2. Inhibitory effects of melittin on osteosarcoma xenografts in nude mice and its action on vasculogenic mimicry

Objective: To investigate the antitumor effects of melittin on UMR-106 cells with osteosarcoma xenografts in nude mice and its action on vasculogenic mimicry (VM) in vitro and in vivo. Methods: A three-dimensional cell culture system was formed on matrigel coats in vitro. Inhibitory action of melittin on the VM of UMR-106 cells was observed in vitro. Osteosarcoma was orthotopically transplanted in nude mice. The mice were randomly divided into three groups: normal saline (NS)-treated group, melittin group (320 μg/kg) and DDP-treated group(2 mg/kg). The drug was administered intratumorally for 10 d. The tumor volume and weight inhibition ratio were calculated, respectively. The VM density in tumor tissues was detected by CD34 and PAS double staining in vivo. The expressions of hypoxia-inducible factor-1α(HIF-1α) and matrix metalloproteinase-2 (MMP-2) proteins were determined by immunohistochemical staining and Western blot. Results: Melittin broke down the mesh and round architectonic of VM in osteosarcoma in vitro. The tumor weight inhibition ratio was 38.92% and the volume inhibition ratio was 43.04%. The VM density and the expressions of HIF-1α and MMP-2 proteins were decreased in the melittin group than that in the NS-treated group (P <0.01). Conclusion: Melittin markedly inhibites the growth of osteosarcoma UMR-106 xenografts in nude mice. The mechanism may be due to down-regulation of HIF-1α and MMP-2 expression and the inhibition of VM.
3.Perinatal outcomes affected by the time of interpregnancy interval after spontaneous abortion

3 months group,6 neonates were admitted to intensive care unit,2 with birth defects and 1 died. Conclusion There was no clear evidence in this study demonstrating any association between adverse perinatal outcomes and pregnancy immediately after spontaneous abortion.
4.A Study on the Biocompatibility between Collagen-Polyglicolic Acid and Bone Marrow Mesenchymal Stem Cells

Objective To investigate the biocompatibility between collagen- polyglycolic acid (PGA) and bone marrow mesenchymal stem cells (MSCs) in vitro to provide some experimental basis for further study in tendon tissue engineering. Methods MSCs were isolated, cultured and characterized. In the experimental group the MSCs were cultured in DMEM containing type-I collagen and PGA suture, and in the control group the MSCs were cultured in DMEM. The cell growth was compared between the two groups, and the cell ultramicroscopic structure of experimental group was observed. Results MSCs grew well in the collagen-PGA scaffold, and 2 weeks after incubation they still kept secretion potential and more than cell 89% vitality, which were not significantly different compared with the control group. There is no statistical difference in the MSCs count in the experimental group during 2 weeks culture, while in the control group MSCs began to proliferate at the 4th day after culture. Conclusion Collagen-PGA has a good biocompatibility with mesenchymal stem cells. It is possible to fabricate a tissue-engineered tendon in vitro using mesenchymal stem cells as seed cells and collagen-PGA as scaffold.
5.Endoscopic sternocleidomastoid muscle amputation for the treatment of congenital muscular torticollis

Objective To investigate curative effects of endoscopic sternocleidomastoid muscle amputation for the treatment of congenital muscular torticollis.Methods A total of 23 children with congenital muscular torticollis were treated in this department.Their age was 1 month ~ 12 years old(median,30 months).A 5 mm trocar was inserted through the right axilla along the cleavage lines.Under the endoscopic visualization,the sarcolemma on the inferior portion of the muscle was bluntly dissected and a subcutaneous space was established by CO_2 inflation at the pressure of 6 mm Hg.Another two 3 mm incisions were made along the cleavage lines at lower lateral part of the neck and the anterior chest wall for the introduction of curved forceps and electric knife,respectively.The sternocleidomastoid muscle was transected with electrocautery and the external fascia within which the sternocleidomastoid muscle resides was also adequately divided. Results The operation was successfully completed under endoscope in all the 23 children.The mean operation time was 51.2 min(range,(35~)135 min) and the intraoperative blood loss was
6.Laparoscopic Splenic Artery Ligation in the Treatment of Hypersplenism and Thrombocytopenia in Children

