1.STUDIES ON THE STRUCTURE,PROLIFERATION AND DISTRIBUTION OF MITOCHONDRIA DURING SPERMATOGENESIS IN MICE AND RATS

This paper presents the observation on the structure,quantity,distribution andmembrane characteristics of mitochondria in spermatogonia,spermatocyte,spermatidand mature sperm during spermatogenesis.They were examined by electron micro-scope (ultrathin section and freeze fracture).Conclusions obtained are as follows:1.There exists regular changes for the proliferation,development and distribu-tion of mitochondria.(a)Quantity of mitochondria:total number of mitochondriain spermatogonia are 80-120 for one cell.They increase to 400-500 in primary sper-matocyte.Spermatid has 200-300 mitochondria.(b)Distribution of mitochondria.Mitochondria in spermatogonia distribute randomly.There are several groups ofmitochondria in spermatocyte.During spermatogenesis mitochondria moved fromhead to back side.In Golgi phase and cap phase spermatid,mitochondria situatedunder the plasm membrane,in mature sperm they arrange regularly outside the densefiber of middle piece of tail.(c)Structure and development:Mitochondria inspermatogonia are round and oval.Cristae arrange parallelly.At spermatocyte stagemitochondria are proliferated by dividing itself.Newly formed mitochondria haveno cristae.In mature sperm mitochondria developed well,but arrangement ofcristae is not regular.2.Membrane of mitoehondria.There are three laminar structure for mitochon-drial membrane.It is thinner than plasm membrane.Protein particles in outermembrane are more than inner membrane.There are a few protein particles in cristae.3.The changes of mitochondria have close relationship with spermatogenesis.(a)Mitochondria proliferate itself rapidly in spermatocyte to match with enlarge-ment of cellular size and content:(b)Mitochondria situated near the Golgi complexin Golgi phase spermatid,after cap formation they moved to tail side to support thetail formation.(c)In mature sperm mitochondria arranged regularly in middlepiece,so sperm can swim quickly.
2.Progress of non-small cell lung cancer in the elderly patients

At present,there is no unified standards of treatment in elderly advanced non-small cell lung cancer (NSCLC),but to elderly patients with higher KPS scores,radiotherapy or chemotherapy alone has gradually become the main treatment measures.The concurrent chemoradiotherapy can benefit to part of the elderly patients,and it is worth further clinical research.Targeted therapy has exactly effect,and the adverse effect is mild,which provides a new treatment model for elderly advanced NSCLC patients.However,the hospital costs are relatively high,which can not be accepted by ordinary family.So targeted drugs and the model of combination treatment can not be applied extensively.
3.Application of the Smart Phone Based SSL VPN Virtual Channel Technology in the Direct Network Report of Epidemic Situation of Infectious Diseases

The paper introduces the definition of smart phone,operating mobile communication network and hidden danger about safety,and definition and advantages of SSL VPN,states the application of SSL VPN technology in the direct network report of epidemic situation information of infectious diseases combining with SSL VPN technology and remote connection,analyzes the work principle and specific operation process,achieves safety login to the China Information System for Disease Prevention and Control with smart phones,and completes normal business data processing.
5.Long-pulsed 1064 nm laser combined with triamcinolone in treatment of keloids

Objective To evaluate the clinical efficacy of long-pulsed 1064 nm Nd ∶ YAG laser combined with intralesional injection of triamcinolone in the treatment of keloids.Methods A total of 78 cases were randomly divided into two groups.The treatment group was treated with long-pulsed 1064 nm Nd ∶ YAG laser combined with intralesional injection of triamcinolone.The control group was only given intralesional injection of triamcinolone.The efficacy and adverse reactions were determined at the end of the first and the second course,respectively,and follow-up for one year was conducted in order to observe the recurrence.Results When the first course was over,cure rate of 21.95 % and efficiency rate of 73.17 % were observed in the treatment group,while they were 10.81% and 45.95 %,respectively,in the control group; the efficacy rate was statistically significant different in these two groups (P<0.05).After two courses,cure rate of 70.73 % and efficiency rate of 100 % were observed in the treatment group while they were 45.95 % and 83.78 %,respectively,in the control group,showing that both of the rates were statistically higher in the treatment group (P<0.05).Conclusions Compared to intralesional injection of triamcinolone,long-pulsed 1064 nm Nd ∶ YAG laser combined with injection of triamcinolone has a more satisfactory effect on keloids.
6.Analysis on drug-resistant genes and homology in nosocomial infection of carba penem-resistant Acinetobacter baumannii

