1.PD-L1 blockade ameliorates suppression of tolerogenic dendritic cells on immunoactivity of T lymphocytes in spleen of zymosan-injured mice

Purpose To investigate the effect of tolerogenic dendritic cells ( DC) on T lymphocytes in the spleen during the develop-ment of zymosan-induced sepsis in mice, and to explore whether PD-L1 blockade could alleviate the immunosuppressive effect of tolero-genic DC on T lymphocytes. Methods Mice sepsis model was established by intraperitoneal injection of zymosan. Splenic DC and T lymphocytes were isolated respectively by using anti-CD11c and anti-CD3 magnetic beads. The expressions of PD-L1, PD-1 and PIR-B on splenic DC were measured, and IL-12 and IL-10 secreted from DC were determined. Mitogen-induced T lymphocyte proliferation and IL-2 secretion were assessed. Anti- PD-L1 antibody was added into mixed culture of tolerogenic DCs with normal Tcells. T cell proliferation and IL-2, IL-12 and IL-10 concentrations in the supernatant of mixed culture were determined. Results At 5 days and 12 days after zymosan injection, the expressions of PD-L1, PD-1 and PIR-B on splenic DC increased greatly, secretion of IL-12p70 re-duced and that of IL-12p40 and IL-10 augmented in DC, which were associated with decrease of T cells proliferation and IL-2 secre-tion. Administrating anti-PD-L1 antibody into the mixed culture of tolerogenic DC and Tcell could alleviate the suppression of DC on T lymphocyte proliferation and secretion of IL-2, and ameliorate the ability of DC secreting IL-12 and IL-10 as well. Conclusions At late stage of zymosan-induced sepsis, the formation of splenic tolerogenic DC resulted in immunosuppression of T lymphocytes. Anti-PD-L1 antibody could improve the immunoactivity of DC and T lymphocyte through intervening PD-L1/PD-1 pathway.
2.A comparative study on rabbit bone mesenchymal stem cells and human amniotic epithelial cells transplantation for rabbit limbal stem cell deficiency

BackgroundLimbal stem cell deficiency usually leads to blindness, and traditional therapy is limited. Recent research demonstrated that bone mesenchymal stem cells ( BMSCs ) and human amniotic epithelial cells(AECs) could differentiate into many kinds of cells including corneal epithelial cells, but the outcome and effect of these cells on corneal stem cell deficiency are still unclear. ObjectiveThis study aimed to observe and compare the effects of rabbit BMSCs and human AECs transplantation for rabbit limbal stem cell deficiency. Methods Eighteen clean New Zealand rabbits were randomly divided into the amniotic stroma(AS) group, rabbit BMSCs group and human AECs group with 6 rabbits for each group. Limbal stem cell deficiency models were established by putting a piece of filter paper that had been soaked in a NaOH solution at the corneal center. Rabbit BMSCs were isolated and purified by density gradient centrifugation combined with the attachment culture method, and human AECs were collected by a sequential trypsin digestion technique,and the third generation rabbit BMSCs and the first generation human AECs were identified with RT-PCR. After that,cells were inoculated onto the denuded AS and grafted to the corneal surface of the experimental animals. Twenty-eight days after cell transplantation, the therapeutic effects were evaluated based on the corneal neovascularization and opacity scores. Corneal histopathological examination and immunohistochemistry were performed to evaluate and compare the effectiveness among AS,rabbit BMSCs and human AECs on corneal stem cell deficiency. The procedure complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. ResultsThe third generation of rabbit BMSCs grew well after 12 hours, and the first generation of human AECs formed a membrane-like monolayer after 48 hours of incubation on AS. Immunohistochemistry staining showed that, 28 days after transplantation, the surface cells of the cornea were positive for cytokeratin 3 in both the rabbit BMSCs group and human AECs group.Compared with the AS group,the corneal neovascularization and opacity grades were significantly decreased in the rabbit BMSCs group( Z=-2. 983, P =0. 003 ; Z =-2. 844, P =0. 004 ) and human AECs group ( Z =-2. 817, P =0. 005 ; Z =-2.041, P =0. 041 ). Histopathological analysis exhibited that stratified corneal epithelial-like cells formed on the corneal stroms 28 days after grafting and no signs of goblet cells and neovascularization were found. Less inflammatory cells and regular collagen fiber could be seen in the rabbit BMSCs group and human AECs group. In addition,clinical observation also revealed that the corneas were much clearer in the rabbit BMSCs group than the human A ECs group( Z =-2. 091 , P=0. 037 ), but the corneal neovascularization score was similar between them (Z = -0. 267,P=-0. 789). ConclusionsRabbit BMSCs and human AECs can differentiate into corneal epithelial-like cells onthedamagedcornealsurfaceandfurtherdemonstrateremarkableinhibitoryeffectsoncornealneovascularization and inflammatory cells. The more dominant and prominent effect is the role played by rabbit BMSCsin the improvement of corneal transparency.
3.Study on Medication Rules of Professor WEI Pin-kang in Treating Gastric Cancer from Phlegm

