1.Hyperthermia: its effectiveness in killing tumor cells and influence on Na~+ -K~+ -ATPase activities in erythrocytes

Objective To investigate the effectiveness of hyperthermia of different degrees in killing tumor cells and its influence on Na+ -K+ -ATPase activities in erythrocytes. Methods Cultured low differentiated stomach gland tumor cells (SGC-170) and large intestine gland tumor cells (LOVO) (1?106?ml-1 each) were mixed with concentrated RBCs respectively and incubated in warm bath of 37℃ , 42℃, 43℃ , 45℃ or 47℃ for 40 min respectively. After hyperthermic treatment tumor cells were isolated from RBCs using density gradient centrifugation and the live tumor cells were counted by typan staining. The isolated tumor cells were then cultured and the clone formation of tumor cells was checked. The cultured tumor cells were marked with 5-bromo deoxyuridine and DNA metabolism was examined. The Na+ -K+ -ATPase activities in RBC after hyperthermic treatment were also determined. Results The amount of tumor cells was significantly decreased by 40 min hyperthermic treatment in a temperature-dependent manner from 42-471 as compared with the control group (37℃) (P

3.Application of temperament combined with social competence in preschoolers with asthma in home care

Objective To explore the application effect of home care on the basis of temperament combined with social competence in preschool children with bronchial asthma.Methods A total of 100 preschool children outpatients with bronchial asthma who came to our hospital from January 1 to February 28,2013 were selected with the method of convenience sampling and were randomly divided into observed group and control group with 50 cases in each group.Cases of the observed group were divided into ordinary,middle,and strengthen subgroups according to temperament and social competence.The control group and the ordinary subgroup received routine care interventions,and the other subgroups were given corresponding enhanced care measures.The attack rate of asthma,medical care,quality of life and lung function during one year follow-up were observed and compared between the two groups.Results Within the first six months of follow-up,patients in observed group got better control in acute attacks than the control group [(2.3±0.4) times vs.(3.4±0.3) times,x2=5.710,P<0.05)].The percentage of forced expiratory volume in the first second and Pediatric Asthma Quality of Life Questionnaire scores in observed group were significantly higher than the control group one years later [(86.9±4.6)% vs.(85.5±3.8)% and (90.1±7.5) vs.(76.5±8.0),P<0.05].Conclusions The application of temperament type combined with social competence showed a good practical value in home care for asthma preschoolers.
4.CHILD'S BEHAVIOR PROBLEMS COMPUTER SCREENING SYSTEM AND IT'S APPLICATION

To programming one Child's Behavior Problems Computer Screening System (CBPCSS), a series of software for analyzing child behavior individual and group sample. According to the world-famous American ACHENBACH child's behavior checklist, to ensure the screening quality and compatibility of cultures, we revised and standardized the norms of different ages in primary schools and nursery schools in various cities based on the principle of cluster stratified sampling. Then we designed CBPCSS carefully. The system can reliably and rapidly screen an individual child behavior and output the behavior factor curve (appearing in front of the profile). With CBPCSS we can observe the child behavior clearly. It takes twenty times shorter than that of manual screening. On the other hand, CBPCSS has a function of group analysis. The clinical practice proved that CBPCSS could substitute for manual screening completely. It is a powerful tool for social, scientific and pediatric medical workers.
5.Effect of Ganoderma lucidum on drug resistance of cisplantin-resistant ovarian cancer cell line

Objective To evaluate the effect of Ganoderma lucidum on drug resistance of the cisplantin-resistant ovarian cancer cell line(A2780CP).Methods The A2780CP cells were randomly divided into control group and test group.The cells in control group and test group were incubated in culture media alone and in culture media containing Ganoderma lucidum 0.5 mg/ml for 48 h respectively.The resistance index(RI) of A2780CP was determined by WST-1 assay.The expression of Akt,Bcl-2 and p53 protein was measured by Western blot.Results The RI was significantly deeressed,the expression of Akt and Bcl-2 protein was down-regulated,while p53 protein expression was up-regulated in test group as compared with control group(P<0.05).Conclusion Ganoderma lueidum can reduce the resistance of A2780 CP cells to cisplantin by down-regulating the expression of Akt and Bcl-2 and up-regulating p53 expression in A2780CP cells.
6.Analysis on current situation and countermeasure of hospital outpatient injection room infection

Objective To explore current situation of hospital outpatient injection room infection and to discusse relevant countermeasures of infection control .Methods A retrospective analysis was conducted in 245 patients admitted and got infected in our hospital outpatient department injection room from October 2012‐to October 2013 .By analyzing its infection type ,treatment time ,age ,etc .,and in winter and items ,air ,medical staff hand in spring and summer and fall of hospital respectively we understand the hospital infection and the seasonal relationship ,the related factors of hospital infection ,and develop targeted and effective inter‐vention measures .Results The outpatient injection room infection were mainly occurred in respiratory system and digestive sys‐tem ,the occurrence rates were 54 .29% (133/245) ,25 .31% (62/245) ,respectively .The infection rate of treatment time> 7 d (2 .74% ) apparently higher than < 7 d (1 .09% ) (χ2 = 56 .246 ,P= 0 .000) .The infection rate of people whose age over 40 (1 .91% )apparently higher than age < 40 (1 .13% ) (χ2 = 12 .316 ,P= 0 .000) .The qualified rate of atmosphere in Winter and Spring (68 .46% ) was significantly lower than summer and autumn (89 .23% ) (P<0 .05) ,the qualified rates of surfaces and hands of medical staffs (84 .62% ,82 .31% ) were significantly higher than summer and autumn(63 .08% ,66 .15% ) .Conclusion The out‐patient injection room infections were related to a variety of factors ,we could carry on the comprehensive analysis and formulate specific interventions to reduce hospital infection .
7.Comparing the characteristics of tissue culturein vitro and different cell types of cancer microenvironment in tissues at different distances from colorectal cancer lesions

