1.Effects of Neuregulin-1 on Calcium Transients in Mouse Ventricular Myocytes

Objective To observe the effects of neuregulin‐1 (NRG‐1)on calcium transients in mouse ventricular myo‐cytes.Methods Nine C57BL/6 mice of male were randomly dived into 3 groups :the blank control group ,NRG‐1 group ,and iso‐proterenol(ISO)group.Their hearts were removed and immediately cannulated via the aorta and retrogradely perfused with en‐zymatic isolation solution to get single ventricular cell by Langendorff system.Myocytes were loaded with the Ca2+ indicator Fluo‐4 and subjected to electrical field stimulation at 0.5 or 1 Hz by using living cells workstation.The change of fluorescene in‐tensity was recorded simultaneously in each group.Results Compared with the blank control group ,NRG‐1 group had signifi‐cantly increased Ca2+ transient amplitude ΔF/F0 (n=10 ,P<0.05) ,and reduced Ca2+ transient recovery time RT50 (n=10 ,P<0.05).There were no significant differences in Ca2+ transient amplitude ,Ca2+ transient recovery time and the time to peak be‐tween NRG‐1 and ISO groups(n=10 ,P>0.05).Conclusion NRG‐1 can increase Ca2+ transient amplitude and Ca2+ transients and reduce the time interval of Ca2+ transients.

Objective To explore the contamination by Legionella pneumophila in cooling water of centralized air condi-tioning system in hospitals and hotels.Methods In Aug2001,the water from cooling towers of centralized air conditioning sys-tems in4hospitals and2hotels were sampled in W city.The membranes filtering the cooling water samples were treated by hy-drochloric acid.After treatment,the mixed treated solution was inoculated into GVPC plate and cultured in candle jar.The susepectable colonies were identified by biochemical test,serological test and PCR test respectively.Results5strains of L pneu mophila in cluding3strains of Lp1type and2strains of Lp5type were isolated from water samples of the cooling towers of centralized air conditioning systems in2hospitals and one star-grade hotel in W city.Conclusion The contamination by le-gionella in water of cooling tower of centralized air conditioning system in hospitals and hotels should be monitored closely.

Objective To compare the efficacy of mechanical ventilation (MV) with low tidal volume (LTV) plus positive expiratory end pressure (PEEP) and conventioal MV in the treatment of traumatic acute respiratory distress syndrome(ARDS). Objective To evaluate the feasibility of cell free fetal DNA(cffDNA)-based noninvasive prenatal diagnosis,we developed a precise technique for fetal sex determining region of Y chromosome(SRY)gene detection using size-fractionated cell-free DNA in maternal plasma.Methods Peripheral blood samples were collected form 117 pregnant women.cffDNA was extracted based on a column absorbent method and isolated by agarose gel electrophoresis.A dulex-polymerase chain reaction(PCR)was used to detected SRY gene and glycerol-dehyde-phosphate dehydrogenase(GAPDH)gene.Results Both SRY and GAPDH gene were detected in 86 cffDNA samples from women bearing male fetuses.And only GAPDH gene was detected in 71 cffDNA samples from women bearing female fetuses.These results had a coincidence whit those of villus or amniotic fluid samples.The specificity and sensitivity reached 100%(117/117)and 100%(66/66),respectively.Conclusion By agarose gel electrophoresis,re-extratedand and dulex PCR,size-fractionated cell-free fetal DNA in maternal plasma can be selective enriched and used to noninvasive prenatal diagnosis of sex-linked disorders and single gene disorders.
6.Correlation between Val279Phe, Ile198Thr mutation in lipoprotein-associated phospholipase A2 gene and cerebral infarction

Objective To investigate the correlation between the mutation in serum lipoprotein-associated phospholipase A2(Lp-PLA2) Ile198Thr, Val279Phe and cerebral infarction in Chinese Han population of Jiangsu Province. Methods One hundred fifty patients with cerebral infarction and 100 healthy controls in Chinese Han population of Jiangsu Province were recruited. The correlation between Val279Phe and Ilel98Thr mutation in Lp-PLA2 gene and cerebral infarction was analyzed using polymerase chain reaction and denaturing high performance liquid chromatography. Results The Val279Phe genotype and the mutant allele frequency in the cerebral infarction group were significantly higher than those in the control group (χ2 were 6. 31and 5. 32, respectively, all P <0. 05), and there was no significant difference between the Ile198Thr genotype and the mutant allele frequency in the control group (χ2 were 0. 039 and 0. 037, respectively, all P >0. 05). Conclusions The Val279Phe mutation in Lp-PLA2 gene may be a genetic risk factor for cerebral infarction in Chinese Han population of Jiangsu Province.
10.Comparison of the Related Substances in Benzalkonium Chloride at Home and Abroad

Objective:To determine the related substances in benzalkonium chloride used as a pharmaceutical adjuvant, and com-pare the quality at home and abroad. Methods:An HPLC method was used with an ODS-HYPERSIL C18 column(250 mm × 4. 6 mm, 5 μm). The detection wavelength was 210 nm and 257 nm. The flow rate was 1. 0 ml· min-1 and the column temperature was 30℃. The injection volume was 20 ml. The mobile phase was A ( dissolving 1. 09 g sodium1-hexanesulfonate and 6. 9 g sodium dihydrogen phosphate in water, adjusting pH to 3. 5 with phosphoric acid and diluting to 1 000. 0 ml) and B ( methanol) with gradient elution. Results:The content of benzaldehyde in the samples at home and abroad was low. The content of benzyl alcohol in the samples from a-broad was qualified, which was significantly higher than that in the domestic samples. The content of benzyl chloride in the domestic samples was higher than that in the samples from abroad. Conclusion:The method is simple and fast, which is suitable for comparing the related substances of domastic and imported samples. At the same time, the study provides basis for enterprises to choose benzalko-nium chloride rationally.
