1.Adipose-derived mesenchymal stem cell transplantation protects the myocardial tissues from acute myocardial infarction

BACKGROUND:Adipose-derived mesenchymal stem cels have rich sources that are easily obtained, which can be used to treat acute myocardial infarction. OBJECTIVE: To investigate the therapeutic effect of adipose-derived mesenchymal stem cel transplantation on acute myocardial infarction in rats. METHODS:Rat models of acute myocardial infarction were made and subjected to adipose-derived mesenchymal stem cel transplantation in comparison with model and control (sham operation) groups. RESULTS AND CONCLUSION:Echocardiography findings showed significant improvement in the left ventricular end-systolic diameter, left ventricular end-diastolic diameter and ejection fraction in the cel transplantation group compared with the model group (P < 0.05). Hematoxylin-eosin staining showed that myocardial infarction was evident in the model group, in which, there were rarely viable myocardial tissues and few vessels in the infarcted region, but in the cel transplantation group, there were evident survived myocardial tissues and transplanted cels. The percentage of infarct size was significantly lower in the cel transplantation group than the model group (P < 0.05). Immunohistochemical staining showed that adipose-derived mesenchymal stem cels were able to survive in the infarcted myocardial tissues, and the expression of cardiac troponin T in the cel transplantation group was significantly higher than that in the model group (P < 0.05). Experimental data show that adipose-derived mesenchymal stem cel transplantation can protect the myocardial tissues after myocardial infarction, and effectively improve the myocardial function.
2.The management and risk factors of pericardial effusion after cardiac surgery

Objective We aimed to recent experience at our hospital in the diagnosis and treatment of pericardial effusion after cardiac surgery and to identify risk factors for its development.Methods We searched our hospital for patient who had cardiac surgery with cardiopulmonary bypass from January 2002 through December 2010.For patient with pericardial effusion,medical records were reviewed to evaluate its manifestations and management.To identify risk factors for effusion,patients with effusions were compared with patients without effusions.All patients had routine postoperative echocardiographic examination.Results Of 22462 patients identified,509 (2.3%) had pericardial effusion.Compared with patients without pericardial effusion,ages,sex,cardiac function and so on were no significant( P > 0.05 ).Body,valve cardiac diseases,and cardiopulmonary bypass time and so on were significant ( P < 0.05 ).Among 509 patients with pericardial effusion,262 patients (51.4%) of whom had specific symptoms.Clinical features of tamponade were documented in 51 patients( 10.1% ).Pericardial effusions were evacuated by echocardilgraphy-guided pericardiocentesis ( n =27,10.3% ) or surgical drainage ( n =15,5.7% ).Pericardial effusion resolved after left thoracocentesis for pleural effusion in 5 patients ( 1.9% ) ; The Other patients with pericardial effusion were treated conservatively.Indeperdent risk factors for pericardial effusion were large body,valve cardiac operations,and prolonged cardiopulmonsry bypass.Conclusion In our study,pericardial effusion occurred in 2.3% of patients,and symptoms were nonspecific.Several factors,mainly related to preoperative characteristics and type of operation,predispose patients to effusion,echocardiography-guided pericardiocentesis is effective and safe in these patients with pericardial effusion.
3.Photoplethysmography Pulse Wave Signal Detection and Pulse Rate Characteristics Based on an in vitro Experiment

Objective To investigate the influence of changes in blood flow parameters on pulse rate characteristics by taking the advantage of controllable parameters in an experimental cardiopulmonary bypass system.Methods A set of human circulatory system equipped with an in vitro wrist model was established.By changing parameters such as the heart rate,wrist flow,pressure,and system compliance,a photoplethysmography pulse wave signal was obtained from the wrist model,and the time-and frequency-domain indices of pulse rate variability were extracted.Results Changes in heart rate,pressure,and system compliance caused a change in pulse shape,but the time domain indices NN50 and PNN50,which indicate pulse rate variability,were zero,and the other indices and frequency domain indices were in the very low value category.Conclusions In the absence of heart rate variability,hemodynamic changes in heart rate,wrist flow,blood pressure,and system compliance did not produce significant pulse rate variability.This study can provide a reference for studies on pulse rate variability and heart rate using more convenient wrist acquisition equipment.

5.Study on the 3,4-Dihydroxyphenylalanine Redox State Characterization Method of Mussel Adhesive Protein.

OBJECTIVETo investigate the feasibility of using liquid chromatography (HPLC) to characterize the 3, 4-Dihydroxyphenylalanine (DOPA) redox state of mussel adhesive protein (MAP).
METHODSThe DOPA and protein contents of MAP were determined by HPLC, Arnow and Bradford methods respectively.
RESULTSWith extended oxidation time, the protein contents of MAP samples remained unchanged whereas the DOPA contents declined. The retention times of main peaks in HPLC for both the accelerated oxidation and retained samples shifted as the storage time extended, which could be related to the changes of sample redox state.
CONCLUSIONSThe redox state of MAP can be characterized by the change of HPLC peak retention time. HPLC can be used in the research on the MAP redox state, which is beneficial to the product development and quality control.
6.Effects of Coating Method on Fabrication Quality of Flexible Silicone Blood Vessels

Objective To investigate the effect of different coating methods on production quality of complex and flexible silicone vascular replicas. Methods Based on models of anterior communicating artery aneurysms, several patient-specific models were made by using spray-coating method and brush-spin-coating method respectively, and two methods for making the same vascular structure were quantitatively compared in terms of thickness growth, circumferential uniformity and light transmittance. Results Brush-spin-coating method was better than spray-coating method in the thickness control and coating uniformity for fabrication of vessels with large curvature, variable diameter and straight tube, and the model had preferably light transmittance and surface smoothness. The relative deviation of thickness by brush-spin-coating method was decreased by 8. 9% , 10. 8% and 16. 9% respectively compared with spray-coating method. Conclusions At present stage, the brush-spin coating method has the advantage of thickness uniformity and light transmittance over the spray-coating method in making silicone phantoms, and it has promising application prospects in fluid mechanics field of in vitro experiment on large vessels.
