1.Effects of Danhong Injection Combined with Low Molecular Heparin on Clinical Symptoms,Serum In-flammatory Factors and Lung Function of Elderly Patients with COPD

OBJECTIVE:To investigate the effects of Danhong injection combined with low molecular weight heparin (LM-WH) on clinical symptoms,serum inflammatory factors and lung function in elderly patients with chronic obstructive pulmonary disease (COPD). METHODS:A total of 146 elderly patients with COPD were randomly divided into observation group and con-trol group,with 73 cases in each group. Both groups received conventional treatment. On this basis,control group was given Dan-hong injection 30 mL,intravenous drip,once a day. Observation group was additionally given LMWH sodium injection 5000 IU, subcutaneously,twice a day,on the basis of control group. Both groups were treated for 14 d. Clinical efficacies of 2 groups were observed and compared. The clinical symptom score,the levels of IL-6,IL-8,TNF-α,FEV1,FEV1/FVC,pa(O2),pa(CO2) and the occurrence of ADR were compared before and after treatment. RESULTS:After treatment,the total response rate of observa-tion group was significantly higher than that of control group(93.15%vs. 78.08%,P<0.05). Before treatment,there was no statis-tical significance between above indexes (P>0.05). After treatment,individual clinical symptom score,total clinical symptom score,the levels of IL-6,IL-8,TNF-α,pa(CO2)in 2 groups were significantly lower than before;the observation group was signif-icantly lower than the control group;FEV1,FEVl/FVC,pa(O2) were significantly higher than before,and the observation group was significantly higher than the control group,with statistical significance (P<0.05). CONCLUSIONS:Based on conventional treatment,Danhong injection combined with LMWH is effective in the treatment of senile COPD,and can significantly improve the clinical symptoms,serum inflammatory factors levels and pulmonary function with good safety.

6.Studies on Effective Composition of Pinecone V.Determination of Lignin in Polysaccharides of Cone of Pinus koraiensis by TLC-UV method

A TLC-UV method for the determination of lignin in polysaccharides of cone of Pinus koraiensis was reported. Active lignin was obtained by acid hydrolysis,and developed on GF-254 TLC-Plate by mixed Solventof dich1oro methane-ethylether. UV absorbance of the eluate was determined at 23Onm. The concentration oflignin was linearly related to absorbance within the range of 2~70pg/ml. The recovery was 100. 2% ~ 101.4% and RSD was 6. 5 %.
7.Study on the amplifying and typing of low copy number DNA

Objective To study the influence on STR typing for low copy number(LCN)DNA using different methods of amplifcation and detection.Methods Control DNA 9947A was diluted and then amplifled with Identifiler~(TM) or DNATyper15~(TM).The heat cycles were set to 28 or 28 add 6.Each template wag amplified three times in parallel,and then the amplified products or the product mixture of three amplifictions were deteced with 310 or 3130 Genetic Analyzer.Results The success rate of STR typing with the method 28+6 cycles was higher than that of 28 cycles.There are no correlation between the allele imbalance or allele dropout and STR locus.With the reduction in the amount of template DNA,allele imbalance and dropout gradually increased,and the allele dropout was more common than allele imbalance when the amount of template DNA Wag very small.When the product mixture of three amplifictions were deteced.the occurrence of allele imbalance and dropout reduce obviously.Conclusion The success rate of STR typing of LCN DNA can be obviously increased by detecting the product mixture of three amplifictions in parallel combined with the 28+6 heat cycle condition.
8.Effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells

Objective To investigate the effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells.Methods From March 2014 to February 2015,breast cancer MCF-7 cells were treated with 99% pure DMY as an inhibitor.MTT assay,flow cytometry and immunocytochemistry were used to analyze the proliferation,apoptosis and protein expression of breast cancer cell MCF-7.Results When the DMY concentration was higher than 20 μg/mL,the inhibitory effect appeared,but not good.When 40 and 80 μg/mL DMY were used,the proliferation of MCF-6 cells were significantly inhibited,and have different degrees of sensitivity to it.When DMY was 80 μg/mL,the IC50 was 226.9 μg/mL.The inhibition rate and IC50 were compared with 0 μg/mL DMY,there was significant difference(P<0.05).Indicating that the relatively high concentration of DMY could significantly improve the patient′s condition of breast cancer;MCF-6 cells treated with 40 and 80 μg/mL DMY could induce G2/M phase arrest in DMY with concentration greater than 20 μg/mL(P<0.01),and showed significant cell apoptosis death phenomenon.At the same time,G0/G1 phase was also blocked and S phase cells decreased significantly,the difference was significant(P<0.05).The positive rate of PCNA in breast cancer MCF-6 cells significantly decreased(P<0.01) when the DMY concentration was higher than 50 μg/mL,the positive rate was> 50%,especially in DMY with 80 μg/mL,the positive rate was 10.00%.Compared with 0 μg/mL DMY,the difference was significant(P<0.05).Conclusion The use of dihydromyricetin in breast cancer patients can effectively inhibit the rapid increase of cancer cells,accelerate apoptosis,slow down the patient′s condition,the effect is outstanding.
9.Study on chemical constituents in stems of Nelumbo nucifera by UPLC-ESI/Q-TOF-MS/MS.

This paper employed UPLC-Electrospray Ionization /Quadrupole-Time of Flight-Mass /Mass Spectrometry( UPLC-ESI/Q-TOF-MS/MS) to analyze the chemical constituents in the stems of Nelumbo nucifera. The stems of N. nucifera were extracted with 75% methanol, and we applied an Agilent Zorbax SB-Aq column (2.1 mm x 100 mm, 1.8 μm) to UPLC analysis with water methanol-water( containing 0.05% formic acid) in gradient as mobile phase. The eluates were then detected by ESI-Q-TOF-MS/MS. Results indicated that 22 benzylisoquinoline alkaloids were indendified. Among them, one alkaloid may be a new compound and a component was found in the Lotus for the first time. We fully identify the composition of the Lotus stems for the first time, Which could provides theoretical foundation for further study and utilization of the medicinal resources.

10.Astrocyte Proliferation and Glial Scar Formation after Spinal Cord Injury (review)

The nature and functions of astrocytes (AS), the pathological changes and roles of AS after spinal cord injury, the experimental methods of inhibiting AS proliferation and glial scar formation, and so on, were reviewed. AS beneficially affects repairing injured spinal cord at different periods of differentiation. However, adult AS secreting the factors formed the chemical glial barrier which severely affects nerve regeneration and hinders axon extension. Because of the static, activated and proliferative AS tending to coexist after spinal cord injury, and complex factors of barrier formation, the current approach taken by a single method is difficult to effectively control the AS proliferation and glial scar formation.
