1.The effect of curcumin on the hyperoxia-induced acute lung injury in neonatal rat

Objective To investigate the effect of curcumin on the hyperoxia-induced acute lung injury in neonatal rat.Methods A model of hyperoxia acute lung injury in neonatal rat was established by exposure to 95% O2 for 3 days.After the model was established,40 rats were randomly divided into the treated group ( n =20) and the control group ( n =20).The 20 injured rats which were treated by curcumin ( 100 mg/( kg· d)were treated group,and the other 20 injured rats were control group.The 20 normal rats were blank group ( n =20).Lung tissues were collected for doing pathological section and measuring the levels of tumor necrosis factor(TNF)-a,interleukin(IL)-1β and myeloperoxidase(MPO) after 4 d and 7 d.Results The gross and micrographic injury of lung was milder in treated group than that in control group.The levels of TNF-α,IL-1β and MPO in treated group were significantly lower than those in control group at 4 d and 7 d ( P <0.05 ).Compared with the blank group,the levels of TNF-α,IL-1β and MPO in control group increased significantly at 4 d and 7 d ( P < 0.01 ).Conclusion Curcumin can offer effective protection against acute lung injury induced by hyperoxia in neonatal rat.
2.Discussion on the effective approaches in cultivating the graduate students' innovations from their own viewpoints

Objective To analyze the viewpoints of the graduate students on the effective ap-proaches in cultivating their innovations. Methods Using Eugenie Creativity Test scale and self-made questionnalre, the graduate students in a medical university were surveyed on their general informa-tion, innovation levels and viewpoints of the important links and effective approaches in cultivating the innovations. The statistical software package-SPSS 16.0 was used for questionnalres. The statistical analysis was based on chi-square test for categorical data in the levels of graduate students' innovation and variance analysis for quantitative data in graduate students' understanding of the importance of cultivating approaches. Results 1 000 questionnalres were given out and effective questionnalre 936 copies were collected. The innovations of the students were malnly medium (557/936, 59.6%) and strong (356/936, 38.0%). They thought research and practice were the most important (F=225.49, P=0.00) in the tralning links and strengthening the tutorial staff was the foremost in the cultivating ap-proaches, which was followed by the sorted tralning (F=50.36, P=0.00). Conclusion The views of the graduate students on the cultivation jobs should be pald attention to. According to this study results, the management of research and practice, tutorial staff and assorted tralning should be fully considered to advance their innovations.
3.Study on the Chromatography Fingerprint of Radix Scrophulariae by RP-HPLC

Objective To establish the chromatography fingerprint of Radix Scrophulariae by RP-HPLC.Methods HPLC was applied in this study.Kromasil KR100-5C18(4.6 mm? 250 mm,5 ? m)column and DAD detector were used with a mixture of methanol and 0.1 % methanoic acid as mobile phase in a gradient mode.Results The chemical substances of Radix Scrophulariae were optimally separated.Conclusion This method is simple,accurate with good reproducibility,thereby can be used specifically for the quality control of Radix Scrophulariae.
4.Effects of miR-200a on the proliferation of lung cancer cells by inhibiting YAP1

Objective: To investigate the effects of miR-200a on the proliferation of lung cancer cells and to identify its direct target genes. Methods:Real-time PCR was performed to analyze the miR-200a expression in 15 paired clinical specimens of non-small cell lung cancer and adjacent noncancerous tissues, human lung cancer cell lines (A549, NCI-H520, and SK-MES-1), and one human normal lung bronchial epithelial cell line (16HBE). The effects of miR-200a on the proliferation of A549 lung cancer cells were detected through CCK-8 method. The candidate target genes of miR-200a were identified by bioinformatics screening and then verified by dual luciferase reporter gene assay, real-time PCR, and Western blot. The effects of YAP1 downregulation on the proliferation of A549 lung cancer cell line were also observed through CCK-8 method. Results:The miR-200a expression in non-small cell lung cancer tissues and lung cancer cell lines was significantly decreased (P<0.01). The upregulation of miR-200a expression could significantly inhibit the pro-liferation of A549 lung cancer cells (P<0.01). Dual luciferase reporter gene indicated that miR-200a could directly affect the 3′-untrans-lated region of the YAP1 gene to inhibit luciferase activity (P<0.01). Real-time PCR and Western blot revealed that the upregulation of miR-200a expression could significantly reduce the mRNA and protein expression levels of YAP1 in A549 lung cancer cells (P<0.01). CCK-8 method indicated that the downregulation of YAP1 could significantly prevent the proliferation of A549 lung cancer cells (P<0.01). Conclusion:MiR-200a inhibits the proliferation of lung cancer cells by targeting YAP1. Thus, miR-200a elicits tumor suppression effects.
5.MRI of acute experimental allergic encephalomyelitis: correlation with pathology

