Objective:To investigate the effects of Gab2 overexpression on the proliferation and migration of human colorectal cancer cell line SW480.Methods: The experimental group (LV-Gab2-GFP group),colorectal cancer SW480 cells were transfected with recombinant lentivirus vector (LV-Gab2-GFP),the negative control group was transfected with negative control lentiviral vector ( LV-GFP) ,and the blank control group without any treatment.The mRNA and protein expression of Gab 2 in cells were identified by RT-PCR and Western blot respectively.Proliferation of the cells was detected by CCK-8 colorimeter and colony forming assay.Wound-healing assay was used to determine the cells migration .Results: RT-PCR and Western blot demonstrated that Gab 2 mRNA and protein expression significantly increased in LV-Gab2-GFP group compared with control groups;overexpression of Gab2 markedly enhanced human colorectal cancer SW 480 cells proliferation and migration compared with control groups .Conclusion:Overexpression of Gab2 accelerates human colorectal cancer SW 480 cells proliferation and migration.
2.Status and influencing factors of self-care for patients with cystostomy and ileal conduit

Objective:To explore the current status and influencing factors of self-care ability in patients with cystostomy and ileal conduit.Methods:Using the convenient sampling method, a total of 160 patients with cystostomy and ileal conduit who visited the Urostomy Clinic of Peking University First Hospital from October 2021 to March 2022 were selected as the research objects. Patients were surveyed using a general information survey questionnaire, Social Support Rating Scale (SSRS) and Urostomy Education Scale. Spearman correlation analysis was used to explore the correlation between social support and self-care in patients with cystostomy and ileal conduit, and multiple linear regression analysis was used to explore the influencing factors of self-care ability in patients with cystostomy and ileal conduit. A total of 160 questionnaires were sent out, and 157 were effectively collected, with an effective recovery rate of 98.13% (157/160) .Results:The total score of Urostomy Education Scale of 157 patients with cystostomy and ileal conduit was [20.00 (4.00, 21.00) ]. The results of multiple linear regression analysis showed that marital status, comorbidities, stoma and peristomal complications, frequency of follow up in urostomy clinic and objective support were the influenting factors for self-care of patients with cystostomy and ileal conduit ( P<0.05) . Conclusions:Self-care of patients with cystostomy and ileal conduit is related to many factors. Medical staff should strengthen the intervention and management of patients with cystostomy and ileal conduit from multiple aspects such as individual and social function, so as to improve their self-care ability.
3.Correlation between urinary incontinence and quality of life among patients with post-laparoscopic radical prostatectomy

Objective? To understand the situation of urinary incontinence and quality of life among patients with post-laparoscopic radical prostatectomy and to analyze the correlation between them. Methods? From September 2015 to October 2016, we selected 115 patients with post-laparoscopic radical prostatectomy at a ClassⅢ Grade A hospital in Beijing for survey by questionnaire with the method of convenience sampling. We collected patients' general information as well as disease information, and evaluated the situation of urinary incontinence (incidence and severity of urinary incontinence) with the International Consultation on Incontinence Questionnaire-Urinary Incontinence Short Form (ICIQ-UI SF) and influence of urinary incontinence on quality of life with the Incontinence Impact Questionnaire (IIQ-7) to analyze the correlation between severity of urinary incontinence and quality of life. Results? Among 115 patients with post-laparoscopic radical prostatectomy, there were 95 (82.6%) urinary incontinence patients including 41 cases (43.1%) with mild urinary incontinence, 47 cases (49.5%) with medium urinary incontinence and 7 cases (7.4%) with severe urinary incontinence. The score of severity of urinary incontinence ranged from 0 to 19.0 with 8.0 for the median and 5.0 for the interquartile range. Spearman correlation analysis showed that the score of IIQ-7 had a positive correlation with the severity of urinary incontinence (r=0.674, P<0.01), and had a negative correlation with the postoperative time (r=-0.215, P<0.05). Multiple linear regression analysis showed that the main influencing factor of quality of life was severity of urinary incontinence (P<0.01). Conclusions? Urinary incontinence is common among patients with post-laparoscopic radical prostatectomy mainly in mild and medium urinary incontinence. The more serious the urinary incontinence is,the greater the influence on quality of life is which points out that the scientific and effective management by medical staff for urinary incontinence can improve patients' quality of life.
4.Best evidence summary of mechanical prevention of venous thrombosis in adult patients undergoing abdominal and pelvic surgery

