Objective To construct a framework and relevant evaluation items of the teaching ability for nursing teachers in colleges or universities.Methods The research was carried out by a variety of means such as theory analysis,literature study,expert consultation,etc,to explore behavior items that could be used to evaluate the teaching ability for nursing teachers in colleges or universities.A questionnaire was formed and a survey was conducted in a total of 194 nursing teachers from ten colleges or universities across the whole nation.Then,factor analysis was performed to build the relevant evaluation items for teaching ability of full-time nursing teacher.Cronbach's α and Spearman correlation were used to test the reliability and validity.Results The relevant evaluation items of teaching ability for nursing teachers contained 31 behavior items from six domains,which were teaching research ability,emotional teaching ability,expressing ability,self-examination ability,integration ability,and organizing ability.Cronbach's α of the total system was up to 0.913 0,Cronbach's α of each domain ranged from 0.681 0 to 0.815 8.Every factor had positive correlation with the totality,and the correlation coefficient of every item with its affiliated factor was greater than other factors.Conclusions The relevant evaluation questionnaire of teaching ability for nursing teachers has excellent reliability and validity.It can provide reference standard to directing admission,training,and teaching evaluation for nursing teachers.

3.Combined reconstruction of the injured posterior cruciate ligament and posterolateral complex of the knee

Objective To discuss the necessity and methods of combined reconstruction of the injured posterior cruciate ligament(PCL)and posterolateral complex(PLC)of the knee. Methods From January 2001 to December 2006,28 patients with injury of both PCL(≥Ⅱ°b)and PLC(>Ⅱ°)of the knee were treated operatively in our department.Eight patients received arthroscopic PCL reconstruction with single bundle of 5-6 strands of hamstring and conservative treatment of PLC during January 2001 to December 2003.Twenty Datients received arthroscopic PCL reconstruction followed by open operative repair of PLC with median 1/3 femoral biceps tendon and gracilis during January 2002 to December 2006. Results All the patients were followed up for 6 to 24 months postoperatively (mean,15 months).By Lysholm scoring,3 cases obtained good results and 5 poor in the first group,while 19 excellent and 1 good in the second group.The mean Lysholm scores were 47.1±8.2 and 86.7±7.2 respectively(P<0.05).According to IKDC scale,3 patients(37.5%)in the first group and 19 patients (95.0%)in the second group were graded normal or nearlv normal(t=19.277,P<0.05). Conclusion Arthroseopic PCL reconstruction with single bundle of 5-6 strands of hamstring plus PLC reconstruction with 1/3 femoral biceps tendon and gracilis is more reliable to restore the stability of the knee than PCL reconstruction only.
4.Protective effects of adenoviral-mediated sphingosine kinase gene on ischemia reperfusion injury of heart

Objective Sphingosine kinase 1 (SPK1) has been identified as a central mediator of ischemia preconditioning, and it has been shown to protect reactive oxygen species (ROS)-induced cardiomyocytes death. The present study aims at investigating the protective effects of adenovirus-mediated sphingosine kinase 1 gene (Ad-SPK1) transfer on ischemia-reperfusion induced cardiac injury. Methods Wistar rats were anesthetized and a left thoracotomy was performed. About 5?108 PFU of Ad-SPK1 in total volume of 100?l was injected intramyocardially into four separate sites of the left ventricular wall with a 30-gauge needle. The control rats received the same injection of adenovirus carrying green fluorescent protein gene (Ad-GFP). Three days later, hearts were isolated and subjected to ischemia/reperfusion (I/R) (30min/30min) ex vivo. Heart performance was evaluated by measuring the left ventricular systolic pressure (LVSP) and left ventricular end-diastolic pressure (LVEDP). The incidence of arrhythmia was recorded. Cardiomyocyte viability was detected by the detection of the release of creatine kinase (CK). The cardiac SPK1 activity was measured using an enzyme method. The forced and the total SKP1 expression was analyzed by immunoblotting with anti-FLAG and anti-SPK1 antibodies. Results The cardiac SPK1 activity was increased by about five-fold by injection of Ad-SPK1, compared to Ad-GFP control group. A more potent performance and a lower incidence of arrhythmia were observed in Ad-SPK1-injected hearts during the reperfusion period, compared with Ad-GFP-injected ones. Enzymatic activity assay showed that creatine kinase release was also less in Ad-SPK1-injected hearts. Conclusion Adenovirus-mediated SPK1 gene transfer efficiently protects heart from injury induced by ischemia-reperfusion.
5.ABC Analysis of Non-antimicrobial Temporary Purchased Drugs in a Hospital in 2014