Objective To investigate the MR imaging findings of acute experimental allergic encephalomyelitis(EAE) in correlation with pathology. Methods An EAE model was induced by intradermal inoculation with guinea pig CNS homogenate in 6 female Lewis rats.Another 6 rats served as control.The clinical presentation and body weight of the animals were recorded daily. Routine MRI,Gd-enhanced MRI were performed when EAE animals showed the initial symptoms. Uhrasmall superparamagnetic iron oxide(USPIO) colloid solution was also administrated intravenously and MRI was performed again after 24 hours. The brain was removed instantly after the second MR imaging. The pathological exams including HE staining,myelin sheath staining and prussian staining were performed.The imaging findings were observed in correlation with pathological results. Results The EAE rats showed decrease of body weight on the 6th to 7th day after inoculation,and the clinical symptoms appeared on the 10th to 11th day after inoculation.Routine MRI did not show any definite abnormalities.The Gd-enhanced MRI found the diffuse thickening and enhancement of brain meninges.The USPIO-enhanced MRI showedareas of low signal intensity at white matter of medulla oblongata on T2WI,and high signal intensity was observed at the corresponding area on T1 WI. Gradient T2 * WI found more foci of low signal intensity in eerebellar white matter besides the lesions in the brain stem.The range of abnormal signal intensity was larger in animal with higher clinical scores than that with lower score.There were no abnormal findings in control animaL The pathological exam found "perivascular cuff" in the brain white matter in EAE animals,some accompanied with adjacent demyelinatian. The prussian staining found blue particles within the cytoplasm of the macrophages around the lesion,which corresponded to the area of low signal intensity on T2WI.Conclusion USPIO-enhanced MRI could reveal acute EAE lesions which were not capable of being shown on routine MRI and Gd-enhanced MRI.It can image the macrophages around the lesions in vivo.USPIO is important for future research and application in MS patients.
6.Opinions on graduate students' common compulsory courses reform of military medical universities

Objective To investigate graduate students' common compulsory courses settings of the Military Medical University and put forward suggestions.Methods With self-made questionnaires,we investigate with simple random sampling all the concerned principals of teaching about their opinions of English,politics and statistics courses.Results Most of the experts thought graduate students should not be required to study English but test should be required(68.9%),that politics course was more than required (76.0%),and the percentage of the experts who thought the contents in graduate and undergraduate courses were less overlapped and more overlapped is respectively 47.9% and 46.6%.Conclusion To satisfy scientific research needs of graduate students in military medical university,the common courses should be reformed,including teaching politics in various methods like lectures,taking software studying and experiment design into statistics,implementing the policy of students taking English test in the exemption of English study,etc.
7.Cleavage of Oncogene ki-rasG12V mRNA by Ribozyme and It' s Specificity

Objective: To design and confirm the cleavage activity of ribozyme Rz217 to oncogene ki-rasG12V messenger RNA and search for a new method for gene therapy targeting oncogene ki-ras. Methods: According to Symon' s principle,design an ribozyme specific for ki-rasc12v mRNA, both the constructs for transcription in vitro of ribozyme Rz217 and ki-ras exonl and the mammalian expression constructs of ribozyme Rz217 were constructed by DNA recombinant technique,ribozyme Rz217 and ki-ras exonl mRNA was obtained by transcription in vitro with T7 and SP6 RNA polymerase. Pancre atic carcinoma cell line PaTu8988 and human hepatocellular carcinomacell line BEL7404 were transfected with Rz217 mammalian expression constructs and the level of endogenous ki-rasG12V mRNA or ki-ras mRNA was determined by semiquantitative RT-PCR. Results: Not only in vitro but also in vivo, ribozyme Rz217 can cleave the mRNA of oncogene ki-ras (G12V) in site-specific manner and can not cleave the mRNA of wild-type ki-ras. Conclusion: Ribozyme Rz217 can cleave oncogene ki-rasc12v mRNA and the cleveage is specific for ki-rasG12V mRNA.
8.Survioin promoter activity in tumor cell lines

