ObjectiveTo observe the improve effect of Yiqihuoxue Chinese materia medica for the ethology of mouse with diabetes mellitus encephalopathy induced by streptozotocin (STZ).MethodsCreating mouse model using 1% STZ intraperitoneal injection. 50 mouses were randomly divided into the normal control group, model control group, large dosis Chinese materia medica group, middle dosis Chinese materia medica group and small dosis Chinese materia medica group. Mouses'behaviors of evading dark reaction and Morris water maze in different groups were tested.ResultsYiqihuoxue Chinese materia medica had not the fuction of lower blood suga to the mouse with diabetes mellitus encephalopathy induced by STZ. But Yiqihuoxue chinese materia medica could improve the ethology show and mental status of mouse with diabetes mellitus encephalopathy induced by STZ obviosly, decrease the lightning stroke frequency when the diabetes mellitus encephalopathy mouses entered the dark chamber, extend the latency period, shorten the diabetes mellitus encephalopathy mouses swimming out time and distance, improve the study and memory ability.ConclusionYiqihuoxue Chinese materia medica had the improve effect for the ethology of mouse with diabetes mellitus encephalopathy induced by STZ.
2.Magnetic compression anastomosis: a promising technique in the further development of minimal invasive surgery

Magnetic compression anastomosis (MCA)involves the use of two rare-earth magnets that attract to each other transmurally between two internal organs, resulting in compression and subsequent fistula formation to create a nonsurgical enteric, vascular, or biliary anastomosis with therapeutic aims. The magnetic vascular coupler allows a sutureless anastomoses between arteries and veins by using interventional radiological techniques. The MCA technique significantly reduces ischemic time during anastomosis, and has the advantages of low invasiveness and simplicity. It is a rather effective method for recanalizing between various hollow viscera, creating anastomosis such as gastrojejunostomy, choledochojejunostomy, or choledochoduodenostomy.Furthermore, this novel technique is considered to be a feasible and alternative reconstructive method for patients who develop significant bile duct complications following hepaticojejunostomy and living donor liver transplantation. MCA is a promising and novel technique in the further development of minimal invasive surgery.
3.Anginogenesis of eutopic and ectopic endometria in endometriosis.

In order to study the angiogenesis in endometriosis, the samples of eutopic and ectopic endometria from patients with endometriosis were quantitatively analyzed by color morphometric image system (CMIS) for vascular surface area, and by examining endometrial blood vessel for microvessel density (MVD). The results showed that within each menstrual phase the vascular surface area and MVD were significantly higher in ectopic endometria with endometriosis than those in eutopic endometria with endometriosis or normal endometrium (P < 0.05). It is concluded that angiogenesis might be involved in the development of endometriosis.

4.Effect of Acupuncture on Spatial Learning and Memory Abilities and Hippocampal CA3 Regional BDNF in Rats with Post-stroke Depression

Objective To investigate the effects of acupuncture at points Baihui (GV20), Yintang (Ex-HN3) and Sishencong (Ex-HN1) on spatial learning and memory abilities and hippocampal expression of brain-derived neurotrophic factor (BDNF) in a rat model of post-stroke depression (PSD). Method Thirty male adult SD rats were allocated, using a random number table, tocontrol, acupuncture and sham operation groups, 10 rats each. A rat model of PSD was made by classical middle cerebral artery occlusion (MCAO) using an intraluminal thread technique, chronic unpredictable mild stress (CUMS) and solitary feeding. At the fourteenth day after the beginning of stress, the acupuncture group received acupuncture intervention for 14 consecutive days. At one week after the end of stress, the Morris water maze test was conducted to assess rat spatial learning and memory abilities. After the completion of the water maze test, hippocampal CA3 regional BDNF expression was determined by immunohistochemical method. Result Rat learning and memory abilities decreased significantly in the control group compared with the sham operation group (P<0.05). Space exploration time and the number of crossing platform reflecting memory abilities decreased significantly in the control group (22.36±6.20 s and 3.72±0.90 times, respectively) compared with the sham operation group (48.73±5.40 s and 8.60±1.10 times, respectively); there were statistically significant differences (P<0.01). Learning abilities, space exploration time, the number of crossing platform and BDNF expression increased significantly in PSD rats treated with acupuncture compared with the control group; there were statistically significant differences (P<0.01). Conclusion Acupuncture can improve spatial learning and memory abilities and raise hippocampal regional BDNF levels in rats with post-stroke depression.
5.Effect of MACC1 down-regulation on proliferation of gastric adenocarcinoma cell line

