1.Preparation of cardiomyopathy model in rats with type 2 diabetes

BACKGROUND: Diabetic cardiomyopathy is closely related to the high incident rate of diabetic heart failure. It is extremely significant to set up the successful dietetic cardiomyopathy model for the study on improving cardiac function.OBJECTIVE: To understand the influence of high blood sugar on myocardium by setting up the model of type 2 diabetes and diabetic cardiomyopathy.DESIGN: Completely randomized design, randomized and controlled study,in which, the experimental animals were taken as the objects.SETTING: Department of endocrinology of a university.METHODS: In the experiment, 99 newly born Wistar male rats were employed, randomized into experiment group (75 rats) and the control group(24 rats). In the control group, abdominal injection was done with sodium citrate buffer and the single abdominal injection was done with strptozocin(STZ)90 mg/kg in the experiment group. Ten weeks later, the glucose tolerance was assayed to determine the set-up of diabetes model. The ultrastructure of myocardium of rats was tested under electronic microscope after being sacrificed.MAIN OUTCOME MEASURES: To observer whether the myofibrillae and mitochondria of myocardiocytes alter or not in diabetic rats.RESULTS: In the experiment group, the fasting blood sugar[(8.87± 0.76)mmol/L] was slightly higher than that[ (5.88 ± 0. 45)mmol/L] in the control group, the blood sugar 2 hours after meal was remarkably higher than that in the control group and the glucose tolerance was abnormal. Under the electronic microscope, the myofibrillae presented focal degeneration and disarranged order of myofilament; mitochondria of myocardiocytes presented swelling and focal proliferation.CONCLUSION: This model is the satisfactory carrier for the study on diabetes Ⅱ and its myocardiopathy, which lays an experimental foundation for the study on cardiac function of type 2 diabetes.
2.Expression of neurotrophins receptor p75NTR in human breast cancer resistant cell line and its correlation with multidrug resistance

Objective To investigate the expression of neurotrophins receptor 75NTR in human breast cancer resistant cell line and its correlation with multidrug resistance. Methods Western blot was used to detect the expression level of p75NTR protein in various breast cancer cell lines and multidrug resistant cell lines. The over-expression vector and siRNA vector of p75NTR were constructed by gene recombination method. Western blot was used to detect the expression levels of p75NTR protein in transfected p75NTR and p75NTR - siRNA breast cancer resistant cell line MDA-MB-231/ADR;the sensitivity of MDA-MB-231 and MDA-MB-231/ADR to different chemotherapeutic anticancer drug (PTX ,ADM ,GEM ,DDP ,OXA) and the multi-drug resistance effects in over-expression and knock-down p75NTR MDA-MB-231/ADR were detected by CCK-8 assay. Results Western blot result showed that the expressions of p75NTR protein in the multidrug-resistant breast cancer cell lines MDA-MB-231/ADR and MCF-7/5-FU were higher than that of other breast cancer cell lines. Over-expression of p75NTRcan up-regulate the expression of p75NTR protein in MDA-MB-231/ADR;the CCK-8 assay indicated that over-expression of p75NTR can effectively enhance the resistance of MDA-MB-231/ADR cells to ADM,GEM and OXA. Conclusion The expression of p75NTR in breast cancer resistant cell line is higher than that of its parental cell line;over-expression of p75NTR can reduce the sensitivity to chemotherapeutic drugs and promote its multidrug resistance.
3.Endoscopic ultrasonography in preoperative assessment of esophageal cancer

Objecfive To explore the value of endoscopic uhrasonography(EUS)in diagnosis and preoperative assessment of esophageal cancer.Methods A total of 57 patients with esophageal cancer were examined by EUS.The image features,depth of invasion and lymph node metastasis obtained by EUS were analyzed and compared with postoperative outcomes.Results Esophageal cancer was characterized as an irregular hypoeehoic mass with partial or total destruction of normal esophageal wall structure under EUS.Compared with post-operative pathological findings,the consistent rates of EUS diagnosis in depth of invasion,T staging,and N staging were 84.2%(48/57),87.7%(50/57)and 84.2%(48/57),respectively.Conclusion EUS exhibited high diagnostic accuracy in T staging of esophageal cancer,which is valuable in managements and prognosis of esophageal cancer.
4.Determination of Selenium Species in Burdock and Panax Notoginseng Using Ultrasonic Assistant Extraction Combined with High Performance Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry

