1.PRELIMINARY OBSERVATION ON ANTIBODY TO RESA IN MALARIA PATIENTS IN XISHUANGBAINA

Antibody to RESA of plasmodium falciparum from malaria patients in Xishuangbaina were tested by RE-SA-IFA. The positive rate of the antibody to RESA in falciparum malaria patients was 22. 0% (9/41), 8. 3% (1/12) in mixed falciparum and virax infections, and negative in 38 vivax patients. TheGMRT of serum titer from patients with positive anti-RESA antibody and from those with negative results by IFA was 1552.1 and 741. 8 respectively (with significant difference statistically). The parasite densities in patients with anti-RESA antibody were lower in comparing with those patients with no anti-RESA antibody. These findings indicated that there was a parallel relationship between ami-RESA antibody liters and the acquisition of clinical immunity in malaria patients.

Objective: To study the effects of parathyroid hormone(PTH) on the calcification characters of cultured human dental papilla mesenchymal(DPM) cells.Methods: DPM cells were cultured up to 35 days in two groups. The control group was cultured in DMEM supplemented with 15% FCS,50 ?g/ml ascorbic acid and 10 mmol/L ?-glycerophosphate. The cells in experimental group were cultured in above mentionned medium containing 33.3 nmol/L of PTH. The medium was changed every 3 or 4 days. [Objective] To study the influence on humoral immunity and ectopic endometrium by infusion of Bushen Tiaojing Decoction (BTD) with the Huoxue Sanjie Suppository (HSS) in anus. [Methods] One hundred and thirty SD rats were allocated to ten groups: normal control (A), model (B), normal dose of HSS (C), normal dose of BTD with HSS (D), 3 doses of BTD (E), 3 doses of BTD with HSS (F), 5 doses of BTD (G), 5 doses of HSS (H), 5 doses of BTD with HSS (I), and Danazol (J) groups. The endometriosis (EM) rats were modeled by endometrium implantation in all groups except those in Group A. Four weeks after the treatment, the contents of immunoglobulin (IgG、IgM、IgA), C_3, C_4 and ALT were tested, and histomorphomological analysis of the thymus, ovary, uterus, EM implants, liver and kidney were performed. [Results] Four weeks after EM implantation, the ectopic endometria were growing well, IgG and C_3 increased, but IgM decreased, and the uterus and ovary significantly enlarged. With the treatment of BTD and HSS or Danazol, the ectopic endometria were atrophied. In Groups F and G, IgG decreased; in Groups H and I, C_3 decreased, and uterus and ovary shrunk in Group I. Danazol can help to prolong the sexual cycle, while BTD and HSS can not. In all the medication groups, no obvious difference was found in ALT of the rats. [Conclusion] Oral administration of BTD with HSS can cause endometrial atrophy and improve humoral immunity in model rats.
4.Effects of FcγRIIb gene modified dendritic cells on immune tolerance of rat orthotopic liver transplantation

Objective:To study the effects of FcγRIIb gene modified dendritic cells on liver function and rejection after orthotopic liver transplantation in rats.Methods:The recombinant lentivirus expression vector TRE-FcγRIIb containing Lewis rat FcγRIIb gene was constructed by gene cloning technique. TRE-FcγRIIb expression virus and TET-on regulatory virus were packaged and co-infected with immature dendritic cells derived from DA rat bone marrow. The expression of FcγRIIb was detected. Donor rats DA and recipient Lewis were paired according to body weight and then randomly divided into three groups. The control group (group A) did not receive any pretreatment. The Lewis rats in group B were treated with cyclosporin A on the 2nd day after liver TX. Immature dendritic cells derived from bone marrow of DA rats were injected intravenously one day before liver TX in FcγRIIb gene modified immature dendritic cells (1×10 6 cells)group (group C). Blood and liver tissue were biopsied 7 days after operation to detect liver function and pathological changes. Results:The abnormal liver function in FcγRIIb gene modified immature dendritic cells group (group C) was significantly lower than that of control group 7 days after operation ( P<0.05), and the liver pathological rejection of FcγRIIb gene modified immature dendritic cells group (group C) was significantly lower than that of control group 7 days after operation ( P<0.05). The average survival days of rats in the control group was 12.8 days; the average survival days of rats in the cyclosporin A group was 65.3 days; the average survival days of rats in the FcγRIIb gene-modified immature dendritic cell group was 58.5 days. Conclusion:FcγRIIb gene modified immature dendritic cells can ameliorate liver dysfunction and acute liver rejection after orthotopic liver transplantation.
5.Effect of statin pretreatment on collateral circulation and prognoses of patients with cardiac large artery occlusive stroke