Objective To analyze the drug resistance genes and homology in nosocomial infection caused by carbapenem‐resistant Acinetobacter baumannii(CRAB) .Methods Forty strains of CRAB were collected and the minimal inhibitory concentration (MIC) was detected by the agar dilution method .The main carbapenemase genes were detected by PCR .The PCR products were performed the DNA sequencing .The enterobacterial repetitive intergenic consensus (ERIC)‐polymerase chain reaction(PCR) was performed to analyze the genotypes and homology of these strains .The plasmid conjugation experiment was carried out to investigate the trans‐mission of resistant genes .Results Forty strains of CRAB maintained the good sensitivity only to polymyxin B (100% ) and cef‐operazone/sulbactam(57 .5% ) .All of them had different degrees of resistance to other drugs .The OXA‐23 and OXA‐51genes were detected by PCR amplified in all of the CRAB strains .The homology analysis showed that the strains could be divided into 4 types according to 80% of cluster similarity coefficient ,the type A1 (19 strains) was the major epidemic strain ,moreover the various de‐partments had cross infection ;the plasmid conjugation experiment failed .Conclusion There is an endemic spread of CRAB in our hospital and its resistant mechanism is mainly related with OXA‐23 gene expression ;in the rational antibiotics use at the same time , the isolation and prevention measures should be done well for avoiding the hospital cross infection .
7.Effects of AMP-activated protein kinase on HMGB1 release from PC12 cells after oxygen-glucose deprivation and reoxygenation and its mediated inflammatory response in BV2 cells

Objective To investigate the effects of adenosine monophosphate-activated protein kinase (AMPK) on high-mobility group box 1 (HMGB1) release from PC12 cells after oxygen-glucose deprivation and reoxygenation (OGD/R) and its mediated inflammatory response in BV2 cells.Methods PC12 and BV2 cells were cultured,respectively.The PC12 cells were used to induce a model of oxygen glucose deprivation for 12 h and reoxygenation for 24 h.After giving 5-aminoimidazole-4-carboxamide (AICAR) 5,50 and 100 μmol/L as well as Compound C 0.1,1 and 10 μmol/L activation or inhibition of AMPK phosphorylation,respectively,methyl thiazolyl tetrazolium (MTT) was used to detect the PC12 cell activity.Enzyme-linked immunosorbent assay was used to detect the HMGB1 release level in the PC12 cell culture media.After OGD/R in each group,the PC12 culture media were acted on normal cultured BV2 cells for 24 h respectively.Westem blotting and Enzyme-linked immunosorbent assay were used to detect the NFκB inhibitory protein (inhibitor of NFκB,IκB) phosphorylation level and TNF-α release level in BV2 cells,respectively.Results After OGD/R,the PC12 cell activity was decreased significantly (68.84%±6.60% vs.100.04% ± 8.82%;P < 0.01);the AMPK phosphorylation level was increased significantly (1.95 ±0.39 vs.1.00 ±0.20;P<0.05),and the extracellular HMGB1 release was increased significantly (287.66 ± 26.42 pg/μl vs.53.05 ± 9.11 pg/μl;P < 0.01).Compared with the OGD/R group,AICAR 100 μmol/L significantly increased the survival rate of PC12 cell after OGD/R (78.6% ± 3.75% vs.68.84% ± 6.60%;P < 0.05),promoted AMPK phosphorylation (3.32 ± 0.66 vs.1.95 ± 0.39;P < 0.01),and reduce the release of extracellular HMGB1 (164.06 ± 12.77 pg/μl vs.287.66 ± 26.42 pg/μl;P <0.01).In contrast,Compound C 10 μmol/L significantly reduced the cell survival rate of PC12 (40.44% ±3.79% vs.68.84% ±6.60%;P <0.01),inhibited AMPK phosphorylation (1.07 ± 0.21 vs.1.95 ± 0.39;P<0.05),and increased the release of HMGB1 (337.97 ± 18.9 pg/μlvs.287.66 ± 26.42 pg/μl;P<0.01).The conditioned medium from the AICAR 100 μmol/L group significantly inhibited IκB phosphorylation (1.68 ±0.51 vs.3.09 ± 0.10;P < 0.05) and reduced the release of TNF-α (669.53 ±38.58 pg/μlvs.841.76 ± 45.82 pg/μl;P< 0.05) in BV2 cells.The conditioned medium from the compound C 10 μmol/L group significantly promoted IκB phosphorylation (4.98 ± 1.24 vs.3.09 ± 0.10;P <0.01) and increased the release of TNF-α (1 035.32 ± 128.06 pg/μl vs.841.76 ± 45.82 pg/μl;P <0.05) in BV2 cells.Conclusions Promoting AMPK phosphorylation activation may reduce the release of HMGB1 from PC12 cells after OGD/R,and inhibit its mediated NF-κB inflammatory pathway and reduce the release of TNF-αin BV2 cells,and thus reducing neuroinflammatory injury.On the contrary,inhibiting AMPK phosphorylation may promote the release of HMGB1 from PC12 cells after OGD/R and aggravate its mediated inflammatory reaction in BV2 cells.
8.Effects of NMDA receptor on glucocorticoid receptor mRNA levels in hippocampus of rats after scald burn stress

