1.Determination of tricin in malt by HPLC

Objective To develop an HPLC method for the quantitative analysis of tricin in malt. Methods HPLC method was used. Analysis was performed on Spherigel C_ 18 column (150 mm?4.6 mm, 5 ?m) with acetonitrile-water-tetrahydrofuran-methanoic acid (26∶74∶10∶1) as mobile phase. The detection wavelength was 350 nm, flow rate was 1.0 mL/min, and the analysic volumn was set at 20 ?L. Results The calibration curve of tricin was linear in the range of 0.22-2.15 mg/L (r=0.999 8). The average recovery rate of tricin was 97.4% (RSD=1.7%) (n=9). Conclusion The method can be used to determine tricin in raw malt quantitatively.
2.Clinical Observation of Shuxuetong Injection Combined with Alprostadil in the Treatment of Acute Cere-bral Infarction

OBJECTIVE:To observe clinical efficacy and safety of Shuxuetong injection combined with alprostadil in the treat-ment of acute cerebral infraction(ACI). METHODS:A total of 123 ACI patients selected from our hospital during Mar. 2014-Sept. 2016 selected as research subjects were divided into observation group (62 cases) and control group (61 cases) according to ran-dom number table. Control group was additionally given Alprostadil injection 2 mL+0.9% Sodium chloride injection 100 mL, ivgtt,qd,on the basis of routine treatment. Observation group was additionally given Shuxuetong injection 6 mL+0.9% Sodium chloride injection 250 mL,ivgtt,qd,on the basis of control group. Both groups were treated for 2 weeks. MPV,FIB content, hemorheology indexes,neurologic impairment degree score before and after treatment as well as clinical efficacy,the occurrence of ADR were compared between 2 groups. RESULTS:Before treatment,there was no statistical significance in MPV,FIB content, hemorheology indexes or neurologic impairment score between 2 groups (P>0.05). Compared with before treatment,MPV,FIB content,hemorheology indexes and modified Rankin scale of 2 groups were all decreased significantly;Barthel index and NIHSS scores were increased significantly,and each aspect of observation group was better than that of control group,with statistical sig-nificance(P<0.05). Total response rate of observation group was 95.16%,which was significantly higher than 85.25% of control group,with statistical significance(P<0.05). No severe ADR was found in 2 groups. CONCLUSIONS:Shuxuetong injection com-bined with alprostadil can significantly improve the neurological function of ACI patients,improve hypercoagulable state by reduc-ing MPV and FIB content,and have good therapeutic efficacy and safety.
3.Adenoviral short hairpin RNA targeting phosphodiesterase 5 attenuates cardiac remodeling and cardiac dysfunction following myocardial infarction in mice.

OBJECTIVETo observe the impact of PDE5shRNA on cardiac remodeling and heart function following myocardial infarction in mice.
METHODSMyocardial infarction (MI) was induced in mice by left coronary artery ligation. Mice were randomly assigned to sham group (n = 6), PDE5shRNA group (n = 12), common adenovirus group (n = 15) and DMEM group (n = 8). Four weeks post-MI, the survival rate was evaluated. Cardiac function was examined by echocardiography. HE staining and Masson staining were used to evaluate the myocardial infarction size and fibrosis. The number of blood vessels was evaluated by immunohistochemistry, PDE5 protein expression in the left ventricular was detected using Western blot, level of cGMP or PKG activity in the left ventricle was evaluated with ELISA.
RESULTSFour weeks post-MI, all mice survived in the sham group, 3(37%) mice died in the DMEM group, 1 (8%) died in the PDE5shRNA group and 5 died in the common adenovirus group (33%). Infarct size was significantly reduced in PDE5shRNA group compared with the common adenovirus group and DMEM group [(25.4 ± 2.9)% vs. (42.0 ± 3.2)% and (43.4 ± 2.6) %, P < 0.05]. Cardiac function was significantly improved in PDE5shRNA group compared to common adenovirus group and DMEM group[LVFS: (21.1 ± 3.7)% vs. (14.2 ± 2.9)% and (14.22 ± 2.91)%, all P < 0.05; LVEF: (48.2 ± 7.1)% vs. (34.6 ± 6.2)% and (38.1 ± 2.8)%, all P < 0.05; LVESD: (3.87 ± 0.45) mm vs.(4.91 ± 0.62) mm and (4.63 ± 0.37) mm, all P < 0.05]. The blood vessel density was also higher in PDE5shRNA group compared with common adenovirus group (infarct area:14.3 ± 2.0 vs. 6.6 ± 1.2, P < 0.05; periinfarct area: 23.6 ± 2.1 vs. 13.7 ± 2.4, P < 0.05). Compared with common adenovirus group, level of PDE5 was significantly downregulated and level of cGMP or PKG was significantly upregulated in PDE5shRNA group (all P < 0.05).
CONCLUSIONSPresent study suggests PDE5shRNA improves cardiac function and attenuates cardiac remodeling through reducing infarction size and cardiac fibrosis and these beneficial effects are possibly mediated by activating cGMP/PKG signaling pathway.