Objective To investigate the effect of statin pretreatment on collateral circulation and prognoses of patients with cardiac large artery occlusive stroke.MethodsFifty-three patients with cardiac large artery occlusive stroke admitted to our hospital from January 2016 to July 2019 were selected. All patients had unilateral middle cerebral artery occlusion. DSA was used to evaluate the collateral flows, and the differences of collateral flows and prognoses in patients took statins before onset were compared with those did not take statins.ResultsAs compared with patients did not take statins, patients took statins had higher incidences of diabetes and coronary heart disease, lower content of low density cholesterol, higher proportion of patients with good collateral circulation (grading 3 to 4), and lower modified Rankin scale scores 3 months after surgery, with statistically significant differences (P< 0.05). Multivariate Logistic regression analysis showed that percentage of patients took statins was independent factors affecting collateral flow in patients with cardiac arterial occlusive stroke(OR=5.000, 95%CI: 1.136-22.006,P=0.033).ConclusionIn patients with cardiac large artery occlusive stroke, statin pretreatment could improve collateral flow and clinical prognoses.
6.Negative result of susceptibility vessel sign in predicting large artery atherosclerosis acute middle cerebral artery occlusion

Objective:To investigate the predictive value of negative result of susceptibility vessel sign (SVS) in etiology types of large vessel occlusion (LVO) stroke.Methods:Sixty-three LVO stroke patients admitted to our hospital from December 2015 to October 2019 were chosen in our study. The clinical data of 63 patients with negative SVS and positive SVS were compared to clarify the independent factors influencing the SVS negative/positive results. The SVS negative rate in large artery atherosclerosis (LAA) patients and non-LAA patients were compared, and the sensitivity, specificity and accuracy of SVS in predicting LAA acute middle cerebral artery occlusion were evaluated.Results:Of the 63 patients, 32 (50.8%) had negative SVS, and 31 had positive SVS; as compared with the positive SVS group, the negative SVS group had significantly higher incidence of hyperlipidemia, lower baseline NIHSS scores, and lower incidence of atrial fibrillation ( P<0.05). Multivariate Logistic regression analysis showed that hyperlipidemia and baseline NIHSS scores were independent factors influencing SVS in patients with LVO stoke. Among the 41 LAA patients, 25 had negative SVS, with a negative rate of 61.0%; among the 22 non-LAA patients, 7 had negative SVS, with a negative rate of 31.8%; significant difference in negative rate was noted between the two groups ( χ2=4.870, P=0.027). The sensitivity, specificity, positive and negative predictive values, and accuracy of negative SVS in predicting LAA stroke was 60.98% ( 95%CI: 44.50%-75.80%), 68.18% ( 95%CI:45.13%-86.14%), 78.12%, 48.39% and 63.49%, respectively. Conclusion:Negative result of susceptibility vessel sign has a high predictive value in acute middle cerebral artery occlusion.
7.Impact of the Varian real-time position management respiratory gating system on radiotherapy planning dosimetry

Objective:To study the impact of the Varian real-time position management (RPM) respiratory gating system on radiotherapy planning dosimetry.Methods:The radiotherapy plans of 40 cases with thoracic or abdominal tumors were retrospectively selected in this study. The motion phantom for quality control was adopted to generate respiratory gating signals, and the 30%-60% stable phase at the end of expiratory was selected as the respiratory gating window. The dose verification for the abovementioned radiotherapy plans was performed using the Portal Dosimetry (PD) system under RPM respiratory gating mode with the Edge accelerator. Afterwards, dose analysis was performed with different γ passing rate criteria and the distribution characteristics of γ values were analyzed. Finally, the verification results between the non-gating mode and the gating mode were compared.Results:Under the respiratory gating mode, the passing rates of all intensity-modulated radiation therapy/volumetric-modulated arc therapy (IMRT/VMAT) plans with or without flattening filters were over 95.5% by γ criteria of (3%, 3 mm) or (3%, 2 mm) and were over 90% by stricter γ criteria of (2%, 2 mm). All plans met the clinical requirements recommended by the American Association of Physicists in Medicine (AAPM). The passing rates of dose verification under non-gating mode were slightly better than those under respiratory gating mode, and the differences between the two modes were statistically significant (3%/3 mm, Z =-1.45; 3%/2 mm, Z =-2.86; 2%/2 mm, Z =-3.70; 1%/1 mm, Z =-4.52; P<0.05). There was no significant difference in the minimum and maximum values of γ and the share of γ > 1.5 of plan verification result under the two modes. However, the average value and standard deviation of the γ were generally smaller under the non-gating mode. Conclusions:The impact of the introduction of RPM respiratory gating technology on dose is clinically acceptable, and the execution of these plans in this gating mode is safe and reliable.
8.Meta-analysis of dosimetric comparison between volumetric-modulated arc therapy and intensity-modulated radiotherapy for breast cancer after modified radical mastectomy

