T),one dinucleotide deletion(1801-02 del AG) and one base insertion(1125 ins GCCATCG),which resulted in eight missense mutations,two nonsense mutations and two frame shift mutations,namely P534R,G343V,R259W,A141T,R401Q,K276E,Y174C,A314P,S108X,Q177X,fs E471 and fs A247.Conclusion The combined DHPLC and sequencing approach may act as a rapid and efficient method for ABCD1 gene mutation analysis in patients and carriers of X-linked adrenoleukodystrophy families.There exist different ABCD1 gene mutations in different pedigrees,and no obvious correlation between the genotype and phenotype has been found.

Objective To investigate the effects of HepG2 and HepG2.2.15 cells steatosis on the mRNA and protein expressions of suppressors of cytokine signaling-3(SOCS-3) and sterol regulatory element binding proteins (SREBP-1c).Methods The cell model of chronic hepatitis B (CHB) combined with nonalcoholic fatty liver disease (NAFLD) was successfully constructed using an oleic acid-induced HepG2 and HepG2.2.15 cells steatosis.Cells were divided into HepG2 cell control group (HepG2 cell control group), HepG2.2.15 cell control group (HepG2.2.15 cell control group), HepG2 cell steatosis group (HepG2 cell steatosis group) and HepG2.2.15 cell steatosis group (HepG2.2.15 cell steatosis group).The expression levels of SOCS-3 and SREBP-1c mRNA were detected by real-time quantitative polymerase chain reaction (PCR).Changes in protein expressions of SOCS-3 and SREBP-1c were measured by western blot.Results SOCS-3 mRNA expression level in HepG2.2.15 cell control group was significantly lower than that in HepG2 cell control group (P<0.01).The level in HepG2 cell steatosis group was also significantly lower than that in HepG2 cell control group (P<0.01).However, the level of SOCS-3 mRNA in HepG2.2.15 cell steatosis group was lower than HepG2.2.15 cell control group with no statistical significance (P=0.173).There was interaction between cells and steatosis (F=25.547, P<0.01).The expression of SREBP-1c mRNA in HepG2.2.15 cell control group was significantly lower than that in HepG2 cell control group (P<0.01), and was significantly higher in HepG2.2.15 cell steatosis group than that in HepG2.2.15 cell control group (P<0.01).There was no significant difference between HepG2 cell steatosis group and HepG2 cell control group (P=1.000).There was interaction between cells and steatosis (F=5.04, P<0.05).Western blot analysis showed that protein levels of SOCS-3 and SREBP-1c in steatosis cells at 48 h and 72 h were significantly higher than those in non-alcoholic steatosis cells.Conclusions Protein expressions of SOCS-3 and SREBP-1c are up-regulated in both steatosis groups.Factorial analysis shows that there is interaction between cells and steatosis.HBV gene could inhibit SOCS-3 mRNA expression and promote the expression of SREBP-1c mRNA in steatosis cells.
4.Effects of Danhongtongjing formula combined with levofloxacin on serum cytokines in patients with chronic prostatitis

Objective To investigate the effect of Danhongtongjing formula combined with levofloxacin on serum cytokines in patients with chronic prostatitis. Methods 74 cases of patients with chronic prostatitis from May 2014 to April 2016 in our hospital were selected,and randomly divided into two groups,37 cases in each groups.The control group treated with levofloxacin,the observation group were treated with Danhongtongjing formula The observation group on the based of control group.The clinical symptoms,TCM syndrome and the changes of SIgA,VCAM-1 and serum VCAM-1 in prostatic fluid of two groups were compared. Results The total effective rate was 94.59% in the observation group and 78.38%in the control group,the difference was statistically significant (P<0.05).After treatment,the NIH-CPSI index of the two groups was significantly decreased,and the observation group was lower than the control group,the difference was statistically significant (P<0.05);the TCM syndrome score of both groups decreased,and the observation group was lower than the control group,the difference was statistically significant (P<0.05).The total effective rate of TCM Syndrome in observation group was 91.89%,which was significantly higher than that of the control group 72.97%,the difference was statistically significant (P<0.05).After treatment,the levels of SIgA,VCAM-1 and serum VCAM-1 in prostatic fluid were decreased,and the observation group was lower than the control group,the difference was statistically significant (P<0.05). Conclusion Danhongtongjing formula combined with levofloxacin in the treatment of chronic prostatitis patients with significant effect,can effectively regulate serum cytokines, control inflammation, improve the treatment efficiency.
5.Effects of functional training on movement performance and balance in elite fencing athletes with patellar tendinopathy

