China Pharmacy 1991;0(01):-
OBJECTIVE:To establish an HPLC method for determining nicardipine hydrochloride(NCD) microspheres in cerebrospinal fluid(CSF) of rabbits by HPLC.METHODS:NCD microspheres were injected into the cistern of rabbits.Cerebrospinal fluid was sampled at different time points and NCD was separated on Agilent-Zorbax column at a flow rate of 1.0 mL?min-1 with column temperature of 30 ℃.The mobile phase consisted of methanol-water(75 :25,pH=6.0) and detection wavelength was set at 236 nm.RESULTS:The linear range of NCD was 2~128 ng?mL-1(r=0.999 8) with recovery of 98.07%.The RSD of intra-day and inter-day were both below 1.47%.The LOD was about 0.5 ng?mL-1.On 3~11 day after medication, the concentration of NCD fluctuated within(14.15?0.37) ng?mL-1.CONCLUSION:The method is simple, rapid and accurate for determination of NCD in cerebrospinal fluid of rabbits.

The patient complained of recurrent sore throat for 2 years, who was diagnosed parapharyngeal abscess or tonsillitis for four times during June 16, 2012 to April 16, 2013. Special physical examination: left or right lateral pharyngeal wall is slightly elevated. Routine blood test showed increasing white blood cells and neutrophils. Oropharyngeal CT showed right lateral pharyngeal wall swelling and abscess formation? Repeated puncture showed no obvious purulent secretions. Symptoms were improved after anti-inflammatory treatment, but it recurrently happened later. Bilateral tonsillectomy was performed under general anesthesia on April 29, 2013. Pathological report (May 6, 2013) showed: (left) chronic tonsillitis with lymphoid hyperplasia; chronic inflammation in (right) tonsil tissue, and salivary gland tissue is also observed, considering as the hyperplasia of ectopic salivary gland tissue and interstitial lymphocytic oinfiltration.

Chinese Pharmacological Bulletin 1987;0(02):-
The compounded prescription of Chinese crude drug is a key problem in the study of traditional Chinese medicine.A new research field that focuses on the group of effective fractions of Chinese crude drugs becomes more important as the result of combination of the theorem of traditional Chinese medicine and techniques of modern medicine.Up to date, the researches on this field are preliminary, remaining a lot of subjects to be explored.

Objective To observe the effects of fluvastatin on proliferation and apoptosis of HL-60 cells, and to offer the theoretical evidence for tumor treatment. Methods HL-60 cells were divided into:fluvastatin groups(0.5,5.0,10.0 and 20.0 ?mol?L-1),HL-60 control group, positive control group(treated with 10.0 ?mol?L-1ATRA).The live cell number was counted for cell proliferation assay. The growth inhibitory rate of HL-60 cells was detected using CCK-8 kit.The cell cycle distribution and apoptotic rate were measured using flow cytometry assay. Results Compared with control group, after HL-60 cells were treated with 0.5,5.0,10.0 and 20.0 ?mol?L-1of fluvastatin for 1-4 d,the number of live cells decreased in different level(P

Objective To investigate the relationship between the expression ofα1-antichymot-rypsin (α1-ACT)in plasma and postoperative cognitive dysfunction(POCD).Methods The cognitive function of 64 patients undergoing operation under general anesthesia was evaluated by a battery of cognitive assessment instruments(BCAI)and the levels ofα1-ACT were measured at the time 1 d pre-operatively and 7 d and 3 months postoperatively. The patients were randomly divided into two groups:30 patients in ulinastatin group (group U)were injected with 10 000 U/kg ulinastatin, which was dissolved in 100 ml saline, within 20 min at the time of 30 minutes before induction;while control group (group C)were injected with the same volume saline. Results There was no significant differ-ence between the expression of α1-antichymotrypsin in patients with POCD and in control patients at the time before operation.Compared with control patients, the levels of α1-ACT in patients with POCD at the time of 7 d and 3 months postoperation were significantly higher (P <0.05).The levels of α1-ACT at 7 d postoperation were significantly higher than that preoperatively in both groups (P <0.05).The incidence of POCD in group U were significantly lower than that in group C,at the time of 7 d and 3 months after surgery (P <0.05).Conclusion POCD is associated with high expression of plasma α1-ACT.

Objective To observe the effect of fluvastatin(Flu) on differentiation and induction of human promyelocytic leukemia HL-60 cells and provide the theoretical foundation for treatment of human promyelocytic leukemia. Methods The cultured HL-60 cells were treated with 20 ?mol?L-1 Flu,the morphological changes of the cell differentiation were examined. The NBT reduction capability was detected in HL-60 cells treated with Flu for 72 h.After HL-60 cells were treated with 20 ?mol?L-1 Flu for 5 d,they were stained with non-specific esterase and the percentage of differentiation cells was analyzed. Results The HL-60 cells treated with Flu showed smaller cell body, reducing proportion of nucleus to cytoplasm, the nucleus tortuosity, fold or sublobe. There were specific granules and vacuoles in cytoplasm, displaying that some cells had differentiated to relative mature cells. As compared with control group, the NBT reduction capability in HL-60 cells treated with Flu for 72 h was significantly higher than that in control cells(P

Objective To develop a highly sensitive luminescent reagent for bloodstain testing at forensic crime scenes. Methods Based ontheprincipleof ECL luminescence and the ping-pong conjugate activation principle of chemical electronic chain, this project developed a new type of highly sensitive luminescent reagent for bloodstain testing by usingthe uniform design of experimental methods to optimize the conditions andsynthesize several new compounds. Results The bloodstain testing luminescent reagentdeveloped in this project has high sensitivity andlongluminescence time.In the case of blood samples diluted by 1,000 times, reading the fluorescence withChemiScope 3300 chemiluminescence imaging system, the maximumvalue of gray scale reached 56,and the luminescence time lasted for 10 minutes. Conclusion The project has successfully developed a highly semitivebloodstain testing reagentthat could be applied to crime scene investigation.

OBJECTIVE:To optimize the water-extraction technology of Compound kuhuang formulation. METHODS:The wa-ter-extraction technology of Compound kuhuang formulation was optimized by orthogonal design, with the total contents of matrine and oxymatrine, the total content of prim-O-glucosylcimifugin and 5-O-methylvisamminol,the content of berberine hydrochloride and extract yield, and with the extraction time, amount of water(3 reflux extractions)and soaking time as the factors;and verifi-cation tests were conducted. RESULTS:The extraction time had significant effect on comprehensive evaluation value (P<0.05). The optimal water-extraction technology was as follows as extraction time of 120 min, water amount of 8,6,and 6 times as the amount of herbs, and soaking time of 20 min. In verification tests, the average total content of matrine and oxymatrine was 3.152 6 mg/g(RSD=1.03%,n=3),that of prim-O-glucosylcimifugin and 5-O-methylvisamminol was 4.977 2 mg/g(RSD=2.27%,n=3),the average content of berberine hydrochloride was 3.345 0 mg/g (RSD=1.19%,n=3) and the average extract yield was 49.23%(RSD=2.43%,n=3). CONCLUSIONS:The optimal water-extraction technology of Compound kuhuang formulation is stable and feasible.
