1.Establishment of a monitoring database of historical Oncomelania hupensis environments at grass-root level for schistosomiasis prevention based on Google Earth

Objective To establish a monitoring database of historical Oncomelania hupensis environments at grass?root lev?el for schistosomiasis prevention based on Google Earth(GE),so as to improve the management efficiency of historical O. hu?pensis environments. Methods GE was labeled with the information about the historical O. hupensis environments through add?ing landmark,path and polygon. Meanwhile,POCO web album was used to storage the practice situation pictures of the environ?ments,which could be imported into GE so that the environments could be monitored dynamically. Results There were 553 historical O. hupensis snail environments in Yangjian Town,Xishan District,Wuxi City and the accumulative area was 506 000 m2. Among these environments,224(40.5%)were Class IV environments,accounting for 40.5%of accumulative area. Conclu?sions The monitoring database of historical O. hupensis environments based on GE at country level is established successfully and it can be used to manage the historical snail environments visually and monitor the changes dynamically.
2.Ultrasonic features of acute acalculous cholecystitis developing at different stages of acute pancreatitis

Objective Acute pancreatitis exhibits different clinical and ultrasonic features in patients complicated with acute acalculous cholecystitis ( AAC) at different stages .The aim of this study was to analyze the ultrasonic characteristics of acute pancreati-tis complicated with AAC at different stages . Methods We retrospectively analyzed the clinical data about 41 cases of acute pancrea-titis with moderate to severe AAC .According to whether AAC developed within or after 2 weeks of the onset of acute pancreatitis , we divided the patients into an early-stage group (n=18) and a late-stage group (n=23).We recorded the gallbladder size, gallbladder wall thickness , fluid around the gallbladder , biliary sludge deposition and the Murphy′s sign by ultrasonography , obtained AAC-related clinical and laboratory data concerning body temperature , Murphy′s sign, WBC count and C-reactive protein level , and analyzed the ultrasonic features of AAC at different stages in the acute pancreatitis patients. Results All the patients experienced a fever of >38.5℃, 38.89%with chills in the early onset group and 47.83%in the late onset group .Increases were observed in patients of the early-and late-stage groups in the WBC count ( 94.44%vs 82.61%) , the C-reactive protein level ( 100%vs 91.30%) , and the fluid volume around the gallbladder (94.44%vs 60.86%, P<0.05), but incidence rate of gallbladder wall thickening was significantly lower in the former than in the latter group (11.11%vs 78.26%, P<0.01). Conclusion AAC developing at different stages of acute pancreatitis has different ultrasonic features , with higher incidence rates of fluid around the gallbladder in the early stage and gallbladder wall thickening in the late stage.
3.Effects of frequency on osteoblast apoptosis induced by tensile strain

Objective To investigate the effect of frequency on osteoblast apoptosis induced by tensile strain.Methods MC3T3-E1 cells were applied with 1% biaxial tensile strain at the frequency of 1,2,3,4,5 Hz,re spectively for 1 hour per day in 8 days.The survival rate of the cells was determined by activity of lactate dehydrogenase (LDH).Annexin V-FITC/ PI Flow cytometry was used to test cell apoptosis.Real-time RT-PCR was used to detect the gene level of apoptosis markers caspase-3,-9 as well as Bcl-2 and Bax,and Western blotting was used to test protein expressions of caspase-3,-9.Results Different loading frequencies had no effect on osteoblast activity of LDH.There was no significant difference in the total apoptosis rate of flow cytometry at different frequencies.However,the frequency of 2 Hz could induce early osteoblast apoptosis.Tensile strain at the frequency of 2 Hz could significantly increase the expression of caspase-3,-9 gene and protein,and induce cell apoptosis with the up-regulation of the Bax/Bcl-2.Conclusions Osteoblast apoptosis and death cannot be induced by 1% biaxial tensile strain at the frequency of 1-5 Hz,but the frequency of 2 Hz can induce the early apoptosis of osteoblasts by up-regulating the expression of Bax/BCI-2.
4.Identification of novel epithelial ovarian cancer biomarkers by cross-laboratory microarray analysis.

The purpose of this study was to pool information in epithelial ovarian cancer by combining studies using Affymetrix expression microarray datasets made at different laboratories to identify novel biomarkers. Epithelial microarray expression information across laboratories was screened and combined after preprocessing raw microarray data, then ANOVA and unpaired T test statistical analysis was performed for identifying differentially expressed genes (DEGs), followed by clustering and pathway analysis for these DEGs. In this work, we performed a combination analysis on microarrays from three different laboratories using gene expression data on ovarian cancer and obtained a list of differential expression profiles identified as potential candidate in aggressiveness of ovarian cancer. The clustering and pathway analysis explored the different molecular basis of different ovarian cancer stages and potential important regulatory pathways in ovarian cancer development. Our results showed that combination of microarray data from different laboratories in the same platforms may overcome biases derived from probe design and technical features, thereby accelerating the identification of trustworthy DEGs, and demonstrating the advantage of integrative analysis in gene expression studies on epithelial ovarian cancer research.
5.Application of improved TOPSIS based on entropy weight to evaluating township health education program