Background and purpose:Cancer microenvironment has become a hot topic of cancer research. It is important in the initiation of colorectal cancer. This study aimed to discuss the correlation between the characteristics of tissue culturein vitro and different cell types in cancer microenvironment.Methods:Samples were collected at different distances from the colorectal cancer lesions, which were named as positions 1, 2 and 3 from distal to proximal. Tissues were cut into 1-2 mm3 forin vitro culturing. HE staining was used to observe the structure of crypts. Immunohistochemistry was used to detect the expressions of Cyclin D1 (CD1), CD133, cytokeratin18 (CK18), vimentin and α-smooth muscle actin (α-SMA).Results:Position 3 grew faster than position 2 and position 1. As getting closer to the colorectal cancer lesions, expressions of CD1, CD133, vimentin and α-SMA were increased while expression of CK18 was decreased.Conclusion:The tissue structure and the expression of different cell types in cancer microenvironment changed more seriously as get-ting closer to the colorectal cancer lesions. This indicated that the change of cancer microenvironment may contribute to the initiation of colorectal cancer.
8.Virtual touch tissue imaging quantification in differential diagnosis of benign and malignant TI-RADS 4 thyroid nodules

Objective To observe the value of virtual tough tissue imaging quantification (VTIQ) in differential diagnosis of thyroid imaging reporting and data system(TI-RADS) 4 thyroid nodules.Methods A total of 185 patients with 192 TI-RADS 4 nodules were included in this study.The nodules were divided into three groups according to the maximum size as follows:Group Ⅰ,the maximum size≤0.6 cm;Group Ⅱ,0.6 cm< the maximum size≤ 1.0 cm;Group Ⅲ,the maximum size > 1.0 cm.Shear wave velocities (SWV) of nodules were measured by means of VTIQ technique.With pathological diagnosis as the gold standard,SWV value of benign and malignant nodules were analyzed and ROC curve was drawn to assess the diagnostic efficiency.Results By the ROC curve test,at SWV cut-off values of 2.44 m/s for group Ⅰ and group Ⅱ,2.49 m/s for group Ⅲ,the sensitivity were 79.0 %,76.0 %,88.6%,specificity were 88.6%,89.5 %,93.7 %,accuracy were 83.5 %,81.8 %,90.1%,Youden index were 0.68,0.66,0.82,respectively.Conclusions VTIQ can reflect the hardness of TI-RADS 4 nodules,the value of the differential diagnosis of such nodules is high,convenient,noninvasive and not limited by the size of nodules.
9.Risk assessments and countermeasures on ultrasound-guided radiofrequency ablation for benign thyroid nodules

Objective To investigate the value of preoperative risk assessments and countermeasures on ultrasound-guided radiofrequency ablation for benign thyroid nodules.Methods The preoperative risk assessments of 311 benign solid thyroid nodules in 274 patients were divided into level 0-Ⅳ,among which 38 nodules on level 0,69 nodules on level Ⅰ,36 nodules on level Ⅱ,59 nodules on level Ⅲ,109 nodules on level Ⅳ.Treatments were performed by moving shot technique or multiplanar shot technique,using Leverage Displacement or Hydrodissection to prevent the adjacent vital structures from thermal injuries.Results 311 thyroid nodules were completely ablated.In ablating the 59 nodules on risk level Ⅲ,thermal injuries were effectively avoided in 30 cases by using Leverage Displacement,while recurrent laryngeal nerve injuries happened in 2 cases using Hydrodissection for 29 nodules.The difference between these two methods on risk level Ⅲ had no statistical significance(P =0.237).To avoid thermal injuries during ablation for 109 nodules on risk level Ⅳ,using Leverage Displacement for 102 nodules,recurrent laryngeal nerve injuries happened in 1 case,while using Hydrodissection for 7 nodules,recurrent laryngeal nerve injuries happened in 2 cases.The difference between the two methods on risk level Ⅳ had statistical significance(P =0.010).No thermal injury happened during ablation for 143 nodules on risk level 0-Ⅱ.Conclusions Preoperative risk assessment on thyroid nodules is helpful in preventing potentially avoidable complications.Proper use of Leverage Displacement can protect adjacent vital structures from thermal injuries during ablation for nodules,which is easy and simple to handle and has a certain practical application value.
10.Approach to CDISC SDTM Implementation for Clinical Trials Data Submission

This study was aimed to discuss the ways of converting clinical data entered in the Case Report Form (CRF) to Study Data Tabulation Model (SDTM) database format in clinical research of traditional Chinese medicine (TCM). Currently, there were three approaches for implementing SDTM, which were the pure SDTM approach, submission-only approach and database-only approach. This article compared and analyzed advantages and disadvantages of three approaches and introduced experiences of the Clinical Evaluation Center of China Academy of Chinese Medical Sciences using database-only approach for SDTM implementation. The results showed that pure SDTM approach can maximally embrace SDTM standards. However, the current process and software system should be modified. Therefore, it was time-consuming and expensive. The submission-only approach was the most economic way in the application of SDTM standards. However, the data quality and traceability may not be guaranteed. The database-only approach built the study database based on the SDTM standard by writing transformed program before data entry while the data collection system was not SDTM-compatible. It was concluded that database-only approach for implementation SDTM was a suitable and practical way to TCM clinical research.
