Objective To study the effect of ATRA and IFN? on growth and differentiation of SH-SY5Y cell line and the possible molecular mechanisms.Methods The expression of TrkA mRNA was detected by RT-PCR.Trypan blue exclusion was used to detect the antiproliferative effect of ATRA,IFN?and ATRA +IFN? on SY5Y cells.Morphologic changes were observed using phase-contrast microscopy.Results TrkA mRNA was upregulated by ATRA and IFN?.ATRA and IFN? could induce morphological differentiation and inhibit growth of SY5Y cells.The combination of ATRA+IFN? enhanced the induced differentiation and growth inhibition of SY5Y cells.Conclusion Synergistic effects of ATRA and IFN? on growth inhibition and inducing differentiation in SY5Y cells are observed and this may result from upregulating the expression of TrkA mRNA.

Perioperative pain management play a critical role in rapid recovery and long term outcome in patient undergoing total knee arthroplasty.With the advancement of peripheral nerve block, technique such as femoral nerve block, sciatic nerve block, abductor canal block have play an important role in perioperative pain management.Local infiltration analgesia have also gained popularity.The ultimate goal of perioperative pain management is to ensure analgesia effect and maintain good motor function of lower extremity.We are still in searching of a safe, effective, analgesia without motor block.Currently multimodal analgesia seems to be the most favorable choice.

Objective To investigate the effect of neonatal mechanical ventilation on lung function and recurrent respiratory diseases of patients.Methods Lung function and recurrent respiratory diseases were studied in 16 children who had been ventilated at birth and were followed up in 8 years later.Results Fifty percentage of them had recurrent respiratory diseases and 75 % of had lung function abnormalities, in which residual volume/ total lung capacity (RV/ TLC) was 56.25 %,43.7 % (Vmax 25),specific airway resistance (sRaw) and specific airway conductance (sGaw) 50 % respectively. Objective:To observe the repair effect of nano hydroxyapatite/magnesium(nHA-Mg)porous composite material modified by poly lactic acid/glycolic acid copolymer(PLGA) on the jaw bone defect of rabbits,and to elucidate the mechanisms preliminary.Methods:The mandibular defect models of 10 mm×5 mm×1 mm in the 18 rabbits were established.Nine rabbits were selected and implanted with nHA-Mg composite materials modified by PLGA in the left as experimental group and the right as blank control group;the other nine were implanted with nHA-Mg in the left as positive control group and the right as blank control group.The rabbits were sacrificed at 4, 8, 12 weeks (respectively 3 of experimental group and positive control group every time)and the mandibular defect areas were intercepted and observed by imaging and histological examination;the sizes of the newborn trabecula area and residual materials in experimental group and positive control group were compared.Results:Compared with positive control group and blank control group, the percentage of newborn trabecula area of the rabbits in experimental group was increased (P<0.05),and there was no statistic difference between positive control group and blank control group (P>0.05).The imaging results showed that the new bone formation can be observed in experimental group and better than positive control group and blank control group.The paraffin section results indicated that there were visible vascular tissue and newborn trabecula,the osteoblasts gathered around the bone trabecula;a lot of pits were located in the bone trabecula, and the pits contained osteocytes in experimental group.As the prolongation of time,thickened newborn trabecula, dense arrangement and trend of converting to lamellar bone were observed in experimental group.The hard tissue section results showed that the remaining amount of materials in experimental group was more than that in positive control group.Conclusion:nHA-Mg porous composite materials modified by PLGA can effectively reduce the rate of degradation in the body, promote osteogenesis and guide the bone regeneration.

Objective To study the clinical and electrophysiological features of the patients with hereditary neuropathy with liability to pressure palsy ( HNPP) diagnosed by gene analysis.Methods Seven patients from two HNPP families were assessed on medical history, physical examination, electrophysiology findings and gene analysis.Results A clinical manifestation of acute, painless, recurrent peripheral nerve palsies was typical for HNPP.Median, ulnar and peroneal nerves were usually affected.Electrophysiology study revealed that prolonged distal motor latency and slowing nerve conduction velocity were prominent.Gene studies exhibited a deletion of the peripheral myelination protein 22 gene in all the seven patients.Conclusions HNPP usually affects areas where nerves are subject to entrapment, and many episodes are preceded by minor compression on the affected nerve.As a reliable screening tool in detecting HNPP, the electrophysiological study shows that segmental demyelination is most commonly seen at common nerve entrapment sites.

