1.Effect of Chuankezhi Injection Combined with Chemotherapy on Quality of Life in Senile Patients with Non-small Cell Lung Cancer

Objective To observe the effect of Zusan Li acupoint injection with Chuankezhi injection ( CI) combined with chemotherapy on the quality of life in the stage III or IV senile patients with non-small cell lung cancer (NSCLC) . Methods We adopted research methods for the prospective, randomized, and controlled trial. A total of 62 qualified patients completed the treatment program, and were randomly divided into two groups. Thirty-two patients in the test group received CI combined with chemotherapy and syndrome differentiation treatment, and 30 patients in the control group were given chemotherapy and syndrome differentiation treatment. We used the WHO Quality of Life Older Module Scale ( WHOQOL-OLD) , Functional Assessment of Cancer Therapy-Lung ( FACT-L V4.0) and the Brief Fatigue Inventory ( BFI) to evaluate and analyze the quality of life before and after treatment. Results (1) In the WHOQOL-OLD evaluation, the total scores and the scores of dimensions of sensory abilities, autonomy, social participation, and past, present and future behavior were much improved in the test group after treatment ( P﹤0.01 compared with those before treatment) , while the control group only had statistically significant difference ( P﹤0.01) in the improvement of social participation and the total scores. The test group had higher scores difference in autonomy than the control group (P<0.01) . (2) In the FACT-L V4.0 evaluation, the total scores and the scores of domains of physiological condition, emotional status, functional status, and additional attention were much improved in the test group ( P﹤0.01 compared with those before treatment) , and the control group had an improvement in the total scores and scores of emotional status and additional attention after treatment ( P<0.01) . The test group had higher scores difference in physiological condition, functional status, and additional attention than the control group (P<0.01) . (3) In BFI evaluation, the degree of fatigue and fatigue influence were improved in both group after treatment ( P﹤0.01 compared with those before treatment) , and the improvement in the test group was superior to that in the control group ( P<0.05) . Conclusion Zusan Li acupoint injection with CI can improve the quality of life of senile NSCLC patients treated with chemotherapy in dimensions of sensory abilities, autonomy, social participation, and past, present and future behavior, and in domains of physiological conditions, functional status, additional concern, and it can also improve the degree of fatigue and fatigue influence.
2.The prevalence, awareness, and control of hypertension in Xianghe county of Hebei province

Objective To investigate the prevalence,awareness,control status and associated risk factors of hypertension in a rural population in Xianghe county in North China.Methods A total of 830 adults (aged≥35) from Xianghe county were examined during July to August,2011.Blood pressure was obtained using a standardized sphygmomanometer after a 5-minute sitting rest.Information on gender,age,education level,marital status,smoking,drinking,income,family history of hypertension and use of antihypertensive medications was obtained.Results A total of 42.4% of all subjects had hypertension.Among those with hypertension,the awareness rate was 54.8% and 50.0% of the patients were taking antihypertension medication with the control rate of 11.9% [BP < 140/90 mm Hg (1 mm Hg =0.133 kPa)].Lower age and education level,lower body mass index,negative family history of hypertension were associated with poor awareness of hypertension and worse compliance with the treatment.Older age,positive family history and alcohol consumption were associated with poor blood pressure control.Conclusions Hypertension is highly prevalent in Xianghe rural area.The awareness,treatment and control rate are all low.There is an urgent need for comprehensive strategies to improve prevention,screening,and treatment of hypertension in rural China.
3.Effect observation of batroxobin for treatment on above moderate degree of sudden hearing loss

Objective To evaluate the effect of treatment with batroxobin on above moderate degree of sudden hearing loss (SHL) ,in order to achieve better clinical therapy .Methods The clinical records of 56 patients (57 ears) with above moderate de‐gree of SHL hospitalized from October 2010 to October 2011 were retrospectively studied .57 ears contained 22 ears of flat type and 35 ears of total type ,33 ears were treated with conventional drugs(conventional group) ,while 24 ears were treated with batroxobin and conventional drugs(combination group) .Results The differences of the total effective rate were statistically significant be‐tween conventional group and conventiona group(P=0 .012 8) and between flat type group and total type group(P=0 .012 6) . Conclusion The combination therapy with batroxobin and conventional drugs can significantly increase the total effective rate of the patients with above moderate degree of SHL ,which is superior to conventional drugs .The prognosis of patients with total type is poor than flat type .
4.Application of flipped classroom combined with problem-based learning in the teaching of diagnostic audiology

