2.Adrenal cortical and medullar hyperplasia--a retrospective analysis of 6 cases.

The features of the symptoms, laboratory tests and pathological characteristics of adrenal cortical and medullary hyperplasia were studied. In 6 cases of hypercatecholaminenia, plasma norepinephrine (NE), epinephrine (E), catecholamine (CA) and 24-h urinary vanillylmandelic acid (VMA), 17-hydroxycorticosteroid (OHCS) and 17-ketosteroid (KS) were determined. Adrenal glands were examined by CT scan and 131I-MIBG imaging. Pathological examination was performed after operation. The results showed that in 6 cases of hypercatecholaminenia (3 men and 3 women) aged from 34-50 years, the clinical features were just like "pheochromocytoma", for example, episodic headache, perspiration, palpitation, pallor, apprehension, nausea, tremor, anxiety and so on. Plasma levels of CA, NE and E were elevated in all 6 cases. 24-h urinary samples obtained at the onset revealed elevated VMA in 1 case. 24-h urinary cortisol was obviously elevated in all 6 cases. 24-h urinary 17-OHCS, 17-KS was normal. B-type ultrasound, CT, MRI and 131I-MIBG revealed 9 lateral adrenal gland diffuse or nodular enlargement in 6 cases. Pathologic examination showed adrenal cortical and medullary hyperplasia. Clinically, adrenal cortical and medullary hyperplasia resembled "pheochromocytoma". The most significant feature of this disease was both elevated plasma CA and 24-h urinary cortisol obviously. Pathologic examination showed adrenal cortex nodular hyperplasia and medullar diffuse or limit hyperplasia. Whether it is an independent disease or symptoms of the other disease has not final conclusion up till now.

3.The expression of ET-1 and ETA/B receptors mRNA in benign prostatic hyperplasia and its significance

Objective To investigate the expression of endothelin 1 (ET 1) and ETA/B receptors mRNA in prostatic tissues and its clinical significance in patients with benign prostatic hyperplasia (BPH). Methods Immunohistochemical staining and RT PCR were used to detect the expression level of ET 1,ETAR mRNA and ETBR mRNA,respectively.The detection results were analyzed in correlation with the clinical parameters of BPH patients. Results The expression level of ET 1 and ETA/BR mRNA in BPH tissue(integral optical densities,0.94?0.08,0.64?0.08,0.97?0.08,respectively)was higher than that in normal prostatic tissue(0.57?0.06,0.37?0.05,0.51?0.04,respectively).In BPH group,the expressed quantities of ET 1 and ETAR mRNA showed positive correlation with IPSS,prostatic volume,prostatic urethral length,prostatic urethral pressure and maximum urethral pressure of BPH patients,but it showed negative correlation with maximum flow rate and average flow rate of them. Conclusions Increased expression of ET 1 and ETAR might play a role in bladder outflow obstruction(BOO) of the patients with BPH.
4.Effect of Sirolimus-eluting stents implantation on platelet activation under dual anti-platelet therapy

Objective To investigate the changes in platele aciveation in terms of platelet fibrinogen receptor expression(PAC-1),P-selectin(CD62P),Thromboxane B2(TXB2),Endothlin(ET-1),Fibrinogen(FIB)in patients undergoing elective Sirolimus-eluting stents implantation and dual anti-platelet therapy.Methods Forty in-hospital patients with stable coronary heart disease(CHD)were enrolled in this study.Dural anti-platelet therapy with aspirin and clopidogrel were prescribed at least three days before operation to achieve sufficient inhibition of platelet activation in all patients.Peripheral artery blood samples were collected before the procedures,immediately and at 6 hours and 24 hours after the procedure,levels of PAC-1,CD62P,TXB2,ET-1 and FIB were measured.All patients were followed up for 6-12 months.Results Expression of PAC-1 and CD62P was increased significantly after stenting compared with pre-stenting and the peak appeared at 6 hours post-operation(PAC-1,P=0.001,CD62P,P=0.007,PAC1+CD62P,P=0.003).TXB2 was also increased and the peak appeared at 24 hours post operation(P=0.021).Plasma levels of ET-1 and FIB showed no difference before and after stenting.After CAG,expression of PAC-1 and CD62P,levels of TXB2,ET-1 and FIB had no significant differences compared with pre-CAG.No MACE happened in all patients.Conclusion The routine dual anti-platelet therapy cannot suppress platelet activation after elective coronary Sirolimus-eluting stents implantation.Plasma FIB and ET-1 levels were not affected by stents implantation.
5.Diagnosis and Surgical Operation of the Lateral Patellar Compression Syndrome

