1.Effects of professional benefit recognition to stress coping of tuberculosis nurses

Objective To investigate the effects of professional benefit recognition to stress coping of tuberculosis nurses. Methods A total of 82 tuberculosis nurses were assigned into experimental and control groups by random digits table method with 41 cases each. The experimental group was given four times of professional benefit recognition interventions in additional to regular training, while control group was just given regular training. The two groups were asked to complete the Professional Benefit Recognition questionnaires and Stress Coping Style questionnaires before the intervention and 1 month after the intervention, respectively. Results Before the intervention, the scores of the Professional Benefit Recognition questionnaires and Stress Coping Style questionnaires between two groups had no significant difference (P>0.05). After the intervention, the Professional Benefit Recognition questionnaires scores of experimental group increased from ( 2.93 ± 0.40) points to (3.81±0.38) points that was significantly better than that of control group(2.99±0.39) points, and there were significant differences (t=49.337,-60.834, P<0.05). The immature and mixed Stress Coping Style questionnaires scores of experimental group: self- accusation, illusion, avoidance, rationalization were (0.51 ± 0.13) , (0.51 ± 0.24), (0.56 ± 0.21), (0.51 ± 0.16) points, which were significantly lower than before the intervention (0.70 ± 0.09), (0.65 ± 0.10), (0.67 ± 0.14), (0.67 ± 0.08) points and that of control group (0.63 ± 0.16), (0.65 ± 0.18), (0.68 ± 0.18), (0.64 ± 0.13) points, with the difference being significant (t=-4.071--2.541, P<0.05). The mature coping scores of the experimental group:problem solving, appealing were (0.70 ± 0.10), (0.67 ± 0.13) points, which were significantly higher than that of control group (0.63 ± 0.12), (0.58 ± 0.15) points and that of experimental group before the intervention (0.55 ± 0.12), (0.51 ± 0.11) points, with the difference being significant (t=2.308, 2.098, P <0.05). Conclusions Professional benefit recognition interventions can improve professional benefit recognition and stress coping style, is beneficial to nurses' physical and mental health and nursing quality.
2.Progress in the relationship between mutant SOD1 and amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis(ALS)is a progressive neurodegenerative disorder.Mutations in the Cu,Zn-superoxide dismutase(SOD1)gene cause ALS by an unknown gain-of-function mechanism.Current researches show that diminishing SOD1 mutation enzymatic activity does not account for motor neuron lose in ALS,while direct toxicity of variant SOD1 protein or aggregation of SOD1 protein may impart cell function and cause motor neuron death.Though motor neuron death is selective,other cells also participate in this course.The author reviewed recent advances in the relationship between mutant SOD1 and Amyotrophic Lateral Sclerosis.
3.Effect of interventional treatment on absolute alcohol in different types of liver cancer

Objective To investigate the effect of routine ultrasound examination and interventional treatment of absolute alcohol in different types of liver cancer.Methods Thirty cases of liver cancer patients who were treated in the 3rd People' s Hospital of Yangquan from May 2010 to May 2014 were selected as ours subjects.They were performed ultrasound exam.The ultrasound characteristic was recorded.Patients were divided into intrahepatic cholangiocarcinoma (ICC) group (11 cases),hepatocellular cholangiocarcinom (HCC) group (14cases) and mixed cholangiocarcinoma group(5 cases) based on pathology results.Nine HCC postoperative recurrence patients and 7 ICC postoperative recurrence patients were treated with absolute alcohol and the clinical effect was observed.Results There were significant differences in the three groups in terms of gender,boundary definition and tumor location (x2 =8.01,6.16,7.32 ; P < 0.05).While the distribution of ultrasound echo intensity was not statistically significant(x =3.44,P > 0.05).The effective rate of interventional treatment of absolute alcohol in HCC patients was 88.89%,and 42.86% in ICC group,and the difference was statistically significant (x2 =3.88,P < 0.05).Conclusion The ultrasound characteristics of different pathological liver cancer are different.Ethanol interventional therapy on HCC patients is significantly better than the effect of ICC patients,suggesting that ICC postoperative recurrence patients can be treated combined with other methods to help improve the effect.
5.Cluster analysis of cystic echinococcosis in Non Tibetan Plateau regions