Objective:To retrieve, evaluate and summarize the relevant evidence of mechanical prevention of venous thromboembolism (VTE) in adult patients undergoing abdominal and pelvic surgery, so as to provide a scientific basis for clinical practice.Methods:We systematically retrieved and screened the guide network and database for evidence on mechanical prevention of VTE in adult patients undergoing abdominal and pelvic surgery, including guidelines, evidence summary, clinical decision-making, systematic review, and expert consensus. The search time limit was to build the database until January 19, 2020. Two researchers who had undergone evidence-based training selected appropriate evaluation tools according to different types of literature to evaluate the quality of the literature, and to extract, translate and integrate recommendations.Results:A total of 24 articles were included, including 7 guidelines, 2 evidence summaries, 1 recommended practice, 12 systematic reviews, and 2 expert consensus. The evidence was summarized from 6 aspects, namely admission assessment, contraindications, applicable population, starting time, duration and patient education, and a total of 10 recommendations were formed.Conclusions:This study summarizes the best evidence for mechanical prevention of VTE in adult patients undergoing abdominal and pelvic surgery, and provides evidence-based evidence for standardizing clinical mechanical prevention.
5.Construction of ostomy nurse-led clinics in tertiary hospitals in China

Objective:To investigate the construction of ostomy nurse-led clinics in tertiary hospitals in China.Methods:Using the convenient method, managers of ostomy nurse-led clinics in tertiary hospitals in 31 provinces, autonomous regions and municipalities in China in August 2020 were selected as the survey objects. Questionnaire for Survey on Status Quo of Ostomy nurse-led clinics in ClassⅢ Grade A hospital in China was used to investigate. A total of 367 questionnaires were recovered in this survey, 313 valid questionnaires were recovered, and the effective recovery rate was 85.29%.Results:A total of 313 tertiary hospitals have set up ostomy nurse-led clinics. Among the ostomy nurse-led clinics, 86.58% (271/313) of the ostomy specialist nursing outpatient visits were independent visits by nurses. The opening period of stoma clinic was (6.27±4.08) years and the weekly opening time was (5.30±3.93) half-days. The average monthly outpatient number was 100, with a minimum of 4 and a maximum of 3 000. In terms of job management, 50.48% (158/313) of the stoma specialist nursing outpatient jobs were part-time, managers were mostly undergraduates (74.44%, 233/313) , the professional title of managers was mostly nurse in charge (48.24%, 151/313) , 38.98% (122/313) of hospitals had promotion system and 36.74% (115/313) of hospitals had performance management system. There were large differences in environmental layout, facilities and equipment, goods management and specialist management and there was no unified standard.Conclusions:There are great differences in the construction of ostomy nurse-led clinics in tertiary hospitals in China, which need to be further standardized and unified. It is suggested that relevant departments should introduce relevant policies to encourage the establishment of full-time nurse posts in ostomy nurse-led clinics, guarantee the training, promotion and performance reward of visiting nurses and fully encourage the subjective initiative of ostomy specialist nurses to promote their career development.
6.Interpretation of International Ostomy Guideline (Second Edition) by World Council of Enterostomal Therapists

Stoma refers to the channel established by surgery to excrete the excreta in the body, which changes the excretion mode of patients, and has a serious impact on the physiological, psychological, spiritual, social and other aspects of patients. The World Council of Enterostomal Therapists released International Ostomy Guideline (Second Edition) in 2020, covering four aspects, including the education and scope of practice, holistic approach to care, preoperative nursing needs and postoperative nursing needs, and a total of 15 recommendations. This article focuses on the interpretation of eight recommendations on the two aspects of preoperative and postoperative nursing needs, with a view to promoting the reform of clinical practice and providing evidence-based basis for clinical nurses. At the same time, it is suggested that clinical nurses should make targeted transformation according to specific clinical situations to improve the quality of clinical nursing.
7.Study on the correlation between glycolipids and prostate volume in patients with benign prostatic hyperplasia