Objective To study the activity of the survivin gene promoter in several tumor cell lines and evaluate the possible application of this promoter in tumor gene therapy. Methods ①The expressions of survivin gene in A549,MDA-MB231 and HepG2 cell lines were detected by RT-PCR and Western blotting.②Tumor cells(A549,MDA-MB231,HepG2) were transiently transfected by reporter plasmids containing different length of survivin promoter using lipofectamine.And 48 h later,the level of reporter gene expression was analyzed.Results There were different levels of survivin expression in A549,MDA-MB231 and HepG2 cell lines.Transient transfection assay approved that pLuc-surP-987,pLuc-surP-596,pLuc-surP-269 and pLuc-surP-158 showed high activity and 269 bp survivin promoter demonstrated the highest activity. Conclusion In transcriptional level,survivin promoter can activate the reporter gene in several tumor cell lines.It is a potential candidate promoter in tumor gene therapy.
9.Effects of atorvastatin on liver cystathionine-?-synthase of apoE~(-/-) mice

Objective To explore the influence of homocysteine(Hcy)on liver cystathionine-?-synthase(CBS)and methylenetetrahydrofolate reductase(MTHFR)system in apoE-/- mice,and determine the effects of atorvastatin and/or folate/vitamin B12 on liver CBS and MTHFR system.Methods Eighty male 6-week-old apoE-/- mice were randomly divided into two groups:65 mice were fed with a chow diet containing 2%(wt/vol)L-methionine(homomethionine group)and 15 mice were fed with normal saline(control group).Two months later,the 60 mice survived in homomethionine group were subdivided into four groups:group Ⅰ(untreated),Ⅱ(3 mg/kg atorvastatin),Ⅲ(3 mg/kg atorvastatin+2 mg/kg folate+30 ?g/kg vitamin B12)and Ⅳ(2 mg/kg folate+30 ?g/kg vitamin B12).After one month,Western blotting was performed to detect the liver CBS and MTHFR system protein expression in each group.Results The relative expression of liver CBS and MTHFR was significantly lower in group Ⅰ than in control group(P
10.Analysis of Effects of Different Doses of Rosuvastatin Calcium on Serum Lipids in Elderly Patients with Coronary Heart Disease and Hyperlipidemia

Objective:To investigate the effects of different doses of rosuvastatin on lipid levels in elderly patients with CHD complicated with hyperlipidemia.Methods:120 elderly patients with coronary heart disease admitted in our hospital were retrospectively chosen.By lipid examination,they were all hyperlipidemia.According to the dose of rosuvastatin,they were divided into three groups,and 40 cases in each group.They were given 5 mg,10 mg and 20 mg treatment,respectively.After 2 months,the changes of blood lipid level and the clinical effective rate were compared before and after the treatment.Results:①The blood lipid levels HDL-C [(1.17 ± 0.62) mmol / L] was significantly higher than the other two groups.LDL-C [(3.67 ± 0.83) mmol / L],TC [(4.36 ± 0.96) mmol / L] and TG [(1.68 ± 0.94) mmol / L] were significantly lower than those of the other two groups (P <0.05).②The effective rate of treatment in the 20mg group (97.5%) was significantly higher than that in the 5mg group (87.5%) and the 10mg group(85.0%).Conclusions:Rosuvastatin can treat coronary heart disease complicated with hyperlipidemia in elderly patients.The dosage of 20 mg can improve the blood lipid level of the patients,is an effective and convenient method,and with high therapeutic efficiency.