Objective:To investigate the teaching effect of flipped classroom combined with problem-based learning (PBL) in the teaching of the Diagnostic Audiology course.Methods:A total of 72 undergraduate students majoring in audiology and speech rehabilitation in the classes of 2019 and 2020 in Chongqing Medical University were enrolled as subjects, and three chapters of the Diagnostic Audiology course were selected for teaching reform. The 34 students in the class of 2019 were enrolled as control group and received lecture-based learning (LBL), and the 38 students in the class of 2020 were enrolled as experimental group and received flipped classroom combined with PBL. The teaching effect was evaluated by comparing the two groups in terms of classroom test scores, classroom participation scores, degree of satisfaction with teaching effect, and overall satisfaction with teaching. SPSS 23.0 software was used to perform the t-test, the chi-square test, and the Mann-Whitney U test. Results:The experimental group had a significantly higher classroom test score than the control group (25.95±1.21 vs. 23.21±1.55, P<0.001). In terms of classroom participation scores, the experimental group had significantly higher scores of participation in class discussion, cooperative and collaborative abilities, and questioning skills than the control group (all P<0.001). In terms of the degree of satisfaction with teaching, compared with the control group, the experimental group had significantly higher degrees of satisfaction in the dimensions such as interest in learning, mastery of theoretical knowledge, teamwork and collaboration abilities, self-learning ability, and self-presentation ( P=0.005, 0.009, 0.001, 0.016, and 0.005). In addition, the experimental group had a significantly higher degree of overall satisfaction with teaching than the control group ( P=0.006). Conclusion:Flipped classroom combined with PBL has a good teaching effect in the Diagnostic Audiology course and thus holds promise for further application.

Objective:To summarize the clinical manifestations and gene mutation features of patients with nucleotide excision repair (NER) disorders.Methods:A retrospective analysis was made on clinical data of patients with NER disorders who were admitted to the Chinese People′s Liberation Army General Hospital from October 2008 to February 2022 and diagnosed in the Outpatient Department of Beijing Children′s Hospital, Capital Medical University from October 2015 to February 2022.Literature on previously reported Chinese patients with NER disorders was reviewed.Results:(1)A total of 16 patients with NER disorders were enrolled, including 6 males and 10 females.The onset age was 7.5 (4.0, 12.0) months and the age at diagnosis was 42.0 (21.5, 77.0) months.There were 3 types of NER disorders: Cockayne syndrome (CS) in 13 cases, Xeroderma Pigmentosum (XP) in 2 cases and Cerebro-Oculo-Facio-Skeletal syndrome (COFS) in 1 case.Four disease-causing genes were detected: CSA gene in 11 cases, CSB gene in 3 cases, XPG gene in 1 case, and XPD gene in 1 case.The first symptoms of the 16 patients were photosensitivity and developmental delay, and neurological symptoms were observed in all the 3 NER disorder types.XP and CS patients had skin symptoms.CS patients presented typical facial features, visual and auditory impairment, microcephaly and changes in neuroimaging features.COFS patients showed intrauterine growth retardation.(2)Results of literature review: a total of 96 Chinese patients reported were retrieved, involving 6 disease types, including CS in 45 cases, XP in 44 cases, trichothiodystrophy in 4 cases, COFS in 1 case, XP-CS in 1 case, and ultraviolet sensitive syndrome in 1 case.Nine mutated genes were identified: CSA in 33 cases, XPA in 15 cases, CSB in 13 cases, XPV in 10 cases, XPC in 9 cases, XPG in 7 cases, XPD in 7 cases, XPF in 1 case, and MPLKIP in 1 case.The common symptoms were growth failure (62 cases), skin photosensitivity (61 cases), typical facial features (52 cases), mental retardation (49 cases) and microcephaly (48 cases). Among 36 cases had imaging data 33 cases(91.7%)had calcification of basal nucleus or globus pallidus.Three cases had intrauterine growth retardation and microcephaly during pregnancy. Conclusions:Patients with such prenatal manifestations as intrauterine growth retardation and microcephaly or with typical symptoms like skin photosensitivity, typical facial features, growth failure, mental retardation, hypertonia, and calcifications of basal ganglia should be suspected of NER disorders.Early genetic testing is recommended to confirm the diagnosis.
7.Effect of Yifei Sanjie Pills (益肺散结丸) Combined with Cisplatin on Plasma Exosomal miRNAs in Lewis Lung Cancer Tumor-derived Mice

