1.Protective M Against Candida albicans :an Experimental Study

Objective To study the protective effect of mon oclonal antibodies(McAb)against Can-dida albicans in systemic candidiasis.Methods Monoclonal antibody was produced wi th hybridoma technique.The effect of McAb on experimental mo use systemic candidiasis was observ ed,expecially on the following as-pects:the survival time of mice,colony forming unit(CFU)of Candida albicans and the histopathologic changes.Results Three types of McAb against the cell w all antigen of Candida albicans were generated,which were designated as 1B5,3E8and 4C7.P rolonged survival time,decreased C FU of Candida albicans in kid-neys,liver and brain,and alleviate d histopathologic changes could be s een in experimental mice treated wit h McAb 1B5and 3E8.McAb 1B5could recog nize a cell wall component of Candida albicans(MW:32000),and inhibit the adherence of Candida albicans to human buccal epithelial cells and umbilical vein endothelial cells.Conclusion McAbs 1B5and 3E8,are the protective monoclonal antibodies against Candida albicans,which inhibit the adherence of Candida albicans to epithelial cells and endothelial cells,thus reducing the invasiveness of the pathogen.
2.Effects of Hedgehog on proliferation and apoptosis of breast cancer cells and its influence in Cx32 and Cx43 expressions

Objective:To study the effects of Hedgehog signal transduction pathway on the cell proliferation, apoptosis and connexin 32 (Cx32)and connexin 43 (Cx43)membranous distribution of breast cancer cells,and to explore its mechanism in the cell proliferation and metastasis of breast cancer.Methods:The breast cancer MCF-7 cells at logarithmic growth period were divided into cyclopamine groups and blank control groups. The MCF-7 in cyclopamine groups were treated with 5,10,20,30 and 40μmol·L-1 for 24,48 and 72 h;MTT assay was applied to detect the inhibitory rate of proliferation of MCF-7 cells. After the MCF-7 cells were treated with 0 (negative control group)and 25μmol·L-1 cyclopamine for 48 h,flow cytometry was employed to determine the apoptotic rate and to analyze membranous distribution of Cx32 and Cx43 in the MCF-7 cells.Results:Compared with blank control group,the inhibitory rates of proliferation in cyclopamine groups were increased (P<0.05), and the inhibitory effect of proliferation was increased with the increasing of cyclopamine doses and prolongation of treatment time.After treated with 25μmol·L-1 cyclopamine,the apoptotic rate of MCF-7 cells was higher than that in blank control group (P<0.05).The positive expression rates of Cx32 and Cx43 48 h after treatment were higher than those in negative control group (P<0.05).Conclusion:Hedgehog signal transduction pathway can inhibit the apoptosis and mediate membranous distribution of Cx32 and Cx43 in breast cancer cells.
3.Medication Analysis of 564 Cases with Cataract Extraction

OBJECTIVE:To get to know the preoperative and postoperative medication of patients with cataract.METHODS:The preoperative and postoperative medication of 564 cases with cataract extraction in our hospital from September 2004 to October 2004 was analyzed statistically.RESULTS:Antibacterial eye gutta was mainly used before cataract extraction while compound eye gutta containing antiseptic drugs and glucocorticosteroid were chiefly used after cataract operation.CONCLUSION:The preoperative and postoperative medication for patients with cataract in our hospital was reasonable.
4.Clinicopathological analysis on angiolymphoid hyperplasia with eosinophilia

Objective To analyze the clinicopathological features of angiolymphoid hyperplasia with eosinophilia (ALHE). Methods The pathological specimens of 7 cases of ALHE collected in our department from 1950 to 1999 were sectioned, stained and observed. Results There were 3 pathological characteristics in ALHE: ①massive hyperplasia of capillaries in the dermis; ②the endothelial cells proliferated and swelled, projecting into vascular cavity like tombstones; ③mixed infiltration of lymphocytes and eosinocytes in the vessels. Conclusion ALHE is a disease with local benign proliferated vessels, whose etiology and pathogenesis is still unknown. It is necessary to grasp the pathological changes of ALHE to distinguish it from other diseases.
5.Temperature gradient and elasticity gradient over the microwave-heated region in porcine liver in vitro

Objective To explore the characteristics of changes in temperature and stiffness within the microwave-induced ablation region.Methods A type of Thy-ablation microwave antenna was used upon a design of various combination of ablation duration and power to induce heated region in samples of fresh porcine liver.Three special sites were set to assess the corresponding temperature and stiffness in each heated region.The temperature was continuously measured by using electronic thermometer with microwave on going,while stiffness was determined 5min after the ending of ablation by using real-time ultrasonic elastography,strain ratio,a quantitative indicator.Results The SR values and temperature determinations decreased progressively from the central to the peripheral area of the heated region.There was a linear regression between the temperatures (X) and SRs (Y),with Y =0.666 X-37.17.A significant correlation exists between X and Y,with the correlation coefficient being 0.956(P <0.001).The stiffness and temperature at the central sites are variable with the changes of both ablation time and power,but little changed at the peripheral sites of heated-region,which we name a phenomenon due to the ending effect of microwave propagation.Conclusions Temperature and stiffness increase considerably following microwave radiation but they are heterogeneously distributed within the ablated region in porcine liver.The characteristic gradient changes are confirmed of both temperature and the elasticity.It is presumable that good recognition and understanding and full utilization of these characteristics are conducive to reasonable control of tumor microwave ablation therapy.
6.Elastographic characterization of microwave-induced lesions in porcine liver