Objective To understand the endemic characteristics and regularity of cystic echinococcosis by evaluating and classifying its endemic situation in Non Tibetan Plateau regions,so as to provide the evidence for formulating effective preventive and control measures. Methods The prevalence data of cystic echinococcosis in 174 counties(cities,districts,banners)in Non Tibetan Plateau regions from a national survey were collected and analyzed by the sample cluster method in 2012. Results The 174 counties(cities,districts,banners)could be classified into 4 clusters by spatial distribution. The first cluster with human high prevalence rate,low infection rate of livestock,and positive rate of dog stool antigen included 3 counties. The second cluster with high infection rate of livestock,low prevalence rate of human,and positive rate of dog stool antigen included 20 counties(cit-ies). The third cluster with high positive rate of dog stool antigen,low prevalence rate of human,and low infection rate of livestock included 39 counties(cities,districts,banners). The fourth cluster with low rates of the above 3 indices included the rest 112 counties. Conclusions The results of the cluster analysis conform to the current epidemiological status of cystic echinococcosis in the Non Tibetan Plateau regions. The epidemiological characteristics and geographical distributions of the four area types will pro-vide a basis for the classified management and guidance of cystic echinococcosis control in these areas.
6.Prokaryotic expression and polyclonal antibody preparation of nonstructural protein 2 transactivated protein of hepatitis C virus

Objective To express nonstructural protein 2 transactivated protein (NS2TP) of hepatitis C virus (HCV) in the prokaryotic expression system and prepare polyclonal antibody,and to study the expressions in different liver tissues.Methods NS2TP gene was amplified by polymerase chain reaction (PCR) technique and cloned into the prokaryotic expression vector pET-32a(+),which was transformed into E.coli BL21.The protein was induced by isopropyl thiogalactose (IPTG) and analyzed with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and confirmed by Western blotting assay.The recombinant protein were expressed and purified in a large amount.The rabbit was immunized with the purified protein to prepare polyelonal antibody.The liver tissues of patients with chronic HCV infection and healthy controls were detected by immunohistochemistry method.Results The recombinant NS2TP protein (relative molecular mass:33×103 ) and polyclonal antibody with high titer and specificity were successfully prepared.NS2TP expressions in the liver of patients with chronic HCV infection were higher than those of healthy controls,and were mainly distributed in the nucleus of hepatocytes.Conclusions The NS2TP expression level and intracellular location in liver tissue of patients with chronic HCV infection are understanded,which could bring new clues for further study of the biological function of NS2TP and the pathogenesis of HCV infection.

8.Development of Unidirectional Negative Corona Discharge Ion Mobility Spectrometer for Detection of Explosives

Abstract Ion mobility spectrometry ( IMS) based explosives detectors is wieldy deployed at the check points of airport, etc, so the development of new ionization method to replace the radioactive 63 Ni ion source in IMS is highly demanded. In this study, a novel negative corona discharge was developed for ion mobility spectrometer for rapid detection of trace explosives, which was running in the unidirectional mode to efficiently remove nitrogen oxides and ozone produced from the discharge process. The diameter of target electrode was 3 mm, the distance between the needle and the target electrode was 2 mm, the discharge voltage was 2400 V and the flow rate of drift gas was 1200 mL/min. Under the optimized conditions, the dominant reactant ions was O-2(H2O)n and could be used to directly detect the common explosives, such as trinitrotoluene (TNT), ammonium nitrate (AN), nitroglycerin (NG), pentaerythritol tetranitrate (PETN) and hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX). The limit of detection of TNT was 200 pg/μL. These results indicated that negative corona discharge ionization source, with the advantanges of the high sensibility, simple structure and no radioactivity, could be used as a potential and promising ionization source for ion mobility spectrometry to detect explosives.
9.Transplantation of bcl-2 gene-modified bone marrow mesenchymal stem cells improves cardiac function and angiogenesis in rabbit ischemic car-diac insufficiency model