ObjectiveTo investigate the tumor inhibition effect and mechanism of Yifei Sanjie Pills (益肺散结丸, YSP) on lung cancer. MethodsLewis lung cancer tumor-derived mice were established and divided into four groups including model control group, cisplatin group, cisplatin + YSP low-dose group and cisplatin + YSP high-dose group, with 12 mice in each group. The corresponding interventions were given for 14 days. The tumor volume was measured on the 0th, 3rd, 7th, 10th and 14th days of administration to evaluate the tumor growth. The plasma and tumor tissue were collected on the 15th day. Plasma from the model group, the cisplatin group and the cisplatin+YSP high-dose group were selected, and plasma exosomes were extracted; the differences in miRNA expression among the groups were detected and analyzed by second-generation sequencing technology, and the potential mechanism of action of YSP was investigated by principal component analysis, biofunctional enrichment analysis and miRNA-target gene regulatory network analysis. Quantitative real-time PCR was used to detect the expression of miRNA-615-3p in tumor tissues, and the relationship between miRNA-615-3p and overall survival of lung cancer were analyzed using the Kaplan-Meier plotter (kmplot.com) database. ResultsCompared to that of the model control group, the tumor volume of the cisplatin group on day 10, and the cisplatin + YSP low- and high-dose groups on day 7, 10, and 14 were significantly reduced (P<0.05 or P<0.01). Compared to that of the cisplatin group, the tumor volume of the cisplatin + YSP low- and high-dose groups on day 10 and 14 was significantly reduced (P<0.05). The principal component analysis of miRNA expression profiles showed significant differences in miRNA expression between different intervention groups. There were 21 differentially expressed miRNAs between the model control group and the cisplatin group, 50 differentially expressed miRNAs between the model control group and the cisplatin+ YSP high-dose group, and 6 differentially expressed miRNAs between the cisplatin group and the cisplatin+ YSP high-dose group. Biological function enrichment analysis showed that the differentially expressed miRNAs were mainly involved in the regulation of signaling pathways related to cell growth, proliferation, differentiation, autophagy and other biological activities. The miRNA-target gene regulatory network showed the top 20 genes that were targeted, among which there were proven miRNAs and genes related to lung cancer, and miRNAs that needed further investigation. The expression of miRNA-615-3p in tumor tissues decreased significantly in the cisplatin group and cisplatin+YSP high-dose group compared to that of the model group(P<0.05 or P<0.01). The miRNA-615-3p was negatively correlated with the survival prognosis of lung cancer(P<0.05). ConclusionCisplatin combined with YSP can effectively inhibit the proliferation of Lewis lung cancer tumors, and the tumor-suppressive effect is related to the regulation of multiple miRNAs, especially the downregulation of miRNA-615-3p expression.
8. Clinical application of contrast-enhanced ultrasound combined with modified labeled method in locating sentinel lymph nodes of breast cancer

Objective:
To explore the clinical value of contrast-enhanced ultrasound combined with modified labeled method in labeling sentinel lymph nodes of breast cancer in comparison with nano-carbon stained method.
Methods:
Eighty female breast cancer patients who underwent surgery in the First Affiliated Hospital of Soochow University between July 2017 and April 2019 were enrolled. Sentinel lymph nodes in all patients were labeled by contrast-enhanced ultrasound combined with modified labeled method and nano-carbon stained method, respectively. The consistency of first lymph node labeled by the two methods was judged, the number of SLNs labeled by two methods was counted, and the pathology of the labeled SLN was compared with that after axillary dissection.

9.Retrospective multicenter nested case-control safety study of Ilaprazole sodium for injection

OBJECTIVE To understand the safety of Ilaprazole sodium for injection in clinical practice. METHODS From Jan. 1st 2019 to Feb. 29th 2020, the data of 3 926 valid hospitalized patients receiving Ilaprazole sodium for injection were collected prospectively from 5 third-level hospitals through CHPS, and the post-marketing safety analysis was performed by using retrospective multicenter single cohort study. At the same time, a nested case-control study (the ratio of trial group and control group was 1∶4) was used to confirm the baseline stability of this study cohort and the correlation between adverse reactions and Ilaprazole sodium for injection. RESULTS Among 3 926 patients, 3 patients experienced 5 adverse drug events after using Ilaprazole sodium for injection, with the incidence of 0.076%. There was no serious adverse event, and the occurrence time was 2 days after medication; adverse drug events mainly include elevated liver function indicators (alanine transaminase, aspartate transaminase, total bilirubin), which were mild and untreated, and all adverse drug events were improved. The results of the nested case-control study showed that the trial group and the control group belonged to the same background baseline, and the occurrence of adverse drug events was more closely related to Ilaprazole sodium for injection. CONCLUSIONS The overall safety of Ilaprazole sodium for injection is relatively high, and the occurrence of adverse events is more related to it.
10.Supervillin regulates convergence and extension movements via Wnt/β⁃catenin pathway in zebrafish embryos

Objective :
To explore the role and molecular mechanism of Supervillin in zebrafish embryo development.
Methods :
Clustal Omega and DNAman software were used to analyze and compare the homology of amino acid sequences of Svil and SVIL in zebrafish and human. The expression pattern of Svila (subtype of Svil protein in zebrafish) during early embryonic development was analyzed by RT⁃PCR and in situ hybridization. Svila expression in zebrafish was inhibited by injecting morphinos ( MO) , and morphological changes of embryos were observed. The expression and nuclear localization of β ⁃catenin protein were detected by Western blot , and the expression of Wnt/β⁃catenin target gene was detected by RT⁃PCR.
Results :
During the early embryonic development of zebrafish , Svila was maternally expressed and showed an upward trend with the development process. The expression of Svila was reduced by MO , and the development of zebrafish embryos was distorted and the body axis was bent , which might be related to the blocked movement of concentrated extension of embryos. Further studies showed that Svila expression affected β⁃catenin nuclear transport and Wnt/β⁃catenin signaling pathway activation.
Conclusion
Svila regulates the concentrated extension movement of zebrafish embryos by activating the Wnt/β⁃catenin signaling pathway.