Objective To investigate the effect of functional training on knee pain,functional movement screen (FMS) score and balance in Chinese elite fencing athletes with patellar tendinopathy.Methods Twenty-four fencing athletes with a diagnosed patellar tendinopathy were randomized into a treatment group (TG) and a control group (CG),each of 12.Both groups were given routine physical therapy,while TG received motor function training in addition for eight weeks.Both groups completed the numerical rating scale (NRS),FMS and balance test before and after the intervention.Results After the intervention,the average PRS and FMS of TG were 2.08± 1.24 and 16.25±0.97 respectively,which significantly outperformed those of TG before the intervention and those of CG after the intervention (P<0.05).Moreover,TG indicated superior results in parameters of static postural balance including center of pressure,total length of swinging pathway,maximal length of swinging pathway,and area of swinging pathway when compared to TG before the intervention and CG after the intervention(P<0.05).Conclusion The motor functiontraining is effective in improving functional movement and balance in elite fencing athletes with patellar tendinopathy.
6.Prenatal molecular diagnosis of four fetuses at high risk for X-linked adrenoleukodystrophy

Objective To investigate methods for prenatal molecular diagnosis of fetuses at high risk for X-linked adrenoleukodystrophy(X-ALD).Methods The amniotic fluid was obtained and genomic DNA was isolated from amniotic fluid cells.Maternal contamination was evaluated by paternity test.PCRRFLP,sequencing and denaturing high performance liquid chromatography(DHPLC)were used to detect the ABCD1 gene of fetal genome.Results In the pedigree 1,the PCR product(799 bp)of the fetus 1 and her father(normal control)could be digested with BcnI. No P560L mutation,which was present in the index patient,was detected in the ABCD1 gene from the genomic DNA of the fetus 1 using direct sequencing.In the pedigree 2,the PCR product(232 bp)of the fetus 2 and her father could not be digested with MaeI and no Q177X mutation,which was present in the propositus,was detected in the ABCD1 gene from the genomic DNA of the fetus 2 using direct sequencing.In the pedigree 3,the PCR product(271 bp)was digested with AciI.the pattern of the fetus 3 and the propositus being the same,and the R617C mutation was found in the ABCD1 gene from the genomic DNA of the fetus 3 using direct sequencing.In the pedigree 4,the PCR product(269 bp)was analyzed with the DHPLC,and the pattern of elution peaks of the fetus 4 and her father was similar,but different from that of the propositus.No K276E mutation was detectable in the ABCD1 gene from the genomic DNA of the fetus 4 by using direct sequencing.Judging from the sex of the fetuses,fetuses 1 and 2 were normal homozygotes,fetus 3 was an ALD hemizygote,and fetus 4 was a normal hemizygote.Conclusion A new protocol for X-ALD prenatal molecular diagnosis is proposed,which would ensure the accuracy of prenatal diagnosis.
7. Effect of clinical first-line Antiviral Agents on outcome of chronic hepatitis B treatment

8.Influence of separating professional title appraisal and appointment on nursing human resources allocation

Objective To discuss the application of implement the separation of professional title appraisal and appointment in nursing human resource allocation .Methods Nursing department confirmed the nursing staff professional title ratio in nursing unit;reasonably deployed the nursing staff at the post;established the quantitative assessment criterion of professional title main performance; strengthened the management after appointment .The main performance quantitative assessment of senior professional titles staff and the nursing quality check scores of nursing unit before and after the implementation were compared .Results After the implementation , night shift number per capita per year , published paper number , section office quality control number and teaching students number of intermediate title staff were higher than before the implementation , and the differences had the statistically purport ( t=-16.98 ,-8.73 ,-23.51 ,-25.33;P<0.001 ) .The night shift number per capita per year ( 22.12 ±2.20 ) , number of papers published in statistical source journals (1.17 ±0.39), nursing department quality control number (7.71 ±1.96), teaching students number (9.94 ± 1.08 ) , teaching ward round presided number or business seminar number of senior title staff ( 12.17 ±0.39 ) were higher than before the implementation (11.06 ±7.90),(0.67 ±0.48),(4.00 ±1.96),(6.13 ±1.41), (6.20 ±1.26 ), and the differences were statistically significant ( t =-5.23, -3.27, -5.33, -8.62,-17.57;P<0.005 ) .The inspection scores of ward management ( 97.27 ±1.07 ) , inpatient nursed quality (97.43 ±0.95), disinfection and isolation (98.19 ±0.71), emergency medication and devices (99.17 ± 0.83 ) , nursing medical records ( 97.26 ±1.09 ) and patients'satisfaction ( 98.27 ±1.14 ) were higher than before the implementation ( 91.32 ±1.58 ) , ( 90.02 ±2.94 ) , ( 92.46 ±1.95 ) , ( 93.09 ±1.74 ) , ( 92.29 ± 1.63),(92.32 ±1.51), and the differences were statistically significant (t=-19.88,-15.38,-17.67,-20.12 ,-16.21 ,-20.16 ,P<0.001 ) .Conclusions Implementing the rational allocation of nursing human resource by professional title appraisal and appointment separation is a new management method .The method plays an important role in strengthening the stability of nursing clinical team , strengthening the key speciality and nursing discipline construction and the improvement of the nursing work quality , which is worth popularizing and applying in other hospitals .
9. Effect of transforming growth factor-β1 on HBV replication and antigen synthesis in HepG2.2.15 cells with steatosis