Objective To investigate the changes of glucocorticoid receptor (GR) gene expression in the hippocampus of rats following scald burn stress and the role of N methyl D aspartate (NMDA) receptor in this change. Methods Adult male Wistar rats inflicted with 30% TBSA full thickness scalding were applied as severe scalding stress model. GR mRNA levels in the hippocampus were detected with RT PCR. Results A significant decrease of GR mRNA levels was observed in the hippocampus 2 h after the scalding stress. The decrease could be inhibited when MK 801, an NMDA receptor antagonist, was administered prior to stress, and be augmented with the administration of NMDA, an NMDA receptor agonist, but not be affected by normal saline. Conclusion NMDA receptors are involved in the scalding stress induced down regulation of GR gene expression in the rat hippocampus.
9.Mini-invasive surgical treatment of acute Achilles tendon rupture by Achillon

Objective To evaluate the clinical efficacy of Achillon,a novel guide for Achilles tendon suture produced by Newdeal Ltd in France,in the mini-invasive surgical treatment of acute Achilles tendon rupture. Methods Between December 2010 and March 2011,7 patients(7 feet) with acute Achilles tendon rupture were treated in our department.They were 5 males (5 feet) and 2 females (2 feet),with an average age of 34.2 years (range,21 to 53 years).The interval between injury and operation ranged from 1 to 11 days(3.8 days on average).A longitudinal incision approximately 2.0-3.5 cm in length was made around the ruptured Achilles tendon for minimally invasive repair after insertion of the Achillon.Postoperative rehabilitation was carried out. Results All the wounds healed at the first stage.All patients were followed up for 5 to 11 months, with an average of 7.2 months. All patients regained normal weight-bearing walking 3 months after operation without any re-rupture. According to the ankle-hindfoot scoring system of American Orthopaedic Foot and Ankle Society (AOFAS),they scored 93.2 points.By the visual analogue scale(VAS),they scored 1.2 points at the operation site. Conclusion The surgical treatment of acute Achilles tendon rupture with Achillon has advantages of mini-invasion,a low rate of incision problems and quick function recovery.
10.Effect of Yuzhenxifeng Decoction on the brain ferritin in PD mouse models

Objective To study the effect of Yuzhenxifeng Decoction on the brain ferritin in PD mouse models .Methods All ex‐perimental animals were devided into 4 groups:the control group ,the model group ,the positive drug group and TCM group .Prepare PD model mice with MPTP ,then use the immunohistochemical staining technique to observe the change of expression of Fn .Results the results showed that compared with the model group ,movement coordination disorder of rats in TCM group were relieved ;the Fn level of the model group on 6th ,13th ,20th were higher than control group(P<0 .05) ,and were 2 .21 times ,1 .15 times and 0 .36 times higher respectively .Compared with model group mice ,expression of Fn were enhanced in the treatment group .Conclusion Yuzhenxifeng Decoction can improve the expression of Fn in the brain ,which provide the basis for further study on mechanism of the treatment of Parkinson′s disease .
