1.Expression of Cytokine in Peripheral Blood of Newborns with Hypoxic-Ischemic Encephalopathy and Its Clinical Significance

Objective To observe the expression of CD_(25),CD_(25) mRNA,interleukin-1?(IL-1?) in peripheral blood of newborns with hypoxic-ischemic encephalopathy(HIE) and its clinical significance.Methods The peripheral blood mononuclear cells(PBMC) of newborns with HIE and normal controls were isolated by routine Ficoll-Hypaque,and the level of CD_(25) induced and non-induced by phytohernagglutinin(PHA) were detected by biotin-streptavidin(BSA),CD_(25) mRNA was detected by real-time PCR and IL-1? in serum was detected by enzyme-linked immunosorbent assay on the first,third and seventh day after birth.The final expressive level of CD_(25) mRNA was based on rate of CD_(25) cDNA and gluceraldehyde phosphase dehydrogenase(GAPDH).Results On the first day,the positive rates of CD_(25) induced and non-induced by PHA and IL-1? were(3.6?1.1)%,(20.9?4.8)%,(17.7?8.2) ng/L,respectively.There were significant differences in HIE group and normal controls(P0.05).Conclusions There is obvious inflammatory(rea)ction in babies with HIE.The main character of PBMC of newborns is all infantile and undifferentiated,which surface sign is infantile,and lower level of CD_(25) is expressed.However,the level of CD_(25) mRNA can be expressed consistently during the course of disease.The immunity and immune regulative response in newborns is gradually built up during the growth and can participate in recondition of brain injury caused by HIE.
2.Efficacy of a second dose measles-mumps-rubella vaccine in children in Xicheng District, Beijing, China in 2016

Objective To analyze the efficacy of a second dose of measles-mumps-rubella (MMR) vaccine in children aged 6 to 7 years in Xicheng District, Beijing, China in 2016.Methods Serum samples were collected in duplicate from 41 children aged 6 to 7 years before and one month after immunization with a second dose of MMR vaccine.ELISA was performed to detect IgG antibodies against measles, rubella and mumps in serum samples.Results The positive rates of antibodies against measles, rubella and mumps in the 41 children before immunization with a second dose of MMR vaccine were 95.12%, 90.24% and 92.68% and the antibody titers were (825.88±499.91), (53.83±32.48) and (697.83±535.82) IU/ml, respectively.The positive rates of the three antibodies were 100%, 97.56% and 100% after revaccination with MMR vaccine and the antibody titers were (1 102.98±356.77), (95.08±26.88) and (3 383.85±1 903.06) IU/ml.Titers of antibodies against measles, rubella and mumps increased significantly following revaccination (P<0.05), while no significant difference in the positive rates of the antibodies was observed before and after revaccination (P>0.05).Conclusion The second dose immunization of MMR vaccine achieves a high successful rate in children in Xicheng District of Beijing.It would be an effective immunization strategy to protect children from measles, rubella and mumps.
3.Immunohistochemical analysis of OTUB1 in 90 cases of ovarian mucinous tumors

Background and purpose:The ovarian mucinous tumor is one of the major subtypes of the ovari-an epithelial cancer. Ubiquitination is one of the main post-translational modifications, which has proven to be involved in tumorigenicity. Deubiquitinase is the protein enzyme that could reverse the process of ubiquitination to affect the initiation and progression of malignancies. This study aimed to analyze the expression and clinical application of deubiquitinase OTUB1 (OTU deubiquitinase, ubiquitin aldehyde binding 1) in ovarian primary mucinous tumors. Methods:This study collected 90 cases of ovarian primary mucinous tumors during 2010-2015 in Obstetrics and Gynecology Hospital of Fudan University, and then collected the clinicopathological information and performed the immunochemistry.Results:Fourteen out of 90 cases were ovarian primary mucinous cystadenoma, 17 were borderline mucinous tumor (intestinal type and intraepithelial carcinoma), and 59 were ovarian primary mucinous carcinoma. The expression rate and intensity of OTUB1 were much higher in malignant cases than those in benign ones (P<0.05). The expression rate and intensity of OTUB1 were much higher in cases with mucinous intraepithelial carcinoma than those in cases with intestinal type borderline mucinous tumor (P<0.05). The expression rate and intensity of OTUB1 increased with the advance of FIGO staging (P<0.05). The expression rate and intensity of OTUB1 were much higher in cases with involved fallopian tubes than those in cases without involved fallopian tubes (P<0.05). The expression rate and intensity of OTUB1were much higher in cases with involved uterus and omentum than those in cases with-out involvement (P<0.05). The expression rate and intensity of OTUB1were much higher in cases with lymph node metastasis than those in cases without involvement (P<0.05).Conclusion:There is significant difference in OTUB1 expression between ovarian primary mucinous carcinoma and benign mucinous cystadenoma. It is highly correlated to FIGO staging and invasion and metastasis of tumor. OTUB1 could be used in differential diagnosis and in monitoring the tumor initiation and progression in ovarian mucinous carcinoma.

