1.Advances in research on pericolonic tumor deposits in colorectal cancer

It has been a problem in clinical practice that recurrence and distant metastasis of patients with colorectal cancer undergoing surgery.Recently,pericolonic tumor deposits has attracted extensive attention of scholars,and they think pericolonic tumor deposits may be closely related with colorectal cancer postoperative recurrence and metastasis.This review aims to introduce concept of pericolonic tumor deposits,pathological characteristics and its important role in clinical staging and prognosis,so as to provide clinical doctors medical advice to set more reasonable treatment and achieve the purpose of reducing tumor recurrence or metastasis for patients with colorectal cancer.
2.Progress in the research on the vector for mediating RNA interference in mammals

RNA interference (RNAi) is a process to inhibit specific gene expression via the degradation of the target mRNA induced by the double strand RNA. The use of RNAi in mammals as a tool to study gene function has rapidly developed in recent years. Here we described the novel progress and applications of the vector for mediating RNAi in mammals.
3.Activity of CDK1 in S phase cell checkpoint

Objective To investigate the phosphorylation and dephosphorylation of CDK1 based on the specific cell cycle apoptosis in Molt-4 cells and active variety of CDK1 in cell cycle specific apoptosis.Methods The exponential phase of growth Molt-4 cells (the human acute lymphoblastic leukemia cell line) were induced with dose response and time course of Camptothecin (CPT).The specific cell cycle apoptosis was detected with API method,then cell apoptosis was verified with post sorting confocal method.Meanwhile,the phosphorylation and dephosphorylation of CDK1 were detected by the protein electrophoretic analysis (Western blot).Results The specific cell cycle apoptosis occurred on exponential phase of growth Molt-4 cells after CPT treatment.When Molt-4 cells occured S-phase apoptosis, the apoptosis cell phosphorylation of CDK1-Thr161 band was more narrow than that of control cells, the apoptosis cell phosphorylation of CDK1-Thr15 band almost had the same band with control cells.Conclusion Cell apoptosis frequently developed in different cell cycle phase. API assay is quick and efficient method to analyze specific cell cycle apoptosis. When cell apoptosis take place in S-phase,the phosphorylation activity of CDK1 is inhibited.

In recent years,the role of phosphodiesterase 5(PDE5)has been highlighted in the development and progression of neurological disease.PDE5 inhibitors show significant effect of neruoprotection,which may be related with some effects such as resistance to stroke,anti-oxidation,inhibition of neuroinflammation and amelioration of cognitive deficits.Based on the domestic and overseas researches about PDE5,this review systematically summarized the neuroprotection of PDE5 and their related mechanisms.
5.Clinical research for thulium laser vaporization enucleation of the prostate combined with traditional transurethral resection of prostate for therapy benign prostate hyperplasia larger than 80 millilitres

Objective To evaluate the feasibility and the safety of thulium laser vaporization enucleation of prostate(TVEP) combined with traditional transurethral resection of prostate (TURP) for therapy benign prostate hyperplasia (BPH) with its volume larger than 80 millilitres.Methods Twenty-five BPH patients (volume larger than 80 millilitres) underwent TVEP combined with TURP.The operation time,intraoperative blood loss,enucleation time,cutting time,bladder irrigating time,catheterization time,perioperative and 6 months' follow-up data such as the international prostate symptom score,quality of life score,the maximum urinary flow rate,the residual urine volume and so on were observed.Results The operation time was (66 ± 26) min.The enucleation time was (25 ± 9) min.The cutting time was (32 ± 8) min.The intraoperative blood loss was (140 ± 25) ml.The bladder irrigation time was (3.0 ± 1.0) d.The catheterization time was (5.7 ± 1.0) d.After 6 months,the maximum urinary flow rate,residual urine volume,international prostate symptom score and quality of life score were improved:(18.7 ± 1.7) ml/s vs.(6.8 ± 1.7) ml/s,(18.9 ± 1.8) ml vs.(65.7 ±8.1) m1,(8.7 ± 1.6) scores vs.(25.7 ±4.3) scores,(1.7 ± 1.2) scores vs.(4.7 ± 1.1) scores,and there were significant differences (P < 0.05).Urethral stricture developed in 2 patients and epididymitis happened in 3 patients.No blood transfusion events and transurethral electric cutting syndrome occurred.Conclusion TVEP combined with TURP for therapy BPH larger than 80 millilitres is safe,and the incidence of complications is low.
6.Research progress in radiotherapy for liver tumors reported in 2013 ASTRO Annual Meeting

