2.Nursing cooperation during posterior cruciate ligament reconstruction using LARS artificial ligament under arthroscopy

Objective To investigate the nursing experience during posterior cruciate ligament(PCL)reconstruction using LRAS artificial ligament under arthroscopy.Method The experience in nursing 51 cases of PCL reconstruction using LARS artificial ligament under arthroscopy from 2009 to 2012 was retrospectively summarized.Results It showed that KDC score of the 39 cases was excellent and 12 cases good,with the excellent and good rate of 100.0%.The average operation duration was(70.0±6.0)min and the average blood lose(50±3.6)ml.Conclusion Sufficient preparation before operation,much familiarity with LARS artificial ligament,skilled and accurate surgical cooperation are important factors for the success of PCL reconstruction using LARS artificial ligament under arthroscopy.
3.Management of 240 cases of non-functioning renal graft

Objective To explore the methods for treating no-functioning renal graft.Methods The management of 240 cases of non-functioning renal graft between 1978 and December 2003 was retrospectively analyzed.Results Twenty-nine recipients with renal failure one month after renal transplantation were subjected to removal of the renal graft. One year after renal transplantation, the recipients with renal failure received resection of the renal graft, 4 were treated with renal artery embolism, and 114 did not receive special treatment.Conclusion The non-functioning renal graft required removal at early stage of renal transplantation. At later stage, no symptomatic cases of post-transplanted renal failure required no management after stopping immunosuppressant. The appropriate management is reserved for the symptomatic cases.
4.Measurement of the Parameters for Tinnitus Masking Therapy

ObjectiveTo search for the optimum external masking time and tonal models for patients with tinnitus.MethodsTwenty-seven patients with cochlear tinnitus were selected respectively from three different out-patients departments and divided into three groups. Nine patients in every group were treated with different masking parameters according to factorial design which had three masking noises, i.e. narrow band noise (NBN), wide band noise (WBN), speech noise (SN),and three masking time ranges, i.e. 15,30,45 minutes. The results before and after treatment were compared and analysed with analysis of variance (ANOVA). ResultsThe treatment effect when masking for 15 minutes was worse than that for 30,45 minutes, but there was no difference between 30 and 45 minutes. The NBN had the best effect when tinnitus signal was single, the WBN was effective for tinnitus with multiple sounds. ConclusionNBN or WBN masking for 30 minutes showed better therapeutic effect.
5.Recombinant CXCR4 gene lentivirus transfection of rat bone marrow mesenchymal stem cells in vitro

BACKGROUND:Chemokine receptor 4 (CXCR4) is the main homing regulating factor for mesenchymal stem cells in vivo. How to improve the self-expression of CXCR4 is crucial for regulating the homing ability of stem cells in vivo targeting organs. OBJECTIVE:To construct lentiviral vectors carrying CXCR4 or green fluorescent protein (GFP) gene, and to observe their effects on growth and secrete function of bone marrow mesenchymal stem cells. METHODS:Lentiviral vectors were used to over-express CXCR4 in rat bone marrow mesenchymal stem cells. GFP gene was transfected as a control. Transfection efficiency was analyzed using fluorescence microscopy, RT-PCR and western blot assay for detecting the expression of CXCR4 or GFP in cells at both the mRNA and protein levels. Effects of CXCR4 expression modified on the cellular proliferation of bone marrow mesenchymal stem cells were assayed with flow cytometry. Effect of CXCR4 regulation on secretion function of bone marrow mesenchymal stem cells was determined by protein analysis from cellsupernatant. RESULTS AND CONCLUSION:CXCR4 gene or GFP gene could be transfected into bone marrow mesenchymal stem cells safely and effectively with lentivirus transfection system. Analysis of CXCR4 expression at the mRNA and protein levels confirmed the transfection efficiency. Effects of CXCR4 over-expression on the cellular proliferation of bone marrow mesenchymal stem cells and effects of CXCR4 regulation on secretion of protective factors from bone marrow mesenchymal stem cells were improved. The proliferation ability of bone marrow mesenchymal stem cells with CXCR4 over-expression was improved, which was approved with flow cytometry (P<0.05). And it was helpful for bone marrow mesenchymal stem cells to secrete some protective factors. Many protein and soluble factors were at higher levels, and those were good at cells proliferation and immunoloregulation in bone marrow mesenchymal stem cells (P<0.05). The method of lentiviral transfection is a safe and effective way to modify bone marrow mesenchymal stem cells. We can improve the ability of bone marrow mesenchymal stem cells in the survival and emiocytosis ability through genetic engineering.

Objective:To investigate the modulating effects of anti-epidermal growth factor monoclonal antibody Cetux- imab(C225)on the ehemosensitivity and radiosensitivity in a Docetaxel-resistant human lung adenocarcinoma cell line SPC-A-1/doeetaxel.Methods:Radiosensitivity of SPC-A-1/docetaxel was determined by clone formation experiment and quantified by calculating the enhancement ratio(ER).The growth inhibition of SPC-A-1/docetaxel cell line caused by C225 or combination of C225 and Docetaxel in different orders was detected by MTT assay.The effect of C225 on cell cy- cle distribution and apoptosis was determined by flow cytometry.Results:C225 combined with radiation significantly de- creased the number of the cell clones than radiation alone;the D_0 values were 1.73 Gy for the former and 2.39 Gy for the latter,and the enhancement ratio was 1.38.C225 alone at concentration up to 1000?g/ml for 48h had neither cytotoxic nor eytostatie effect on SPC-A-1/docetaxel in vitro.C225 administration followed by Docetaxel significantly decreased the ICw of Docetaxel(85.2?g/ml vs 128.7?g/ml).Flow cytometry demonstrated that C225 exposure induced apoptosis of SPC-A-1/docetaxel cells in a time-dependent manner.The cell in the G_0/G_1 fraction increased from(43.80?4.46)% to (60.50?6.57)%(P
7.The Role of VCAM-1/VLA-4 in the Activation of Allogenic T Cells by Murine Macrophages

