OBJECTIVE:To study the influencing factors of protocol deviation in drug clinical trial implementation, and pro-vide reference for improving the quality of drug clinical trial. METHODS:Quality verification was conducted for the drug clinical trial projects in the First Affiliated Hospital of Chongqing Medical University during 2010-2016,and protocol deviations in each year were retrospectively studied, classified and analyzed. Category, frequency, international and domestic pilot projects and the differences of protocol deviation after full-time research nurse participating in trail management were explored, and the measures were put forward. RESULTS:27 drug clinical trials were implemented in our hospital during 2010-2016,including 949 cases,176 cases with protocol deviation, accounting for 18.55%. Deviation protocol in drug clinical trial was decreased year by year for 7 years. The categories were mainly incompleteness of observation/records (30.11%),checking omission/broaden the window(28.41%),adverse drug events and improper combined medication (14.20%) and omission in drug management (11.93%). The proportion of protocol deviation with full-time research nurse participated was lower than the projects without full-time research nurse(11.11% vs. 28.67%,P<0.01),and proportion of deviation protocol in international multi-center project was lower than the domestic projects(6.60% vs. 28.84%,P<0.01). CONCLUSIONS:It is suggested to pointedly strengthen the weak links of drug clinical trial. For example, clinical trial institutions should establish the clinical trial data retention system, electronic data should be timely backed up in a different places, etc. Besides, clinical trial institutions should equip professional full-time research nurses as much as possible, learn the rigorous scientific experimental design, standard operational procedures and the authenticity of data pro-cessing from the international multi-center clinical trial projects to effectively reduce the incidence of deviation protocol and im-prove the quality of drug clinical trials.

Chinese Pharmacological Bulletin 1987;0(03):-
The effects of TSCP and TSCC on aggregation and adhesion of blood platelets were measured by turbidimetric method and salzman method respectively. TSCP and TSCC strongly increase mouse and rat platelet aggregation and platelet adhesion. These effects are important for blood coagulation. cAMP and TXB2 levels in plasma and platelet were determind by RIA TSCP significantly increased the TXB2 level in plasma and platelet. This effect may be the main mechanism of increasing platelet aggregation by TSCP.

Objective To investigate the clinical manifestions, pathological features and diagnosing methods of Lafora disease. Methods The chnical and pathological features of 5 patients with Lafora disease who were diagnosed by axillary biopsies were systemically studied. The specimen were stained by HE, PAS and AB-PAS methods. Results Four of 5 cases had an onset during adolescence and 1 during adulthood. All cases presented with progressive generalized tonic-clonic seizure, myoclonus and dementia. Emotional disturbance, dysarthria and ataxia appeared in the early course of the disease. Lafora bodys were identified in myoepithelial cells and duct cells of both eccrine sweat glands and apecrine sweat glands in the biopsies of axillary skin. Conclusions Lafora disease could be confirmed by round and oval periodic acid-Schiff- positive inclusions in skin biopsy specimen combined with the proper clinical settings. Both axillary and other skins can be chosen as the sites of biopsy.

Objective To investigate the effects of exercise on learning and memory after cerebral ischemia. Methods Ninety Sprague-Dawley rats were randomly divided into a sham-operated group, a model group and a treadmill training group ( n=30 for each) , which were further subdivided into groups to receive 7 days, 14 days or 28 days of training with 10 rats in each. The training was treadmill running at 10 m/min for 30 min a day. Cerebral ische-mia was induced in the model and training groups using permanent, bilateral common carotid artery occlusion. The training began three days after the operation. Morris water maze tests were used to measure the rats′ learning and memory ability, and Nissl staining was employed to detect the survival rates of pyramidal neurons in the CA1 area of the hippocampus. Results The average escape latency in the treadmill trained group had shortened significantly by day 7, then further by days 14 and 28. It was significantly shorter than the model group′s average at each time point. The average platform crossing time increased significantly compared with the model group′s average. Few dead neu-rons were observed in the sham-operated group. On days 14 and 28 the average survival rate of pyramidal neurons in the model group was significantly lower than in the treadmill training group or the sham-operated group, though there was no significant difference on day 7. Conclusion Treadmill training can improve learning and memory after cere-bral ischemia, at least in rats. Better effects can be observed after longer training.

BACKGROUND: The osteogenesis effects of concentrated growth factors (CGF) gel have been studied, but little has been reported on CGF liquid.OBJECTIVE: To explore the bone mass changes after CGF is used in the dental implant surgery with guided bone regeneration (GBR) in patients with upper anterior tooth labial bone defect, and to evaluate the effect of CGF liquid combined with Bio-Oss bone meal on bone augmentation.METHODS: Fifty patients with single anterior tooth labial bone defect were enrolled and randomly divided into experimental and control groups (n=25 per group). Patients in both groups underwent dental implant surgery with GBR.CGF liqiud, Bio-Oss bone meal and biological membrane were used in the experimental group, and Bio-Oss bone meal and biological membrane used in the control group.RESULTS AND CONCLSUION: All patients healed well according to the successful criteria. Six months postoperatively, there was no significant difference in the thickness changes of labial bone between two groups (P > 0.05). From 6 months to 12 months after operation, the thickness changes of labial bone in the experimental group was significantly less than that in the control group (P < 0.05). Our findings indicate that CGF liquid combined with Bio-Oss bone meal makes good effect on dental implantation of patients with anterior tooth labial bone defect. CGF liquid also contributes to reduce the labial bone resorption of the implant after functional loading.