Objective To investigate the effectiveness and feasibility of laparoscopic splenic artery ligation in the treatment of hypersplenism and thrombocytopenia in children. Methods From August 2014 to December 2014, four children with hypersplenism and three children with ITP (idiopathic thrombocytopenic purpura) were treated in our hospital.Laparoscopic splenic artery ligation was performed in all the patients.During the operation, the gastric colon ligament was cut with an ultrasonic knife, and the gastric wall was suspended to expose the pancreas and the spleen.The splenic artery was ligated at the upper edge of pancreas closing to the splenic hilum.The splenic venous branches were dissected and ligated at the splenic hilum by using the Hem-o-lok, which leaded to an area of splenic infarction more than 50%. Results The laparoscopic splenic artery ligation was successfully performed in all the patients, without conversion to open surgery.The operative time was 120-150 min ( mean, 126 min) , and the intraoperative blood loss was 10-20 ml ( mean, 15 ml) .No patient underwent blood transfusion.The length of postoperative hospital stay varied from 4 to 11 days (mean, 6.6 days).The splenic length significantly decreased postoperatively [(13.6 ±2.6) cm vs.(15.1 ±1.7) cm, t=3.199, P=0.049], and so was the splenic thickness [(3.8 ±1.0) cm vs.(4.1 ±0.8) cm, t =3.703, P=0.034].Partial splenic infarction and decreased splenic blood flow were found in the 7 patients by postoperative ultrasound.After surgery, four children suffered from fever, which were 38.8 ℃, 39.0 ℃, 38.6 ℃, and 39.2 ℃, with the duration of fever of 2, 4, 8, and 5 days, respectively.All the patients were followed-up for 6 -10 months (mean, 8.4 months).The complete blood cell count was within normal range. Conclusion The laparoscopic splenic artery ligation is an effective treatment for hypersplenism and thrombocytopenia in children.
8.Bioinformatics analysis and expressed level of histone methyltransferase genes in Lonicera japonica.

Twenty-three histone methyltransferase genes were obtained from transcriptome dataset of Lonicera japonica. The nucleotide and proteins characteristics, subcellular localization, senior structural domains and conservative forecasting were analyzed. The result of phylogenetic tree showed that 23 histone methyltransferases were mainly divided into two groups: lysine methyltransferase and arginine methyltransferases. The result of gene expression showed that 23 histone methyltransferases showed preference in terms of interspecies and organs. They were more expressed in buds of L. japonica than in L. japonica var. chinensis and lower in leaves of L. japonica than in L. japonica var. chinensis. Eight genes were specific expressed in flower. These results provided basis for further understanding the function of histone methyltransferase and epigenetic regulation of active ingredients of L. japonica.

9.Multi-slice Spiral CT Study of Liver Graft Regeneration after Living Donor Liver Transplantation

Objective To study the effect of liver transplantation using right liver lobe Ⅴ and Ⅷ segments with middle hepatic vein(MHV) and without main MHV on the regeneration of liver transplantation. Methods 25 pair of donors and receptors of liver transplantation were divided in to two groups:with M HV group (group A, n= 14) and without main MHV (group B, n=11). The volumes of Ⅴ and Ⅷ segments of liver were measured in donors presurgicaUy,half month and 3 month in receptors postsurgically, the rates of regeneration of the Ⅴ and Ⅷ segment were calculated half month and 3 months postsurgically and compared between two groups. Results In group A,the regenerate rates of the Ⅴ,Ⅷ segment were 0.360±0.043 and 0.853±0.059 half month after surgery,0.253±0.043 and 0.708±0.059 three months after surgery respectively. In group B, the regenerate rates of the Ⅴ,Ⅷ segment were 0.306±0.049 and 0.815±0.066, respectively half month after graft, 0.161±0.049 and 0.627±0.066, respectively three months after surgery. There were no statistically significant differences in regenerate rates of the Ⅴ, Ⅷ segment of graft be-tween the two groups (P= 0.685 ,P>0.05 and P= 0.738, P>0.05). Conclusion The right lobe living donor liver transplantation, maintaining the MHV or reconstruct MHV tributaries without MHV has similar effects on the regeneration of Ⅴ,Ⅷ segment of graft.

Objective To emphasize the correlation and concordance of electronic medical record through analysis of frequently asked problems on handwritten medical record.Methods The problems of electronic medical record were classified and found the typical cases to analyze.Results In order to improve the quality of medical record,the quality control of electronic medical record must be strengthened and the phenomenon that some doctors pay attention to hand-written med-ical record more than electronic medical record must be overcame.Conclusion The normative management of electronics medical is strengthened that is fundamental guarantee for paper-based medical records.
