Objective To investigate effect of monosialotetrahexosy-1 ganglioside (GM1) on the autophagic neuronal death induced by spinal cord injury.Methods Ninety SD rats were randomly divided into sham group,isotonic saline group and GM1 group,with 30 rats per group.Spinal cord injury at T10 segment was induced by Allen method in the isotonic saline group and GM1 group.Expression of endogenous LC3 was detected by florescence microscope.Ratio of LC3-Ⅱ and LC3-Ⅰ,and expression of Beclin-1 were detected by Western blot.Results LC3 was significantly up-regulated in the isotonic saline group,as compared to the sham group (P ＜0.05).Also,an up-regulation of LC3 and a decline of autophagic body formation were observed in GM1 group,as compared to isotonic saline group (P ＜0.05).Expressions of LC3-Ⅱ and Beclin-1 and ratio of LC3-Ⅱ to LC3-Ⅰ were significantly increased in the isotonic saline group,as compared to the sham group (P ＜ 0.05).Expressions of LC3-Ⅱ and Beclin1 and ratio of LC3-Ⅱ to LC3-Ⅰ significantly were decreased in the GM1 group,when compared to the isotonic saline group (P ＜ 0.05).Conclusion GM1 inhibits autophagic neuronal death after spinal cord injury and hence exerts protective effect on the neurons.

[Objective]To establish a reproducible rat model of disc degeneration by puncturing the annulus with needles of defined gauges.[Method]Twenty skeletally mature male Sprague-Dawley rats were used to determine the depth of puncture by measuring anular thickness of lumbar disc.Twelve rats were carried out transperitoneal ventral surgery and anular stab was performed using a 21-gauge needle to make a full-thickness or partial-thickness stab incision in the L3、4,L4、5 and L5、6 anteriorlateral disc.The L6S1 level was used as a control.Magnetic resonance images scans of the stabbed discs and control discs were performed at 4,8-week time point and after MRI histological analysis were performed.[Result]The depth of puncture was respectively partial-thickness 1.5 mm and full-thickness 2.3 mm.The stabbed discs exhibited a decrease signal intensity from T2-weighted images starting at 4-week time point in full-thickness stab incision and 8-week in partial-thickness.Histological analysis revealed disc degeneration in both stab incision groups at 4-week time point and full-thickness stab incision displayed more severe degenerative changes compared with that of partial-thickness.[Conclusion]Full-thickness or partial-thickness anulus stab incision with needles of defined gauges is reliable,effective and reproducible to establish a rat model of intervertebral disc degeneration.

METHODSColorectal cancer cell lines with stable Sam68 over-expressing and low Sam68 expression were established to test the effect of Sam68 in the proliferation, invasion, and migration of the cancer cells using colony formation, MTT and Transwell assays.

RESULTSSW480 and Ls174t colorectal cell lines over-expressing Sam68 showed significantly enhanced cell proliferation, invasion and migration (P<0.05). Conversely, the low Sam68 expression in SW620 and HCT116 colorectal cell lines significantly suppressed the cell proliferation, invasion and migration (P<0.05).

CONCLUSIONThe expression of Sam68 can promote the proliferation, invasion and migration of colorectal cancer cells lines in vitro.

OBJECTIVE: To explore the effect and indications of radiofrequency ablation for the treatment of obstructive sleep apnea-hypopnea syndrome. METHOD: Multilevel temperature-controlled radiofrequency therapy of soft palate, uvula, inferior turbinate, and tonsils were applied to 74 adults with obstructive sleep apnea-hypopnea syndrome (OSAHS). There were 16 mild, 23 moderate, and 35 severe cases respectively in this study. Evaluation of mucosal injury and effect of radiofrequency therapy on pain, speech and swallowing were performed early after operation. The volume of targets and length of soft palate and uvula were measured three months after operation. Polysomnography, Epworth Sleepiness Scale and Snoring Scale Score questionnaires were reevaluated six months after operation and compared with the results of pre-operation. Treatment outcome measurements were mainly based on polysomnography. RESULT: By our definition, 5 of 74 patients (6.76%) have been cured and 42 of 74 (56.76%) had improved totally. Mean Apnea-Hypopnea Index (AHI) decreased significantly and mean lowest oxygen saturation value increased significantly postoperatively (P < 0.01). The total effective rate of the patients, whose obstructive sites were all treated by radiofrequency, was remarkably higher than that of the ones, whose obstructive sites were only partly treated by radiofrequency (P < 0.01). The total effective rate of the former was 72.92%. Patients showed a significant decrease in mean score on ESS and SSS postoperatively (P < 0.01). No significant complications were observed in most patients. There were little influence on pain, speech and swallowing. The volume or length of targets decreased obviously three months after operation. CONCLUSION Radiofrequency can reduce the volume of tissue. The short-term outcomes of radiofrequency were satisfying if obstructive sites had been all treated. This study demonstrates that the characters of radiofrequency are as follows: minimally invasive, safe, efficient, repeatable and multilevel applicable. Temperature-controlled radiofrequency therapy is a safe and effective procedure for hypertrophic infraturbinal when used separately, or as a part of a the combined approach for complex syndromes.

Cholelithiasis, a prevalent disease of the digestive system, is characterized by its intricate and diverse mechanisms, which are influenced by a complex interplay of genetic, environmental, lifestyle, and other factors. Recently, with the widespread application of molecular biology techniques, the role of the biliary tract microecological environment in the pathogenesis of gallstones has garnered increasing attention. This review includes the most recent and pertinent literature on the association between biliary tract microecology and gallstones, summarizing the latest research advancements in this field. Furthermore, it delves into the role of the biliary tract microecology in the formation of both cholesterol and pigment gallstones.

The development of gastritis is associated with an increased risk of gastric cancer. Current invasive gastritis diagnostic methods are not suitable for monitoring progress. In this work based on 78 gastritis patients and 50 healthy individuals, we observed that the variation of tongue-coating microbiota was associated with the occurrence and development of gastritis. Twenty-one microbial species were identified for differentiating tongue-coating microbiomes of gastritis and healthy individuals. Pathways such as microbial metabolism in diverse environments, biosynthesis of antibiotics and bacterial chemotaxis were up-regulated in gastritis patients. The abundance of Campylobacter concisus was found associated with the gastric precancerous cascade. Furthermore, Campylobacter concisus could be detected in tongue coating and gastric fluid in a validation cohort containing 38 gastritis patients. These observations provided biological evidence of tongue diagnosis in traditional Chinese medicine, and indicated that tongue-coating microbiome could be a potential non-invasive biomarker, which might be suitable for long-term monitoring of gastritis.