Objective To investigate the effect of Shugan-Yishen formula on Tamoxifen-resistant MCF-7 cell line(LCC9) through HER2-ERK/MAPK-ERα pathway. Methods Four serums were prepared with the method of serum pharmacology:the serum of Shugan-Yishen formula, Shugan-Yishen formula plus Tamoxifen(TAM), blank and TAM alone. After LCC-9 cells were affected in the above-mentioed serums in vitro, the inhibition rate and the level of mRNA as well as the protein expression of HER2, ERα, ERK/MAPK, p-ERK/MAPK were detected. Results MTT showed that TAM alone group、Shugan-Yishen formula group and the Shugan-Yishen formula plus TAM group had higher inhibition rates at 48th hour, the inhibition rates was(6.61±0.129)%, (47.43±2.24)%, (54.19±3.364)%, compared to the blank group, Shugan-Yishen formula group and the Shugan-Yishen formula plus TAM group had a stronger inhibition on LCC9 at 48th hour (P<0.05), and the combination group was better than Shugan-Yishen formula alone(P<0.05), there was no difference between TAM group compared to the blank group(P>0.05). RT-PCR result showed that compared to the blank group, the expression levels of ESR1 mRNA of group of Shugan Yishen formula plus TAM werehigher by and large at 48th hour (P<0.05), the expression of ESR1 mRNA was(2.282±0.185);and the levels of ERBB2, MAPK3 mRNAwere lower(P<0.05), which levels were(0.71±0.095), (0.336±0.058). Western blotting result showed that Shugan-Yishen Formula plus TAM group could markedly decrease the expression of HER2, p-ERK/MAPK of LCC9 at 48th hour (P<0.05), which the expressions were(0.708± 0.072), (0.228±0.034), and the expression of ER-αelevated(P<0.05). There was no significant difference on ERK/MAPK(P>0.05). Conclusions ①The combination of Shugan-Yishen formula with tamoxifen can inhibit the LCC9 cells. The inhibition effect reaches the peak at 48th hour. ②The mechanism of Shugan-Yishen formula plus TAM combination effects on human breast cancer TAM-resistant cell line LCC9 may improve the sensitivity of ERαby inhibiting the expression of HER2, ERK/MAPK.

Conclusion:The method has the advantages of high specificity, good reproducibility and high precision, and can be used as a method for the determination of chloramphenicol and hydrocortisone in chlor-amphenicol hydrocortisone ear drop.

Objective To explore the effects of HMGCoA agents and ACE inhibitor on serum inflammatory markers in patients with acute coronary syndrome (ACS). Methods The 60 patients with ACS were randomly divided into two groups, one treated with lipid lowered by HMGCoA agents and the other treated with HMGCoA agents added with ACE inhibitor. After 4 months, serum levels of some inflammatory markers were measured by means of ELISA. Results Serum levels of certain inflammatory markers were significantly higher in the ACS group than in the control group and became significantly lower 4 months later in the follow-up group. There were no differences in all the inflammatory markers between the two groups receiving different therapies 4 months later. Conclusion Serum levels of some inflammatory markers may have certain diagnostic value for ACS and may reflect the stability of the disease. HMGCoA agents seem to have no effect on inflammatory responses in a short period.

Objective To explore the influence of fluoxertin on level of IL-2 ,IL-6 and TNF-α in brain tissue and serum of rats with post-stroke depression(PSD) .Methods 60 rats were divided into sham operation group ,depression group ,stroke group ,PSD group and fluoxertin group ,12 cases in each group .ELISA was used to detect the level of IL-2 ,IL-6 and TNF-αin brain tissue and serum .Results The level of IL-2 ,IL-6 and TNF-αin depression group ,stroke group and PSD group were significantly increased and higher than that in sham operation group and fluoxertin group(P<0 .05) .There was no significant difference between sham op-eration group and fluoxertin group(P>0 .05) .Conclusion Fluoxertin could significantly decrease the level of IL-2 ,IL-6 and TNF-αand play an important role in therapy of PSD .