Vascular cell adhesion molecule 1 (VCAM - 1) is a member of immunoglobulin superfamily. The principal ligand for VCAM - 1 is integrin ?4|?1/VLA - 4(very late antigen 4). In recent years, VCAM - 1 was found to be expressed on macrophages and dendritic cells, but little is known about its identity on these professional antigen presenting cells (APC). In the present study we analyzed VCAM - 1 expression on macrophages by fluorescence - activated cell sorting (FACS) and found that VCAM- 1 was constitutively expressed on macrophages and its expression level was up-regulated by soluble tumor associated antigen (TAA: freeze - thaw lysates of FBL - 3 tumor cells) and TNF - a. In macrophages and allogenic T cells mixed lymphocyte reaction (MLR) assays, we observed that blocking VCAM - 1/VLA - 4 interaction with anti - VCAM - 1 or anti - VLA - 4 mAbs caused significant inhibition of the proliferative response and inhibition of IL - 2 production. These findings suggest that VCAM - 1 on macrophages not only allows for increased cell-to-cell contact through adhesive interaction but also plays a role in the costimulation of T cells via its interaction with VLA-4.
8.The Roles of MHC Class I Molecule and ICAM-1 in the Induction of CTL by IL-2, IL-4 or IL-6 Gene-Transfected Tumor Vaccines

In the present study, the expression of MHC class I molecule and ICAM - 1 on the surface of B16 melanoma cells were observed, and their roles in the induction of CTL were investigated. The results showed that both MHC class I and ICAM - 1 expression increased after IL-2, IL-4, or IL-6 gene transfection. The splenocyte CTL activity was enhanced significantly after in vivo immunization with cytokine gene - transfected B16 melanoma cells. The CTL induction was partly inhibited by anti-ICAM-1 mAb and was completely abolished by anti-MHC class I mAb. These results suggested the increased immunogenicity of IL-2,IL-4 or IL-6 gene-transfected B16 melanoma cells may be related to the upregula-tion of ICAM-1 or MHC class I molecules after cytokine gene transfection.
9.Analysis of pathogen distribution and drug resistance of 1676 cases of urinary tract infection

Objective To retrospectively analyze the pathogenic bacterial distribution and drug resistance in the patients with u-rinary tract infection in our hospital to provide a basis for clinically rational use of antibacterial drugs .Methods The midstream u-rine of the patients with urinary tract infection in our hospital during 2013-2015 was performed the bacterial culture .The bacterial identification and drug susceptibility test were performed by using the Vitek-2 Compact automatic microorganism identification in-strumen .The data were statistically analyzed by adopting the WHONET 5 .6 software .Results Among 11130 urine culture sam-ples ,1676 strains of pathogenic bacteria were cultured with the detection rate of 15 .1% ;among them ,1332 strains were Gram-negative bacteria and accounted for 79 .5% ,275 strains were Gram-positive bacteria and accounted for 16 .4% and 69 strains were fungi and accounted for 4 .1% .The top 3 pathogenic bacteria of detection rate were in turn Escherichia coli ,Klebsiella pneumoniae and Enterococcus faecalis .Six hundreds and eleven strains of ESBLs bacteria were detected ,in which Escherichia coli ,Klebsiella pneumoniae accounted for 88 .5% and 10 .2% respectively .Escherichia coli had higher sensitivity to carbapenems ,fosfomycin ,ami-kacin ,etc .(>95 .0% ) .Klebsiella pneumoniae had higher sensitivity to carbapenems (>90 .0% ) .No vancomycin ,teicoplanin and linezolid resistant Enterococcus faecalis was detected .One strain of vancomycin resistant enterococcus faecium was detected .These pathogens mainly derived from the urology surgery department ,the constituent ratio of female was higher than that of male .Conclu-sion The pathogens of urinary tract infection in this hospital is dominated by Gram-negative bacteria ,Escherichia coli is mainly pathogenic bacterium .Multidrug resistant species and number are continuously increased .Clinicians should rationally and correctly use the antibacterial drugs based on the drug sensitivity test results for improving the effect of clinical anti-infection treatment .
10.Factors for anemia after kidney transplantationA data analysis of 826 patients from one institute during 10 years

BACKGROUND: Anemia after kidney transplantation has a clinical incidence rate of 30%-40%, is the important risk factor for cardiovascular diseases and kidney failure after kidney transplantation and is also the independent prediction index of patient's death. OBJECTIVE: To analyze the factors related to anemia after kidney transplantation. RESULTS AND CONCLUSION: In the anemia group (n = 225, 27.2%, aged 26-65 years), the incidence rate of anemia in female and male patients was 23% and 37%, respectively (P < 0.05), 46 patients had hypertension and used angiotensin-converting enzyme inhibitor or angiotensin Ⅱ receptor antagonist and 16 patients had chronic erosive gastritis or upper gastrointestinal tract ulcer, with the human survival rate of 85.3% and kidney failure rate of 25.3%. In the non-anemia group (n = 601, 72.8%, 405 males, 196 females, aged 18-71 years), 35 patients had hypertension and used angiotensin-converting enzyme inhibitor or angiotensin Ⅱ receptor antagonist and 14 patients had chronic erosive gastritis or upper gastrointestinal tract ulcer, with the human survival rate of 92.1% and kidney failure rate of 12.6%. There was significant difference in above-mentioned indices between anemia and non-anemia groups (P < 0.05). These results suggest that gender, age, kidney function, digestive tract disease history, and drug application are closely related to anemia after kidney transplantation.