Objective To study the effects of MACC1 down-regulation on the growth of gastric adenocarcinoma cells. Methods siRNA (MACC1-siRNA1 and MACC1-siRNA2) that can transiently silenced MACC1 was designed, syn?thesized and transfected into MGC-803 cells by lipofectamine 2000. Non-specific siRNA was transfected to be used as nega?tive control. The efficiency of MACC1 depletion was determined by Real-time quantitative PCR. MTT, colony formation and flow cytometry assay were performed to examine cell proliferation. The expressions of MACC1, P21,CDK4, CCND1 and c-myc were determined by Western blot. Results Compared with cells in negative control group, transiently silencing MACC1 decreased the expression of MACC1 in MGC-803 cells shown by Real-time PCR. MACC1 downregulation drastical?ly changed the proliferation, colony formation and cell cycle of gastric adenocarcinoma cells in vitro ( P<0.05). The expres?sions of MACC1 , CDK4, CCND1 and c-myc proteins in cells of MACC1 silence group were much lower while P21 expres?sion level was much higher than those in negative control. Conclusion Down-regulation of MACC1 result in blocking cell cycle, inhibiting proliferation of MGC-803 cells. So it may serve as a promising target in the treatment of gastric cancer.
6.Clinical Study of Western Medicine Combined with Traditional Chinese Medicine on Aged Diabetic Mellitus

Objective To study the clinical effect of herbs of replenishing qi, nourishing yin and promoting blood combined with western medicine on symptoms and hemorrheology indexes of aged diabetic mellitus (DM). Methods Sixty cases with DM-2 were randomly divided into two groups. The treatment group was given herbs combined with western medicine and the control group was given western medicine treatment only. The treatment course was 1 month. The clinical symptoms, blood glucose and hemorrheology indexes were observed in both groups. Results There were 11 marked effective cases, 15 effective cases and 4 ineffective cases in treatment group, while 2, 12 and 16 cases in control group. The total effective rate was 86.7% in treatment group and 46.7% in control group. There was a significant difference between the two groups. The whole blood viscosity, platelet aggregation, D-dimer and fingernail microcirculation were significantly improved in the treatment group. Conclusion The herbs of replenishing qi, nourishing yin and promoting blood combined with western medicine can improve the clinical symptoms and hemorrheology of aged DM.
7.Efficient amplification of NK cells and their anti-tumor cytotoxic activity in vitro

Objective:To efficiently amplify NK cells and determine their cytotoxic activity against a variety of tumor cell lines in vitro,thereby providing evidence for potential clinical application.Methods: PBMCs were isolated from adult peripheral blood and co-cultured with K562 cells that were genetically modified to express 4-1BBL,IL-15 and IL-21 on the surface for 15 days to effectively amplify NK cells.The total cell number and Purity of CD3-CD56+ cells were measured.Granzyme B and perforin expression of the amplified NK cells were detected by flow cytometry and real-time PCR.The anti-tumor effect on different cancer cells was evaluated.Results: This method obtained a more than 1.1×1010 CD3-CD56+ NK cells with 95% purity over a 15 day amplification procedure.The expanded NK cells could efficiently release granzyme B and Perforin.The cytotoxicity against different tumor cells was followed the order from strong to weak:gastric,pancreatic,cervical,ovarian and renal cancer cells,with the highest activity against gastric cancer cell line A549 (90% at E∶T=10∶1) (P<0.05).A time-dependent killing effect of activated NK cells on cervical,liver and pancreatic cancer cells was observed.Conclusion: This amplification procedure can consistently generate large amounts of pure NK cells with effective cytotoxic function against a variety of tumor cells.
8.The treatment of neodymium-ythrium aluminum garnet vitreolysis for anterior vitreous opacity after implantation of intraocular lens

