1.Recent advances in association studies of estrogen system and non-small cell lung cancer

Lung cancer, a malignant cancer with highest death rate, severely threats the people's health. 80% lung carcinomas are non-small cell lung cancer (NSCLC). Recent studies have found that estrogen system has close relationship with the biological behaviors of NSCLC. Meanwhile, the level of estrogen influences the progression of disease and the prognosis of lung cancer patients. As a result, estrogen pathway has become a hot target in NSCLC treatment. This paper reviews the recearch advances of estrogen system in NSCLC treatment.
2.Role of cell adhesion molecules in bone metastasis of tumor

Bone metastasis is one of the most common complications of malignant tumors.Cell adhesion molecules(CAMs),including cadherin family,integrin family,selectin family,immunoglobulin superfamily,and hyaladherin,play an important role in bone metastasis of tumor through homotypic and heterotypic adhesive interactions.In this review,molecular and cellular mechanisms underpinning bone metastasis associated with CAMs,the role in a multi-step bone metastatic cascades will be discussed.
3.The value of sequential, alternating and consolidation chemotherapy in advanced non-small-cell lung cancer

A platinum-based doublet with a third-generation agent represents the standard first-line treatment for advanced non-small cell lung cancer patients with good performance status (PS). Traditional chemotherapy provides response rates of 20 %-40 % and a median survival of 8-10 months. In an attempt to improve its outcome, alternative schedules have been proposed, namely sequential, alternating, and maintenance therapy. Sequential chemotherapy with a platinum-based doublet followed by a single agent is feasible in patients with good PS; The use of sequential single agents is an option for elderly and frail patients unsuitable for a platinum-based combination. Based on trials published so far, it is unlikely that an alternating chemotherapy strategy will be proved superior to standard chemotherapy in patients with good PS.Consolidation chemotherapy may provide additional benefit for patients achieving disease control after standard first-line chemotherapy.
4.Relation between mutation of DNA-Ligase Ⅳ gene and radiosensitivity in human nasopharyngeal carcinoma cell lines

Objective To define the correlation between mutation of DNA-LigaseⅣ gene and radiosensitivity.Methods Nasopharyngeal squamous carcinoma cell line (CNE), lung adenocarcinoma cell line (SPC-A1) and breast adenocarcinoma cell line(MCF-7) after irradiation were assessed with specific biological parameters. Polymerase chain reaction, cloning and sequencing techniques were used to determine the sequence of DNA-LigaseⅣ gene in these three cell lines. Then the impact of homologous change and mutation on hydrophicity-hydrophobicity of genic products was analyzed.Results The surviving fractions derived from irradiation were different with more radiosensitivity in the CNE cell line than in the others. In three cell lines, the homology of LigaseⅣ gene were: 99.95%,99.99%,99.98%, respectively. Some mutations including transversion and transition were detected and led to alterations in the hydrophicity-hydrophobicity function of products. Higher radiosensitivity of CNE was associated with amino-acid substitutions: 313aa His→Arg,538aa Gly→Arg,579aa Lys→Arg and 585aa Asn→Ser.Conclusion These results suggest that LigaseⅣp play an important role in the ligation of DNA double strand breaks and certain mutations bring about changes in radiosensitivity.
5.Experimental study on prevention of non steroidal anti-inflammatory drugs induced small intestinal damage by metronidazole and rabeprazole

Objective To establish rat model of small intestinal damage by administrating low dose of diclofenac in short-term and to observe the preventive effect on non steroidal anti-inflammatory drugs (NSAIDs) induced small intestinal damage by metronidazole and rabeprazole.Methods Sixty-four rats were randomly divided into control,model,metronidazole and rabeprazole groups with 16 each.The rats in four groups were then further separated into acute stage (T1) and subacute stage (T2) groups.The rats were lavaged with metronidazole (50 mg/kg) and rabeprazole (15 mg/kg) before modeling.The rats in control,model,metronidazole and rabeprazole groups were lavaged with 1ml of distilled water, 7.5 mg/kg diclofenac,50 mg/kg of metronidazole plus 7.5mg/kg of diclofenac,15 mg/kg of rabeprazole plus 7.5mg/kg of diclofenac twice a day,respectively.The rats in T1 group were sacrificed on day 1 and T2 group on day 5.Results Administration of diclofenac induced multiple lesions in the small intestine, such as obviously erythema,erosion,ulcer and cystoid expansion.The lesions in T2 of model group was more seriously than that in T1 group(P<0.05).And the lesions in model group was also more seriously than that in metronidazole and rabeprazole groups (both T1 and T2 groups,P < 0.05).Serum concentration of NO in T1 of model group was obviously lower than that in control group (P<0.05),but it was higher in T2 of model group compare to the control group (P<0.05).There were no significant difference among metronidazole,rabeprazole and model groups in serum level of NO (P>0.05). Conclusions The lesions of small intestinal can be induced by administration of diclofenac.The serum level of NO presents a tendency with decrease before increase afterwards.The metronidazole or rabeprazole has preventive effect on small intestinal lesions.
6.Clinical significances and relationship between vascular endothelial growth factor expression and microvessel density in gastric carcinoma