Objective To explore a more objective scheme for comprehensive evaluation of township health education program. Methods Based on the indicators and grading results of 17 township hospitals (named from A to Q) in Xiangshan County, we calculated the weight of each indicator by entropy weight method and sorted the township hospitals by the improved TOPSIS. Then we compared the ranking with the annual assessment results in 2017 to verify the rationality of this scheme. Results The entropy weights of"printed materials of health education","play of audio-visual materials","health education bulletin board","public health consultation" ,"health knowledge lecture" ,"individualized health education" and"awareness rate of health knowledge"were 0.122 0, 0.125 3, 0.134 6, 0.157 2, 0.157 0, 0.179 8 and 0.124 1, respectively. Through the improved TOPSIS, 17 hospitals were sorted as follows: P, L, O, C, G, A, H, N, B, E, M, Q, K, J, F, D, I. Compared with the annual assessment rankings which were P, L, A, C, B, O, G, H, K, N, D, E, M, J, I, F, Q, thirteen of them were different. Conclusion "Printed materials of health education","awareness rate of health knowledge" and"play of audio-visual materials" have little room for improvement, while"individualized health education" should be focused on in the future. The improved TOPSIS based on entropy weight could objectively evaluate the quality of health education service.
6.H2 receptor mediates the protective effect of histamine against the cellular edema and viability reduction induced by oxygen-glucose deprivation in rat hippocampal slices.

AIMTo determine the effect of histamine on ischemia-induced cellular edema and viability reduction in rat hippocampal slices, and the involved subtypes of histamine receptor in this effect.
METHODSIn vitro ischemic injury of hippocampal slices was induced by oxygen-glucose deprivation (OGD). The slice injury was determined by real-timely measuring the changes of light transmittance (LT) for the cellular edema in CA1 region of the hippocampal slice, and by detecting the product of 2, 3, 5-triphenyltetrazolium chloride (TTC), formazan, for the slice viability. The effect of histamine at various concentrations on the slice injury was observed, and the blockage by antagonists of histamine receptors was also investigated.
RESULTSHistamine (0.01-10 micromol x L(-1)) inhibited the peak value of LT during OGD in hippocampal slices and improved the reduced viability after OGD. Diphenhydramine (0.1-10 micromol x L(-1)), an H1 receptor antagonist, did not affect the effect of histamine, while cimetidine (0.1-10 micromol x L(-1)), an H2 receptor antagonist, partly abolished the protective effect of histamine.
CONCLUSIONHistamine protects hippocampal slices against ischemia-induced cellular edema and viability reduction; this effect might be mediated via, at least partly, H2 receptor.

7.Assessment of a capsid-modified E1B 55-kDa protein-deficient adenovirus vector for tumor treatment

ONYX-015 and H101 are E1B 55-kDa protein-deficient replicating C group adenoviruses that are currently in clinical trials as antitumor agents. However, their application in cancer gene therapy is limited by the native tropism of C group adenoviruses. This is in part due to low expression of the C group adenovirus receptor (coxsackievirus-adenovirus receptor, CAR) on malignant tumors. An H101-based chimeric virus vector containing sequences encoding the Ad35 fiber domain instead of the Ad5 fiber (H101-F35) was constructed. This modification allowed infection of tumor cells through CD46, a membrane protein over-expressed on tumors. The CAR and CD46 RNA expression was evaluated by RT-PCR method. H101-F35 conferred a stronger cytocidal effect than H101 and ONYX-015 in tumor cell lines that lacked CAR expression (MDA-MB-435 and MCF-7), while the cytocidal effect of H101-35, H101 and ONYX-015 was similar in high-level CAR expressing cancer cell lines (A549, NCI-H446, Hep3B, LNCaP, ZR-75-30 and Bcap-37). In an MDA-MB-435 xenograft mouse tumor model, tumor growth in mice receiving H101-F35 was significantly inhibited compared with mice injected with H101. These results suggest that the chimeric oncolytic adenovirus H101-F35 vector might be a useful candidate for gene therapy of cancer.
8.Detection of co-infection with Lyme spirochetes and Spotted fever group rickettsiae in a group of Haemaphysalis longicornis

Objective The present study was conducted to investigate the infection of Lyme disease, Spotted fever, Ehrlichiosis (anaplasmosisin) in wild animals and ticks in the mountain areas of Zhejiang province. Methods Nested polymerase chain reaction was used to amplify specific DNA sequences of Lyme spirochetes, Spotted fever group rickettsiae, Ehrlichia (anaplasma) from samples of mice and ticks. Results 14 positive samples were identified from 121 mice and 105 groups of ticks. Among mice samples, one positive 5S-23S rDNA intergenic spacer of Borreia burgdorferi and two 5' fragments of Ehrlichia (anaplasma) 16S rDNA were obtained. 11 positive results were detected from tick samples including three 5S-23S rDNA intergenic spacer regions of Borreia burgdorferi and eight 5' fragments of Spotted fever group rickettsiae outer member protein A gene. One group of adult ticks, Haemaphysalis longicornis, which had been collected from eastern mountain area were detected to have co-infected with Lyme spirochetes and Spotted fever group rickettsiae. The positive sequences of 5S-23S rDNA intergenic spacer and ompA gene were tested and analyzed as Lyme spirochetes while rickettsia which was closely related to Borrelia valaisiana and R. massilliae. Conclusion This was the first report about co-infection of Lyme spirochetes and Spotted fever group rickettsiae found in the same group of adult Haemaphysalis longicornis. It is very important to strengthen the surveillance program on tick-borne infectious disease and their pathogenic in vectors, wild animals and targeted high risk groups and to differentiate the clinical manifestation and diagnosis to extend the knowledge of tick-borne infectious diseases in Zhejiang.
9.Epidemiological investigation of risk factors for cataract among people aged 50 and over in Tacheng area, Xinjiang