Objective To explore the medication rules of Professor WEI Pin-kang in treating gastric cancer; To provide references for clinical medication. Methods Gastric cancer cases treated by Professor WEI Pin-kang in TCM outpatient department of Shanghai Changzheng Hospital from January 2008 to December 2015 in WEI Pin-kang TCM Studio Outpatient Medical Cases (including electronic database and paper medical cases database). TCM Inheritance Support System V2.5 was used to build database. Frequency analysis, association rule analysis, complex system entropy clustering and other data mining methods were used to analyze frequency, medicine association rules and core medicine combination. Results Totally 305 clinical prescriptions including 185 kinds of Chinese materia medica were collected. The Chinese meteria medica with the high frequency were Pinellinae Rhizoma Praeparatum, Arisaematis Rhizoma Preparatum and Galli Gigerii Endothelium Corneum, etc. High frequency Chinese materia medica were with the properties of warm and cold, and with the flavors of bitter, spicy and sweet, to Stomach Meridian of Foot-Yangming, Spleen Meridian of Foot-Taiyin, and Liver Meridian of Foot-Jueyin. The medicine combinations with high frequency were Pinellinae Rhizoma Praeparatum and Arisaematis Rhizoma Preparatum,Pinellinae Rhizoma Praeparatum and Galli Gigerii Endothelium Corneum, and Galli Gigerii Endothelium Corneum and Arisaematis Rhizoma Preparatum. The common medicine pairs were Aurantii Fructus and Aurantii Fructus Immaturum and Scorpio and Scolopendra. Analysis showed 36 association rules of the confidence > 0.99. 70 core medicine combinations and 11 new prescription were obtained. Conclusion Professor WEI Pin-kang treats gastric cancer based on phlegm theory. The dissolving phlegm and eliminating stagnation medicine, regulating qi-folwing for harmonizing stomach medicine and heat-clearing and detoxifying medicine are commonly used.
4.A study on the standard of multi-slice spiral CT on evaluation of resectability of vascular invasion of pancreatic carcinoma

Objective To investigate the ability of different standards on the evaluation of vascular invasion of pancreatic carcinoma. Methods The MSCT imagings of 56 cases of pancreatic carcinoma confirmed by surgery were analyzed retrospectively. The vascular invasion degrees of 5 large vessels around the pancreas were evaluated according to Loyer classification standard, Lu classification standard and the Changhai standard, the surgical result was used as the gold standard. The accuracy of different standards was evaluated, and the Kappa coefficient were calculated. Results The accuracy, sensitivity, specificity, positive predictive value and negative predictive value of Loyer classification standard were 86.79%, 86.27%, 86.90%, 59.46% and 96.60%, respectively, and the Kappa coefficient was 0.623. The accuracy, sensitivity, specificity, positive predictive value and negative predictive value of Lu classification standard were 93.21%, 84.31%, 95.20%, 79.63% and 96.46%, respectively, and the Kappa coefficient was 0.777. The accuracy, sensitivity, specificity, positive predictive value and negative predictive value of Changhai standard were 95.36%, 84.31%, 97.82%, 89.58% and 96.55%, respectively. The Kappa coefficient was 0.841. Conclusions The Changhai standard was feasible to evaluate the vascular invasion of pancreatic carcinoma.
5.Efficacy and safety of oxaliplatin or irinotecan combined with capecitabine in the treatment of metastasis colorectal carcinoma

Objective To evaluate the efficacy and safety of oxaliplatin (L -OHP) or irinotecan ( CPT -11 ) combined with capecitabine (CAP) in the treatment of metastasis colorectal carcinoma.Methods Sixty -six patients with metastasis colorectal carcinoma were recruited in this study.There were 31 cases in CAP plus L -OHP group and 35 sub-jects in CAP plus CPT -11 group.All of the included cases were treated with 4 cycles of chemotherapy.The clinical efficacy and chemotherapy related side effects were compared between the two groups .Results The objective response rate (ORR) were 19.4% and 20.0% in the CAP plus L -OHP and CAP plus CPT -11 group respectively without statistical difference (P >0.05).The grade Ⅲ to Ⅳ adverse reactions in the CAP plus CPT -11 group and CAP plus L -OHP group were 51.6% and 60.0%, without statistical differences in two groups (P >0.05) .But diarrhea incidence in the CAP plus CPT -11 group (22.9%) was much higher than that in the CAP plus L -OHP group (3.2%) (P <0.05). Conclusion There was no statistical difference on clinical efficacy between the two chemotherapy regimens, but there is a higher incidence of serious diarrhea in CAP plus CPT -11 regimen.
6.Combined anterior and posterior surgery for treatment of cervical fracture-dislocation in patients with ankylosing spondylitis.