A high performance liquid chromatography-inductively coupled plasma mass spectrometric ( HPLC-ICP-MS) method for analysis of selenocystine (SeCys2), selenite (SeⅣ), selenomethionine (SeMet) and selenate ( SeⅥ) was described. Chromatographic separation was performed using 8 mmol/L citric acid solution (pH 5). The potentially interfering 40Ar40Ar+ at selenium masses of m/z 80 was reduced using CH4 as a reactive cell gas in the dynamic reaction cell. The selenium species was extracted using ultrasonic extraction system with a mixture of protease K and lipase. The limits of detection for SeCys2 , SeⅣ, SeMet, and SeⅥwere observed as 0. 088, 0. 033, 0. 30, and 0. 17 ng/mL, respectively. The correlation coefficients were greater than 0 . 9995 . The recoveries were observed in the range of 95 . 1%-114 . 6%. The developed method was successfully applied for the determination of selenium species in Burdock and Panax notoginseng.
5.Effects of apM1-contained recombinant adenovirus on malonaldehyde and superoxide dismutase levels in HUVEC cells

Objective To evaluate the effects of exogenous adenovirus-mediated gene transfer of human apM1 gene (Ad-apM1) on malonaldehyde (MAD) and superoxide dismutase (SOD) levels in human umbilical vein endothelial cells (HUVEC). Methods HUVEC proliferation was measured by MTT after infection by AdapM1. Adiponetin protein level in cell culture medium of HUVEC cells infected with Ad-apM1 was detected by double antibody sandwich ELISA. The levels of MAD and SOD were measured by chromatometry. Results MTT assay showed that Ad-apMl had no effect on HUVEC proliferation. Human adiponectin protein levels in cell culture medium significantly increased after HUVEC was infected with Ad-apM1 for 24, 48, and 72 h, along with decreased MAD and increased SOD ( all P<0.05 ). MAD levels markedly increased and SOD levels decreased after HUVEC were incubated with 100 μmol/L H2O2 for 6, 12, and 24 h, and these reactions were reversed by AdapM1 transfection (all P<0.05 ). Conclusions HUVEC infected with Ad-apM1 effectively secrete human adiponectin. Ad-apM1 exerts antioxidation effect and antagonizes H2O2 -induced endothelial injury.
6.Expression and clinical significance of Topo Ⅱα , COX-2 in breast cancer tissues

Objective To explore the expression of Topo Ⅱα and COX-2 in breast cancer tissues and investigate their correlations to clinicopathologic feature of breast cancer. MethodsThe expression of Topo Ⅱα and COX-2 in 50 specimens of breast cancer and their normal tissues were detected by immunohistochemistry. Their correlation to clinicopathologic features of breast cancer were analyzed.ResultsThe positive rates of Topo Ⅱα were 64 % (32/50) and 22 % (11/50) and COX-2 were 68 % (34/50) , 14 %(7/50) in breast cancer and normal tissue (P<0.05). The expression of Topo Ⅱα was correlated to degree of differentiation (P <0.05), not correlated to patient age, tumor size, clinical stage and lymph node metastasis(P >0.05). The expression of COX-2 was correlated to tumor size, degree of differentiation, clinical stage and lymph node metastasis (P <0.05), not correlated to patient age (P >0.05). Conclusion Topo Ⅱα and COX-2 expression can be used as an indicator for predicting the differentiation, infiltration and metastasis characteristics of breast cancer.
7.Experimental Study on CompoundWeifu in Gout Treatment