With the development of radiotherapy techniques,the efficacy of radiotherapy for liver tumors has been improved,and radiothera-py can be considered as an alternative treatment for patients who are not suitable for surgery.The progress in liver tumor radiotherapy repor-ted in the 55th annual meeting of the American Society for Therapeutic Radiology and Oncology (ASTRO)in 2013,particularly the efficacy and safety of stereotactic body radiation therapy for liver tumors,is reviewed.In the future,radiotherapy may be increasingly used as an ef-fective method in the treatment of liver tumors.The main studies reported at the meeting are sorted as follows in this article.
7.Protective role and immunoregulatory effect of CXCR3 in hepatic ischemia-reperfusion injury

Objective To investigate the role and action mechanism of chemokine (C-X-C motif)receptor 3 (CXCR3)in hepatic ische-mia-reperfusion injury (IRI).Methods Forty-eight mice were divided into operation group and sham-operation group.The operation group was treated to establish a mouse model of IRI.Liver tissues were obtained at 3,6,12,and 24 h after IRI,with 6 mice at each time point.The expression of chemokine (C-X-C motif)ligand 9-1 1 (CXCL9-1 1 )and their receptor CXCR3 were measured by real-time PCR and Western blot.The effect of CXCR3 was blocked by its specific antagonist C6.Hepatic injury was estimated based on the activity of hepatic transaminase and morphological indices.The distribution of subsets of infiltrating T cells was analyzed by flow cytometry.All data were expressed as mean ±standard deviation.Comparison between groups was made by one-way analysis of variance.Results Compared with the sham-operation group,the operation group had significantly upregulated expression of CXCL9-1 1 and CXCR3 at all time points after IRI (P<0.05).Blocking CXCR3 significantly protected liver function and morphology (P<0.05).Antagonist C6 significantly re-duced Th1 cell infiltration (P<0.01),but significantly increased Treg infiltration (P<0.01).Conclusion CXCR3 is an ideal therapeu-tic target in IRI treatment due to its relationship with immunoregulation.
8.Expression of Ezrin and inducible nitric oxide synthase and their correlation in colonic adenoma

ObjectiveTo investigate the expression of Ezrin and inducible nitric oxide synthase (iNOS) and their correlation to malignant proliferation of colonic adenoma. MethodExpressions of Ezrin,iNOS and nuclear Ki-67 antigen were detected by immunohistochemistry in tubular adenoma (60 cases),villous adenoma(40 cases) and malignant (30 cases ) pleomorphic adenoma. ResultsExpressions of Ezrin,iNOS in tubular adenoma [58.3% (35/60), 50.0% (30/60)], villous adenoma [72.5% (29/40), 70.00(28/40)]and malignant pleomorphic adenoma [96.7%(29/30),93.3%(28/30)]were gradually increased ( X2 = 11.600,P = 0.005; X2 = 8.451, P = 0.015 ). There was a significant positive correlation between the expression of Ezrin and iNOS (r = 0.765, P < 0.01 ). Nuclear Ki-67 antigen proliferation index was increased with the increasing of Ezrin and iNOS expressions. ConclusionOverexpression of Ezrin and iNOS may promote proliferation and malignant transformation of colonic adenoma.

Objective For analysis of the ability to resist the shear stress and anti-calcification of endothelial cells (EC), and analysis of migrating and self-repairing ability of myofibroblasts. Methods (1) The fresh bovine pericardial patches were acellularized, tanned and modified. (2) Autologous myofibroblasts and ECs were seeded onto the patches of Group A sequentially; Group B, unseeded group. Then the patches of both groups were implanted to the porcine abdominal aortic wall separately. (3) The retrieved specimens were sent for thickness, calcium content, scanning electron microscopy (SEM) and histological examination. Objective To evaluate the efficacy and safety of composite milk clotting enzyme capsules in children with functional dyspepsia.Methods A multicenter,randomized,open-label clinical trial was carried out in 4Guangzhou hospitals during Feb.2012 to Mar.2013.Children with functional dyspepsia on basis of Rome Ⅲ criteria were enrolled and treated with composite milk clotting enzyme capsules.Total symptom score and drug-related adverse reactions were evaluated after one-week and two-week therapy.The treatment outcome were divided into very effective,fairly effective and ineffective according to the symptom score changes.Superiority test was done for effectiveness of the treatment based on efficacy difference by 10% regarded as superiority.Results Two hundred and one children were enrolled.One hundred and ninety-six children completed the study as planned.After two-week therapy,better effective rate and total effective rate were 68.88% and 87.76%,higher than those of one-week therapy (27.04%,76.02%) (U =2.935,P < 0.05).The lower limits of 95 % confidence interval of difference in symptoms improve rate,better efficacy rate and symptom score decrease rate between two-week and one-week therapy were more than 10%,and two-week therapy was superior to one-week therapy.No significant drug-related adverse reaction was found during the study.Conclusions Composite milk clotting enzyme capsules can improve functional dyspepsia symptoms significantly.The effectiveness of two-week of treatment is better than that of one-week treatment.No obvious drug-related adverse reaction is observed.