Objective:To investigate the prevalence of cataract and its risk factors in the Jiu Shi Hospital of Xinjiang Production and Construction Corps and Han and Kazak people aged 50 years old and over in Emin County, Tacheng area, Xinjiang.Methods:A cross-sectional study was conducted.An epidemiological survey on the prevalence of cataract among people aged 50 years old and over was performed by the Department of Ophthalmology of North China University of Science and Technology Affiliated Hospital in the Jiu Shi Hospital of Xinjiang Production and Construction Corps and Emin County from October 2015 to June 2018.The basic sampling units (BSUs) were divided according to the administrative regions, with about 1 000 people in each administrative region.With a population exceeding 1 500, sample units would be divided into more than two BSUs according to the population.Xinjiang Tacheng area was divided into 240 BSUs, and 12 BSUs were selected by random number table method for inspection.With the lens opacity classification system Ⅱ (LOCS Ⅱ) as the diagnostic standard, the prevalence rate of cataract in different nationalities and different ages in this area was evaluated, and the relationship between age, nationality, sex, hypertension, diabetes, smoking, hyperlipidemia, coronary heart disease, ultraviolet irradiation time and the prevalence rate of cataract was analyzed.This study protocol was approved by the Ethics Committee of the Jiu Shi Hospital of Xinjiang Production and Construction Corps (No.SW-201501-04).Results:The prevalence rate of cataract among people aged 50 and over in the Jiu Shi Hospital of Xinjiang Production and Construction Corps and Emin County was 76.34%.The prevalence rate of cataract in 50-<60-year-old group, 60-<70-year-old group, 70-<80-year-old group and ≥80-year-old group was 23.30%, 77.40%, 89.60% and 100.00%, respectively.There were significant differences in the prevalence among people between different ages, different nationalities, with and without hypertension, with and without diabetes, with and without hyperlipidemia (all at P<0.01). There was no significant difference in the prevalence among people between different sexes, with and without coronary heart disease, smoking or not, different ultraviolet irradiation time (all at P>0.05). Multiple logistic regression analysis showed that age (50-<60 years old: [ OR=39.93, 95% CI: 20.77-76.77; 60-<70 years old: OR=3.81, 95% CI: 2.05-7.08; 70-<80 years old: OR=1.72, 95% CI: 0.92-3.22]), hypertension ( OR=1.54, 95% CI: 1.13-2.11) and hyperlipidemia ( OR=1.41, 95% CI: 1.10-2.03) were independent risk factors for cataract. Conclusions:The incidence of cataract in the Jiu Shi Hospital of Xinjiang Production and Construction Corps and people aged 50 and over in Emin County is higher than that in other parts of China, in which age, hypertension and hyperlipidemia are independent risk factors for cataract.
10.HS-SPME-GC-MS Analysis on Volatile Components in Various Processed Products of Achyranthes Bidenta-tae Radix

Objective: To analyze and compare the difference in volatile components in Achyranthesbidentatae Radix, salt-pro-cessed product of Achyranthes bidentatae Radix and wine-processed product of Achyranthes bidentatae Radix. Methods:Headspace sol-id-phase microextraction ( HS-SPME) was used to extract the volatile components from Achyranthes bidentatae Radix and the two pro-cessed product of Achyranthes bidentatae radix. The chemical compositions were analyzed by gas chromatography-mass spectrometry ( GC-MS) , and their relative mass fraction of each component was determined by an area normalization method. Results:A total of 38 chemical components were identified, and 23, 16 and 15 components were respectively identified from Achyranthes bidentatae Radix, the salt-processed product of Achyranthes bidentatae Radix and the wine-processed product of Achyranthes bidentatae Radix accounted for 79. 14%, 49. 80% and 44. 57% of the total content of volatile components, respectively. There were 4 types of common volatile chemical constituents in the three products. Compared with Achyranthes bidentatae Radix, 10 types of new compounds were produced and 17 types of old compounds disappeared in the salt-processed product, and 9 types of new compounds were produced and 17 types of old compounds disappeared in the wine-processed product. Conclusion:Different processing methods have some influence on the type and the content of the volatile components in Achyranthes bidentatae Radix. Using HS-SPME-GC-MS combination technique can quickly and effectively obtain the volatile components information for Achyranthes bidentatae Radix, which can provide scientific reference for the quality evaluation and application of the processed products of Achyranthes bidentatae Radix.