Objective To evaluate the correlation between vascular endothelial growth factor(VEGF) expression and microvessel density(MVD) in gastric carcinoma (GC) and the relationship of VEGF and MVD with clinicopathologic characteristics and prognosis of GC. Methods The expression of VEGF and intratumoral microvessel density (MVD) in one hundred and sixteen resected specimens of the patients with GC were observed and counted, and the relationship of VEGF and MVD with clinicopathologic factors and prognosis of GC were analysed. Results The expression of VEGF was found in 60.34% of the specimens. The MVD value was much higher in VEGF(+) group than that in VEGF(-) group (26.16±8.50 and 19.22±8.20, respectively, P<0.01). Expression of VEGF was significantly higher in patients with lymph node metastasis than that in patients without lymph node metastasis (p<0.05).Expression of VEGF was highly correlated with the stage of tumor (P<0.05). MVD correlated with lymph node metastasis (P<0.01) and abdomen metastasis (P<0.01) and increased with the TNM stage (P<0.01). The total five-year survival rate in VEGF(-) group and low MVD group were significantly higher than that in VEGF(+) group and high MVD group respectively(both P<0.01). Multivariate analysis indicated that the expression of VEGF and MVD were independent prognostic factors of GC. Conclusions The expression of VEGF and MVD can reflect the malignant degree of GC. They may serve as the prognostic factors and guide the decisions on the therapy.
7.One stage surgical correction of congenital heart disease and tracheal stenosis in infants

Objective To review and evaluate the effect of one stage surgical correction of congenital heart disease with tracheal stenosis in infants.Methods Between August 2001 and December 2005,six infants with congenital heart disease with tracheal steno- sis were repaired at one stage operation.The age was 24 d~3 y[mean(13.16?12.03)months]and the weight was 4.2~10.0 kg [mean(7.98?2.03)kg].There were 3 patients with tetralogy of Fallot,1 with pulmonary atresia;2 with ventricular septal defect, 1 with coarctation of aorta;and 1 with pulmonary sling.The congenital heart disease were repaired underwent low temperature and car- diopulmonary bypass,and the tracheal stenosis were corrected simultancously.Results There was one death postoperative because of right and left branch stenosis.One patient with pulmonary sling died of granulation tissue on the patch 3 months after operation.All other 4 patients recovered uneventfully.These patients were followed up from 6 month to 4 years.There were no complications.CT scan shows that the tracheal anastomosis was patent without any stenosis.Conclusion One stage repair of congenital heart disease with tracheal stenosis in infants may have a satisfactory result.To deal with the tracheal stenosis is difficult,the operative methods de- pends on the length of stenosis.The principles of surgical methods consist of using autologous material,maintaining its contour and function of respiratory epithelium,and maintaining its growth.
8.Docetaxel for 15 patients with paclitaxel-resistant advanced non-small cell lung cancer

Objective To determine the efficacy and toxicity of docetaxel in patients with paclitaxel-resistant advanced non-small cell lung cancer (NSCLC). Methods The clinical data from 15 patients with NSCLC who were admitted in the Shanghai Chest Hospital from January 2005 to May 2008 were retrospectively analyzed. The effects and toxicities of the second-line treatment were assessed. The progression-free survival time(PFS) and overall survival time(OS) were analyzed. Results The disease control rate was 66.7 %, with a progression-free survival time of 6 months, and a overall survival time of 17.3 months. The 1-year survival rate was 63.3 %. The toxic effects were as expected. Conclusion The doeetaxel-based agent is active in patients with paelitaxel-resistant advanced NSCLC.
9.Simultaneous Determination of the Content of 4 Psoralen Compounds in Buwu Tincture by HPLC

OBJECTIVE:To establish a method for simultaneous determination of 4 psoralen compounds in Buwu tincture. METHODS:HPLC was performed on the column of Dikma Diamonsil C18 with mobile phase of acetonitrile-0.2% Acetic acid by gradient elution at flow rate of 1 ml/min,detection wavelength was 246 nm,column temperature was 30 ℃,and the injection vol-ume was 15 μl. RESULTS:The linear range was 13.00-208 μg/ml for angelicin,26.00-416 μg/ml for bavachin,24.50-392 μg/ml for psoralidin and 37.88-606 μg/ml for isobavachalcone,respectively(r≥0.999 6);RSDs of precision,stability and reproducibility tests were less than 2.00%;recoveries were 95.22%-97.23%(RSD=0.87%,n=6),100.24%-104.64%(RSD=1.62%,n=6), 102.28%-104.39%(RSD=1.47%,n=6)and 97.68%-100.17%(RSD=0.97%,n=6),respectively. CONCLUSIONS:The method is simple and reproducible,and can be used for the quality control of Buwu tincture.