ObjectiveTo observe the effect of the treatment of neodymium-yttrium aluminum garne (Nd:YAG) vitreolysis for the anterior vitreous opacity after implantation of intraocular lens.MethodsForty-nine eyes of 47 patients with the anterior vitreous opacity after implantation of intraocular lens received the slit lamp examination, optical coherence tomography (OCT), and B-scan. The anterior vitreolysis and posterior capsulotomy were performed simultaneously with Nd:YAG laser. The outcomes of visual acuitiy changes and complications were studied. ResultsIn 49 eyes, 46 had anterior vitreous opacity associated with posterior capsule opacification, and the other 3 without obvious posterior capsule opacification. In all patients, the visual acuity improved significantly without any complications after the laser procedure (t=32.50, P=0.007). After Nd:YAG laser treatmen, transparent area was found in anterior opaque vitreous in 21 eyes (42.86%) within 15 minutes, and in 47 eyes (95.92%) within 24 hours. No complication occured in or after the operation. ConclusionsIn the patients with visual deterioration after implantation of intraocular lens, the prescence of anterior vitreous opacity should be concerned. Opening the opaque anterior vitreous with Nd:YAG vitreolysis is effective for the patients with the anterior vitreous opacity after implantation of intraocular lens.
9.Retrospective Study on the Protocol Deviation in Drug Clinical Trial Implementation of Our Hospital

OBJECTIVE:To study the influencing factors of protocol deviation in drug clinical trial implementation,and pro-vide reference for improving the quality of drug clinical trial. METHODS:Quality verification was conducted for the drug clinical trial projects in the First Affiliated Hospital of Chongqing Medical University during 2010-2016,and protocol deviations in each year were retrospectively studied,classified and analyzed. Category,frequency,international and domestic pilot projects and the differences of protocol deviation after full-time research nurse participating in trail management were explored,and the measures were put forward. RESULTS:27 drug clinical trials were implemented in our hospital during 2010-2016,including 949 cases,176 cases with protocol deviation,accounting for 18.55%. Deviation protocol in drug clinical trial was decreased year by year for 7 years. The categories were mainly incompleteness of observation/records (30.11%),checking omission/broaden the window(28.41%),adverse drug events and improper combined medication (14.20%) and omission in drug management (11.93%). The proportion of protocol deviation with full-time research nurse participated was lower than the projects without full-time research nurse(11.11% vs. 28.67%,P<0.01),and proportion of deviation protocol in international multi-center project was lower than the domestic projects(6.60% vs. 28.84%,P<0.01). CONCLUSIONS:It is suggested to pointedly strengthen the weak links of drug clinical trial. For example,clinical trial institutions should establish the clinical trial data retention system,electronic data should be timely backed up in a different places,etc. Besides,clinical trial institutions should equip professional full-time research nurses as much as possible,learn the rigorous scientific experimental design,standard operational procedures and the authenticity of data pro-cessing from the international multi-center clinical trial projects to effectively reduce the incidence of deviation protocol and im-prove the quality of drug clinical trials.
10.Prostate cancer: diagnostic value of quantitative analysis by dynamic contrast-enhanced MR at 3.0 T

Objective To investigate the diagnostic value of the quantitative analysis parameters of dynamic contrast-enhanced MRI (DCE-MRI) for prostate cancer.Methods MR examination were performed in 26 patients and correlations were made with surgical pathology following radical prostatectomy.According to the localization of pathologic specimens,ROI were drawn on areas of cancerous regions,noncancerous regions in peripheral zone and central gland to measure the values of Ktrans,kep and Ve.The values of the three parameters in different tissue were compared with Kruskal-Wallis H and Mann-Whitney U.Results Twenty six patients had prostate cancer confirmed by pathology.Data from 468 assessable regions of prostate were acquired,including 115 cancerous regions and 142 noncancerous regions in central gland,68 cancerous regions and 143 noncancerous regions in peripheral zone.Prostate cancer showed a multifocal distribution.The K,kep and Ve values were (1.04 ± 0.26)/min,(1.43 ± 0.46)/min,(0.76 ±0.12) respectively for cancerous regions in central gland,and (0.82 ±0.19)/min,(1.12 ±0.26)/min,(0.75 ± 0.14) respectively for noncancerous regions in central gland,(0.95 ±0.31)/min,(1.31 ±0.51)/min,(0.76 ± 0.13) respectively for cancerous regions in peripheral zone and (0.32 ± 0.07)/min,(0.52 ± 0.13)/min,(0.64 ± 0.14) respectively for noncancerous regions in peripheral zone.The differences among the three parameters were statistically significant (x2 =316.008,297.985,63.681,P <0.01).The Ktrans,kep values of cancerous regions were significantly higher than the corresponding values of noncancerous regions,respectively.Conclusion Quantitative analysis parameters of DCE-MRI contribute to the diagnosis of prostate cancer,including cancer located in central gland.