OBJECTIVE:To provide reference for scientific and standardized management of temporary purchased drugs. METHODS:By using ABC analysis method,the data of non-antimicrobial temporary purchased drugs in a hospital in 2014 were analyzed statistically with ABC analysis in terms of quantity,the number of types and departments. Class A drugs were analyzed es-pecially. RESULTS:A total of 152 kinds of drugs were purchased temporarily,including 20 464 boxes,20 kinds of class A drugs, which accounted for 70.56%. Main types were antitumor drugs and nutraceuticals by pharmacology;26 departments temporarily purchased drug,mainly in tumor department and hematology department. CONCLUSIONS:The types of temporary purchased drugs meet the clinical demand in the hospital basically. The necessary type A drugs used frequently can be used as reference of tem-porary drug purchase;the temporary purchased drugs can be managed scientifically and normatively based on ABC analysis.
6.Correlation between exposure to polycyclic aromatic hydrocarbons during pregnancy and placental mitochondrial DNA copy number

Objective:
To examine the correlation between exposure to polycyclic aromatic hydrocarbons ( PAHs ) and placental mitochondrial DNA ( mtDNA ) copy number among pregnant women, so as to provide the evidence for evaluation of potential effects of PAHs exposure during pregnancy on offspring health.
Methods:
A total of 200 pregnant women delivered at a tertiary hospital in Urumqi City during the period from January to October 2019, and their newborns were recruited, and grouped according to the time of delivery, including the heating group [delivery during the heating period ( from January to April ) ] and the non-heating group [delivery during the non-heating period (from July to October) ]. Subjects'age, ethnicity, educational level and type of home heating were collected, and the total concentration of 16 PAHs was determined in the blood samples of pregnant women and their babies using gas chromatography-mass spectrometry. Placental DNA was extracted, and placental mtDNA copy number was measured using real-time fluorescence quantitative PCR assay. In addition, the correlation between PAHs concentration and placental mtDNA copy number was examined using the Spearman rank correlation analysis.
Results:
There were 100 subjects in the heating group, which had a median age of 29 ( interquartile range, 3 ) years and had a mean gestational age of ( 275.06±0.72 ) days, and there were 100 subjects in the non-heating group, which had a median age of 29 ( interquartile range, 4 ) years and had a mean gestational age of ( 276.82±0.66 ) days. The total concentration of PAHs in the blood of pregnant women [15.71 (4.30) vs. 12.98 (5.49) μg/L; P<0.05 ], the total concentration of PAHs in neonatal blood [ 14.29 (4.25) vs. 11.24 (5.09) μg/L; P<0.05 ] and the placental mtDNA copy number [4.67 (1.18) vs. 4.51 (0.62); P<0.05] were all higher in the heating group than in the non-heating group. Spearman rank correlation analysis revealed that the total concentration of PAHs in the blood of pregnant women and neonates was positively correlated with placental mtDNA copy number ( rs=0.240, P=0.001; rs=0.273, P<0.001 ), and the total concentration of PAHs in the blood of pregnant women was positively correlated with the placental mtDNA copy number in the heating group ( rs=0.245, P=0.014 ), while the PAHs concentration in the neonatal blood was positively correlated with the placental mtDNA copy number in the non-heating group ( rs=0.292, P=0.003 ).
Conclusions
Exposure to PAHs positively correlates with placental mtDNA copy number among pregnant women, and there is a correlation between maternal exposure to PAHs and neonatal oxidative stress.
7.Studies on Pharmacodynamics of Duodongan Oral Solution and Its Acute Toxicity

Objective: To investigate the pharmacodynamics and acute toxicity of Duodongan Oral Solution. Methods: The test of walking time and raising the front limbs of mice Step down test were used to observe the improvement of Duodongan Oral Solution on the spontaneous activity inhibition and memory acquisition disturbance of mice. Results: Duodongan Oral solution could obviously inhibit the spontaneous activity of mice, and its action is strengthened with the increase of dosage. The step down test showed that the oral solution could prolong the latent peroid and decrease the error rate in 5 minites, which indicated that the drug obviously improved memory acquisition disturbance. The acute toxic test indicated that the maximum tolerance dose is 159 times as much as the clinical dose a day. Conclusion: This oral solution has much low toxicity, and is safety for oral administration.
8.Solubility and Apparent Oil/Water Partition Coefficient of Sulfamethazine