Objective To study the function difference of T helper cells(Th)and dendritic cells (DCs) in patients with different hepatitis B virus (HBV) genotypes. AIM: The cytoplasm Ca 2+, expression of Bcl-2 and Bax and germ cell apoptosis of testes in adult male rats with varicocele were observed to investigate the relationship between apoptosis and infertility. METHODS: Thirty-five healthy male Wistar rats were divided into two groups randomly: varicocele group (VG, n=20) and sham operation group (SOG, n=15). Bilateral testes were removed after ten weeks. One part of it was homogenized, the other part was performed for tissue sectioning. The atomic absorption method was used to detect the cytoplasm Ca 2+. The germ cell apoptosis was detected by the TdT-mediated dUTP-biotin nick end labeling (TUNEL) technique. The expression of Bcl-2/Bax was detected by immunohistochemical method. RESULTS: The cytoplasm Ca 2+ in testes of rats with varicocele was decreased obviously. The apoptosis indexes of germ cells and Bax expression in VG were significantly increased as compared with those in the SOG, while Bcl-2 expression was obviously decreased. CONCLUSION: The results showed that apoptosis was increased in testes of VG rats, suggesting that male infertility with varicocele may be caused by some apoptosis agents such as high temperature, testes toxin and oxygen free radical. The cytoplasm Ca 2+ and change of expression of Bcl-2/Bax may contribute to germ cell apoptosis and cause male infertility.
6.A randomized controlled comparison between digital three-dimensional reconstruction and percutaneous balloon kyphoplasty

BACKGROUND:Experience of the surgeon has a direct effect on the operation time and the repair effect of percutaneous baloon kyphoplasty. Digital technology-assisted rehabilitation program opened up new ideas. OBJECTIVE:To compare the differences of operation time and repair effect between digital technology and percutaneous baloon kyphoplasty.
METHODS:A total of 64 patients treated with percutaneous baloon kyphoplasty in the Department of Orthopedics of the Second People’s Hospital of Huaian from January 1, 2013 to November 20, 2014 were enroled in this study. Using a randomized controled method, according to the order of admission, odd number received digital technology-assisted percutaneous baloon kyphoplasty (experimental group). Even number received percutaneous baloon kyphoplasty (control group). Onset age, preoperative visual analogue scale score, operation time, 3-day postoperative visual analogue scale score and the decreased value of visual analogue scale score were compared and analyzed between the two groups.
RESULTS AND CONCLUSION:Significant differences in operation time, 3-day postoperative visual analogue scale score and the decreased value of visual analogue scale score were detected between the two groups (P < 0.05), and above indexes were better in the experimental group than in the control group. These findings confirm that digital technology-assisted percutaneous baloon kyphoplasty could significantly shorten operation time, optimize repair program, obtain ideal repair effect and elevate patient satisfaction.
7.Reconstruction and measurement of a digital dental model using grating projection and reverse engineering.

OBJECTIVEThis work lays the foundation for establishing a digital model database with normal occlusion. A digital dental cast is acquired through grating projection, and model features are measured through reverse engineering.
METHODSThe grating projection system controlled by a computer was projected onto the surface of a normal dental model. Three-dimensional contour data were obtained through multi-angle shooting. A three-dimensional model was constructed, and the model features were analyzed by using reverse engineering. The digital model was compared with the plaster model to determine the accuracy of the measurement system.
RESULTSThe structure of three-dimensional reconstruction model was clear. The digital models of two measurements exhibited no significant difference (P > 0.05). When digital and plaster models were measured, we found that the crown length and arch width were not statistically different (P > 0.05), whereas the difference between the crown width and arch length was statistically significant (P < 0.05).
CONCLUSIONThe reconstruction of a digital model by using the grating projection technique and reverse engineering can be used for dental model measurement in clinic al and scientific research and can provide a scientific method for establishing a digital model database with normal occlusion.

8.Changes in pain threshold and glial cell line-derived neurotrophic factor in rat model of trigeminal neuralgia.

OBJECTIVEThis research aims to study the changes in pain threshold and glial cell line-derived neurotrophic factor (GDNF) in a Sprague Dawley (SD) rat model oftrigeminal neuralgia.
METHODSA total of 36 male SD rats were randomly divided into three groups: operative, sham-operative, and control. In the operative group, a chronic constriction injury (CCI) was caused by placing loose chromic gut ligatures around the right infraorbital nerve (ION). In the sham-operative group, the right ION was subjected to the same procedure, but without ligation. In the control group, the right ION was not subjected to any treatment. The pain thresholds of the three groups were recorded at different times after the operation. The GDNF expression in each group was analyzed via immunohistochemical staining.
RESULTSAn allodynia to mechanical stimulation in the region of the ligated ION was observed starting on the 2nd week after operation. Pain thresholds started to increase gradually from the 6th week and returned to the original level at the 10th to 12th week after operation. Cells that expressed the GDNF markedly increased in number in the operative group with changes observed at different times.
CONCLUSIONWe use chronic constriction injury to the infraorbital nerve (CCI-ION) to establish a trigeminal neuralgia-like animal model in SD rats. GDNF may play a role in regulating pain by promoting the restoration and regeneration of nerve fibers.

10.Clinical analysis of 17 patients with relapsing polychondritis

Objective To enhance the level of the diagnosis and treatment of relapsing polychondritis(RP).Methods Clinical data of 17 patients of RP were studied retrospectively. Results Of the patients,ear was involved in 16 cases, nose in 10 cases, laryngotracheobronchial region in 9 cases, eyes in 8 cases, joints in 4 cases, cardiovascular system in 1 case.Of 17 cases-1 died from respiratory complications. Fifteen patients had recurrence over twice,1 patient had sixth recurrence. Conclusions RP is rare autoimmune system disease, early clinical manifestation are nontypical, misdiagnosis and missed diagnosis. Corticosteroids, immunosuppressive agents, dapsone and surgical operation are used in these patients to control symptoms.
