1.Optimization of Prokaryotic Expression Conditions of Human β2-microglobulin in E. Coli and Its Purification.

To obtain recombinant human β2-microglobulin (rhβ2M) with properties of good solubility and high purity from E. coli, prokaryotic expression conditions were optimized and protein purification was performed in this study. After testing the effect of different IPTG concentrations, temperatures and induction times on the production of rhβ2M, the optimum expression conditions were determined, i. e. joining IPTG to final concentration being 0.8 mmol/L and inducing time 6 h and at temperature of 25 degrees C. Under the optimum induction conditions, the ratio of soluble rhβ2M to soluble bacterial protein was 63.7%. After purified by Ni Sepharose 6 Fast Flow, the purity of rhβ2M achieved a greater value of 95%. Western blot analysis revealed that rhβ2M possessed the antigen property that specifically interacted with anti-β2M antibody.

2.Clinical effect of Chinese compound medicine lotion in flat wart

Objective To explore the effect of compound Chinese medicine lotion in the preparation room of the Third Affiliated Hospital of Jinzhou Medical University on the patients with flat wart.Methods Confocal laser scanning microscopy and clinical diagnosis were used to collect 100 cases of flat warts from March 2013 to March 2015.The patients were randomly divided into the treatment group ( Chinese compound lotion) and the control group ( tretinoin cream) with 50 cases in each group.Self-made compound Chinese medicine lotion or Victoria A acid cream for 20 days, after the treatment, the curative effect and relapse rate of the two groups were compared.Results The total effective rate of the treatment group (94.0%) was significantly higher than the control group (76.0%), the difference was statistically significant (P<0.05).The relapse rate in the treatment group was 4.3%, significantly lower than that in the control group the rate was 28.9%, the difference was statistically significant (P<0.05).Conclusions Self-made Chinese compound lotion on flat wart patients have a good effect, and its curative effect on verruca plana is better than tretinoin.
3.Effect of lentinan polysaccharides on hippocampus in Alzheimer 's disease rats

Objective To evaluate the effect of lentinan polysaccharide (LPS) on Alzheimer's disease (AD) rats, and providing a new idea for prevention and treatment of Alzheimer's disease (AD).Methods 96 SPF rats were randomly divided into control group, model group, LPS low dose-treated group, LPS middle dose-treated group, LPS high dose-treated group and positive control group(piracetam treated), each had 16 rats.The model of AD was induced by injection of Aβ25-35(80 pmol/μL), rats in four treatment groups were treated with LPS (200 mg/kg), LPS (400 mg/kg), LPS (600 mg/kg) and piracetam(600 mg/kg) orally once a day for 60 days while control group and model group were given equal-dose saline.After treatment, the ability of navigation and memory were evaluated by water maze test (MWM), and then rats in all groups were sacrificed to obtain the hippocampus.HE staining was used to observe the histopathological changes, the expressions of TACE,Aβ,BACE1 and PP2A protein in hippocampus were detected by Western blot.Results Compared with the model group, high dose LPS could significantly improve the ability of learning and memory (P<0.01) and improve the ability of locating navigation (P<0.01).HE staining results showed that LPS treatment could repair the rat hippocampus injury induced by Aβ25-35 .Western blot analysis showed that the levels of Aβ, and BACE1 were down-regulated in LPS group compared with the model group, while the protein level of PP2A and TACE was up-regulated.Conclusion LPS has a protective effect on Aβ25 -35 induced AD model, and it is expected to be a strategy for treatment of AD .
4.Comparison of the radiographic results of total knee arthroplasty with or without using electromagnetic navigation

Objective To compare the lower limb alignment and prosthesis position after total knee arthroplasty (TKA) with or without using electromagnetic navigation.Methods Sixty-four patients (100 knees) underwent TKA under electromagnetic navigation,while 62 patients (100 knees) underwent conventional TKA.Three months after operation,the mechanical axial line angle and prosthesis position (angels α,β,γ) were measured via the full-length radiograph of both lower limbs and anteriorposterior and lateral Xrays of the knee.Results The average mechanical axial line angle and angle α were 1.20°±1.92°and 89.33°±1.64° in navigation group,respectively,and 2.31°±2.25° and 88.68°±2.57° in conventional group.And the differences were significant with regard to the above two indexes between two groups.The average angle β and angle γ were 89.64°±1.47° and 90.86°±2.37° in navigation group,respectively,and 89.26°±2.05° and 90.59°±3.44° in conventional group.However,the differences were not significant with regard to the above two indexes between two groups.After operation,mechanical axial line angle error was within ±3°in 86% of patients in navigation group,while in 79% of patients in conventional group; there was no significant difference between them.The angle α error was within 90°±3° in 92% of patients in navigation group,while in 77% of patients in conventional group; there was significant difference between them.However,there were no significant differences in percentage of patients whose errors of angle β and angle γ were within 90°±3° between two groups.Conclusion With using electromagnetic navigation in TKA,more precise prosthesis position and the mechanical axis can be achieved compared with the conventional technique.And its advantages mainly contribute to the coronal plane of the femoral side rather than the tibial side.
5. The effect of cardiac autonomic function on skeletal function and weakness in elderly diabetic patients

Objective:
To study the effect of cardiac autonomic nerve function on body function and weakness in elderly diabetic patients.
Methods:
A total of 100 patients diagnosed as diabetic autonomic neuropathy in the endocrine ward of Dalian Friendship Hospital from August 2017 to August 2018 were enrolled. The demographics, clinical data, and Activities of Daily Living (ADL) scale of each patient were recorded, and the Instrument Activities of Daily Living (IADL) and the weak score were used to record heart rate variability in each patient. Aweak score of 5 was used as boundaries. Patients with weak score greater than or equal to 5 were divided into weak group, and those less than 5 were divided into non-weak groups. Each indicators were compared between the two groups. The statistically significant variables were used as covariates, and the weak scores were used as dependent variables to perform direct and indirect Logistic regression analysis of covariates and dependent variables and the receiver operation characteristic (ROC) curve.