Objective:
To investigate the effect of transforming growth factor-β1 (TGF-β1) on HBV replication and protein expression in HepG2.2.15 cells with steatosis, as well as the association of TGF-β1 with suppressor of cytokine signaling-3 (SOCS-3) mRNA and sterol regulatory element-binding protein-1c (SREBP-1c) mRNA during the steatosis of HepG2.2.15 cells.
Methods:
The cells were divided into HepG2/HepG2.2.15 cell control groups (C1/C2 groups) and HepG2/HepG2.2.15 cell steatosis groups (F1/F2 groups). 5 ng/ml TGF-β1 was added to the two cell systems for intervention to establish TGF-β1 intervention groups (T1/T2 groups) and steatosis+TGF-β1 intervention groups (TF1/TF2 groups). A time-resolved fluorescence analyzer was used to measure HBsAg and HBeAg, and quantitative real-time PCR was used to measure HBV DNA, SOCS-3 mRNA, and SREBP-1 mRNA. A one-way analysis of variance and a factorial analysis were used for the statistical analysis of data.

10.Amplitude-integrated electroencephalogram combined with cranial magnetic resonance imaging for diagnosis of brain injury and prediction on neurobehavioral development in premature infants with asphyxiation

Objective:To study the predictive value of the amplitude-integrated electroencephalography (aEEG) within 6 hours and 3 days after birth and magnetic resonance imaging(MRI) on the adverse neurobehavioral development of asphyxiated preterm infants at the correction age of 6 months.Methods:From December 2017 to June 2019, 50 asphyxiated preterm infants who were delivered at the obstetrical department transferred to the division of neonatology in the Third Affiliated Hospital of Anhui Medical University were monitored by aEEG within 6 hours after birth, then once a day for at least 4 h. MRI was administered at 40 weeks of corrected age, neuromotor developmental function of the infants was assessed by the Geisel developmental diagnostic scale at 6 months of corrected age, then the infants were divided into good prognosis group and poor prognosis group according to the assessment results. SPSS 19.0 software was used for statistical analysis.The software of SPSS 19.0 was used to analyze the data.Independent sample t-test and χ 2 test were used to analyze the difference between the two groups.The relationship between aEEG grading and MRI, and their predictive value for adverse neurobehavioral development were analyzed at 6 months of corrected age. Results:The degree of white matter damage( H=24.896) and intracranical hemorrhage( H=29.245) of premature infants with different aEEG clinical grades were different (both P<0.01) on MRI. The sensitivity of aEEG within 6 hours and 3 days after birth on predicting poor prognosis was 96.2% and 97.8%, the specificity was 56.2% and 62.5%, the negative predictive value was 98.2% and 99.0%, the positive predictive value was 37.8% and 52.3%, the correct index was 52.4% and 60.3%, respectively. The aEEG was combined with MRI, the sensitivity (90.0%, 97.0%), specificity (89.0%, 99.0%), negative predictive value (99.2%, 99.5%), positive predictive value (80.6%, 88.5%), and correct index (79%, 96%) were all improved. Conclusion:The combination of aEEG grading and MRI can improve the prognostic value on neurodevelopmental prognosis, and provide a better evaluation basis for clinical follow-up and intervention of asphyxiated premature infants with brain injury.