Objective To explore the diagnosis and therapies of the lateral patellar compression syndrome.Methods This group included 21 cases(30 knees) of lateral patellar compression syndrome,with the average 45 years old(2 males and 19 females of 36~58 years old).All of the patients had obvious Clinical sings and symptoms,whose auxiliary examination was negative.It was almost as same as the Merchant classificasion of the lateral patellar compression syndrome.We adopt the lateral retinaculum release and debridement cartilage defects and drilling.Results All of the definitely diagnosised patients were reviewed with an average of 4 years follow-up visiting(range from 0.5 to 15 years) after operation.The rate of fineness is 96.7% in accordance with Ye Qibin's synthesis estimation for the ailing knee.Conclusion The lateral patellar compression syndrome is a common disease.The lateral retinaculum release and debridement cartilage defects and drilling is a good therapy with minor injury and significant effects for the severe or recurrent attacking lateral patellar compression syndrome.
6.Construction and identification of recombinant lentivirus vector for microRNA-223 overexpression and suppression.

OBJECTIVETo construct microRNA-223 overexpression and suppression lentivirus vectors and determine their effects after infecting oral squamous cell carcinoma (OSCC) cell line.
METHODSLentivirus vectors GV229 and GV232 were cut by the restriction sites of Age I and EcoR I and connected to the target gene, which contained mature microRNA-223 and microRNA-223 oligonucleotide. Real-time polymerase chain reaction (PCR) method was used to detect the microRNA-223 expression level after infecting the recombinant lentivirus vector into the OSCC cell line.
RESULTSThe successful construction of microRNA-223 recombinant lentivirus vectors was confirmed by the PCR method and DNA sequencing. HN-30 cell infected with microRNA-223 overexpression vector showed a significant increased in microRNA-223 expression, whereas HN-30 cell infected with microRNA-223 inhibitor vector suppressed microRNA-223 expression.
CONCLUSIONThe microRNA-223 overexpression and suppression lentivirus vectors are successfully constructed. These vectors could alter the expression level of microRNA-223 in OSCC cell line significantly, and provide a stable cell line for functional studies in the future.

7.Effects of environmental intervention on growth hormone and insulin-like growth factor-1 in very low birth weight infants in NICU

Objective To investigate the effects of environmental intervention on growth hormone (GH) and insulin-like growth factor 1(IGF-1) in very low birth weight infants in NICU.Methods 60 cases of very low birth weight infants were randomly divided into the experimental group and control group (30 cases in each group) in NICU from June 2012 to June 2013.By adjusting the light,reducing the noise,focusing the nursing time,the levels of growth hormone and insulin-like growth factor-1 were detected and the growth indexes were measured including body weight,length,and head circumference on the 7th day and 14th day.Results The indicators of growth and development in the experimental group on the 7th day and 14th day after birth were significantly better than those of the control group.The IGF-1 level in the experimental group on the 7th day was significantly higher than that of the control group.The GH and IGF-1 level in the experimental group was significantly higher than that of the control group.Conclusions Effective environmental intervention in NICU can improve the GH and IGF-1 lev-el of very low birth weight infants and improve their quality of life.
8.Relationship between motor complications of Parkinson's disease and phosphorylated GluR1Ser845

Objective To explore the relationship between motor complications of Parkinson's disease(PD)with chronic L-dopa treatment and striatal phosphorylated GluR1Ser845.Methods The hemi-parkinsonian rat model was produced by injecting stereotaxically 6-OHDA to right medial forebrain bundle(MFB).Then the hemi-parkinsonian rat was treated intraperitoneally with L-dopa methylester(25 mg?kg-1?d-1,twice one day)for 22 days and rotational duration and frequency of off period were respectively estimated.After they were sacrificed,their subcellualr distribution of GluR1 and GluR1Ser845 phosphorylation was observed with immunofluorescence and Western blot.Results After the chronic treatment with L-dopa methylester,PD rats displayed shortened rotational duration and increased frequency of off period,which was similar to fluctuations of the symptoms and on-off phenomenon in PD patients.In the lesioned striatum of PD rats,the amount of GluR1 and phosphorylated GluR1Ser845 in striatal membrane was reduced to 73.0%?4.8% and 42.0%?5.6%,respectively,compared with those non-lesioned.After chronic treatment of PD rats with L-dopa,the amount of GluR1 and phosphorylated GluR1Ser845 in striatal membranes were increased to 104.0%?5.5% and 112.0%?3.4%.These unique changes occurred only in parvalbumin-positive interneurons.Conclusions These findings suggest that subcellular distribution and phosphorylation state of GluR1Ser845 in parvalbumin-positive interneurons may play a significant role in the pathogenesis of motor complications of chronic L-dopa treatment for PD.
