Vaccination is the primary strategy for the prevention and control of pandemic influenza. Because influenza virus is highly variable across strains, universal influenza vaccines need to be developed to address this problem. This review describes the research progress in conserved epitopes of influenza virus, the advances in the research and development of universal influenza vaccines based on the relatively conserved sequences of NP, M2e, HA2, and headless HA, the mechanisms of cross-protection, and the methods to improve cross-protection.

2.Acute adverse effects of radiotherapy on HIV-positive patients with malignancy

Objective To observe and evaluate acute toxicities in a series of human immunodeficiency virus (HIV)-positive cancer patients receiving radiation therapy.Methods The study retrospectively reviewed the acute radiation reaction of radiation therapy of 14 HIV seropositive patients diagnosed with carcinoma between Feb 2008 and Dec 2013 at the Yunnan Tumor Hospital during the radiotherapy period and 1 month following treatment.Acute adverse effects were classified according to the site of radiation therapy and analyzed using the Common Terminology Criteria for Adverse Events (CTCAE) version 3.0.Results Seven patients experienced interruptions or delays in treatment,and 2 stopped treatment entirely.The most common acute adverse effects were skin reactions and mucous membrane reactions,including dermatitis,stomatitis or diarrhea.Eight patients had grade 3 acute adverse effects,including 6 patients with grade 3 skin reactions and 2 patients with grade 3 mucosa reactions.Conclusions Radiotherapy is an effective treatment for HIV positive patients with tumors,however it frequently induced severe acute radiation responses.

5.Effect of Gleditsia sinensis stings on growth inhibition and expression of PCNA and p53 in mice bearing uterine cervical carcinoma (U14).

OBJECTIVETo study the antitumor effect of the stings of Gleditsia sinensis on mice bearing uterine cervical carcinoma (U14) and the expression of PCNA (proliferating cell nuclear antigen) and p53.
METHODThe effect of the ethanolic extract of G. sinensis stings on the inhibition rate of solid tumor and the life span of ascites tumor were calculated by the animal tumor model experiment in vivo. The positive cell numbers of PCNA and mutant p53 protein were measured by immunohistochemical SP method.
RESULTAs compared with the control group, the ethanolic extract of G. sinensis stings (250, 500 and 1 000 mg x kg(-1) body weight, p.o.) and CTX (25 mg kg(-1) body weight, i.p.) administration significantly reduced the tumor weight of solid tumor and increased the life span of ascites tumor harboring mice (P < 0.01). The inhibition rate of solid tumor and the rate in life span were up to 47.44%, 59.49%, 63.92%, 73.42% and 52.21%, 67.26%, 78.76%, 95.58% respectively. Meanwhile,the expression of PCNA and mutant p53 protein also suppressed by ethanolic extract (P < 0.05, P < 0.01).
CONCLUSIONThe stings of G. sinensis showed antitumor activity and its possible mechanism might be related with the expression inhibition of PCNA and mutant p53 protein.

6.The changes of GAP-43 expression in spiral ganglion after exposed to noise

OBJECTIVE To understand the synaptic remodel in inner ear after hearing injury by investigating growth associated protein 43(GAP-43) expression in the inner ear hair cells after exposed Kangmin mice to neural injury noise stimulation to induce permanent threshold shift(PTS) or temporary threshold shift(TTS).METHODS To compare the expressions of GAP-43 of NMDAR signal pathway during neural injury stimulation by immunocytochemistry.RESULTS The sustained GAP-43 increase occurs after 7d and 14d of PTS group,and there is not remarkable difference between them.Compared with normal control,the change is not significant in TTS group.CONCLUSION The increase of GAP-43 in spiral ganglion after 7d trauma reveals the synaptic remodel in inner ear.
7.Analysis of intractable cough in 128 children.

