass spectrometry

Objective To identify the pathogenic microorganism by MALDI-TOF MS.Methods A total of 560 strains were resuscitated,which included 260 gram-positive bacteria strains,180 gram-negative bacteria strains,60 yeast-like-fungi strains and 60 enteropathogenic bacteria strains.Comparing MALDI-TOF MS with Vitek2 Compact,the discordant results were validated by 16S rDNA sequencing.Results Comparing MALDI-TOF MS with Vitek2 Compact,the coincidence rate was 94.6% (246/260) for gram-positive bacteria,96.7% (174/180) for gram-negative bacteria,95% (57/60) for yeast-like-fungi,and 93.3%(56/60) for enteropathogenic bacteria Fifteen strains were validated by 16S rDNA gene sequencing.Comparing with sequencing,the coincidence rate of two methods was 66.7% ( 10/15 ) for MALDI-TOF MS and 26.7%(4/11 ) for Vitek2 Compact,respectively.Conclusion MALDI-TOF MS shows rapid turnaround time and modest reagent costs,and it will be another effective tool for microorganism diagnosis.
8.Polymorphic Analysis of DYS390 and DYS389Ⅰ/Ⅱ in Han Nationality in Liaoning Province in China

Objective: Our aim was to investigate the polymorphism and forensic meaning of the Y-chromosomal loci DYS390 and DYS389Ⅰ/Ⅱ in Han nationality in Liaoning Province. Methods: By amplifying these loci with polymorase chain reaction (PCR) followed by polyacrylamide gel electrophoresis and silver staining, the allele frequencies of these loci among 98 unrelated men of Han nationality were investigated. Results: Five alleles with different frequencies from 0.010 to 0.480 were detected of DYS390, and three alleles with different frequencies from 0.224 to 0.439 were detected of DYS 389Ⅰ. Locus DYS 389Ⅱ was detected in five alleles and the frequencies ranged from 0.020 to 0.367. The probability discrimination of these loci were 0.637, 0.644 and 0.723. These loci were found to generate at least 31 different haplotypes and the power of discrimination was 0.948. Conclusion: The loci DYS390 and DYS389Ⅰ/Ⅱ are very important when detect mixed samples, especially in deficiency cases of paternity test.
9.Experimental observation of electrical stimulation on cultured nervous system cells

ObjectiveTo explore the influence of electrical stimulation on nervous cells.MethodsOn 5th day the brain primitive nervous cells of rat brain, in vitro culturing, were divided into 4 groups, in order of control group, electrical pole control group, low stimulation group, and high stimulation group. The cells were stimulated with electro-acupuncture apparatus for 7 days, 30 minutes once, twice a day. On 10th day, the cells were observed microscopically for 3 days and taken photos.ResultsElectrical stimulation is helpful to raise the survival rate of cell.Conclusions Electrical stimulation influences morphological change of nervous cells during culturing.
10.Clinical pathological analysis of 11 cases of pancreatic solid-pseudopapillary tumors

Purpose To investigate the clinicopathlogic characteristic, diagnosis and histogenesis of pancreatic solid-pseudopapillary tumors ( SPPT) . Methods Combined with relevant literature, the clinical history, histopathological features and immunohistochemi-cal characteristics were analyzed in 11 cases of SPPT. Results There were 10 female patients and only 1 male in total 11 cases, aged from1 7 to 60 years (mean 33). The sizes of tumors were from 3. 2 to 10. 0 cm. Histologically, they were composed of papillary and microcysticsolid structures. Pseudopapillary with a fibrovascular core was remarkable. Immunohistochemically, the tumors expressed EMA (1/11), vimentin (10/11), NSE (11/11), Syn (7/11), CgA (1/11), CD56 (11/11), CD10 (11/11), PR (9/11), CD99 (9/11),α-AT (11/11),β-catenin (11/11), E-cadherin (11/11), Cyclin D1 (11/11), c-Myc (11/11). 6 patients were followed up for a period of 20 to 112 months, and they were all alive and had no recurrence and metastasis. Conclusions SPPT is a tumor with low malignancy of the pancreas that most frequently affect young females. SPPT may be derived from multipotent stem cells and closely related withβ-catenin signaling pathway. Pathological morphology and immunohistochemistry are very important to the diag-nosis and differential diagnosis of SPPT.
