1.Relationship between gut microbiota and Kawasaki disease

Kawasaki disease is a disease based on systemic vasculitis, which has become the most common disease in children with acquired heart disease, but its etiology and pathogenesis remain unclear.The homeostasis of the gut microbiota plays an important role in the maturation of the body′s immune system and immune regulation.The disorder of the gut microbiota is involved in a variety of inflammatory and immune diseases in children.Studies have found that there are changes in the composition of gut microbiota in children with Kawasaki disease.Gut microbiota disorders are related to the occurrence and development of Kawasaki disease, with the immune system function affected.Various mechanisms such as intestinal mucosal barrier dysfunction, immune inflammation, oxidative stress and metabolite effects promote the pathogenesis of Kawasaki disease.This article reviews the research on the relationship between gut microbiota and Kawasaki disease.

3.Periplocin of cortex periplocae inhibits Stat3 signaling and induces apoptosis in human hepatocellular carcinoma cell line SMMC-7721

Objective To investigate the effect of periplocin of cortex periplocae (CPP) on Stat3 signaling and its probable molecular mechanism of inducing apoptosis and anti-tumor activity. Methods Cell proliferation was detected by MTT. Cell apoptosis and cell cycle were investigated by flow cytometry. Expression of Stat3 protein in SMMC-7721 cells was analyzed by Western blot. Mcl-1, Survivin and XIAP mRNA expressions were measured by RT-PCR. Results CPP inhibited the proliferation of SMMC-7721 cells significantly, induced their apoptosis and arrested their cell cycle at G2/M phase. Decreased expression of Stat3 protein in the cell nucleus was observed after CPP treatment, but no significant changes were found in cytoplasma. Mcl-1, Survivin and XIAP mRNA expression levels were decreased in a time-dependent manner. Conclusion CPP inhibits cell proliferation and induces apoptosis by inhibiting Stat3 signal transduction in human hepatocellular carcinoma cell line SMMC-7721.

5.A pilot study to evaluate the invasiveness of mandible ameloblastoma on ultrasonography

Objective To evaluate and demonstrate ultrasonographic characteristics of mandibular ameloblastoma with active proliferation. Methods Nineteen subjects with ameloblastomas in the mandibles were examined with ultrasonography. In addition to sizes,internal echoes,the research was focused on blood flow of the tumors and destruction of mandibles. Ultrasonographic appearances of the tumors were compared with histopathological findings. Sensitivity and specificity of Doppler flow signals and destruction of mandibles for prediction of active tumor proliferations were calculated. Results Fifteen cases (79%)showed no or minimal flow signals on color Doppler flow imaging(CDFI), whereas the remaining 4 lesions demonstrated abundant flow signals,in which 3 cases were confirmed histopathologically with active tumor proliferations. The sensitivity and specificity of the Doppler flow signals for prediction of active tumor proliferations were 100% and 94%, respectively. Fourteen cases (74%) showed the mandibular cortices were thin but continuous,among the remaining 5 lesions showed local cortices destruction,3 of them were confirmed histopathologically with active proliferations. The sensitivity and specificity of the destruction of cortical bone for prediction of active tumor proliferations were 100% and 88 %, respectively. Conclusions Ultrasonography can be used as a new effective supplementary diagnostic method for mandibular ameloblastomas. CDFI of tumor vascularity and destruction of cortical bone could be used to predict active tumor proliferations and invasiveness.

0 05); The protein expression of erbB2 in carcinomas of palate and floor of mouth were obviously higher than the expression in lip cancer and gingival carcinoma( P 0 05).Conclusion The amplification and overexpression of c erbB2 and p53 play important roles in the formation and development of OSCC, the overexpression of c erbB2 protein has more important significance in tumorigenesis of carcinomas of palate and floor of mouth; Not only transcriptional regulation but also translational modulation exists on the expression of c erbB2 and p53. 8.Sodium nitrite enhanced the potentials of migration and invasion of human hepatocellular carcinoma SMMC-7721 cells through induction of mitophagy.

Nitrites play multiple characteristic functions in invasion and metastasis of hepatic cancer cells, but the exact mechanism is not yet known. Cancer cells can maintain the malignant characteristics via clearance of excess mitochondria by mitophagy. The purpose of this article was to determine the roles of nitrite, reactive oxygen species (ROS) and hypoxia inducing factor 1 alpha (HIF-1 α) in mitophagy of hepatic cancer cells. After exposure of human hepatocellular carcinoma SMMC-7721 cells to a serial concentrations of sodium nitrite for 24 h under normal oxygen, the maximal cell vitality was increased by 16 mg x (-1) sodium nitrite. In addition, the potentials of migration and invasion for SMMC-7721 cells were increased significantly at the same time. Furthermore, sodium nitrite exposure displayed an increase of stress fibers, lamellipodum and perinuclear mitochondrial distribution by cell staining with Actin-Tracker Green and Mito-Tracker Red, which was reversed by N-acetylcysteine (NAC, a reactive oxygen scavenger). DCFH-DA staining with fluorescent microscopy showed that the intracellular level of ROS concentration was increased by the sodium nitrite treatment. LC3 immunostaining and Western blot results showed that sodium nitrite enhanced cell autophagy flux. Under the transmission electron microscopy (TEM), more autolysosomes formed after sodium nitrite treatment and NAC could prevent autophagosome degradation. RT-PCR results indicated that the expression levels of COX I and COXIV mRNA were decreased significantly after sodium nitrite treatment. Meanwhile, laser scanning confocal microscopy showed that sodium nitrite significantly reduced mitochondrial mass detected by Mito-Tracker Green staining. The expression levels of HIF-1α, Beclin-1 and Bnip3 (mitophagy marker molecular) increased remarkably after sodium nitrite treatment, which were reversed by NAC. Our results demonstrated that sodium nitrite (16 mg x L(-1)) increased the potentials of invasion and migration of hepatic cancer SMMC-7721 cells through induction of ROS and HIF-1α mediated mitophagy.

10.Origin and trend of the upper eyelid artery in the eyelid region of a skull model after blood reperfusion: an anatomical observation

BACKGROUND: Upper eyelid flap grafting-related vessels such as superficial temporal artery, supratrochlear artery, supraorbital artery trunk are reported. Upper eyelid artery dissection is becoming more and more important for the surgery on the eyelid, but there is a lack of anatomical analysis of upper eyelid artery. OBJECTIVE: To measure the anatomical position of the upper eyelid artery in the eyelid region, and to provide anatomical basis for adjacent flap grafting. METHODS: Twenty adult skull specimens were dissected, and a reference coordinate system was made based on the inner canthus connection for the X axis, and the center line for the Y axis. The red lactoprene was injected into the skull model via common carotid artery.The locations A-E of the upper eyelid artery in the eyelid area were measured. RESULTS AND CONCLUSION: The upper eyelid artery in the eyelid area was mainly from the supratrochlear artery and the supraorbital artery, generally paralleling to the X axis. The upper eyelid branch originated from the supratrochlear artery was located at the projection of the inner canthus, with a total length of 24.50 mm, and a diameter of 0.51 mm, extended to the outer canthus and the diameter of the vessel gradually reduced. The upper eyelid branch originated from the supraorbital artery was located at pupil and inner canthus junction 1/2 projection. The total length of the blood vessels was about 23-24.6 mm, and the diameter of the blood vessels was (0.55±0.05) mm. In the current study, we obtained the surface projecting of upper eyelid artery in the eyelid area by establishing the skull model of blood perfusion, which provides an anatomic basis for upper eyelid flap grafting.