[ ABSTRACT] AIM:To investigate the effects of transplantation of bone marrow mesenchymal stem cells ( BMSCs) modified by bcl-2 gene on myocardial cell apoptosis, angiogenesis and cardiac function in the rabbit after acute myocardial in-farction ( MI) .METHODS:The rabbit BMSCs were isolated, cultured and purified in vitro.The BMSCs were transfected with adenovirus or adenovirus-Bcl-2.The rabbit model of MI was established by ligation of left anterior descending branch. The rabbits were injected with Ad-Bcl-2-BMSCs ( MI+Bcl-2-BMSCs group) , Ad-BMSCs ( MI+BMSCs group) and DMEM ( MI group) in infarction marginal zone 2 weeks after ligation.The cardiac function was evaluated by echocardiography.The apoptosis of myocardial cells was measured by TUNEL.The mRNA expression of VEGF was detected by real-time PCR.The expression of CD31 was examined by immunohistochemical staining, and new blood capillaries were counted at 4 weeks after BMSCs transplantation.The correlation of the above values with cardiac function was analyzed.RESULTS: The cardiac function was better, the apoptotic rate was lower, the mRNA expression of VEGF and the capillary density were higher in both MI+Bcl-2-BMSCs group and the MI+BMSCs group than those in MI group, and those in MI+Bcl-2-BMSCs group in-creased more obviously .The left ventricular ejection fraction ( LVEF) had a negative correlation with the myocardial cell ap-optosis rate.A positive correlation with the mRNA expression level of VEGF and the capillary density was also observed. CONCLUSION:The transplantation of BMSCs modified by bcl-2 gene significantly reduces the myocardial cell apoptosis, promotes angiogenesis, improves heart function of the rabbits with MI.
10.Effects of perindopril at different doses on cardiac function and ACE2/Ang-(1-9)/Ang-(1-7) axis of ischemic cardiac dysfunction rabbits

[ ABSTRACT] AIM:To investigate the different dose of perindopril on cardiac function in the rabbits with ische-mic cardiac dysfunction .METHODS:Male rabbits weighing 2.5~3.0 kg ( n=30) were randomly divided into 3 groups (n=10):high dose perindopril group (HD group), low dose perindopril group (LD group) and cardiac dysfunction group (CD group).The Left anterior descending coronary artery of the rabbits was ligatured for model preparation .In HD group, the rabbits were treated with perindopril split normal saline solution (1 g/L)2 mL· kg-1 · d-1 .In LD group, the rabbits were treated with perindopril split normal saline solution (0.33 g/L)2 mL· kg -1 · d-1.In CD group, the rabbits were treated with normal saline solution 2 mL· kg-1 · d-1 .Four weeks after treatment , the cardiac function was measured via echocardiography , the mRNA expression of angiotensin-converting enzyme 2 ( ACE2 ) and angiotensin type 2 receptor (AT2R) was analyzed by real-time PCR, serum angiotensin (Ang)-(1-9) and Ang-(1-7) levels were detected by ELISA. RESULTS:Compared with CD group , the cardiac function of the 2 groups treated with perindopril was significantly im-proved (P<0.01), and more improvement in HD group was observed than LD group (P<0.05).The serum angiotensin ( Ang)-(1-9) and Ang-(1-7) level and the mRNA expression of ACE 2 and AT2R in the 2 groups treated with perindopril were significantly improved (P<0.01).Compared with LD group, the mRNA expression of ACE2 and AT2R and the ser-um levels of Ang-(1-9) in HD group were significant improved (P<0.05), while no difference of serum Ang-(1-7) level was observed.Correlation analysis revealed that the improvement of the cardiac function was associated with serum Ang -(1-9) level, mRNA expression of ACE2 and AT2R (P<0.01), but has no significant correlation with serum Ang-(1-7) lev-el.CONCLUSION:High dose of perindopril may improve more cardiac function in ischemic cardiac dysfunction model in rabbits.The mechanism may relate to increasing serum Ang-(1-7) level to activate AT2R.