Objective:
To evaluate the significance of bacteria detection with filter paper method on diagnosis of diabetic foot wound infection.
Methods:
Eighteen patients with diabetic foot ulcer conforming to the study criteria were hospitalized in Liyuan Hospital Affiliated to Tongji Medical College of Huazhong University of Science and Technology from July 2014 to July 2015. Diabetic foot ulcer wounds were classified according to the University of Texas diabetic foot classification (hereinafter referred to as Texas grade) system, and general condition of patients with wounds in different Texas grade was compared. Exudate and tissue of wounds were obtained, and filter paper method and biopsy method were adopted to detect the bacteria of wounds of patients respectively. Filter paper method was regarded as the evaluation method, and biopsy method was regarded as the control method. The relevance, difference, and consistency of the detection results of two methods were tested. Sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of filter paper method in bacteria detection were calculated. Receiver operating characteristic (ROC) curve was drawn based on the specificity and sensitivity of filter paper method in bacteria detection of 18 patients to predict the detection effect of the method. Data were processed with one-way analysis of variance and Fisher's exact test. In patients tested positive for bacteria by biopsy method, the correlation between bacteria number detected by biopsy method and that by filter paper method was analyzed with Pearson correlation analysis.

Primary hyperparathyroidism is a disease with a large potential population. Some cases of primary hyperparathyroidism are non-primary, preventable and curable at early stage, requiring long-term follow-up after surgery. Therefore, all-round and full-cycle management are necessary for primary hyperparathyroidism, which involves an enhancing focus on etiological prevention, early detection, prompt diagnosis, timely intervention, multi-disciplinary standardized diagnosis and treatment, and postoperative scientific management. Meanwhile, implementing a "12+5+1" multidisciplinary joint diagnosis and treatment model, along with a two-way referral model, to achieve the transition from a disease-oriented diagnostic and treatment model to a patient-oriented, all-round and full-cycle interdisciplinary management model. This management can reduce the incidence and recurrence rate of primary hyperparathyroidism, and related osteoporosis or osteopenia, fractures, nephrolithiasis, metastatic vascular calcification, and systemic abnormal migratory calcium deposits, improve the overall quality of life and prognosis of patients.
9.One case report of repeated chest tightness and palpitation cured after strengthening calcium supplementation

The incidence of parathyroid hyperfunction is high and its clinical manifestations are diverse. Some patients develop chest tightness and palpitations as the main discomfort, which may be caused by the hypocalcemia and hypercalcemia related to negative calcium balance and parathyroid hyperfunction. We report a case of 53 years old male with parathyroid hyperfunction who was diagnosed with osteoporosis before and received conventional regular supplementation of vitamin D and calcium supplements. However, his condition worsened and he developed chest tightness and palpitation. After 1 month of sufficient supplementation of calcium, the symptoms of chest tightness and palpitation disappeared completely. Then we continued to provide the patients enough vitamin D and calcium supplementation actively. After 1 year of follow-up, the patient's condition was stable. His discomfort of chest tightness and palpitation never recurred, and all the bone metabolism indicators returned to normal.
10.Correlation between sleep midpoint and sleep quality in type 2 diabetic patients with insomnia

Objective:To analyze the correlation between sleep midpoint and sleep quality in insomnia patients with type 2 diabetes mellitus (T2DM).Methods:By adopting current situation investigation research, total of 150 T2DM patients hospitalized in the Department of Endocrinology, the First Affiliated Hospital of Guizhou University of Traditional Chinese Medicine from November 2021 to July 2022 were selected as the research objects. The general information questionnaire, Pittsburgh Sleep Quality Index (PSQI), anxiety scale (SAS) and Depression Scale (SDS) were used to investigate, and then analysis the datum.Results:Among 150 T2DM insomnia patients, 41 cases (27.33%) were in the early midpoint sleep group, 37 cases (24.67%) were in the middle midpoint sleep group, and 72 cases (48.00%) were in the late midpoint sleep group. There were significant differences in the distribution of sex, age and BMI level among different sleep midpoint groups ( χ2=7.24, 13.36, 15.93, all P<0.05). The scores of time to fall asleep at the midpoint of sleep in the 3 groups were (2.12 ± 1.25), (2.65 ± 0.79), (2.33 ± 1.02), the difference was significant ( F=2.14, P<0.05); the daytime disability scores in the 3 groups were (1.39 ± 1.36), (2.16 ± 1.12), (1.85 ± 1.32), the difference was significant ( F=3.17, P<0.05). Logistic regression analysis of disorder showed that the time to fall asleep ( OR=4.922, P<0.05) and daytime disability ( OR=4.043, P<0.05) had significant influence to the middle midpoint of sleep group when the early midpoint of sleep group as the control, while the male ( OR=2.182, P<0.05), 50 - 70 years old ( OR=5.005, P<0.05) and BMI over fat side ( OR=3.488, P<0.05) had significant influence to the late midpoint of sleep group. Conclusions:Medical staff should pay attention to the sleep quality of T2DM patients, pay attention to the sleep midpoint of patients, and improve patients′cognition of healthy sleep patterns.