Objective:To compare the dosimetric differences between volumetric-modulated arc therapy (VMAT) and intensity-modulated radiation therapy (IMRT) on planning target volume (PTV) and organ-at-risk (OAR) for breast cancer after modified radical mastectomy, aiming to provide evidence-based reference for clinical practice.Methods:According to strict inclusion and exclusion criteria, literature search was performed in PubMed, Cochrane Library, FMRS, CNKI, Wanfang Data and VIP full text databases from the inception of databases up to March 2020. The controlled clinical trials of dosimetric comparison between VMAT and IMRT for breast cancer following modified radical mastectomy were selected. The meta-analysis was performed using Stata14 software.Results:The meta-analysis included 281 patients from 13 observational studies. Compared with IMRT, VMAT significantly increased the PTV dose coverage D 95%( P<0.001) and significantly improved the PTV homogeneity index (HI, P<0.001) and conformity index (CI, P=0.004). Compared with IMRT, VMAT more effectively decreased the ipsilateral lung V 20Gy (WMD=1.332, P=0.027) and contralateral lung V 10Gy ( P=0.003). There were no significant differences in theD mean, V 5Gy, V 10Gy and V 30Gy of the ipsilateral lung, D mean and V 5Gy of the contralateral lung, D mean, V 10Gy and V 30Gy of the heart between VMAT and IMRT (all P>0.05). Compared with VMAT, IMRT reduced the cardiac V 5Gy ( P=0.001). However, sensitivity analysis of included literature on cardiac V 5Gy showed that the P value was reversed, indicating that the stability of the results was poor. VMAT significantly shortened the delivery time ( P<0.001) and the number of monitor units ( P<0.001) compared to IMRT. Conclusion:Compared with IMRT, VMAT can achieves superior target dose coverage, HI and CI, better protection for the ipsilateral and contralateral lung, fewer monitor units and shorter delivery time.
9.Difference analysis of chemical components in roots and rhizomes of 3 medicinal plants of Gentiana from the same origin

OBJECTIVE To provide reference for exploring alternative resources of Gentiana rigescens from the plants of Gentiana. METHODS The contents of four components (gentiopicroside, swertiamarin, swertioside and amarogentin) in the roots and rhizomes from 3 plants of Gentiana (G. rigescens, G. cephalantha, G. delavayi) were determined by high-performance liquid chromatography (HPLC). The chemical compositions in the above roots and rhizomes were identified by ultra-performance liquid chromatography electrospray ionization quarter-time of flight mass spectrometry (UPLC-ESI-Q-TOF-MS), and the differences were analyzed by principal component analysis (PCA). RESULTS Four active components such as gentiopicroside, swertiamarin, swertioside and amarogentin were detected in the roots and rhizomes of G. rigescens and G. cephalantha, and the contents of the four components were similar in both. The contents of gentiopicroside in the root and rhizome of G. cephalantha and G.rigescens were more than four times of the limit standard of the Chinese Pharmacopoeia (Part Ⅰ) in 2020; However, only swertiamarin, swertioside and amarogentin were detected in the roots and rhizomes of G.delavayi, and the contents of swertioside and amarogentin were 34.12 and 8.81 times of those of G. rigescens, respectively. Additionally, G. rigescens and G. cephalantha contained xantones, G. delavayi contained flavonoids. PCA showed that there was a small difference between G. rigescens and G. cephalantha; however, there was a big difference between G. delavayi and G. rigescens. CONCLUSIONS The difference between the roots and rhizomes of G. cephalantha and G. rigescens from the same origin is small and there is substitutability; while the difference in the chemical components from roots and rhizomes between G. delavayi and G. rigescens is great and G. delavayi cannot be used as medicine instead of G. rigescens.
