Objective To analyze the factors of Protein A/G affinity column, influencing purification effect, and get the best condition to improve application effect of Protein A/G affinity column. Methods We Purified anti-HCV-IgG serum with different disposal modality, different sample input modality and different application number of affinity column before detecting and analyzing the purified samples. Results The Protein A/G affinity column had the best purified effect after using saturated ammonium sulfate to first purification, which increased the affinity column adsorption effect within 30 minutes adsorption. Conclusion Using antibody with first purification and adding the adsorption time could improve utilization rate of affinity column and prolong the service life.

ObjectiveTo investigate the effect of early electro-acupuncture treatment on prevention of post-operative adhesions.Methods40 rabbits were randomly divided into 5 groups: normal control group, model group 1, model group 2, early electro-acupuncture group and later electro-acupuncture group with 8 animals in each group. Muscle adhesion model was established by operation. The early electro-acupuncture group was received the treatment from the 11th to 20th day after the operation; later electro-acupuncture group was from the 21st to 30th day after the operation. The weight, gait and range of motion (ROM) of animals were observed and analyzed.ResultsThe standing, walking and ROM of animals of the early electro-acupuncture group improved; but only few gait indexes improved and ROM not changed in the later electro-acupuncture group.ConclusionThe early electro-acupuncture treatment can prevent the post-operative adhesion of muscles, but not having effect on formed adhesion.

Objective To establish a method for determination of total hardness in drinking water by using automatic potentiometric titrator. Methods Dynamic equivalence point titration(DET)and monotonic equivalence point titration(MET)mode was used to determine the high total hardness and low total hardness of drinking water samples respectively. Results Used DET mode to determine the high total hardness, the relative standard deviation(RSD)was 0.69%~1.72% and the recovery rate was 101.5%~102.2%.Used MET mode to determine the low total hardness,RSD was 3.49%~4.00% and the recovery rate was 95.8%- 103.6%.Conclusion This method is rapid, simple, accurate, precise and applicable to the determination of total hardness in drinking water in low and high levels.

Objective To analyze the characteristics of gastric stromal tumors(GST)under endoseopic ultrasonography(EUS)according to its aggressive risks. Methods The clinical data of 36 patients with GST,who underwent surgery from July 1997 to July 2007,were analyzed retrospectively. All the patients underwent EUS before operation and were classified according to Fleether's 4-tier system to predict the aggressiveness of the tumors. The features of the tumor under EUS including its size, ulceration, border, echo charateristies and growth pattern were recorded and the difference between each tumor group were analyzed by ANOVO and rank sam test. Results The mean maximal diameter of GST Was 7.3 cm(range 1.0-20.0 cm),and the size of tumors with hiisher aggressiveness risk Was significantly larger than that with lower risk(P<0.01).The mucosal ulceration, obscure border, irregular shape and echo heterogeneity were more commonly seen in the groups with higher risk(P<0.05).There Was no difference in the internal echo and growth pattern of GST between different groups. Conclusion EUS features are useful in differentiation of the aggressiveness risk of the GST.and guide the management of the tumors.

Objective To investigate whether valproic acid (VPA) affect spatial learning memory and senile plaques in the APP/PS1 double transgenic AD mouse model of different gender. MethodsTwenty 3-month old APP/PS1 double transgenic AD mice, male and female mouse evenly, were randomly divided into VPA-treated and saline-treated groups ( 10 for each group). 30 mg· kg-1 · d-1 of VPA and the same amount of saline were peritoneally injected into mice for 4 weeks. Morris water maze was conducted to check the effect of VPA on the capability of spatial learning and memory of AD mouse model. Immunohistochemical staining was used to examine the effect of VPA on the morphological changes in the brains of mice. ResultsVisible platform test showed that VPA-treated and saline-treated mice had similar escape latency (P>0.05) and path length (P>0.05) ,the swimming speed between male and female mice had no difference (P>0.05). Hidden platform test showed that VPA treated mice had a significantly shorter latency (P<0.01) and path length (P<0.01) to reach the platform compared with saline-treated mice. Meanwhile, both in VPA-treated and control groups, the male mice had a shorter correlation escape latency and path-length than female mice had(P<0.05 ). Immuohistochemical staining showed that the number (11.23±3.78) of senile plaques (SP) in the cerebral cortex and hippocampus of VPA-treated male mice were notably decreased than that(28.17 ±3.46) in the control group ( t= 14.67, P<0.01 ),furthermore, the number of SP in the cerebral cortex and hippocampus of VPA-treated male mice was significantly reduced,as compared with which (20.36 ±4.21)in the VPA-treated female mice(P<0.05). ConclusionVPA can significantly lower formation of SP, and remarkably improve the capability of spatial learning and memory of APP/PS1 transgenic mice, which have gender difference.

The new generation cyclooxygenase (COX-2) inhibitor could reduce the gastrointestinal side effect of NSAID drugs, but eventually increase the cardiovascular risk, because its selective inhibition of COX-2 induces the imbalance between PGI2 and TXA2 and the reduction of vasodilatory NO. Under pathological conditions, active oxygen species (O2-*2, etc) were used to induce endo- thelial dysfunction, activate NF-κB to induce expressions of pro-inflammatory cytokines IL-1β and TNF-α, increase ET-1, TXA2 with vasoconstrictor effect, reduce PGI2 and NO with vasodilatory effect, generate further oxidative damage together with NO, and reduce the bioavailability of NO. NO-NSAIDs and NO-Coxibs drugs raised the level of NO by introducing NO-donor (ONO2). NSAIDs drugs enhanced the anti-inflammatory activity of COX-2 and reduced gastrointestinal side effects by inhibiting selectively COX-2. If antioxidant structures with active ingredients of traditional Chinese medicines were introduced to improve the antioxidant activity of NSAIDs, they could scavenge the active oxygen species to protect the normal function of vascular endothelia and enhance the bioavailability of NO, which is conducive to enhance the cardiovascular safety of cyclooxygenase (COX-2) inhibitor.

Journal of Kunming Medical University 2016;37(6):117-122
Objective To investigate the clinical characteristics of hodgkin's lymphoma (HL) patients . Method The clinical data of HL patients in our hospital from January 1, 2003 to December 31, 2012 were analyzed retrospectively. The contents of investigation included gender, age, pathological type, first symptom, the incidence locations, clinical stages, extranodal involvement locations, ECOG and treatments. Results A total of 143 cases of HL were included, the M/F ratio was 2.5:1 with a median age of 37 years (11~73years) . The most frequent subtypes of HL were mixed cellular (34.3%) and nodular sclerosis (28.0%) . The nodal lymphomas accounted for 97.2%, while the extranodal lymphomas occupied 2.1%. The nodal lymphomas with extranodal invasion accounted for 29.5%, and the common locations were spleen (14.4%), lung (5.8%) and bone (5.0%) . The most clinical stage of all patients was stage Ⅰ/Ⅱ (60.8%), The differences were no significant in gender, pathological type, first symptom, the incidence locations and the effective rate of the first-line treatment (P>0.05), while were statistically significant in B symptoms, ECOG and treatments betweenⅠ/Ⅱgroups andⅢ/Ⅳgroups (P<0.05) . Conclusions In this group, the incidence of HL patients were mainly found in middle-aged people, males were lager than females. The pathological type, first symptom and the incidence locations had no obvious effect on clinical stage. The clinical stage had no obvious influence on the effective rate of the first-line treatment.