OBJECTIVE:To determine the equilibrium solubility and the apparent partition coefficient of sulfamethazine(SM2) in a series of phosphate buffer solutions of different pH so as to provide a basic study for the exploitation of SM2 preparation.METHODS:A series of buffer solutions of different pH were prepared.The apparent solubility was determined by saturation method;Ko/w was calculated with concentration ratio of SM2 in n-octanol and water phase after partition equilibrium.RESULTS:The maximum equilibrium solubility was 1.916 g?L-1 at pH 2 and 1.375 g?L-1 at pH 9,and the maximum of apparent partition coefficient was 3.9070 at pH 8.CONCLUSION:The equilibrium solubility and apparent partition coefficient of SM2 are correlated to pH of the medium.SM2 dissolved preferably when pH6.8,but SM2 was more distributed in the lipid phase and easier to be absorbed by body when pH=3~8.
9.Value of thyroid imaging reporting and data system and shear wave elastography for diagnosis of thyroid microcarcinoma.

OBJECTIVE:
To investigate the value of each single and combined applications of ultrasound thyroid imaging reporting and data system (TI-RADS), ultrasonic shear wave elastography in the diagnosis of thyroid microcarcinoma (TMC).
METHOD:
323 patients with thyroid tiny nodules confirmed by surgery and pathology were analyzed prospectively. Then their TI-RADS and shear wave elastography features were summarized. SWE elasticity indices (mean, minimum and maximum value of interest) of nodules were measured. The surgical pathology results were regarded as the gold standard to calculate the sensitivity, specificity, accuracy, positive predictive value and negative predictive value of TI-RADS, shear wave elastography and their combination in the diagnosis of TMC. The receiver operating characteristic (ROC) curve of TI-RADS, shear wave elastography and combined application of both were drawn. The area under the ROC curve were calculated. Furthermore, the value of combined application of both examinations in the diagnosis of thyroid microcarcinoma (TMC) was investigated.
RESULT:
From 323 nodules,253 were thyroid carcinomas and 70 were benign. Areas under the ROC curves (AUC) of SWE parameters were 0. 648,0. 629 and 0. 605. For SWE, the optimal cut-off value according to ROC curve was SWE_max = 65.50 kPa and SWE_mean = 37.50 kPa. Generally, the sensitivity, specificity, accuracy, PPV and NPV were 88.14%, 82.86%, 87.00%, 94.89% and 65.91% for TI-RADS, and 49.01%, 74.29%, 54.49%, 87.32% and 28. 73% for SWE. There were statistically significant differences in the sensitivity, accuracy, PPV and no statistically significant differences in the specificity, and NPV. The sensitivity,specificity, accuracy, PPV and NPV were 95.26%, 67.14%, 89.16%, 91.29% and 79.66% for combined application of both. The area under the ROC curve of TI-RADS, shear wave elastography and their combination were 0.763, 0.616, 0.804. For nodules ≤ 5 mm, the sensitivity, specificity, accuracy, PPV and NPV were 91.67%, 66.67%, 87.07%, 92.44% and 64.29%. However, for nodules 5-10 mm,the sensitivity,specificity, accuracy, PPV and NPV were 98.50%, 67.44%, 90.91%, 90.34% and 93.55%.
CONCLUSION
The diagnose of TMC can be further improved by combined application of ultra-sonic TI-RADS and SWE.

10.Novel small-molecule CDK2-cyclinA2 inhibitors: design, synthesis, and biological evaluation

Objective · To design and synthesize a series of benzenesulfonamide derivatives, test their inhibitory activity to CDK2-cyclinA2 kinase, and investigate the structure-activity relationship. Methods · Virtual screening was executed via computer-aided drug design according to the ATP binding site in CDK2-cyclinA2 protein crystal. A series of benzenesulfonamide derivatives were designed and synthesized on the basis of the interaction modes between the lead compound and the CDK2-cyclinA2. The biological evaluation of compounds was made through the CDK2-cyclinA2 in-vitro kinase activity detection system. Results · Twenty-nine new benzenesulfonamide compounds were prepared, and their inhibitory activity to CDK2-cyclinA2 was elicited. WZ-026 had the highest inhibitory parameter, which half maximal inhibitory concentration (IC50) was 3.81 μmol/L. Conclusion · By multipurpose utilization of virtual screening, chemical synthesis, and biological activity test, a benzenesulfonamide compound WZ-026 was found, which has great inhibitory activity towards CDK2-cyclinA2. Preliminary structure-activity relationship of compounds was obtained.