4.Study on fast quality control in extracting process of Paeonia lactiflora using near infrared spectroscopy.

OBJECTIVETo set up a novel method for fast analysis of active components in water extracting process of Paeonia lactiflora with near infrared spectroscopy (NIRS).
METHODHPLC was used as the reference method to determine the content of Paeoniflorin and a multivariate calibration model based on PLS algorithm was developed to analyze the correlation between the spectra and the corresponding values determined by the reference method.
RESULTThe correlation coefficient of the calibration model was 0.996 2, and the predicted coefficient was 0.989 5. The RMSEC and RMSEP were 0.109 g x L(-1) and 0.138 g x L(-1), respectively, and the RSEP was 5.6%.
CONCLUSIONThe method mentioned above is proved to be convenient, rapid and nondestructive, accurate and reliable, and is applicable for fast analysis and monitoring of active components in extraction process of traditional Chinese medicine.

For the quality management system of herbal medicines, intermediate and finished products it exists the " short board" effect of methodologies. Based on the concept of process control, new strategies and new methods of the production process quality control had been established with the consideration of the actual production of traditional Chinese medicine an the characteristics of Chinese medicine. Taking Banlangen granule as a practice example, which was effective and widespread application, character identification, determination of index components, chemical fingerprint and biometrics technology were sequentially used respectively to assess the quality of Banlangen herbal medicines, intermediate (water extraction and alcohol precipitation) and finished product. With the transfer rate of chemical information and biological potency as indicators, the effectiveness and transmission of the above different assessments and control methods had been researched. And ultimately, the process quality control methods of Banlangen granule, which were based on chemical composition analysis-biometric analysis, had been set up. It can not only validly solute the current status that there were many manufacturers varying quality of Banlangen granule, but also ensure and enhance its clinical efficacy. Furthermore it provided a foundation for the construction of the quality control of traditional Chinese medicine production process.

6.Research on technological process of two-pot countercurrent extraction of hydroxysafflor yellow A.

OBJECTIVETo study the optimum technical conditions of extracting Hydroxysafflor yellow A (HSYA) from Carthmus tinctorius by multi-stage countercurrent extraction technology.
METHODThe effects of extraction time of each stage, extraction temperature, ethanol concentration and solid-liquid ratio (g x mL(-1)) on extraction yield of HSYA were studied by orthogonal test design and the comparison of other extraction methods were presented.
RESULTExtraction time and solid-liquid ratio had significant influence on the extraction yield, and the optimum parameters were as follows: Extraction time of each stage was 120 min, solid-liquid ratio was 1 : 10 (g x mL(-1)), ethanol concentration was 30%, and extracted at room temperature. Under the optimum conditions, the extraction yield of HSYA was 1.56% and the purity of the extract was 6.06%. Compared with the traditional extraction method and the ultrasonic extraction method of the pharmacopoeia, the extraction yield was increased by 6.12% and 9.09%, the purity of extract was increased by 42.9% and 27.0%, respectively.
CONCLUSIONThe multi-stage countercurrent extraction technology has many advantages such as simple operation, less solvent consumption, higher extraction yield and purity of extract and it has wide industrial application prospect.

7.The impact of astragaloside Ⅳ on the experimentally atherosclerotic formation in ApoE-deficient mice

Objective To explore the impacts of astragalosideⅣ( AST-Ⅳ) on the experimentally atherosclerotic formation in ApoE-deficient mice. Methods ApoE-deficient mice were randomly divided into AST-Ⅳ and control groups, both groups were fed with high-fat diet, and were killed after 12 weeks of treatment. The plaque areas were measured; the percentage of CD4 +CD25 + T cells were determined by flow cytometry;the levels of supernatant cytokine such as interferon ( IFN )-γ, interleukin ( IL )-10 and transforming growth factor-β (TGF-β) were measured by enzyme-linked immunosorbent assay (ELISA). Results The plaque area of AST-Ⅳ group was ( 815. 78 ± 165. 43 )μm2 , which was smaller than that in control group of (2152. 13 ± 525. 47)μm2. The percentage of CD4 +CD25 +Treg accounted for (9. 86 ± 1. 78)% of CD4 + cells, which was higher than that in control group (2. 52 ± 1. 43) %. Compared to con-trol group, the supernatant levels of IL-10 and TGF-βwere significantly higher, and IFN-γwas significantly lower in AST-Ⅳ group (P<0. 05). Conclusions By influencing the regulatory T cell-mediated immune tolerance, AST-Ⅳ might inhibit atherosclerosis in mice.
