Objective: To explore the effects of Lycium barbarum polysaccharide (LBP-X) on IL-2 and TNF-? gene expressions in human peripheral blood mononuclear cells. Methods: RT-PCR (reverse transcription-polymerase chain reaction) was used to detect the gene expressions of IL-2 and TNF-?. Results: LBP-X at 5-40 mg/L upregulated the levels of IL-2 and TNF-? mRNA in human peripheral blood mononuclear cells after optimal time culture. Conclusion: The immunoenhancing effect of Lycium barbarum polysaccharide may be associated with its stimulation of IL-2 and TNF-? gene expressions.

ObjectiveTo investigate the influence of overtaking of liquor on the expression of caspase-3 in the mouse seminiferous tubule.MethodsAdult mice were given liquor through the stomach.The dose was 4g/kg body weight per day for 5 continuous days.Hybridization in situ was performed for detecting the caspase-3 expression in the mouse seminiferous tubule.ResultsIn the control group,there were few Caspase-3 positive cells in the mouse seminiferous tubule.In the group of five days taking liquor,the caspase-3 positive cell count increased obviously.In the group of ten days taking liquor,the caspase-3 positive cell count was maximum.ConclusionAlcohol could induce the apoptosis of spermatogenic cell.

Objective To investigate the pro-apoptotic effects of an ?1-adrenergic inhibitor,doxazosin,in HeLa cells and the role of transcription factor activator protein-2? (AP-2?) in the process.Methods The HeLa cells were treated with doxazosin (0,10,40 or 60 ?mol/L) for 16 h,and cell apoptosis was detected by flow cytometric analysis.After doxazosin (60 ?mol/L) treated the HaLa cells,the cells transfected with AP-2? overexpressing constructs or an antisense oligonucleotide against AP-2? for 16 h,the apoptosis was analyzed with FCM.The expression of AP-2? and caspase-3 in above 3 groups of cells was detected by relative quantitative RT-PCR and Western blot analysis,respectively.Results Doxazosin increased the apoptotic rate and total cell death rate of the HeLa cells,and upregulated the expressions of AP-2? and caspase-3 in a dose-dependent manner.Overexpressing AP-2? improved the increased apoptotic rate and total cell death rate induced by doxazosin,and enhanced the increased expression of caspase-3.Whereas doxazosin-induced apoptosis and the total cell death in HeLa cells were decreased by antisense AP-2?,and antisense AP-2? in part abolished the increased effects of doxazosin on caspase-3 expression.Conclusion Doxazosin induces apoptosis in HeLa cells in a dose-dependent manner,and transcription factor AP-2? is functionally involved in this process.

The hereditary diffuse gastric cancer (HDGC) is associated with CDHI (E-cadherin gene)germline mutations, and accounts for about one-third of the familial gastric cancer (FGC) cases. The other two thirds FGC cases which are not fit the diagnosis standard of HDGC remain lack of clear molecular diagnosis basis. The current disposal of prophylactic gastrectomy in FGC according to E-cadherins genetic counseling, has clinical significance only in part of HDGC.

Objective: To study the antitumor effect of Lycium barbarum polysaccharide (LBP X) and its immunity regulating effect on S 180 bearing mice. Methods: S 180 bearing mice were used as animal model. The effect of LBP X on tumor weight and immune function was observed. Results: LBP X could inhibit the growth of S 180 effectively and increase thymus index, the macrophage phagocytic function, anti SRBC antibody secreted by the spleen cells, lymphocyte proliferation and CTL activity and decrease LPO markedly in S 180 bearing mice. Conclusion: The mechanism of tumor inhibition by LBP X was due to enhance the immune function.

Objective To observe the infection status of Helicobacter pylori (HP) in gastric cancer patients and to explore its relationship with the clinical characteristics of gastric cancer. Methods Paraffin sections of surgical specimens from 794 patients who underwent gastrectomy in Changzheng Hosptial or Changhai Hosptial were detected with the modified Giemsa staining to identify the status of HP infection. The association of HP infection status with the clinicopathological features was analyzed by Chi-square test. The relationship between HP infection status and prognosis of gastric patients was analyzed by Kaplan-Meier method and proportional hazard regression model. Results HP infection was significantly associated with Lauren classification, differentiation degre e, and lymph-node metastases of gastric cancer (P<0. 05). The median survival time of the HP positive patients (68. 5 months) was significantly longer than that of HP negative patients (36. 4 months) (P < 0. 003). For patients undergoing curative surgeries, those with HP infection had a better prognosis compared with those without HP infection (P=0. 034). For patients with intestinal-type gastric cancer, those without HP infection had a poorer prognosis compared with those with HP infection (P = 0. 006); there were no differences in patients with diffuse-type gastric cancer (P = 0. 922). Conclusion Our results indicate that HP infection status is not an independent factor for prognosis of gastric cancer patients. HP positive patients with intestinal-type gastric cancer may have a better prognosis.

