2.Detection of rpsL drug-resistant gene in L-form of Mycobacterium tuberculosis by PCR-SSCP
Chinese Journal of Clinical Laboratory Science 2006;0(02):-
Objective To study the relationship between mutation of rpsL drug-resistant gene in L-forms of Mycobacterium tuberculosis and drug-resistance to streptomycin in pneumoconiosis patients. Methods A total of 52 clinical isolated strains of Mycobacterium tuberculosis L-forms were collected from 97 pneumoconiosis patients. The mutation of rpsL gene was detected by PCR-SSCP,and the drug-resistance to streptomycin was performed by routine antimicrobial susceptibility test(AST).Results The results of drug susceptibility test showed that 26 in the 52 clinical isolated strains were drug-resistant to streptomycin. The streptomycin-resistant rate was 50.00%(26/52).The gene mutation rate of rpsL detected by PCR-SSCP was 40.38%(21/52).The coincidence rate of two experimental results was 80.77%(21/26).Conclusion High detectable rate of streptomycin-resistant strains in Mycobacterium tuberculosis L-forms was found by PCR-SSCP.The application of PCR-SSCP may possess important value for guiding clinical medication of pneumoconiosis patients complicated with tuberculosis among coal workers

OBJECTIVEThis work lays the foundation for establishing a digital model database with normal occlusion. A digital dental cast is acquired through grating projection, and model features are measured through reverse engineering.
METHODSThe grating projection system controlled by a computer was projected onto the surface of a normal dental model. Three-dimensional contour data were obtained through multi-angle shooting. A three-dimensional model was constructed, and the model features were analyzed by using reverse engineering. The digital model was compared with the plaster model to determine the accuracy of the measurement system.
RESULTSThe structure of three-dimensional reconstruction model was clear. The digital models of two measurements exhibited no significant difference (P > 0.05). When digital and plaster models were measured, we found that the crown length and arch width were not statistically different (P > 0.05), whereas the difference between the crown width and arch length was statistically significant (P < 0.05).
CONCLUSIONThe reconstruction of a digital model by using the grating projection technique and reverse engineering can be used for dental model measurement in clinic al and scientific research and can provide a scientific method for establishing a digital model database with normal occlusion.

Objective To study the effectiveness and safety of combined laparoscopy and gonadotropin-releasing hormone agonist (GnRH-a) in the treatment of endometriosis (EM).Methods From January to December 2010,198 patients with EM undergoing treatment in Department of Obstetrics and Gynecology,Beijing Anzhen Hospital were randomly divided into three groups, which include 52 cases treated by only laparoscopy in laparoscopy group; 76 cases treated by laparoscopy combined with domestic Leuprolide acetate with dose of 3.75 mg every 28 days in Leuprolide acetate group; 70 cases treated by laparoscopy combined with imported Goserelin acetate with dose of 3.6 mg every 28 days in Goserelin acetate group. The efficacy, pregnancy rate and adverse reactions were compared among the three groups. Results Thirteen cases lost following up,including 3 cases in laparoscopy group,6 cases in Leuprolide acetate group and 4 cases in Goserelin acetate group.(1) Effective rates:effective rates were 47 % (23/49)in laparoscopy group,77% (54/70)in Leuprolide acetate group and 74% (49/66)in Goserelin acetate group.Compared with laparoscopy group, the effective rate of Leuprolide acetate group and Goserelin acetate group was significantly elevated (P < 0.05).There was no statistically significant difference between Leuprolide acetate group and Goserelin acetate group (P > 0.05).(2) Recurrence rate:recurrence rate were 33% (16/49) in laparoscopy group,13% (9/70)in Leuprolide acetate group and 12% (8/66)in Goserelin acetate group.Compared with laparoscopy group, the recurrence rate of Leuprolide acetate group and Goserelin acetate group was significantly declined (P < 0.05).There was no statistically significant difference between Leuprolide acetate group and Goserelin acetate group (P > 0.05).(3) Pregnancy rate:the number of patients require fertility were 28 cases in laparoscopy group,39 cases in Leuprolide acetate group and 35 cases in Goserelin acetate group. After 2 years follow up,pregnancy rate of 62% (24/39) in Leuprolide acetate group and 60% (21/35)in Goserelin acetate group were high than 39% (11/28)in laparoscopy group significantly, which did not reached significant difference (P > 0.05).(4) Adverse drug reaction:rates of a adverse reactions were 21% (15/70) in Leuprolide acetate group and 20% (13/66) in Goserelin acetate group, including irregular vaginal bleeding associated with low estrogen level. There was no significant difference in adverse reactions (P > 0.05).Conclusions Compared with laparoscopy alone, laparoscopy combined with GnRH-a is more effective in treatment of,which exhibit lower recurrence rate, higher pregnancy rate and fewer adverse reactions.Domestic Leuprolide acetate have similar safety and efficacy compared with imported GnRH-a.

Objective To discuss the relation between level of Hs-CRP and glycometabolic disturbance in patients with acute coronary syndrom. Methods 155 patients with acute coronary syndrome were divided into 3 groups according to the WHO standard of diabetes diagnosis, normal glucose tolerance group(group A,44 cases),impaired glucose tolerance group(group B,60 cases),diabetes group(group C,51 cases). All cases were checked by the 75g glucose tolerance test, blood Glucose, insulin, HbA1c,lipid,hypersensitive C reaction protein(hs-CRP). 95 patients were checked by coronary angiography(CAG). Statistic comparison were measured among three groups including the levels of hs-CRP,insulin,HbA1c,lipid and coronary artery lesions. Results The levels of Hs-CRP,PBG, FINS, PINS,HbA1c,TC,LDL-C in both group B and C were higher than those in group A;The levels of TG,PINS were not found significant differences between group B and C. The level of Hs-CRP was correlated with the levels of PBG,PINS,HbA1c,TC,LDL-C. The coronary artery lesions showed by CAG in group B and C were more severe than those in group A,coronary multivessel changes in group B and C were more frequent than in group A,it had no significant difference between group B and C.Conclusion Compared with the patients of normal glucose tolerance, patients with glycometabolic disturbance have higher levels of hs-CRP,their coronary artery lesions are more severe.

10.Use of Green Fluorescent Protein to Study the Relationship between Legionella pneumophila and Its Protozoan Host

Microbiology 1992;0(02):-
Protozoans, the natural host of the facultative intracellular pathogen Legionella species, play an important role in survival, proliferation, virulence and stress resistance of Legionella species. By repeating transformation and selection,a spontaneous mutant of plasmid over expressing green fluorescent protein was obtained. This mutant replicates and is maintained stably in Legionella cells. The colonies of L. pneumophila harbouring the mutated plasmid were intense green in colour even under the daylight. After feeding BF1 strain of Tetrahymena thermophila with transformed L. pneumophila, the intracellular dynamic of changing of bacterial shape, bacterial proliferation and lysis of the host cell due to the bacterial proliferation were observed clearly under fluorescent microscopy. Thus, the present paper provides a simple and intuitionistic strategy for investigating the ecological and cellular relationship between L. pneumophila and its host.