OBJECTIVETo discuss the pathological characteristics of cervical spinal fracture complicating ankylosing spondylitis (AS) and the effect of combined anterior and posterior operation.
METHODSEighteen AS patients with traumatic cervical fracture-dislocation were treated operatively from January 2000 to January 2006. The symptom duration of AS was 14.5 years on average. Three cases had undergone osteotomy in lumbar spine. There were 4 cases of Grade A, 3 cases of Grade B, 9 cases of Grade C and 2 cases of Grade D according to Frankel's score. There were 15 cases of Grade III dislocation and 3 cases of Grade II. All patients underwent surgical procedures by combined anterior and posterior approach.
RESULTSThere were 4 anterior-posterior procedures, 8 anterior-posterior-anterior procedures and 6 posterior-anterior procedures. Seven patients had one stage operation and 11 cases underwent two stage surgeries. There was certain extent of neurological improvement in 14 incomplete paraplegic patients, but no improvement in 4 complete paraplegic patients. The follow-up period was 21.2 months on average and the time for bone fusion was 3.6 months. There were 4 complications during operation and a long-term complication in follow-up.
CONCLUSIONSThe study suggests that anterior combined with posterior approach makes the spine stable and relieves the pressure immediately. It is a reasonable surgical strategy for treatment of cervical spinal fracture-dislocation with AS.

It is necessary to study on the core collection for the development of the germplasm resources of medicinal plants.

8.Parallel Mining of Gene Expression Differences of Nucleotide Excision Repair Gene XPA in Human Skin Microarrays based on GEO Database

Objective To make a parallel mining the data of expression differences of a crucial gene XPA involved in nucleotide excision repair pathway of human skin microarrays by bioinformatics from the system level.Methods Using the ScanGEO, the data of microarrays which included the significant differences expression level of XPA were screened and analyzed from 59 human skin samples in the GEO database. Results There were 7 samples with the down-regulated expression of XPA: cutaneous malignant melanoma, epidermal injury model, DNA damage and UV radiation, foreskin fibroblast response to Toxoplasma gondii RH type 1 (ROP5) mutant infection, interleukin-20 subfamily cytokines effect on epidermal keratinocytes, Egr-1 overexpression effect on skin fibroblasts in vitro: time course, in vitro model for inflammatory dendritic cells.Present expression down. Conclusion Based on the GEO database and ScanGEO, high-throughput shared data can be screened and analyzed efficiently.
9.Elevated Resting Heart Rate is Associated with Dyslipidemia in Middle-aged and Elderly Chinese

Objective To study the relationship between resting heart rate and blood lipid level.
Methods A total of 9 415 subjects aged≥40 years were included in the present study. Their resting heart rate was monitored and their serum levels of triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) were measured to define dyslipidemia according to the 2007 Chinese Guidelines on Prevention and Treatment of Dyslipidemia in Adults.
Results The subjects were divided into group A with their resting heart rate<70 beats/min, group B with their resting heart rate=70-79 beats/min, group C with their resting heart rate=80-89 beats/min, and group D with their resting heart rate≥90 beats/min. High TG, TC, and LDL-C were presented across the resting heart rate (Ptrend<0.01). Multiple logistic regression analysis revealed that the risk of high TG and TC was higher in subjects with their resting heart rate≥90 beats/min than in those with their resting heart rate<70 beats/min (OR=1.42;95%CI:1.16-1.74 and OR=1.33;95%CI:1.09-1.64, respectively).
Conclusion Elevated resting heart rate is associated with high TG and TC in middle-aged and elderly Chinese subjects.
10.Comparison of Liver Transplantation and Liver Resection for Hepatocellular Carcinoma Patients with Portal Vein Tumor Thrombus Type I and Type II

Purpose:
The aim of this study was to compare the efficacy of liver transplantation (LT) and liver resection (LR) for hepatocellular carcinoma (HCC) patients with portal vein tumor thrombus (PVTT) and to investigate risk factors affecting prognosis.
Materials and Methods:
A total of 94 HCC patients with PVTT type I (segmental PVTT) and PVTT type II (lobar PVTT) were involved and divided into LR (n=47) and LT groups (n=47). Recurrence-free survival (RFS) and overall survival (OS) were compared before and after inverse probability of treatment weighting (IPTW). Prognostic factors for RFS and OS were explored.
Results:
Two treatment groups were well-balanced using IPTW. In the entire cohort, LT provided a better prognosis than LR. Among patients with PVTT type I, RFS was better with LT (p=0.039); OS was not different significantly between LT and LR (p=0.093). In subgroup analysis of PVTT type I patients with α-fetoprotein (AFP) levels >200 ng/mL, LT elicited significantly longer median RFS (18.0 months vs. 2.1 months, p=0.022) and relatively longer median OS time (23.6 months vs. 9.8 months, p=0.065). Among patients with PVTT type II, no significant differences in RFS and OS were found between LT and LR (p=0.115 and 0.335, respectively). Multivariate analyses showed treatment allocation (LR), tumor size (>5 cm), AFP and aspartate aminotransferase (AST) levels to be risk factors of RFS and treatment allocation (LR), AFP and AST as risk factors for OS.
Conclusion
LT appeared to afford a better prognosis for HCC with PVTT type I than LR, especially in patients with AFP levels >200 ng/mL.