Objective To observe the clinical efficacy of midnight-noon ebb-flow needling based on individualized timing in treating senile chronic gastritis. Method Seventy-eight elderly patients with chronic gastritis were randomized into a treatment group of 39 cases and a control group of 39 cases. The treatment group was intervened by midnight-noon ebb-flow needling based on individualized timing, while the control group was by midnight-noon ebb-flow needling based on local mean solar time, 10 d as a treatment course. The clinical efficacies were observed after 2 treatment courses. Result The total effective rate was 94.9% in the treatment group versus 89.7% in the control group, and the difference was statistically insignificant (P>0.05); the recovery rates were respectively 46.2% and 28.2%, and the difference was statistically significant (P<0.05). Conclusion Midnight-noon ebb-flow needling based on individualized timing can produce a significant efficacy in treating senile chronic gastritis, and is worth promoting in clinic.

Objective To investigate the effects of 1 ,25-dihydroxyvitamin D3 on the expression of urinary Nephrin in patients with diabetic nephropathy .Methods According to albumin excretion rate ,40 type-2 diabetic nephropathy patients were divided into 2 groups ,10 healthy volunteers served as control .The treatment groups received 1 ,25-dihydroxyvitamin D3 0 .25 μg/d for 12 weeks .The levels of blood calcium ,phosphorus ,serum creatinine ,24-hour urinary protein excretion were detected .Enzyme-linked immunosorbent assay was used to detect urinary Nephrin in different groups .Results The expressions of urinary Nephrin were sta-tistically different among NC group ,DN1 group and DN2 group .1 ,25-dihydroxyvitamin D3 treatment significantly decreased the ex-pression of urinary Nephrin in DN1 patients ,whereas no statistically changes were observed in DN2 group .Conclusion In diabetic nephropathy patients ,urinary Nephrin is supposed to be effective an marker of disease progression .1 ,25-dihydroxyvitamin D3 may be effective to prevent podocyte injury in early-stage diabetic nephropathy .

Objective　To prepare monoclonal antibody (McAb) pool against Bacteroides fragilis and Clostridium perfringens and evaluated its effect in rapid diagnosis in surgical infection. Methods　After the preparation of monoclonal antibodies against B. fragilis and C. perfringens, 191 specimens collected from infectious patients was detected with indirect immunofluorescent antibody (IFA) and enzyme labeled antibody (ELA). The results were compared with routine cultural method (CM). Results　Among the 191 specimens, 53 (27.7%), 55 (28.8%) and 25 (13.1%)strains of B. fragilis were detected with IFA, ELA and CM respectively, and for C.perfringens, 12(6.3%), 11(5.8%), and 6(3.1%) strains were found. The detectable rate of anaerobic bacteria by IFA and ELA was higher than by CM, but no difference was found between these 2. Conclusion　The self-made McAb pool against B.fagilis and C.perfringens might be a rapid, specific, sensitive and simple McAb pool for detecting B.fragilis in surgical infection and C.perfringens in wartime.

Objective To explore the changes of the cardiac enzymes in the critical patients after operation and to analyze their relationship with the patient's condition level and the prognostic.Methods Fittyseven critical patients after operation were enrolled to observation group,and 55 small operation patients acted as control group.Their cardiac enzymes in three days after operation were detected and compared.Results The cardiac enzymes of the observation group increased significantly when compared with the control group(aspartate aminotransferase (AST) (153.1 ± 211.0) U/L vs (54.7 ± 39.0) U/L,t =3.404,P ＜ 0.01) ; lactic acid dehydrogenase(LDH) (381.0 ± 216.1) U/L vs (194.6 ± 75.3) U/L,t =6.050,P ＜ 0.01) ; creatine kinase (CK) (882.9 ±630.7) U/L vs (130.1 ± 71.8) U/L,t =8.796,P ＜0.01) ;creatinine kinase,MB isoenzyme (CK-MB)(314.3 ± 164.2) U/L vs (164.9 ± 87.7) U/L,t =5.979,P ＜ 0.01) ; α-hydroxybutyrate dehydrogenas(α-HBDH) (410.1 ± 327.6) U/L vs (27.8 ± 36.3) U/L,t =8.605,P ＜ 0.01) ; The fatality ratio was 24.6％ (14/57) in the observation group.The cardiac enzymes of the survival group existed significant difference compared with the death group(AST:(81.3 ± 40.5) U/L vs (373.6 ±342.1) U/L,t =5.585,P ＜0.01) ;LDH:(303.6±134.0) U/Lvs (618.6±250.1) U/L,t=6.064,P＜0.01) ;CK:(658.5±328.0)U/Lvs (1572.0±829.4) U/L,t =6.001 P＜0.01) ;CK-MB:(303.2±184.3) U/Lvs (738.6±445.6)U/L,t=5.242,P＜0.01) ;α-HBDH:(254.1±118.6) U/Lvs (499.3 ±147.6) U/L,t=6.320,P＜0.01).Conclusion Elevated myocardiac enzymes are common in the critical patients after operation.The change of the cardiac enzymes is related with the degree of disease and the wound.