OBJECTIVETo disclose the etiological factors and to investigate the therapeutical effect on nasal disorders in 128 children with intractable cough.
METHODSOne hundred and twenty-eight children with intractable cough were consulted by ENT doctors using electronic nasopharyngoscope, for those children with nasal diseases, relevant treatment was given.
RESULTSAmong 128 children, one hundred and sixteen had nasal disorders. Upper airway cough syndrome (UACS) was diagnosed in 92 cases (71.9%), nasal disorders without UACS was diagnosed in 24 cases (18.7%), no nasal disorders were found in 12 cases (9.4%). Among children with UACS, allergic rhinitis was diagnosed in 76 cases, rhino-sinusitis in 39 cases, adenoid hypertrophy in 54 cases, and chronic rhinitis in 8 cases. However, for those children without UACS, 10 cases had chronic rhinitis and 14 cases had allergic rhinitis. Clinical symptoms, signs and the findings of electronic nasopharyngoscope were analyzed in 92 children with UACS. The results showed that regular cough occurred in 62 cases, nasal disease in 65 cases, subjective feeling of postnasal discharge in 18 cases, mouth breathing or snoring in 32 cases, purulent secretion in nasal meatus in 61 cases, retropharyngeal folliculosis in 58 cases, purulent secretion in pharynges detected from pharynx in 41 cases, and purulent or viscosity secretion were found in 91 cases by first electronic nasopharyngoscopy. After 14 days of treatment, the children with UACS (90/92) had significant higher remission rate (chi² = 32.21, P = 0.000) in cough than those who had nasal disorders but without UACS (13/24).
CONCLUSIONSThe nasal disorders, especially for UACS, were common in children with intractable cough. The therapy to nasal disorders could release the chronic cough symptoms in children with intractable cough.

8.Inhibition of HCV replication by HCV specific U1 small nuclear RNA chimeric ribozyme in vivo.

OBJECTIVETo determine the advantage of U1 small nuclear RNA as a ribozyme vector (U1-Rz) to inhibit HCV replication in vivo.
METHODSThe 3rd stem-loop was substituted by HCV core specific ribozyme to construct an U1-Rz eucaryotic expression plasmid. Then it was co-transfected with pCMV/T7-NCRC Delta-luc into Huh7 cell line mediated by lipofectin. The cell lysis supernatant was subjected to Western blot and lumenometer to determine the luciferase levels.
RESULTSA U1 snRNA chimeric ribozyme was constructed successfully. Both Rz and U1-Rz inhibited luciferase expression in Huh7 by 48.64% and 87.46%, respectively.
CONCLUSIONRz has more efficacy in cells when using U1 snRNA delivery system. U1 can be an efficient vector for HCV specific ribozyme.

9.Expressions of metastatic tumor antigen 1 and hypoxia-inducible-factor l alpha in lung cancer and their clinical significance.

OBJECTIVETo explore the correlation of the expressions of metastatic tumor antigen 1 (MTA1) and hypoxia-inducible-factor l alpha (HIF-1alpha) to the clinical features of lung cancer.
METHODSThe expressions of MTA1 and HIF-1alpha proteins in 59 lung cancer patients were detected by immunohistochemistry.
RESULTSPositive staining of MTA1 and HIF-1alpha was found in 54.24% and 50.85% of the patients, respectively. MTA1 expression was not found to correlated to any of the clinical parameters. HIF-1alpha expression was significantly lower in poorly differentiated than in moderately and well differentiated lung cancer (P<0.05), and a positive association between the expressions of MTA1 and HIF-17alpha were noted (P<0.05).
CONCLUSIONMTA1 and HIF-1alpha overexpression occurs in lung cancer possibly as an important factor of lung carcinogenesis. MTA1 may promote lung carcinogenesis by enhancing HIF-1alpha protein activity.

10.A sero-epidemiologic survey on human plague in source of three rivers area Qinghai from 2005 to 2007

Objective To explore the characteristics of human plague using a sero-epidemiologic method in the source of the three rivers area in Qinghai for possible plague control strategies. Methods Investigate human plague sero-epidemiologically in the source of 4 counties in the three rivers area in Qinghai. The human serum would be tested to confirm the sew-positive rate for plague F1 antibody using indirect hemagglutination assay(IHA). Results A total of 2508 local participants were tested in 4 counties, the overall plague sero-positive rate was 2.31%(58/2508). This represents a statistically significant difference with 4 counties(X2=19.30,P<0.01). The sew-positive rate for males and females were 2.54% (32/1261) and 2.09% (26/1247), respectively. There were no statistically significant differences between males and females(X2= 0.65,P 0.05). The sero-positive rate in herdsman, cadre, Tibetan, Hart nationalities were 3.54% (44/1243), 6.47% (11 / 170), 2.40% (56/2335) and 1.47% (2/136), respectively. The sero- positive rate increased with age. The highest titre for human plague serum antibody was 1 : 640. Conclusion There were occult infections of plague in the population on source of three rivers area in Qinghai. Sero-epidemiologic data revealed that the human plague sero-positive rate was closely correlated with the local animal plague.