Objective To investigate the elasticity reduction of liver tissue due to microwave ablation and the relevance to the histographic damages. Methods An experimental study using fresh porcine liver was designed. Elasto-ultrasonography scanning both in color display and strain ratio calculations was conducted before and 5 min after microwave ablation ( 2450 MHz) in manner of antenna insertion under ultrasound guidance to determine the alterations of the liver elasticity, in correspondence with the histopathologic assessment of each ROI. Results Elasto-ultrasonography showed a significant elasticity reduction and hardness augment of the targeted liver tissue and the corresponding histopathology revealed increases in the amount of massive coagulative necrosis and coking of liver cells after microwave irradiation,in proportional to the applied field power and working time. Conclusions Elasto-ultrasonography helps to demonstrate microwave-induced lesion in porcine liver got rapidly hardened. It is possible to estimate the tissue necrosis to the changing of tissue hardening.
8.Transduction efficiency of green fluorescent protein gene in leukemia cells K562

Objective To investigate the gene transduction efficiency of lentiviral vector in leukemia cells to provide key basis for leukemia gene therapy. Methods A third-generation self-inactivating(SIN) lentiviral vector system based on human immunodeficiency virus type 1(HIV-1) was used to improve transduction efficiency.The transduction efficiency of the HIV-1-based vector was compared directly with the moloney murine leukemia virus(MLV) SIN vector in human leukemia cell line K562.The expression of green fluorescent protein(GFP) in cells was observed by fluorescence microscopy and flow cytometry(FCM) to detect the percentage of gene trasduction cells.Results The GFP expression in K562 cells was observed qualitatively by fluorescence microscopy.At the same gene transduction conditions,the GFP marker gene expression intensity and GFP positive cells in leukemia cells transduced with HIV vectors were significantly higher than those transduced with MLV vectors.Initial transduction efficiencies were almost 100% for the HIV and less than 40% for the MLV vectors. Objective To study the mechanisms of Chinese medicine Euphorbia kansui induced liver damage. Methods 40 mices were selected and radomly divided into four groups according to the dose of Chinese medicine Euphorbia kansui used,there were 4 g/kg group(1/8 LD50 ),2 g/kg group(1/16 LD50),1 g/kg group(1/32 LD)50,and control group,which were given saline. After continuous administration for 21 d by ig,the body weight and liver coefficient of mice in all groups were determined,liver histological changes were detected by HE staining. The serum enzymatic activity of alanine aminotransferase (ALT)and aspartate aminotransferase (AST),the levels of superoxide dismutase (SOD)and glutathione peroxidase (GSH-Px),the content of malondialdehyde (MDA),superoxide anion radical (O2-·)and hydroxyl radical (OH·)were all measured by ultraviolet spectrophotometry. The changes of reactive oxygen species (ROS)content were detected by flow cytometry. Results With the increase dose of Chinese medicine Euphorbia kansui,the body weight of rats were decreased,and liver coefficient were increased(P<0.05). The serum ALT,AST activity were significantly increased,SOD and GSH-Px activity were significantly decreased(P<0.05),MDA content were increased significantly,radical O2-·, OH·and intracellular ROS levels were all increased (P<0.05 ). Conclusion Chinese medicine Euphorbia kansui induced liver injury though lipid peroxidation.
10.Androgen receptor up-regulates EphA3 expression in androgen-dependent prostate cancer cell lines

Objective To evaluate the relationship between liver cell type A receptor (EphA) expression and androgen receptor (AR) signaling in androgen-dependent prostate cancer cells. Methods RT-PCR and Western blot assay were used to determine mRNA and protein levels of EphA3 and AR in prostate cancer LNCaP and 22Rv1 cells, respectively. The variations of EphA3, AR and prostate specific antigen (PSA) expressions were also measured in these cells after dihydrotes?tosterone (DHT) treatment for 48 h. The constructed EphA3-Luc (-789-+146) luciferase reporter plasmid was co-transfect?ed with pcDNA3.1(+)-AR or siAR in 22Rv1 cells to analyze the effects of different AR expression levels on EphA3 tran?scription activity. Results The expression pattern of EphA3 was similar to AR, showing a lower level in prostate stromal cell line WPMY-1 and a higher level in prostate cancer cell lines LNCaP and 22Rv1. When stimulated with 10 nmol/L DHT, the expression levels of AR, PSA and EphA3 were significantly increased in 22Rv1 cells, and the protein levels of these genes were also increased in LNCaP cells. Moreover, AR expression levels markedly influenced the activity of EphA 3 pro?moter. Conclusion AR up-regulates EphA3 expression by increasing the activity of EphA3 promoter.