This article was aimed to study the action of antigout effects of compoundWeifu. Through the acute gouty arthritis models of rabbits and rats induced by micro-crystal sodium urate (MSU). The histopathological changes of synovial tissues among rabbits were observed. And the contents of TNF-α, IL-1β, IL-6 in rabbit serum were determined. The swelling degree of the rats’ feet and the content of prostaglandin E2 (PGE2) from the inflammatory exudate foot induced by MSU were also determined. The antigout effects of compoundWeifu were observed. The results showed that the grade of the vascular congestion, swelling and inflammatory cell infiltration in synovial tissues in the high-dose and medium-dose CompoundWeifu group had significantly improved (P <0.05). And the levels of TNF-α, IL-1β, IL-6 in serum were significantly decreased (P< 0.05, orP < 0.01). It can obviously reduce the swelling degree induced by MSU, and the level of PGE2 in the inflammatory exudate foot (P< 0.05, orP < 0.01). It was concluded that CompoundWeifu had obvious antigout effects in the study.
8.Endoscopic diagnosis and treatment of gastritis cystica profunda with early gastric cancer

Objective To study the relationship between gastritis cystica profunda (GCP)and early gastric cancer (EGC),and to explore the diagnosis and treatment of GCP.Methods Clinical data of 17 patients,who were pathologically diagnosed as having GCP,including clinical symptoms,white light endo-scopic findings,magnifying endoscopic features,EUS outcomes and histopathological characteristics were an-alyzed.Results All 17 patients of GCP had no specific clinical symptoms or history of gastric surgery prior to the operation.Eight lesions were located at cardiac,4 at gastric body,4 at gastric antrum and 1 at gastric angle.As to the morphology,3 cases were type Ⅰ,3 type Ⅰ +Ⅱc,9 type Ⅱa +Ⅱc,1 type Ⅱc and 1 type Ⅱb +Ⅱc.The mean lesion size was 1.8 cm.Of 16 patients with preoperative EUS examination,11 patients were found to have cystic changes,with the diagnostic rate being 68.8%(11 /16).All 17 patients underwent ESD,14 (82.4%,14 /17)of which were found to have EGC.Postoperative pathology confirmed low grade intraepithelial neoplasia in 3 cases,high grade intraepithelial neoplasia in 7,intramucosal carcino-ma in 4,submucosal carcinoma in 3 patients.Conclusion EUS plays an important role in the diagnosis and treatment of GCP.And GCP is highly correlated with EGC,which should be managed in time.
9.Preoperative evaluation of risk factors of complete resection in early gastric cancer with endoscopic submucosal dissection: a prospective study

Objective To prospectively evaluate the risk factors of complete resection in early gastric cancer (EGC) with endoscopic submucosal dissection (ESD),and to guide the choice of treatment methods.Methods This study prospectively evaluated the endoscopic features of 66 EGCs,including the lesion size,presence or absence of ulceration,the extent of differentiation,invasion depth and entire margins of the EGC,then compared them with postoperative pathologic results and analysed these factors.Results The lesion size of the high grade intmepithelial neoplasia (H) group and the intramucosal carcinoma (M) group were mainly less than 30 mm (90.9% vs.88.5%),but 57.1% of the submucosal carcinoma (SM) were more than 30 mm.There was a significant difference between any two of three groups (P < 0.05).Fourteen EGCs who got ulceration without invasion beyond mucosal muscularis underwent ESD successfully,and the basal or dissected margin had no residual tumor cells confirmed pathologically.And no tumor cell infiltration or lymph node metastasis was discovered.Of 45 EGCs with ESD,the underestimation rate for horizontal extent determined by white light and chromoendoscopy was higher than that of magnifying endoscopy with narrow-band imaging (ME-NBI) (15.6% vs.2.2%,P <0.05).Diagnostic accuracy for the extent of differenciation by conventional endoscopy was 93.9% (31/32,P > 0.1),but it's unable to determine the extent of differentiation by ME-NBI.The accuracy of the group H was 84.8% (28/33),that of M was 57.7% (15/26),that of SM was 71.4% (5/7),and there was a significant difference between group H and group M (P < 0.05).Conclusion To achieve complete resection of EGC with ESD,the lesion more than 30 mm,presence of ulceration,undifferentiated type,deep infiltration should be considered as the risk factors,and it's also important to identify the horizontal extent of EGC to avoid unnecessary operation.