To study the clinical correlation between fasting plasma glucose, lipid metabolism, prostate-specific antigen and prostate volume in patients with benign prostatic hyperplasia, and to explore the combined effect as diagnostic indicators. A total of 108 patients with benign prostatic hyperplasia treated in Beijing University of Chinese Medicine Third Affiliated Hospital from June 2021 to March 2023 were retrospectively analyzed as the hyperplasia group, and 98 healthy physical examination personnel were selected as the control group during the same period. Compare the differences in levels of fasting plasma glucose (FPG), total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), small and dense low-density lipoprotein cholesterol (sdLDL-C), homocysteine, lipoprotein a (LPa), prostate specific antigen (PSA), and free prostate specific antigen (fPSA) between two groups of patients. Using Pearson analysis method to analyze the correlation between the above indicators and the size of prostate volume in patients with benign prostatic hyperplasia; using multiple linear regression to analyze the influencing factors of prostate volume enlargement; draw receiver operating characteristic (ROC) curves and analyze the application value of individual and combined detection of HDL, FPG, PSA, and fPSA. The results showed that there were significant differences in HDL, FPG, PSA, and fPSA levels between the control group and the proliferative group( P<0.05). The size of prostate volume is negatively correlated with HDL( r=-0.183, P<0.05) and positively correlated with FPG ( r=0.202, P<0.05), PSA( r=0.412, P<0.05), and fPSA( r=0.425, P<0.05). The results of multiple linear regression analysis showed that HDL( P=0.000), FPG( P=0.048), PSA( P=0.044), and fPSA ( P=0.012) were risk factors for increased volume of benign prostatic hyperplasia; ROC curve analysis shows that the AUC of HDL, FPG, PSA, and fPSA combined detection is 0.823, which is better than individual detection. In conclusion,HDL, FPG, PSA, fPSA has close correlation with hyperplasia of prostate, the joint detection may has better prediction for benign prostatic hyperplasia.

9.Research progress on self-management of stoma patients

With the development of medical technology, the survival rate of cancer patients continues to improve, and the number of stoma patients is increasing day by day. Self-management refers to patients monitoring and managing their own diseases to maintain and improve physical health. Good self-management ability can enhance patient treatment compliance, reduce the incidence of stoma and surrounding complications, improve health outcomes, and raise patient quality of life. This article reviews the research progress on self-management of stoma patients, with the aim of providing reference for research on self-management of stoma patients.
10.Construction of position responsibilities index system of wound, ostomy and continence nurses based on Delphi method

Objective:To construct a position responsibilities index system of wound, ostomy and continence nurses (WOCN) based on the Delphi method.Methods:Through literature review, semi-structured interviews, and investigation and research on the current working situation of WOCN, the expert inquiry questionnaire was self-designed with reference to opinions of the expert coordination group. From January to April 2019, a total of 45 senior experts including professional doctors, WOCN representatives, representatives of international Enterostomal Therapist Training Program (ETNEP) and chairmen of various provincial professional committees in China were invited to conduct 3 rounds of Delphi expert consultation, and construct the position responsibilities index system of WOCN.Results:The positive coefficients of the 3 rounds of expert consultation were respectively 100.0% (45/45) , 93.3% (42/45) and 100.0% (42/42) , and the authority coefficient of the experts was (0.92±0.16) . The final position responsibilities index system of WOCN included 5 first-level indexes such as clinical practice, education, research, coordinated management and professional promotion and 33 second-level indexes.Conclusions:The index system constructed in the study clarifies the position responsibilities of WOCN, which helps to provide a certain theoretical basis for the work, performance and classification of specialist nurses.
